ABSTRACT

【Objective】 To investigate the expression of transcription factor POU domain class 2 transcription factor 2 (POU2F2) in clear cell renal cell carcinoma (ccRCC) and human renal cancer cell lines (786-O and ACHN) and its effects on the cells’ biological behaviors such as proliferation, migration and invasion in vitro. 【Methods】 The mRNA expressions of POU2F2 in ccRCC tissues, adjacent normal tissues, cell lines 786-O and ACHN were detected with real-time polymerase chain reaction (qRT-PCR). The protein expression of POU2F2 in ccRCC tissues and adjacent normal tissues were detected with immunohistochemistry. The effects of knockdown of POU2F2 on the mRNA and protein expressions of epithelial mesenchymal transformation (EMT)-related tumor markers were detected with qRT-PCR and Western blot. 【Results】 The mRNA expression of POU2F2 in ccRCC tissues was significantly higher than that in adjacent normal tissues, and was correlated with patients’ gender, WHO/ISUP nuclear grade and TNM stage. The protein expression of POU2F2 was significantly higher in ccRCC tissues than in adjacent normal tissues, and was correlated with tumor pathological grade and TNM stage. The mRNA expression of POU2F2 was significantly decreased in 786-O cells after sh-POU2F2-1013 plasmid transfection (P<0.05); the proliferation ability, clonal formation rate, migration ability and invasion ability were significantly reduced (P<0.05). Knockdown of POU2F2 down-regulated the mRNA and protein expressions of MMP2, MMP9 and Twist in 786-O cells, while up-regulated E-ca expression. 【Conclusion】 The mRNA expression of POU2F2 was significantly up-regulated in ccRCC tissues and renal cancer cells. Knockdown of POU2F2 inhibited the proliferation, migration and invasion of cells in vitro, and slowed or inhibited the occurrence and development of renal cancer.

ABSTRACT

Objective To evaluate the effects of nutritional intervention on the incidence of malnutrition in patients on maintenance hemodialysis (MHD).Methods Totally 196 patients on MHD were randomly assigned to nutritional intervention group (n =98) and control group (n =98).Nutritional intervention or free diet were given in these two groups.The incidence of malnutrition in these two groups was recoded.Results The baseline age,biochemical indicators,and incidence of malnutrition were matched in these two groups (P >0.05).After 6 months,although the number of the patients with malnutrition [35.7% (35/98)] was slightly increased in the intervention group,it showed no significant difference when compared with the baseline level [33.7% (33/98),P >0.05].In the control group,however,the incidence of malnutrition [59.2% (58/98)] significantly increased (P <0.05).The incidence of malnutrition was significantly lower in the intervention group than in the control group (P < 0.05).Conclusion Nutritional intervention can lower the incidence of malnutrition in patients on MHD.

ABSTRACT

Objective To evaluate the efficacy and safety of sunitinib as post-operative adjuvant therapy in patients with high-risk renal cell carcinoma (RCC).Methods A total of 60 patients with resected,histologically confirmed clear cell RCC were enrolled in this study.Patients received orally sunitinib either at a dose of 50 mg on treatment schedule (once daily for 4 weeks followed by 2 weeks off) or at a dose of 37.5 mg once daily for three 6-week cycles from 1 month after surgery.Results All the 60 patients tolerated Sunitinib treatment well and no patient discontinued treatment due to adverse events.Most adverse events were grade Ⅰ to Ⅱ.The most frequently reported adverse events were neutropenia (56.7%),thrombocytopenia (53.3%),leucopenia (48.3%),hand-foot syndrome (46.7%) and hypertension (36.7%).The most frequently reported grade 3 or 4 toxicities were thrombocytopenia (25.0%),neutropenia (15.0%),hand-foot syndrome (11.7%) and leucopenia (8.3%).The majority of adverse events occurred within the first 1-2 cycles of sunitinib treatment,and was ameliorated 1 month after 3 cycles finished.No irreversible adverse event was observed.As of April 5,2012,no recurrence occurred in patients except one death due to cerebrovascular accident unrelated to treatment,with both 6-month and 9-month disease-free survival rate of 100%.Conclusions Myelosuppression occurred less frequently in high-risk RCC patients treated with sunitinib as operative adjuvant therapy than in advanced RCC patients,with a better benefit trend.However,long-term follow-up data are needed to further confirm the efficacy of sunitinib in the adjuvant setting.

