ABSTRACT

OBJECTIVE@#To investigate the correlation of iron metabolic parameters with platelet counts in blood donors.@*METHODS@#A total of 400 blood donors who met requirements of apheresis platelet donation were collected, and their hematological parameters were analyzed. The donors were divided into low ferritin group and normal group, the differences of hematological parameters between the two groups were compared, and the correlation of iron metabolic parameters and routine hematology parameters with platelet counts were analyzed.@*RESULTS@#Whether male or female, low ferritin group had higher platelet counts than normal group (P < 0.01). Among the iron metabolic parameters, the platelet counts was negatively correlated with serum ferritin (SF), serum iron (SI), and transferrin saturation (TSAT) (r =-0.162, r =-0.153, r =-0.256), and positively correlated with total iron binding capacity (TIBC) and unsaturated iron binding capacity (UIBC) (r =0.219, r =0.294) in female blood donors. Platelet counts was also negatively correlated with SF, SI and TSAT (r =-0.188, r =-0.148, r =-0.224) and positively correlated with UIBC (r =0.220) in male blood donors. Among the routine hematology parameters, platelet counts was negatively correlated with mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCH), mean corpuscular hemoglobin concentration (MCHC), and reticulocyte hemoglobin equivalent (Ret-He) in female blood donors (r =-0.236, r =-0.267, r =-0.213, r =-0.284). Platelet counts was also negatively correlated with MCH, MCHC and Ret-He in male blood donors (r =-0.184, r =-0.221, r =-0.209).@*CONCLUSION@#In blood donors with low C-reactive protein level, the lower the iron store capacity, the lower the iron utilization, and the platelet counts tends to rise.

ABSTRACT

Objective To investigate the differences between the sequences of papA and papG of UPECA030 strain and the related genes, to better understand the genetic variation of UPEC4030 papA and its combination forms with papG so as to identify if it was a new genotype. Methods Cloning and sequencing methods were used to analyze the sequences of papG and papA of UPEC4030 strain and to compare their related sequences. Results Through sequence analysis of papA, it was revealed that there was a 722 bp gene,encoding 192 amino acid polypeptide. The overall homology of the papa genes between UPEC4030 and the standard strains of ten F types were 36.11%-77.95 % and 22.20%-78.34% at nueleotide and deduced amino acid levels. Homology between the sequences of reverse primers and the corresponding sequence of UPEC4030 papA was 10.00%-66.67%. The results confirmed that UPECA030 strain contained a novel papA variant. Through sequence analysis of UPEC4030 papG, we revealed a 1100 bp gene, encoding 337 amino acid polypeptide. The homology of the papG genes between UPEC4030 and UPEC IA2, the standard strain, was 99.00 % at nucleotide level and 99.11% at deduced amino acid and UPEC4030 strain carried class I] genotype of papG. Conclusion UPEC4030 strain contained an unknown papA variant or the new genotype and carried class II genotype of papG. The pathogenic mechanism and epidemiology call for further study.

ABSTRACT

Klebsiella pneumoniae was cultured followed by the preparation and immunoactivity elucidating of its polysaccharide (CPS). The lysis of cell is the first key step in the preparation, under the co-action of trypsin, lysozyme and NP-40, the cell lysed within 2h, then the lysate was concentrated by ultrafiltration which serves as concentrating and partial purifying action simultaneously. Crude CPS was got by ethanol precipitation, then purified through the Ion-exchange and gel filtration, the purity of CPS was judged by the gel filtration and agarose gel electrophoresis. The effect of CPS on the cell immunoactivity was studied in detail, the results show that CPS possesses bidirectional immunoregulation on the spleen cells of mice, that is, low concentration of CPS can stimulate the immune response while the high concentration manifests the inhibition significantly. The investigation results will benefit on the exploitation of the CPS.

ABSTRACT

<p><b>OBJECTIVE</b>To analyse the pathogen of child patients with influenza in Tianjin area.</p><p><b>METHODS</b>The influenza virus isolation was performed by MDCK cells and embryonated eggs. The identification of the isolates was carried out with hemagglutination (HA) and hemagglutination inhibition (HI) tests.</p><p><b>RESULTS</b>Two hundred and thirty-eight throat swab specimens from children with influenza-like illness were collected in Tianjin area from Oct. 2001 to Mar. 2002 and 64 strains (26.9%) of influenza virus were isolated. Data showed that there were 42 strains (65.6%) of A (H3N2) subtype, 13 strains (20.3%) of A (H1N1) subtype and 9 strains (14.1%) of B type in these positive isolates. All the isolated viruses grew very well in MDCK cells and hemagglutinated with human "O" red blood cells, and most (62/64 strains) of them were able to multiply in embryonated chick eggs. However, there were only 3 isolates with HA positive in inoculating embryonated eggs with the specimens. Meanwhile, it was revealed that out of 55 strains of A type viruses, 53 strains (96.4%) were from O to D phase, 2 strain of A (H3N2) were D phase characters and all B type isolated viruses being D phase properties.</p><p><b>CONCLUSION</b>There were three endemic types of influenza viruses-A (H3N2), A (H1N1) and B type in Tianjin area, with A, the main type.</p>
