ABSTRACT

Objective@#To explore the relationship between physical fitness and energy balance related behaviors (EBRBs), and their influencing factors among primary school students, so as to provide a scientific basis for the improvement of physical fitness in primary school students.@*Methods@#By using a random sampling method, a total of 1 451 pupils aged 10-12 were selected from 8 regions of Shannxi Province (Fengxiang, Weibin, Danfeng, Shangzhou, Huazhou, Linwei, Gaoling, Weiyang). According to the total score of physical fitness test, pupils were divided into the excellent good physical fitness group and the pass failed group. The students EBRBs and their influencing factors were investigated cross sectionally, and the Mann-Whitney U test and stepwise linear regression analysis were used to explore the relationship between physical fitness and EBRBs.@*Results@#Breakfasts behavior( r = 0.061 ) and physical activity behavior( r =0.105) among primary school students were positively correlated with total physical scores, with the excellent good physical fitness group (757.56, 768.57)were higher than that of the pass failed group(710.93, 705.67) ( Z= -2.41, -2.69, P <0.05). The screen behavior ( r =-0.065) was negatively correlated with the total physical fitness scores, with the excellent good physical fitness group (681.96) was significantly lower than the pass failed group(747.04) ( Z=2.78, P < 0.05 ). There was no statistically significant correlation between the frequency of beverage behavior and the total score of physical fitness ( P >0.05). The excellent good physical fitness group scored(762.22, 761.19, 758.82, 756.00, 761.20, 755.57, 761.52, 759.48, 781.78) higher than the pass failed group(708.70, 709.19, 710.32, 711.67, 709.19, 711.88, 709.04, 710.01, 699.36) including health beliefs, parental norms and parental role models for breakfast behaviors, and preferences, self efficacy, and self regulation for physical activities, as well as self efficacy, parental role models, family rules for screen behavior ( Z=-2.40, -2.78, -2.35, -2.48, -2.52, -2.27, -2.35, -2.22, -3.65, P <0.05).@*Conclusion@#The physical fitness of primary school students is affected by EBRBs. Parents should model positive behaviors in the family, promote the health behavior of primary school students, improve the physical health of primary school students health.

ABSTRACT

@#Abstract: Objective To analyze the association between drug resistance and the risk of latent tuberculosis infection and disease among household contacts of patients with pulmonary tuberculosis, and to explore whether the compensatory mutation of drug-resistant Mycobacterium tuberculosis will enhance its pathogenicity or transmission ability. Methods The English and Chinese databases, including PubMed, web of science, EMBASE, Cochrane library database, CNKI and Wanfang database, were searched by computer from the time of establishment of the database to January 2022. Cohort studies on the risk of infection and disease among household contacts of patients with drug-resistant and sensitive pulmonary tuberculosis were searched and screened according to the inclusion and exclusion criteria. The data were extracted and evaluated by NOS scale, using stata16.0 software meta-analysis to calculate the combined effect of tuberculosis infection and disease risk of family contacts, and carry out heterogeneity test, subgroup analysis and sensitivity analysis. Results A total of 7 cohort studies involving 9653 TB index cases and 29, 734 house contacts were included. The results of meta-analysis showed that compared with drug-sensitive pulmonary tuberculosis patients, the risk of tuberculosis infection in house contacts of drug-resistant pulmonary tuberculosis patients was increased (OR=1.56, 95%CI=1.25-1.96, P<0.001), but there was no difference in the risk of incidence (RR=1.06, 95%CI=0.80-1.41, P=0.67>0.05). Subgroup analysis showed that the risk of latent tuberculosis infection in house contacts was affected by the study area, and the size of family contacts had an impact on the risk of TB . Sensitivity analysis showed that the results of meta-analysis were robust. Conclusion Compared with drug sensitive TB patients, household contacts with drug-resistant TB patients had a higher risk of tuberculosis, but there was no difference in the risk of TB among the two groups.

