ABSTRACT

Objective Non-motor symptoms are important prodromal characteristics of Parkinson's disease (PD).The siblings and the spouses of PD patients do not have classic motor symptoms.This study aimed to investigate if they have PD prodromal symptoms.Methods A total of 98 PD patients from the Affiliated Hospital of Yangzhou University were recruited between January 2015 and August 2017;256 siblings of these patients were included in a siblings group,87 spouses of PD patients were included in a spouses group and 250 healthy individuals were included in a control group.Various scales were used to assess non-motor symptoms,including depression,anxiety,cognitive function,sleep status,constipation,daytime sleepiness,subjective olfactory disorder,rapid-eye-movement sleep behavior disorder (RBD),and restless legs syndrome (RLS).Results The incidence of anxiety (OR=3.06,95%CI 1.86-5.05,P<0.01),depression (OR=2.16,95%CI 1.16-4.04,P=0.01),RBD (OR=3.83,95%CI 1.79-8.19,P<0.01) and subjective olfactory disorder (OR=4.48,95%CI 2.02-9.90,P<0.01) was higher in the siblings group than the control group.There were not statistically significant differences in constipation,cognitive impairment,sleep disorder,daytime sleepiness,and RLS between the two groups.There were not statistically significant differences in non-motor symptoms between the spouses group and the control group,except that the mild depression (OR=2.58,95%CI 1.07-6.20,P =0.03) in the spouses group was more obvious.Conclusions The siblings of PD patients are more likely to have PD prodromal symptoms compared with those without PD family history,perhaps because PD patients and their siblings have common pathogenic genetic factors and early living environment for neurodegeneration.There are no obvious non-motor symptoms in the spouses of PD patients.It can be concluded that the onset of PD is not related to the family life environment in adulthood.

ABSTRACT

Objective To investigate the correlation between hemorrhagic transformation (HT) and each parameter of blood pressure variability (BPV) in patients with massive cerebral infarction caused by cardioembolic middle cerebral artery occlusion. Methods The patients with massive cerebral infarction caused by unilateral cardioembolic middle cerebral artery occlusion admitted to hospital within 24 h after onset were selected. For patients without revealing HT with head MRI on admission were analyzed retrospectively. According to the results of MRI rescan within 7 days after onset, they were divided into either a HT group or a non-HT group. Al specific parameters of blood pressure were compared and analyzed between the 2 groups, specificaly including initial value (initial), mean value ( mean), maximum value (max), minimum value (min), maximum-minimum difference (max-min), standard deviation (sd), successive variation (sv), and maximum value of sv (svmax) of systolic blood pressure (SBP) and diastolic blood pressure (DBP). Results A total of 146 patients were enroled in the analysis, and 77 (52. 7% ) had HT within 7 d after onset. Al the blood pressure parameters were grouped by quartiles. Multivariate logistic regression analysis showed that after adjusting for age, admission time, baseline National Institutes of Health Stroke Scale score, congestive heart failure, antiplatelet therapy, initial blood glucose, prothrombin time, and international normalized ratio of the patients, SBPmin , SBPmax-min , SBPsv , SBPsvmax , DBPinitial, DBPsd , DBPmax , DBPmax-min , DBPsv , and DBPsvmax had significant independent association with and HT (likelihood ratio test of trend, al P < 0. 05). Conclusions Higher BP level and variability may increase the risk of HT in patients with massive cerebral infarction after cardioembolic middle cerebral artery occlusion. A smooth and slow antihypertensive treatment may need for these patients.

ABSTRACT

Objective:To investigate the neuro protective effects of topiramate on the changes of the nerve function deficiency scores and the influence of it on the content of amino acids neurotransmitter of cerebral cortex of rats with cerebral ischemia.Methods:Male SD rats were subjected to right middle cerebral artery occlusion for 2h and reperfusied for 24 h.Rats in the treatment group were injected with topiramate suspl(8mg/ml,80mg/kg)at the begining of the artery occlusion and the reperfusion.The nerve function deficiency scores were measured.The infarct volume was evaluated through TTC staining.The content of amino acids of cerebral cortex was estimated by the method of high performance liquid chromatography(HPLC)with fluorescent detection.Results:The infarct volume was significantly reduced by 41.8% in the TPMtreated group compared with that in the control group.There were significant differences in the neurological impairment scores between the treatment group and the ischemia group.TPM significantly decreased the content of Glu(P

ABSTRACT

Objective To investigate the effects of Topiramate(TPM) on activity of SOD and concentration of MDA in blood serum and the changes of the nerve function scores after cerebral ischemia/perfusion in rats. Methods Male Sprague-Dawley rats were randomly divided into ischemia/perfusion group, TPM group and sham-operation group. Rat models of transient focal cerebral ischemia were made by 2 h occlusion of right middle cerebral artery occlusion and reperfusion for 24 h.Rats in TPM group were injected TPM(8 mg/ml,80 mg/kg)in the beginning of the artery occlusion and the reperfusion. The rats were sacrificed after they were evaluated by the nerve function deficiency scores. The activity of SOD and concentration of MDA in blood serum were measured.Results The activity of SOD and concentration of MDA in blood serum in ischemia/perfusion group were (157.72?19.04)U/ml and (7.45?0.84 )nmol/ml, those in TPM group were ( 171.25?15.72)U/ml and (( 6.10?0.98) nmol/ml,those in sham-operation group were (179.74?7.95)U/ml and (5.90?0.72 )nmol/ml. ABSTRACT

Objective To investigate the neuroprotective effects of Topiramate (TPM) on rat brain with ischemia-reperfusion damage and its mechanisms.Methods The 30 male SD rats were randomly assigned to sham operated group, ischemia-reperfusion group and TPM treated group.The cerebral ischamia and reperfusion model was made by suture occlusion of right middle cerebral artery(MCAO).Rats in TPM treated group were intraperitoneally injected TPM (80 mg/kg) twice. At 24 h following onset of MCAO,the nerve function score was evaluated with Neurological Grading Scale. The infarction volume was measured with TTC staining. The contents of glutamate (Glu) and gamma-aminobutyric acid (GABA) of cerebral cortex were tested by the high performance liquid chromatography with fluorescent detection.
