ABSTRACT

Myocardial dysfunction is the most serious complication of sepsis. Sepsis-induced myocardial dysfunction (SMD) is often associated with gastrointestinal dysfunction, but its pathophysiological significance remains unclear. The present study found that patients with SMD had higher plasma gastrin concentrations than those without SMD. In mice, knockdown of the gastrin receptor, cholecystokinin B receptor (Cckbr), aggravated lipopolysaccharide (LPS)-induced cardiac dysfunction and increased inflammation in the heart, whereas the intravenous administration of gastrin ameliorated SMD and cardiac injury. Macrophage infiltration plays a significant role in SMD because depletion of macrophages by the intravenous injection of clodronate liposomes, 48 h prior to LPS administration, alleviated LPS-induced cardiac injury in Cckbr-deficient mice. The intravenous injection of bone marrow macrophages (BMMs) overexpressing Cckbr reduced LPS-induced myocardial dysfunction. Furthermore, gastrin treatment inhibited toll-like receptor 4 (TLR4) expression through the peroxisome proliferator-activated receptor α (PPAR-α) signaling pathway in BMMs. Thus, our findings provide insights into the mechanism of the protective role of gastrin/CCKBR in SMD, which could be used to develop new treatment modalities for SMD.

ABSTRACT

OBJECTIVE: To establish a fluorescence identification method of the microscopic characteristics for Plantagin Semen and its adulterants, and to provide technical support for the market supervision and inspection of TCM decoction pieces. METHODS: Under visible and ultraviolet light, comparative study and identification of the Plantagin Semen (seeds of Platago asiatica L. and Platago depressa Willd.) and its adulterants as seeds of Platago major L., fruits of Schizonepeta tenuifolia Briq., seeds of Codonopsis pilosula (Franch.) Nannf, seeds (peeled) of Kochia scoparia (L.) Schrad., fruits of Bupleurum chinense DC. were carried out by means of stereoscopic fluorescence microscopy from aspects of overall surface characteristics, umbilicus characteristics and section characteristics. RESULTS: Under visible light, the surface texture of Plantagin Semen was wavy stripe or fine wrinkle, while the adulterants were wavy stripe, longitudinal edge or texture was not obvious. The umbilicus of Plantagin Semen was located in the center of the ventral surface, while that of adulterants were located at one end except for P. major. In the section of Plantagin Semen, there were obvious direct embryos, in which the adulterants were small or circular embryos except for P. major. Under ultraviolet light, P. asiatica had obvious wavy stripes in surface, orange and light blue-green fluorescence; P. depressa had grid-shaped wrinkles and gray-blue and gray-brown fluorescence; the umbilical fluorescence of Plantagin Semen was strong, and the fluorescence of the adulterants was weak except for S. tenuifolia. There were obvious differences in fluorescence color, embryo size and distribution between the section of Plantagin Semen and adulterants. CONCLUSIONS: The stereoscopic fluorescence microscopy is effect and accurate for the identification of Plantagin Semen.

ABSTRACT

Objective@#To observe the therapeutic effect of dihydroartemisinin on pulmonary fibrosis in rats exposed to dust, and compare the therapeutic effects of dihydroartemisinin and tetrandrine.@*Methods@#Sixty male Sprague-Dawley rats were randomly divided into control group, model group, treatment group 1 and treatment group 2, with 15 rats in each group. The model group and the treatment group were stained with disposable non-exposure silica tracheal instillation method. The drug was administered on the second day after the dust was applied. The treatment group was given with dihydroartemisinin 75 mg/kg, the treatment group was given tetrahexine 22 mg/kg, model group and control group were intragastrically administered with 1 ml of normal saline per 100 g of body mass. The drug was administered for 6 days per week for 28 days. Rats were sacrificed on the 7th, 14th and 28th day after dusting, and the lung tissues of rats were taken, detection of rat lung coefficient, ELISA was used to detect transforming growth factor-1(TGF-1)and Smad 2/3 in rat lung tissues, type I collagen (Col-I) expression level, pathological changes of rat lung tissue, immunohistochemical observation of rat lung tissue TGF-1 and Col-I protein expression. Statistial analysis was proformed with SPSS 19.0. The mean cornparis between graups wad perfomed using a completely randonized one-way (ANOVA).@*Results@#The expressions of TGF-1, Smad 2/3 and Col-I in the lung tissue of the treatment group were significantly lower than those in the model group (P< 0.05), but the difference between the treatment group 1 and the treatment group 2 Not statistically significant (P>0.05). The results of immunohistochemistry showed that the TGF-1 and Col-I proteins in the interstitial of the model group showed strong positive expression. The expression of TGF-1 and Col-I protein in the lung interstitial of the treated group was weaker than that of the model group.@*Conclusion@#Dihydroartemisinin can down-regulate the level of inflammatory cytokines in rat lung tissue. It is possible to inhibit pulmonary fibrosis by intervening in the TGF-β1/Smad 2/3 signaling pathway, but the therapeutic effect between DHA and tetrandrine is not significantly different in a short time.

