ABSTRACT

Objective:To explore the predictive performance of machine learning model based on vascular risk factors in early prediction of vascular cognitive impairment. Methods:From April to September, 2020, 70 subjects were enrolled and collected information of the demographics and vascular risk factors. They were assessed with Montreal Cognitive Assessment (MoCA), and then divided into normal group, vascular mild cognitive impairment (VaMCI) group and dementia group. The differences of vascular risk factors among the three groups were detected with one-way ANOVA, and the significant factors were selected to establish predictive models with support vector machine (SVM) and extreme learning machine (ELM). The predictive performance of two models was compared with Receiver Operating Characteristic Curve. Results:There were 32 cases in the normal group, 23 in VaMCI group and 15 in dementia group. Systolic blood pressure, fasting blood glucose, total cholesterol, low density lipoprotein and blood uric acid were significantly different among the three groups (F > 3.318, P < 0.05). The area under curve was the most (0.911) in SVM model predicting for VaMCI (P < 0.01), and the predictive efficacy was better for SVM model. Conclusion:SVM predictive model based on vascular risk factors may be more effective for predicting VaMCI.

ABSTRACT

METHODS@#To establish the acquired aplastic anemia mouse model through the X-ray irradiation in combination with lymphocytes injection. AA Group: the purified Pan T lymphocytes from the spleen of C57BL/6J mice were enriched and injected to the mice through tail vein(5×10@*RESULTS@#Compared with 4, 5 Gy irradiated mice in AA groups, the survival time of 3 Gy irradiated AA groups was significantly prolonged. 3, 4 and 5 Gy X-ray irradiation combined with Pan T lymphocyte injection could successfully induced severe reduction of red blood cells, blood neutrophils, and platelets, severe reduction of bone marrow nucleated cells, severe bone marrow hematopoietic failure, and the significant expansion of T lymphocytes ratio in the bone marrow. ABSTRACT

Objective:To investigate the feasibility of mandibular angle osteotomy combined with high-density polyethylene (MEDPOR) prosthesis implantation for one-time lower facial contouring of the protuberance and related surgical precautions.Methods:A total of 18 patients who were treated in the oral and maxillofacial surgery of Yangzhou First People's Hospital from June 2010 to August 2016 were selected to complete the mandibular angle osteotomy combined with MEDPOR prosthesis implantation in the lower part of the prosthetic plane. The patients were followed up regularly and the effect of surgery was observed.Results:All 18 patients were operated successfully with an average time of 2 hours. The effect of operation was satisfactory. Except for one patient whose hematoma caused local bruise to subside completely in 2 weeks, all the others were discharged smoothly in one week after operation. After 2 months to 3 years of follow-up, the effect was satisfactory.Conclusions:Mandibular angle osteotomy combined with MEDPOR prosthesis is safe and effective.

ABSTRACT

Objective To study the relationship between myofibril fragmentation index (MFI) of human skeletal muscle and postmortem interval (PMI). Methods The protein concentrations of human right bi-ceps brachii muscle and right quadriceps femoris muscle were obtained at different PMI, and detected at room temperature by biuret method. The MFI of skeletal muscle at 540 nm was measured by ultraviolet spectrophotometer. Regression analysis was performed with time of death as independent variable (x) and MFI as dependent variable (y). Results In early PMI, the MFI of human skeletal muscle increased obviously according to the prolongation of PMI, and peaking by 12 h and then tended to steady. Within 12 h after death, the regression equations of right biceps brachii muscle and right quadriceps femoris muscle were y=32.660+3.227 x(r=0.9879) and y=32.380+3.495 x(r=0.9839), respectively. Conclusion There's high correlation between MFI and PMI. Combining with forensic practice, MFI can be used for the estimation of early PMI (especially in 12 h).