ABSTRACT

The present study investigated the pharmaceutical effect and underlying mechanism of Zexie Decoction(ZXD) on nonalcoholic fatty liver disease(NAFLD) in vitro and in vivo via the LKB1/AMPK/PGC-1α pathway based on palmitic acid(PA)-induced lipid accumulation model and high-fat diet(HFD)-induced NAFLD model in mice. As revealed by the MTT assay, ZXD had no effect on HepG2 activity, but dose-dependently down-regulated alanine aminotransferase(ALT) and aspartate aminotransferase(AST) in the liver cell medium induced by PA, and decreased the plasma levels of ALT and AST, and total cholesterol(TC) and triglyceride(TG) levels in the liver. Nile red staining showed PA-induced intracellular lipid accumulation, significantly increased lipid accumulation of hepatocytes induced by PA, suggesting that the lipid accumulation model in vitro was properly induced. ZXD could effectively improve the lipid accumulation of hepatocytes induced by PA. Oil red O staining also demonstrated that ZXD improved the lipid accumulation in the liver of HFD mice. JC-1 staining for mitochondrial membrane potential indicated that ZXD effectively reversed the decrease in mitochondrial membrane potential caused by hepatocyte injury induced by PA, activated PGC-1α, and up-regulated the expression of its target genes, such as ACADS, CPT-1α, CPT-1β, UCP-1, ACSL-1, and NRF-1. In addition, as revealed by the Western blot and immunohistochemistry, ZXD up-regulated the protein expression levels of LKB1, p-AMPK, p-ACC, and PGC-1α in vivo and in vitro. In conclusion, ZXD can improve NAFLD and its mechanism may be related to the regulation of the LKB1/AMPK/PGC-1α pathway.

ABSTRACT

Three sesquiterpenoids were isolated and purified from the 95% ethanol extract of Atractylodis Macrocephalae Rhizoma by column chromatography on silica gel, Sephadex LH-20, ODS, and high-performance liquid chromatography(HPLC). Their chemical structures were identified on the basis of spectroscopic analysis and physiochemical properties as(7Z)-8β,13-diacetoxy-eudesma-4(15),7(11)-diene(1), 7-oxo-7,8-secoeudesma-4(15),11-dien-8-oic acid(2), and guai-10(14)-en-11-ol(3). Compounds 1 and 2 are new compounds and compound 3 was obtained from Compositae family for the first time. Compounds 1, 2, and 3 showed weak inhibitory activities against sterol regulatory element-binding proteins(SREBPs).

ABSTRACT

Objective:To investigate the effect of Huayu Jiedu prescription medicated serum(HJRMS)on the proliferation, invasion and migration of human lung cancer cells (H1299 cells) and its mechanism. Method:Cell counting kit-8 (CCK-8) method was used to detect the inhibitory effect of HJRMS on the proliferation of lung cancer cells, the effect of HJRMS on the invasion and migration of H1299 cells were determined by Transwell assay and wound healing assay. The protein expressions of Janus kinase 2 (JAK2), signal transduction and activation transcription factor 3 (STAT3), phosphorylated JAK2(p-JAK2) and phosphorylated STAT3 (p-STAT3) were detected by Western blot, the mRNA expression levels of JAK2 and STAT3 were detected by Real-time quantitative polymerase chain reaction(Real-time PCR). Result:① Compared with control group, the proliferation of H1299 cells was significantly inhibited after treatment with 1%~16%HJRMS serum for 24, 48 h, respectively(<italic>P</italic><0.01), and showed a certain concentration dependence. ② After treatment with HJRMS for 24 h, the scratch healing ability of cells in the 4%,8%HJRMS serum groups was inhibited(<italic>P</italic><0.05,<italic>P</italic><0.01). ③ Compared with control group, the membrane permeability of H1299 cells in invasion and migration experiments in 2%,4%,8%HJRMS serum groups was decreased significantly(<italic>P</italic><0.05,<italic>P</italic><0.01). ④ Western blot showed that compared with control group, 4%,8%HJRMS serum groups inhibited the expression of JAK2/STAT3 signaling pathway related proteins (JAK2, p-JAK2, STAT3, and p-STAT3) in lung cancer H1299 cells(<italic>P</italic><0.05, <italic>P</italic><0.01). ⑤ Compared with control group, the mRNA expression levels of JAK2 and STAT3 in lung cancer H1299 cells treated with 8%HJRMS for 24 h decreased significantly (<italic>P</italic><0.05). Conclusion:The HJRMS can inhibit the proliferation, invasion and migration of lung cancer H1299 cells, and its mechanism may be related to the inhibition of JAK2/STAT3 signaling pathway.

ABSTRACT

Objective: Peptidyl alkaloids, a series of important natural products can be assembled by fungal non-ribosomal peptide synthetases (NRPSs). However, many of the NRPSs associated gene clusters are silent under laboratory conditions, and the traditional chemical separation yields are low. In this study, we aim to discovery and efficiently prepare fungal peptidyl alkaloids assembled by fungal NRPSs. Methods: Bioinformatics analysis of gene cluster containing NRPSs from the genome of Penicillium thymicola, and heterologous expression of the putative gene cluster in Aspergillus nidulans were performed. Isolation, structural identification, and biological evaluation of the product from heterologous expression were carried out. Results: The putative tri-modular NRPS AncA was heterologous-expressed in A. nidulans to give anacine (1) with high yield, which showed moderate and selective cytotoxic activity against A549 cell line. Conclusion: Heterologous expression in A. nidulans is an efficient strategy for mining fungal peptidyl alkaloids.

