ABSTRACT

Four main flavonoids of the Chinese medicine Rhododendri Daurici Folium were studied using the density functional theory (DFT) B3LYP method with 6-311 + + G (d, p) basis set.Their activities were analyzed based on molecular structure, bond dissociation energy (BDE) and the energy gap between HOMO and LUMO. As a result, the antioxidant ability order of the four flavonoids compounds is farrerol<kaempferol< quercetin <myricetin base on the work.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate whether the administration of the ultra-filtration extract from Danggui Buxue Decoction (EDBD) was able to protect cardiomyocytes from oxidative injury of rats induced by hydrogen peroxide (H(2)O(2)) and its potential mechanism.</p><p><b>METHODS</b>Myocardial cells from 1- to 2-day-old neonatal rats were cultured in Dulbecco's modified Eagle's medium low-glucose and Ham's F12 medium (1:1), and the cellular injury was induced by H(2)O(2). The ultra-filtration extract mixture from Angelica sinensis and Hedysarum polybotrys was given in three doses of 3.75, 7.5, and 15 mg/mL. Morphological changes of cardiomyocytes were observed by microscope. Survival rate of myocardial cells was assessed using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. The cardiomyocyte damages were estimated by detecting lactate dehydrogenase (LDH) and creatine kinase (CK) releases in the medium, superoxide dismutase (SOD) activities, and intracellular malondialdehyde (MDA) and myeloperoxidase (MPO) contents. The levels of caspase-3 and heat shock protein 70 (hsp70) mRNA expression in cardiomyocytes were measured by reverse transcription polymerase chain reaction.</p><p><b>RESULTS</b>The EDBD could protect the cardiomyocytes from H(2)O(2) injury in a dosedependent manner (3.75, 7.50, and 15.00 mg/mL). The EDBD could significantly decrease LDH and CK leakages and intracellular MDA and MPO contents, increase SOD activity, up-regulate hsp70 expression, and down-regulate caspase-3 expression.</p><p><b>CONCLUSION</b>The EDBD has protection on cardiomyocytes injured by H(2)O(2) through improving cell antioxidant ability, up-regulating hsp70 expression, and inhibiting caspase-3 activity.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the erythrocyte immuno-regulatory effect of Patrinia scabra Bunge extracts extracted by macroporous adsorptive resins in tumor bearing mice.</p><p><b>METHODS</b>Patrinia scabra Bunge was extracted by macroporous adsorptive resins, and the amount of polysaccharides and saponins in the extract were determined. Mice bearing S180 tumor were treated with the extract and their survival prolongation rate, erythrocyte rosette formation rates of C3b receptor (ERR-CR), immune complex (ERR-IC) and tumor cell (ERR-TC), as well as the CD35 and CD44s were observed.</p><p><b>RESULTS</b>Polysaccharide content was 21.4%, saponin 41.8% in the extract. As compared with the model group, the survival rate was increased, the erythrocyte immune function was improved (showed increase of ERR-CR and ERR-TC, decrease of ERR-IC), and the amount of CD35 and CD44s in red blood cell membrane increased in mice after being treated with the extract (P < 0.05 or P < 0.01).</p><p><b>CONCLUSION</b>Extract of Patrinia scabra Bunge extracted by macroporous adsorptive resins can regulate the erythrocyte immune function to a certain extent.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To observe the effect of traditional Chinese herbs Fuzheng Yiliu Granule (FYG)-contained serum from tumor bearing mice on apoptotic rate, free radicals content and mitochondrial membrane potential of hepatoma cell lines H22 in vitro.</p><p><b>METHODS</b>The effect of FYG drug-serum on apoptosis of hepatoma cell line H22 was determined using flow cytometry. The changes of DNA RNA, free radicals and mitochondrial membrane potential (delta psi m) in H22 cell were detected through laser scanning confocal microscope.</p><p><b>RESULTS</b>FYG-contained serum can induce the apoptosis of H22 cell, enhance the free radicals content, and reduce the content of DNA RNA and delta psi m of H22 cell in vitro.</p><p><b>CONCLUSION</b>The apoptosis of hepatoma cell line H22 induced by FYG is probably correlated to the change of free radicals content and delta psi m.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the clinical efficacy of Fuzheng Jiedu Granule (FJG) in coronary heart disease (CHD) patients in long term contact with nickel (Ni) and its effect on electrocardiogram (ECG) and cardial enzymes.