ABSTRACT

Objective To study the association of Hp infection and insulin resistance (IR) with vascular dementia (VaD).Methods Eighty-six elderly VaD patients admitted to our hospital from April 2015 to April 2016 served as a VaD group and 52 healthy subjects undergoing physical examination served as a control group in this study.The patients in VaD group were further divided into Hp-positive group (n =50) and Hp-negative group (n =36).Their fasting blood glucose (FBG) and fasting insulin (FINS) levels were measured with positive C13 urea breath test as the diagnostic index of Hp infection.HOMA-IR=FINS× FBG/22.5 was calculated according to the self balance model analysis.Results The positive Hp rate,serum FBG and FINS levels,and HOMA-IR were significantly higher in VaD group than in control group (58.14% vs 28.58%,x2=10.273,P=0.002;5.53±0.60 mmol/L vs 5.19±0.38 mmol/L,P=0.0004;10.29±4.95 μIU/L vs 8.77±4.02 μIU/L,P=0.0317;2.57±1.34 vs 2.06±0.96,P=0.0179).The serum FBG and FINS levels and HOMA-IR were significantly higher in Hp-positive group than in Hp-negative group (P<0.05,P<0.01).Conclusion Hp infection and IR are closely associated with VaD,and can thus lead to the occurrence or progression of VaD by inducing or aggravating IR.

ABSTRACT

OBJECTIVES@#To explore the correlation between early postmortem interval (PMI) and eight RNA markers of rat's brain at different temperatures.@*METHODS@#Total 222 SD rats were randomly divided into control group (PMI=0 h) and four experimental groups. And the rats in the experimental groups were sacrificed by cervical dislocation and respectively kept at 5 ℃, 15 ℃, 25 ℃ and 35 ℃ in a controlled environment chamber. The RNA was extracted from brain tissues, which was taken at 9 time points from 1 h to 24 h postmortem. The expression levels of eight markers, β-actin, GAPDH, RPS29, 18S rRNA, 5S rRNA, U6 snRNA, miRNA-9 and miRNA-125b, were detected using real-time fluorescent quantitative PCR, respectively. Proper internal reference was selected by geNorm software. Regression analysis of normalized RNA markers was performed by SPSS software. Mathematical model for PMI estimation was established using R software. Another 6 SD rats with known PMI were used to verify the mathematical model.@*RESULTS@#5S rRNA, miR-9 and miR-125b were suitable as internal reference markers for their stable expression. Both β-actin and GAPDH had well time-dependent degradation patterns and degraded continually with prolongation of PMI in 24 h postmortem. The mathematical model of the variation of ΔCt values with PMI and temperature was set up by R software and the model could be used for PMI estimation. The average error rates of model validation using β-actin and GAPDH were 14.1% and 22.2%, respectively.@*CONCLUSIONS@#The expression levels of β-actin and GAPDH are well correlated with PMI and environmental temperature. The mathematical model established in present study can provide references for estimating early PMI under various temperature conditions.

ABSTRACT

AlM:To investigate biological measurement parameters of anterior segment in acute angle- closure glaucoma ( AACG) .METHODS:Forty-six eyes of 46 patients with AACG and 52 eyes of 52 patients with shallow anterior chamber and 50 eyes of 50 normal individuals were examined. The parameters of anterior segment including chamber crowd rate (CCR), lens thickness (LT), lens position (LP) and anterior chamber depth ( ACD ) were measured by A-ultrasound according to different ages in each group. The data were performed statistical analysis in three groups.RESULTS: ln each age range group (≥50 ~ 59 years old, ≥60 ~ 69 years old, ≥70 years old ) , statistically significant differences in three groups ( AACG, shallow anterior chamber group and the controls ) were found in CCR, LT, LP, ACD (P 0. 05 ) between AACG and shallow anterior chamber group in each age range group.CONCLUSlON: CCR can be used as the index of evaluating crowding state of anterior segment in AACG patients and the sensitivity is better than LT and LP.

ABSTRACT

Chinese medicine is a healing medicine which respect the power of human body itself. The essentials of Chinese medicine is promoting health wellness instead of disease management. Modern medicine just realized the limits of its theory. P4 medicine (personalized, predictive, preventive, and participatory/precision) similar to Chinese medicine theory is beginning to representing the pioneer in the Western world. In this review, we summarized different domains of Chinese medicine. Based on the basic of promoting health wellness, we compared the fundamental theory of Chinese medicine to the new merging P4 medicine idea of Western medicine. We also discussed the potential for using modern computational medicine technique to integrate Chinese medicine theory and Western medicine theory.