ABSTRACT

italic>Shiraia bambusiccola is an important medicinal fungus in China. Hypocrellins with perylenequinone skeleton are main bioactive components of Shiraia bambusiccola, which are widely used in food, medicine, pesticide and other fields as natural photosensitizers. For example, "hypocrellin ointment" has already been used clinically. As a rare and vulnerable species, wild Shiraia bambusiccola resources are very limited. Due to the complex structure and chanllenge in chemical total synthesis of hypocrellins, it is urgent to find an effective strategy to rationally utilize its medicinal value while protecting the wild resources. In this study, a candidate gene cluster hpc was identified in Shiraia sp. cfcc 84681 based on careful bioinformatic analysis. A heterologous expression system for hpc gene cluster was successfully constructed and a mutant strain with high yield of hypocrellins was obtained, which mainly produced hypocrellin A and isohypocrellin A. The main ingredients in the mutant strain are consistent with that in the wild Shiraia bambusiccola. These results provide a new strategy to solve the shortage of wild Shiraia bambusiccola resources.

ABSTRACT

Diabetes mellitus (DM) has a high prevalence in patients with pancreatic cancer (PaC), but the prognostic value of DM in PaC remains controversial. Alterations of P-glycoprotein (P-gp) and cytochrome P450 3A4 (CYP3A4) contribute to multidrug resistance and intestinal metabolism in a variety of cancer types, which may be implicated in DM development. This study aimed to explore the potential prognostic value of P-gp and CYP3A4 in PaC patients in the context of DM through long-term follow-up. We retrospectively reviewed the medical records of patients with PaC admitted at The First People's Hospital of Changzhou, Jiangsu, China, from January 2011 to November 2019 and identified two cohorts of adult patients with PaC, including 24 with DM and 24 without DM (non-DM). The baseline clinical characteristics and outcomes were compared. Immunohistochemistry showed that protein expression of P-gp, but not CYP3A, in duodenum tissues was significantly upregulated in PaC patients with DM compared with those without DM. Kaplan-Meier analysis and log-rank test showed that the survival of patients with PaC and DM/high expression of P-gp was not significantly reduced compared with that of patients without DM/low expression of P-gp. These findings suggested that P-gp expression levels were different in the DM and non-DM groups of patients with PaC, but DM and duodenal P-gp levels were not associated with the long-term survival of patients with PaC. It appears that the presence of DM or P-gp expression levels may not serve as effective prognostic markers for PaC.

ABSTRACT

Amphetamines are chemical synthetic drugs that are becoming increasingly popular in China. As a common sample in the inspection of poisons, hair has the advantages of easy storage, good stability, and long detection time compared with traditional human body fluid samples (blood, urine), thus possesses an unique application value in the field of forensic toxicology analysis. By now, methods for detecting amphetamines in human hair have been widely used, and validity of the results has been recognized and adopted by the court. This paper reviews domestic and foreign research progress of the detection of amphetamines in hair samples, including the pretreatment and analytic methods.

ABSTRACT

Objective:To observe the effect of Chaibei Zhixian decoction and peimine on Carbamazepine (CBZ) concentration, P-glycoprotein (P-gp) and multi drug resistance 1(MDR1) expression in the brain tissues of rats with refractory epilepsy, and to understand the contribution of Peimine in the compound prescription to treat the refractory epilepsy. Method:Epilepsy rat models were established by injecting kainic acid (KA) in the lateral ventricle. The successfully modeled rats were randomly divided into model group, CBZ group(0.12 g·kg-1),Chaibei Zhixian decoction+CBZ group(8.39 g·kg-1+0.12 g·kg-1), peimine+CBZ group(0.01 g·kg-1+0.12 g·kg-1) and sham operation group. After 60 days of intervention, the expression levels of P-glycoprotein (P-gp) and MDR1b mRNA in the brain cortex were detected by Western blot and quantitative real\|time fluorescence polymerase chain reaction(Real-time PCR),the contents of CBZ and 10,11-epoxidation of carbamazepine (CBZE) were measured by liquid chromatography-mass spectrometry (LC-MS). Result:Compared with sham group, the expression of P-gp/MDR1 in the cortex of model group was significantly increased (PPPPPPPConclusion:Chaibei Zhixian decoction and peimine may increase the content of CBZ and CBZE in the brain tissues in rats with intractable epilepsy by reducing the expression of MDR1/P-gp in the cortex.

