ABSTRACT

Objective To investigate the expression and significance of monoacylglycerol O-acyltransferase 2 (MOGAT2) in the tissues of pterygium. Methods Immunohistochemistry staining methods were adopted to detect the expression of MOGAT2 in 54 patients of pterygia (primary in 50 patients and recurrence in 4 patients)and 18 patients of normal conjunctival tissues. The semi-quantitative integration method was used to analyze the strength of immunohistochemical expression, and the results were compared and statistically analyzed, and the relationship between the expression of MOGAT2 in the pterygium group and the factors such as age, sex, and clinical pathological stage was analyzed. Results Immunohistochemical staining showed that,in 54 patients of pterygium,MOGAT2 was strongly expressed in 8 patients, positive expression in 29,weakly positive expression in 8 and negative expression in 9,while in 18 normal conjunctival tissues, strongly positive expression of MOGAT2 was found 1 patient, positive expression in 6,weakly positive expression in 3,negative expression in 8,respectively.Rank-sum test showed that the difference in MOGAT2 expression in the pterygium and normal conjunctiva tissues was statistically significant(Z =-2.403,P =0.016).There was no significant difference in the expression of MOGAT2 between the different age and the sex patients with pterygium (all P > 0.05),and there was no significant correlation of MOGAT2 expression with education, occupation, income, protection in outdoor work, smoking, drinking, blood sugar, blood pressure, primary and recurrence pterygium, but significant correlation with the clinical stage of pterygium, that is,MOGAT2 expression in advanced pterygium was overexpressed when compared with tumor tissues in quiescent stage (P < 0.05).Conclusion MOGAT2 is highly expressed in pterygium tissues, and the expression in advanced pterygium was significantly upregulated,of which is related to the formation and development of pterygium.

ABSTRACT

BACKGROUNDNerve growth factor (NGF) is well-known for its important role in the development and maintenance of the nervous system. Along with its neurotrophic role, NGF has been detected in the testis of mouse, rat and human, suggesting an additional non-neurotrophic effect in the male reproductive system. The expression of β-NGF in the undescended testes (cryptorchidism) has not been detected at present. The aim of this study was to evaluate the expression of β-nerve growth factor mRNA and protein in experimental cryptorchidism.

METHODSA unilateral mechanical cryptorchidism model in the Sprague-Dawley rat was established and the expression of β-NGF with histologic changes in experimental cryptorchidism were investigated using one step quantitative real-time reverse transcription-polymerase chain reaction, in situ hybridization histochemistry, immunofluorescence and hematoxylin-eosin staining.

RESULTSThe expression of β-NGF mRNA and protein were both significantly decreased in the development of unmarred testis and cryptorchidism-induced testis, and the decrease of β-NGF in cryptorchidism-induced testis was far greater than that in uninjured testis.

CONCLUSIONFrom this investigation, we confirmed a lower expression of β-NGF in undescended testes than in the development of testis.

ABSTRACT

Objective To discuss the histology change, apoptosis state and Bcl-2,Bax expression after the bone cement leakage into the intervertebral disc in vertebroplasty. Methods Eight majority canis familiaris were studied.Three lumbar intervertebral discs(L2 to L5)in each dog were randomly classified into three groups(control group,PMMA group, and CPC group),the canine discs were stabbed by 18-gauge needle, and 0.1 ml cement was injected into them.Control discs were only stabbed and injected with nothing.Histology of all discs was studied 24 weeks after the operation.Terminal-deoxynucleotidyl transferase mediated nick end labeling(TUNEL)and immunohistochemisty were used to detect apoptosis and Bcl-2,Bax expression in the discs. The data were statistically analyzed by SPSS 12.0.Results Intervertebral disc degeneration was not found in control groups. In bone cement groups, however, ruptured or serpentine patterned fibers, decreased cellularity of the nucleus pulposus and condensed matrix of the nucleus pulposus were found in histologic results. The Bax protein decreased in the order of control group, CPC group, and PMMA group. However, the Bcl-2 protein increased in the order of control group,CPC group, and PMMA group. The histology grade was significantly different among the three groups under ANOVA analysis(P