ABSTRACT

Objective To discuss the application value of multislice CT (MSCT) in diagnosis of ureteral biology variation and diseases.Methods The MSCT data of 51 patients with suspected ureteral biology variation and diseases were retrospectively analyzed.The data of MSCT images included plain scan,renal cortical phase,renal parenchyma phase and excretion phase.The MSCT images of renal cortical phase,renal parenchyma phase and excretion phase were reconstructed respectively to gain the 3D images of renal arteries,veins and urinary passage.Results Among the 51 patients,1 patient with traumatic ureteral rupture,10 patients with ureteral biology variation diseases,14 patients with stone of ureter,2 patients with polyp of ureter,3 patients with non-specificity ureteritis,3 patients with tuberculosis of ureter,1 patient with endometriosis,17 patients with carcinoma of ureter.Satisfied 3D images of renal arteries,veins and urinary passage could be reconstructed.And the imaging diagnosis of 51 patients were all in accord with their clinical diagnosis or pathologic diagnosis.Conclusion MSCT can obtain high qualified images,and this technique should become the preferred inspection item of ureteral biology variation and diseases.

ABSTRACT

<p><b>OBJECTIVE</b>To compare docetaxel plus prednisone with mitoxantrone plus prednisone as first-line chemotherapy for metastatic hormone-refractory prostate cancer (mHRPC).</p><p><b>METHODS</b>From January 2007 through August 2010, 62 patients with mHRPC received 5 mg of prednisone twice daily were randomly assigned to receive mitoxantrone 12 mg/m² every three weeks (group A) or 75 mg/m² every three weeks (group B). The cycles of each regimen were less than 10 times. The primary end point was overall survival. The secondary end points were the prostate-specific antigen (PSA) response rate, the duration of PSA response and the objective tumor response rate (ORR). All the t test, χ² test and Fisher's exact test were performed between 2 groups.</p><p><b>RESULTS</b>Thirty-one patients enrolled in group A received a median 4 cycles of regimen (range 1 - 10), whereas 30 patients enrolled in group B received a median of 7 cycles of regimen (range 2 - 10). There were 45.2% patients in group A and 70.0% in group B had PSA response (χ² = 3.85, P < 0.05). The duration time of PSA response was 121 days (range 20-323 days) in group A and 168 days (range 42 - 447 days) in group B, respectively. The ORR was 15.0(3/20) in group A and 10.3% (3/29) in group B, respectively. The median survival was 511 days (95%CI: 357 - 665 days) in group A and 833 days (95%CI: 634 - 1032 days) in group B, respectively (χ² = 4.20, P = 0.040). The incidence of thrombocytopenia in group A was higher than group B (χ² = 5.60, P = 0.018); the incidences of nausea and vomiting (χ² = 4.32, P = 0.038), diarrhea (P = 0.024), fatigue (χ² = 5.90, P = 0.015), and alopecia (χ² = 5.42, P = 0.020) in group B were higher than group A.</p><p><b>CONCLUSION</b>Docetaxel plus prednisone can lead to superior overall survival and PSA response rate in patients with mHRPC.</p>

ABSTRACT

OBJECTIVE@#To observe the degradation of myoglobin in skeletal and cardiac muscles after death and to explore its usefulness in estimation of postmortem interval.@*METHODS@#Human skeletal and cardiac muscles were collected at 0, 4, 8, 12, 24, 36, 48, 60, and 72 hours after death and stored at 25 degrees C. The protein was extracted and examined and analyzed by Western blot and imaging technique.@*RESULTS@#Myoglobin showed a continuously postmortem degradation in both skeletal and cardiac muscles. There was no observed difference in degradation between the two tissues.@*CONCLUSION@#Changes in myoglobin degradation could be a useful parameter for estimation of the postmortem interval.