ABSTRACT

Six novel monacolin analogs, monacolins V-V (1-6), together with seven known ones (7-13), were isolated from the ethyl acetate extract of red yeast rice. Their structures and absolute configurations were determined by spectroscopic methods, especially 2D NMR (H-HCOSY, HSQC, HMBC, and NOESY/ROESY) and CD spectroscopic analyses as well as chemical derivation. Monacolins V (2) and V (3) represent the first examples of monacolins with 3-hydroxybutyrate substitute. The anti-inflammatory inhibitory activities against the lipopolysaccharide (LPS) induced NO production in BV-2 cells as well as antioxidant activities against rat liver microsomal lipid peroxidation were evaluated.

ABSTRACT

The purpose of this study was to investigate the expression profile of circular RNA (circRNA) in gastric cancer cells MGC-803, SGC-7901 and NCI-N87 with different degrees of differentiation and normal gastric epithelial cells GES-1. High throughput circRNA microarray technique was used to detect the differential expression of circRNA between three kinds of differentiated gastric cancer cells and normal gastric epithelial cells. The interaction of microRNAs (miRNAs) with circRNAs was predicted by bioinformatics software, and circRNA, which might have great significance in gastric cancer, was identified by literature search. The results showed that there were 79 up-regulated circRNAs and 229 down-regulated circRNAs in gastric cancer cells with different degrees of differentiation compared with those in normal gastric epithelial cells. Through bioinformatics software analysis and literature retrieval, it was found that hsa_circ_0001897 was related to the staging and metastasis of gastric cancer, while the miR-150-5p, which combined with it, was closely related to many kinds of digestive tract tumors, such as colon cancer, liver cancer, and cholangiocarcinoma. The miR-16-5p, which has binding sites with hsa_circ_0008106 and hsa_circ_0060456, has been confirmed to be involved in the development of gastric cancer. The above results suggest that hsa_circ_0001897, hsa_circ_0008106 and hsa_circ_0060456 may be closely related to the occurrence and development of gastric cancer by interacting with miRNA.

Objective:To investigate the influence of Portulaca extracts on H1R,PAR-2 and TRPV1 in skins of rats and the regulation mechanism of H1R,PAR-2/TRPV1 itch signal pathway with atopic eczema.Methods:30 SD rats were randomly divided into normal control group,model group and Porulaca group,each group contained 10 rats.In addition to the normal group,2,4-dinitrochloro-benzene was used on the rest of group for making rat model with atopic eczema.After the success of the model manufacture,each group was given corresponding drugs.After the last administration,EASI was evaluated.The levels of H1R,PAR-2 and TRPV1 in rats skins were detected by immunohistochemical method.Ca2+concentration was analyzed by flow cytometry.Results: Compared with normal control group,EASI(P<0.01),the levels of H1R(P<0.05) and PAR-2(P<0.01),and Ca2+concentration(P<0.01) of rats in model group were significantly increased.The levels of TRPV1 were not increased obviously (P>0.05).Compared with model group,EASI (P<0.01),the levels of H1R(P<0.01) and PAR-2(P<0.01),and Ca2+concentration(P<0.05) of rats in Porulaca group were sig-nificantly reduced,and the levels of TRPV1 were not decreased obviously(P>0.05).Conclusion:Purslane could reduce the levels of H1R,PAR-2 and Ca2+concentration to treat the acute eczema.The mechanism may be to lose the activation of downstream molecule TRPV1 and reduce the inflow of Ca2+by reducing the levels of upstream molecules H1R and PAR-2,then to achieve the effect of anti itch.

<p><b>OBJECTIVE</b>To study the association of blood lipids with the development, clinical stage, allergic condition, and pulmonary function of asthma.</p><p><b>METHODS</b>A total of 56 children with asthma who attended the hospital between October 2016 and March 2017 were enrolled as the asthma group, and 46 children who underwent physical examination as the healthy control group. According to the clinical manifestations, the children with asthma were divided into acute exacerbation group (n=24) and chronic persistent group (n=32). According to the results of skin prick test (SPT) and serum IgE measurement, the children with asthma were divided into non-allergic asthma group (n=16) and allergic asthma group (n=38). Fasting blood lipid levels were measured in both asthma and control groups. Pulmonary function tests were performed for asthmatic children.</p><p><b>RESULTS</b>There were no significant differences in blood lipid levels between the asthma and control groups (P>0.05). The acute exacerbation group had significantly lower serum levels of high-density lipoprotein (HDL) and total cholesterol compared with the control group and the chronic persistent group (P<0.05). The allergic asthma group had a significantly lower serum HDL level than the non-allergic asthma group (P<0.05). In asthmatic children aged 6-13 years, the ratios of the measured values to the predicted values for forced vital capacity, peak expiratory flow, and maximal expiratory flow at 50% of vital capacity had a linear regression relationship with HDL and were positively correlated with HDL (P<0.05). Forced expiratory volume in one second and maximal mid-expiratory flow had a linear regression relationship with both HDL and LDL and were positively correlated with them (P<0.05).</p><p><b>CONCLUSIONS</b>Blood lipids are associated with the clinical stage, allergic condition, and lung function of childhood asthma. This indicates that blood lipids may be involved in several aspects of the pathogenesis of childhood asthma.</p>

