ABSTRACT

Objective::To investigate the regulatory effect of Danggui Shaoyaosan (DSS)-containing serum on oxidative stress and inflammation in H2O2-induced SH-SY5Y cells. Method::Methyl thiazolyl tetrazolium(MTT) assay was used to determine the cell activity and construct the H2O2-induced cell damage model with the optimal time and dose. Normal group, model group and high, medium and low-dose DSS groups(2.5%, 5%, 10%) were set up. MTT method was used to detect cell activity, spectrophotometry anti-oxidation indexes of malonaldehyde (MDA), catalase (CAT) and superoxide dismutase (SOD) and glutathione (GSH). Real-time quantitative PCR (Real-time PCR) was used to detect tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β) and interleukin-6 (IL-6) mRNA expressions. And immunofluorescence test was adopted to detect nuclear transcription factor-κB(NF-κB) p65 nuclear translocation of the DSS after the intervention. Result::After 24 h intervention with 250 μmol·L-1 H2O2, SH-SY5Y cell viability was about 55%, which was the best modeling condition. After high, medium and low-dose DSS intervention on H2O2-damaged cell model, compared with the model group, the cell activity showed a dose-dependent increase (P<0.05), MDA was significantly reduced (P<0.05), and antioxidant indexes CAT, SOD and GSH were significantly increased (P<0.05). H2O2 could significantly increase SH-SY5Y cell inflammatory factor TNF-α, IL-6 and IL-1β mRNA expressions, and promote activation of cytoplasmic NF-κB and nuclear translocation. DSS-containing serum showed a dose-dependent inhibition of NF-κB p65 from nuclear, and reduced inflammatory factor levels, such as TNF-α, IL-6 and IL-1β. Conclusion::DSS-containing serum can significantly reduce the oxidative damage in H2O2-induced SH-SY5Y cells by improving their antioxidant status, and reduce the inflammatory response by inhibiting NF-κB signaling pathway.

ABSTRACT

OBJECTIVE: To compare the characteristics of the pepos and seeds of Trichosanthes kirilowii Maxim, of different strains in Changqing district, Jinan, Shandong province, and to discuss the correlation among the characteristics, thus to provide references for germplasm resources evaluation and breeding of the Chinese medicinal materials. METHODS: The external indexes reflecting the characteristics of the pepos and seeds were observed and measured. The data were analyzed by SPSS 17 statistical software. RESULTS: The external indexes of three strains differed significantly. The observed and measured data of the pepo-weight, peel-weight and seed weight of single fruit were the highest for strain II, which indicated that it was an excellent strain. The coefficients of variation of the indexes of different strains were different, with 92.70% between strains and 7.30% within strains. The correlation among the characteristics displayed that when the fruit was larger, the total weight of the pepo, peel and seeds of a single fruit was higher. CONCLUSION: The research provides references for germplasm resources evaluation and breeding of Trichosanthes kirilowii Maxim.

ABSTRACT

<p><b>OBJECTIVES</b>To determine changes of insulin-like growth factor-1 (IGF-1) and IGF binding protein-3 (IGFBP-3) in serum samples from benign prostatic hyperplasia (BPH) patients and to evaluate the value of these molecules as possible etiologic factors for BPH.</p><p><b>METHODS</b>The serum IGF-1 and IGFBP-3 levels were measured with immunoradiometric assay(IRMA) in 64 cases of BPH and in 30 healthy subjects as controls. The cases of BPH were divided into 3 groups according to the prostate volume(PV). There were 18 cases(PV < or = 30 ml) in group A, 24 cases(PV31 approximately 50 ml) in group B, 22 cases(PV > or = 50 ml) in group C.</p><p><b>RESULTS</b>There were no statistical differences between the levels of Both IGF-1 and IGFBP-3 in BPH groups and healthy groups (both P > 0.05), but there were statistical differences among three groups of BPH. Both IGF-1 and IGFBP-3 levels in group C of BPH were significantly higher than those in group A (both P < 0.05). A positive correlation between the serum levels of IGF-1 and PV displayed(r = 0.58), as well as IGFBP-3 (r = 0.48).</p><p><b>CONCLUSIONS</b>Together these observations implicate IGF-1 and IGFBP-3 as important factors during the progression of BPH. It shows the value of non-operation treatment for this disease.</p>