ABSTRACT

<p><b>BACKGROUND</b>Newborn blood vessel formation correlates closely with active cellular proliferation of tumor. The aim of this study is to explore the relationship of expression of COX-2 and VEGF, MVD with CT enhancement appearance of lung cancer.</p><p><b>METHODS</b>Dynamic CT scans were evaluated in 25 patients with lung cancer and 35 patients with pulmonary benign disease who underwent complete resection and did not receive any chemotherapy or radiotherapy prior to operation. Expression of COX-2, VEGF and MVD were detected by immunohistochemistry.</p><p><b>RESULTS</b>The levels of COX-2 (P < 0.05), VEGF (P < 0.05), MVD (P < 0.05) and CT enhancement value (P < 0.01) in lung cancer group were significantly higher than those in plumonary benign tumor group. The levels of COX-2, VEGF and MVD were closely related to CT enhancement value, histological type, clinical stages and lymph node metastasis, but not to differentiation grade of lung cancer.</p><p><b>CONCLUSIONS</b>COX-2, VEGF and MVD may be important molecular biological predictors in early diagnosis and prognosis estimation of lung cancer. Dynamic CT enhancement scans can reflect the vascularity characteristics of lung cancer, and it may be helpful to determine the involvement, metastasis and prognosis of lung cancer.</p>

ABSTRACT

<p><b>AIM</b>To develop an HPLC-MS assay for determination of donepezil in human plasma and to investigate the pharmacokinetics and bioequivalence of donepezil capsule in healthy volunteers.</p><p><b>METHODS</b>A randomized crossover design was performed in 20 healthy volunteers. In the two study periods, a single 5 mg dose of either capsule or tablet was administered to each volunteer. After spiked with the internal standard (phenoprolamine) and treated with saturated sodium bicarbonate, plasma was extracted with ethyl acetate and separated with a C18 reversed phase column. LC-ESIMS was used in the selected ion monitoring (SIM) mode with target ions at m/z 380 for donepezil and m/z 344 for phenoprolamine. The fragmentor voltage was 120 V. The main pharmacokinetic parameters of donepezil and the bioequivalence of its two preparations were calculated.</p><p><b>RESULTS</b>The main pharmacokinetic parameters T1/2, Tmax and Cmax were (63 +/- 10) h, (3.3 +/- 0.4) h and (8.5 +/- 0.4) microgram.L-1 for the capsule; (57 +/- 9) h, (3.4 +/- 1.0) h and (8.1 +/- 1.0) microgram.L-1 for the tablet, respectively. The relative bioavailability of the donepezil capsule was 102% +/- 11%.</p><p><b>CONCLUSION</b>The assay was shown to be sensitive, accurate and convenient. The two preparations of donepezil were bioequivalent.</p>

ABSTRACT

Objective To research the relationship between the expression of cyclooxygenase-2(COX-2),vascular endothelial growth factor(VEGF),microvessel density(MVD)and the CT appearances of lung cancer.Methods CT scans were performed respectively in 35 patients with benign disease and 25 patients with lung cancer confirmed pathology.The immunohistochemistry of all pathologic specimen was analysed using P-V method.The relationship between the expression of COX-2,VEGF,MVD and the CT appearances,histological types,clinic stages,lymph nodes metastasis of lung cancer was evaluated.Results The expression level of COX-2,VEGF,and MVD in lung carcinoma was(64.8?13.8)%,(76.4?14.1)% and(62.5?14.6)/HP respectively,it was higher than that in the lung benign disease group.The level of expression of COX-2,VEGF and MVD was closely related to the CT appearances,histological types,clinic stages and lymph nodes metastasis of lung cancer,but there was no relationship with differentiation of carcinoma.Conclusion COX-2,VEGF and MVD may be important molecular biological predictors in diagnosing lung cancer early and can evaluate prognosis of patients with lung cancer.CT appearances can reflect the vascular characteristics of lung cancer,and can predict the involvement,metastasis and prognosis of lung carcinoma according to the appearances of CT.

ABSTRACT

Objective To research the value of dynamic CT enhancement in diagnosing lung cancer.Methods 35 patients with benign disease and 25 patients with lung cancer underwent dynamic CT scan,the results of CT were analysed in comparison with that of pathological specimens,the relationship between the dynamic CT enhancement phenomena and histological results.Results CT enhanced peak,the enhanced ratio of focus and aorta,the form of enhancement and time-dense curve were different between two groups.CT enhanced peak was related to the histological types and clinical stages of pulmonary cacinoma,and metastasis of lymph nodes.Conclusion Dynamic CT enhanced scan can reflect the characteristic of blood in lung cancer.The invasion and metastasis in lung carcinoma can be predicted according to the CT enhanced peak.