ABSTRACT

Objective To investigate the changes of urinary bladder structure and function of aging rats. Methods The aging group and the young adult group were composed of 24 month old rats and 6 month old rats, respectively. The urodynamic changes were studied with filling cystometry, structural changes by Masson stain and image analysis. Results Filling cystometry revealed that the maximum bladder capacity increased in the aging rats. No difference in leak point pressure was found in the two groups. The occurrence rate of the detrusor instability in aging group was higher than that in young adult group. In bladder structure, fibrous tissue increased significantly but smooth muscle decreased significantly in the detrusor of aging rats. Conclusion Aging may lead to changes of rat bladder structure and function, but its mechanism needs to be further studied.

ABSTRACT

Objective To evaluate objectively the traditional methods on the assessment of bladder outlet obstruction(BOO) due to BPH. Methods Correlation between the urodynamic findings and the traditional diagnostic parameters such as age,IPSS,Vp,Qmax z and PVRr was studied.The clinical prostatic score(CPS),derived from multiple regression of clinical parameters depending on URA,was evaluated. Results The parameters such as age,IPSS,Vp,Qmax z and PVRr were evaluated with reference to pressure flow study.Every parameter alone was not enough for BOO diagnosis. The regression equation was CPS=49.8-3.3 Qmax z+0.5 IPSS+0.2 Vp+7.5 PVRr.Correlation coefficient between CPS and urodynamic findings was 0.629 and was significantly higher than that of any clinical parameter alone.With CPS≥35,the sensitivity was 83.7% and specificity 85.8% for the diagnosis of BOO.With CPS

ABSTRACT

Objective To study the preventive effect of high-dose immuno-chemotherapy on the recurrence and metastasis of renal cell carcinoma(RCC). Methods Overall 18 patients with RCC(13 men and 5 women;mean age,56 years,range,37 to 78 years) underwent radical nephrectomy.These cases were assigned to 2 groups.①Immuno-chemotherapy group included 13 cases (10 men and 3 women),whose tumor stages were T 1 (1 case),T 2 (8) and T 3(4).They were treated with high-dose subcutaneous interleukin-2(IL-2) in combination with interferon-?(IFN-?) and venous infusion 5-fluorouracil(5-Fu) 10 days after operation.②Control group included 5 cases (3 men and 2 women),whose tumor stages were T 1(1 case),T 2(3),T 3(1).They didn’t receive immuno-chemotherapy.The immuno-chemotherapeutic courses consisted of IL-2 (8?10 6 U,3 times/w,weeks 1 and 4;4?10 6 U,3 times/w,weeks 2 and 3);IFN-? 6?10 6 U,3 times/w,weeks 2,3,6 and 7;9?10 6 U,3 times/w,weeks 5 and 8);and 5-Fu (1000 to 1250 mg,once a week,weeks 5 to 8).Such therapeutic cycle could be repeated at 3-month intervals.Follow-up was conducted with hemogram,hepatic and renal function,ultrasonography of liver and kidney,chest X-ray every month after treatment.Long-term outcomes for these patients were analyzed. Results Follow-up of 60 to 87 months was carried out after 1 to 4 therapeutic cycles(mean,1.9 cycles) in the 13 cases of immuno-chemotherapy.Of them,12 cases experienced cancer-free survival.Only 1 case had metastasis in thigh bone 43 months after operation.No severe side effects occurred.5 cases in control group were found with metastasis in 3 to 14 months after operation.The mean survival of control group was 12.8 months (range,6 to 21 months). Conclusions Immuno-chemotherapy with high-dose subcutaneous IL-2 in combination with IFN-? and venous infusion of 5-Fu is safe and effective in prevention of the recurrence and metastasis of RCC.