</p><p><b>METHODS</b>Sixty cases were randomly assigned to two groups with 30 cases in each group, the treatment group treated with FJG plus Western medicine and the control group given Western medicine alone. The therapeutic course in both groups was 4 weeks. Another 15 healthy subjects from the same region were taken as the healthy control. Changes of clinical symptoms and ECG were observed, and the serum levels of creatine kinase (CK), lactic acid (LD), lactic dehydrogenase (LDH), Na+-K+-ATPase, Ca2+-ATPase and Ni were detected before and after treatment.</p><p><b>RESULTS</b>The ameliorative effects on symptoms, signs and ECG in the treatment group were superior to those in the control group (P<0.05). Compared with healthy subjects, the serum levels of CK, LD, LDH and Ni were higher and the serum levels of Na+-K+-ATPase and Ca2+-ATPase were lower in patients before treatment; after treatment, the decrease of serum CK, LD, LDH and Ni levels and increase of serum Na+-K+-ATPase and Ca2+-ATPase were more significant in the treatment group than those in the control group (P<0.05 or P<0.01).</p><p><b>CONCLUSION</b>FJG can decrease the serum Ni level, recover the activity of myocardial enzymes, thus to improve symptoms and abnormal ECG figures in CHD patients.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore the inhibitory effect of Fuzheng Yiliu Granule (FYG) on growth and apoptosis of H22 tumor cell, and expressions of p53 and Caspase-3 gene.</p><p><b>METHODS</b>The tumor inhibitory rate of FYG on H22 tumor cell line was observed in vivo, cell apoptosis rate and cell cycle were determined by flow cytometry (FCM), and expressions of wild type p53 and Capase-3 mRNA were determined by RT-PCR.</p><p><b>RESULTS</b>FYG could inhibit the growth of H22 tumor cell at the dose of 12g/kg, 24g/kg, the maximal inhibitory rate up to 51.24% (P < 0.01). By FCM, it was shown that FYG could significantly enhance apoptosis rate of cell line H22, with the peak reached 15.84% (P < 0.01), and cause the tumor cell cycle being blocked at G0/G1 phase, with decrease of cells in S phase. RT-PCR illustrated that FYG could significantly up-regulate the level of p53 and Caspase-3 mRNA expression.</p><p><b>CONCLUSION</b>FYG can significantly inhibit the growth of tumor cell in mice, its anti-tumor mechanisms may relate to the cell apoptosis induction, cell cycle regulation and wild type p53 and capase-3 gene expression enhancing.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To observe the effect of Fuzheng Yiliu Granule (FZYLG) on cell cycle and nuclear transcription factor-kappa B (NF-kappa B) in tissue of esophageal-gastric carcinoma.</p><p><b>METHODS</b>Seventy-six patients with esophageal gastric carcinoma were randomly divided into two groups, the FZYLG group and the control group. FZYLG was given to the former for 15 days. The tumor tissue in both groups was resected and cell cycle and apoptosis rate as well as NF-kappa B were determined by flowcytometry.</p><p><b>RESULTS</b>Level of NF-kappa B in the treated group was significantly higher than that in the control group (P < 0.05). In the treated group, the percentage of G0/G1 stage cells were significantly increased and that of S stage significantly decreased (both P < 0.05). At the same time, obvious cell apoptosis was found in the treated group, the apoptosis rate of which was significantly higher than that in the control group (P < 0.01).</p><p><b>CONCLUSION</b>FZYLG can increase the NF-kappa B expression, block the proliferation to promote the apoptosis of tumor cells.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore the effect and clinical significance of Fuzheng Yiliu Granule (FYG) on nuclear transcriptional factor-kappa B (NF-kappa B) and cell cycle in breast tumor.</p><p><b>METHODS</b>Fifty-five patients of breast carcinoma were randomly divided into the treated group and the control group, both were treated with conventional chemotherapy, but to the treated group, FYG was given additionally by oral taking. Operation was performed one month later to take out the tumor tissue for detecting NF-kappa B expression and ration of cells in different phases of cell cycle.</p><p><b>RESULTS</b>Compared with the control group, NF-kappa B expression was significantly increased in the treated group, accompanied with increased G0/1 phase cell proportion and decreased S phase cell proportion as well as significantly reduced cell proliferation index (all P < 0.01).</p><p><b>CONCLUSION</b>FYG could enhance the expression of NF-kappa B in breast tumor tissue, raise the proportion of G0/1 phase cells, decrease proportion S phase cells and reduce the cell proliferation index, showing an active action on the patients prognosis.</p>