With the development of molecular biology and genomics,metagenomics is playing a more important role in forensic science and forensic identification.In recent years,as a branch discipline studying the composition profile and diversity of microbe flora as well as studying the interaction within microbe and with environment,the application of metagenomics has gradually risen and brought new opportunities for forensic identification-related area.In this review,strategy of metagenomics and its application in forensic identification including individual identification,origin determination of biological stain in crime scene and drug abuse detection are summarized.This article aims to elucidate the role and application value of metagenomics in forensic science.

ABSTRACT

Objective:The purpose of this study was to explore the predictive value of PNI in surgical site infections (SSIs) for colorectal cancer patients undergoing enterectomies.Methods:A retrospective review of 332 colorectal cancer patients who underwent colorectal surgery between 2014 and 2015 was performed.Univariate and multivariate analyses were conducted to identify risk factors for SSIs,and receiver operating characteristic (ROC) cure was used to quantify the effectiveness of PNI.Results:SSIs were diagnosed in 43 (12.1%) patients,with incisional infection or deep incisional infection in 36 patients and organ/space infection in 7 patients.ROC curve analysis defined a PNI cut-off level of 48 corresponding to postoperative SSIs (area under the curve [AUC] =0.763,78.1 % sensitivity,62.7 % specificity).Furthermore,a multivariate analysis indicated that operation time ≥ 3 h (OR=3.42,95 % CI:1.22-8.78,P =0.04) and PNI < 48 (OR=2.35,95% CI:1.20-4.53,P =0.03) were independently associated with postoperative SSIs.Conclusion:Preoperative PNI is a simple and useful marker to predict SSIs in colorectal cancer patients after enterectomies.

Objective:To explore the reform of procurement management of university under the policy of streamlining administration and delegating power, combination of delegation and management, and optimizing service (delegation, management and services).Methods: Through analyzed the current status of the procurement management of university to explore the method of implementing the policy of "delegation, management and services" from deepening institutional reform, enhancing management efficiency and innovating service level and other aspects.Results: A set of system of procurement institution that conformed with the requirement of "delegation, management and services" has been established. Through formulated prevention and control measure of integrity risk and adopted informatization method to omnibearingly promote the normalization of procurement service.Conclusion: System of science and reasonability and mean of informatization management can ensure the procurement management to preferably serve the teaching and research work of university.

ABSTRACT

Objective To present novel measures in endoscopic removal of foreign bodies in the upper gastrointestinal tract. Methods The clinical trial performed a retrospective analysis of emergency endoscopic management of 251 cases of foreign bodies in the upper gastrointestinal tract from January 2013 to November 2016. The patients underwent gastric endoscopic examinations under local pharyngeal anesthesia or general intravenous anesthesia. Flexible endoscopes and retrieval accessories such as rat-tooth and alligator forceps, polyp snares, polyp graspers, dormia basket, lithotripter or grasping forceps or rubber gloves were used for the endoscopic procedures. We reviewed of patients' general data (age, sex, type of foreign body), type of anaesthesia, foreign bodies' locations, type of accessory, complications, success rate. Results A total of 251 foreign bodies were definitely diagnosed through endoscopy, 248 foreign bodies were successfully removed with compatible devices according to the foreign bodies' location, nature, and size, and 3 cases failed. At the same time, 192 cases had little mucosal laceration and bleeding, and 45 cases were found to have underlying diseases. 1cases had esophageal surgery and 1cases had laryngeal surgery. 1 cases refuse to remove the foreign body because no symptoms and therapheutic risks. Conclusion Emergency endoscopic management is the foremost option for foreign bodies in the upper gastrointestinal tract, for its obvious advantages in safety, efficiency, low cost, and minimal invasion. Endoscopic intervention can also diagnose early underlying digestive diseases. Innovative measures can increase the successful rate of removing foreign bodies with gastric endoscopy.
