ABSTRACT

The aim of this study is to identify the express specificity of bone morphogenetic protein 15 (Bmp15) in porcine. The pBMP15-EGFP reporter vector was constructed from the 2.2 kb fragment of porcine bmp15 promoter to trace the differentiation process of stem cells into oocyte-like cells. We used porcine ovary and Chinese Hamster Ovary cell line (CHO), mouse myoblast cell line (C2C12) and porcine amniotic fluid stem cell (pAFSC) to investigate the expression and regulation of this gene via RT-PCR, immunofluorescence, cell transfection, and microinjection methods. We also used single layer cell differentiation to detect the application potential of bmp15. The results show that bmp15 gene was specifically expressed in the porcine ovary and CHO rather than in C2C12 and pAFSC. In addition, the characteristic of tissue-specific of Bmp15 was detected on CHO instead of other cell lines by transient transfection. We also detected the expression of Bmp15 in oocyte at different development stages by immunofluorescence of fixed paraffin-embedded ovary sections. Furthermore, microinjection results show that bmp15 expressed in oocytes at 18 h of maturation in vitro, and continued up to 4-cell stage embryos. Most importantly, we found that the expression of Bmp15 started at day 12 after inducing pAFSC into oocyte-like cells by transfection; green fluorescent was visible in round cell masses. It indicated that bmp15 has the expression specificity and the pBMP15-EGFP reporter vector can be used to trace Bmp15 action in the differentiation of stem cells into germ cells.

ABSTRACT

Objective To search for new therapeutic apparatus and method of acupuncture. Methods 650-660 nm semiconductor laser and 10.6 um CO2 infrared laser were coupled to generate integrated laser acupuncture. Controllable infrared electric heating device with negative ion smokeless moxa was mounted on this integrated laser acupuncture machine to compose the smokeless and pain-free acupuncture therapeutic apparatus. Results The integrated laser acupuncture has both the conventional acupuncture effect and heated effect generated by laser; Negative ion moxa under the control of infrared electric heating device can be electrically distillatod without open fire at the temperature lowering than the burning point and generate both warming effects and chemical effects. The negative ion moxa composed Tourmaline can send off massive negative ion when burning, which can decrease harmful gas and purify environment. Conclusion The new type of acupuncture therapeutic apparatus conquered insufficiencies of conventional acupuncture as pain, breakdown of needle, bleeding, infection, smoking, bad smell, burning, and blistering, and retain and enforce the acupuncture functions of treatment,health-care, and cosmetology.

ABSTRACT

Objectlve To observe the influence of Tourmaline on peripheral neuropathy with diabetes mellitus(DM).Methods Tourmaline bedding series were applied to pafients with DM peripheral neuropathy, observeA the changes of subjective symptoms and nervous function, and thus analyzed the therapeutic effects of Tourmaline on DM peripheral neuropathy. Results AfarTourmalinewas usedfor halfa year, some symptoms of the diseasc were alleviated, and after one year's usage.symptoms of spontaneous pain, numbness.and burning sensation improved greatly.Body examinations showed tendon reflex.vibration sense, nerve conducfion velocity, and autonomic nerve were improved on different levels, and progress of disease was obviously coming down. Conclusion Tourmaline bedding series has the function of improving peripheral neuropathy, whichis aconvenientand safety healthcaremethodto CalTyon.

ABSTRACT

Objective To observe the functions of far-infrared rays and negative ions in Tourmaline and explore its new healthcare usage. Methods Questionnaires were adopted to obtain changes of diabetic patients in terms of dyskinesia, dysreflexia, abnormal skin and muscle, kinesthesis, touch sensation, algesia, and temperature sensation after using Tourmaline bedding series. Results Diabetic patients in fight and moderate level get improvements after using the series for one year, although the patient in fight level get outstanding improvements. Data showed the shorter therapeutic courses the better therapeutic effects were obtained and the improvements highlighted the changes of pain, ferver, hypesthesia, numbness, hight attack symptoms, and motion abnormality. Motion abnormality, algesia, and temperature sensation are three sensitive indexes of this disease. Conclusion Tourmaline bedding series has the function of improving peripheral neuropathy, which may be related with Tourmaline producing far-infrared rays and negative ions.

ABSTRACT

Objective To evaluate the factors affecting the treatment of old persons diagnosed hypertension. Method A sample of 55566 persons from the community of Yanji area of Shanghai was recruited. Results Among the 19295 persons over sixty there are 5842 patients with hypertension, the morbidity was 30.3%. Among the 36271 persons aged fifteen to fifty-nine there are 3536 patients with hypertension, the morbidity is 9.7%. The ratio of non-treatment in the old age group is 3.1% and in younger group is 8.8%. However, the compliance to the treatment was better in the younger than in old group(73.7% vs 56.2%). The age, hypertension grade and sex were all accounted for the differences in the regularity of the hypertension treatment. Conclusion Further effort on the education of non-old hypertensive patients is essential to decrease the damage from hypertension.