ABSTRACT

Objective To investigate the effect of resveratrol on high glucose-induced oxidative stress and cardiomyo-cyte hypertrophy and possible mechanism .Methods H9C2 cardiomyocytes were divided into normal glucose group , high glucose group , resveratrol group and resveratrol +DAPT group respectively .The cell surface was measured by rhodamine-labeled phalloidin .Reactive oxygen species generation was measured using DCFH-DA.The contents of SOD and MDA were evaluated by colorimetry .The mRNA and protein levels of Notch 1, Hes1, ANP and BNP were evaluated by RT-qPCR and western blot .Results High glucose exposure significantly increased cell surface area ( P<0.01) and enhanced ROS and MDA generation ( P <0.01 ) with concomitant decrease of SOD activity ( P <0.01).It also decreased the mRNA and protein expressions of Notch 1 and Hes1(P<0.05), and increased the ex-pression levels of ANP and BNP ( P<0.05 ) .Resveratrol treatment inhibited cell surface expanding ( P<0.01 ) and decreased ROS and MDA generation (P<0.01) with concomitant enhance SOD activity (P<0.01), as well as in-creased the expression level of Notch 1 and Hes1 ( P<0.05 ) , and decreased the expression of ANP and BNP ( P<0.05).Conclusions Resveratrol may inhibit high glucose induced H9C2 cardiomyocytes hypertrophy by improving oxidative stress and activating Notch 1/Hes1 signaling pathway .

ABSTRACT

AIM:To investigate the expression of aryl hydrocarbon receptor (AhR) in atrial tissues of the patients with rheumatic heart disease (RHD), and the effects of AhR on rheumatic atrial fibrosis.METHODS:Right atrial specimens obtained from the patients with RHD requiring valve replacement surgery were divided into chronic atrial fibrillation (RHD+cAF, n=11) group and sinus rhythm (RHD+sinus rhythm, n=25) group.The patients with congenital heart disease (CHD) and sinus rhythm (CHD+sinus rhythm, n=12) who underwent heart surgery served as controls.The collagen volume fraction in the atrial specimens was examined by Masson`s trichrome staining.The protein expression and distribution of AhR, AhR nuclear translocator (ARNT) and CYP1A1 were detected by the methods of immunohistochemistry and Western blot.The mRNA expression of AhR, ARNT and CYP1A1 was detected by real-time fluorescence quantitative PCR.RESULTS:Compared with CHD+sinus rhythm group, the collagen content and the expression of AhR, ARNT and CYP1A1 were significantly increased in RHD+sinus rhythm group and RHD+cAF group.Compared with RHD+sinus rhythm group, the collagen content and the expression of AhR, ARNT and CYP1A1 were significantly increased in RHD+cAF group (P<0.05).CONCLUSION:The expression of AhR is correlated with the degree of fibrosis.The expression of AhR/ARNT/CYP1A1 is increased in atrial tissues of patients with RHD, suggesting that AhR/ARNT/CYP1A1 should be involved in atrial fibrosis of the patient with RHD.

ABSTRACT

AIM:To investigate the changes of aryl hydrocarbon receptor (AhR) in the process of cardiomyo-cyte hypertrophy induced by high glucose , and to explore its potential mechanisms .METHODS: The rat cardiomyocytes (H9c2 cells) were divided into normal glucose group , high glucose group, DMSO group and resveratrol (an AhR antago-nist) group.The content and distribution of AhR were observed with immunofluorescence staining .The myocardial cells were stained with rhodamine-labeled phalloidin to visualize cytoskeleton , and the cell surface area were determined after im-aging by fluorescence microscopy .The generation of reactive oxygen species ( ROS) in the cardiomyocytes was measured u-sing a fluorescent probe DCFH-DA.The mRNA expression of AhR , CYP1A1, atrial natriuretic peptide ( ANP) and brain natriuretic peptide ( BNP) were evaluated by real-time quantitative PCR ( RT-qPCR).The protein levels of AhR, CYP1A1, ANP and BNP were assessed by Western blot .RESULTS:AhR was constitutively presented in the cytosol un-der normal-glucose condition and was translocated to the nuclei under high-glucose condition .High glucose induced cardiac hypertrophy , and increased ROS generation .Significant reductions in the cell size and ROS generation were observed after treated with resveratrol.The expression of AhR, CYP1A1, ANP and BNP at mRNA and protein levels in high glucose group was increased as compared with normal glucose group and resveratrol group , and the above-mentioned indexes signifi-cantly decreased in resveratrol group as compared with DMSO group .CONCLUSION: High glucose-induced cardiac hy-pertrophy increases AhR expression , which may be involved in the maintenance of glucose homeostasis in the cardiomyo-cytes.AhR translocation to the nucleus induced by high glucose results in the increases in CYP 1A1 expression and ROS generation, which may be an important mechanism of high glucose-induced cardiomyocyte hypertrophy .

ABSTRACT

Objective To evaluate the different effects of laryngeal mask and mask ventilation during bronchoscopy in elder patients. Methods 120 old patients (ASA I-III) were divided into three group according to the table of random number as following:surface anesthesia with autonomous respiration (group I);endoscope mask ventilation (group II);laryngeal mask airway ventilation (group III). Oxygen inhalation through nasal tube in groupⅠ,group II and group III was ventilated with endoscope mask and LMA respectively ,thenwe observed whether there were adverse effects or not during the procedure. Results The adverse reactions of group I were more than other groups. Compared with the group I, variance of MAP,HR, RR, SpO2 in the groupⅡand groupⅢ had less changes, the cases with severe bucking decreased significantly, and satisfaction degree increased markedly, (P <0.01or P<0.05 ) . The incidence rate of resp iratory depression and airway obstruction in groupⅡwas higher than that in groupⅢ (P<0.05). Conclusion Endoscope mask ventilation and LMA can both be adopted in analgesia bronchoscopy for old patient. LMA under general anesthesia to transbronchial lung biopsy would control respiration according to the demand at any time.LMA has more advantage in the operation for the stable respiration and hemodynamics and less complications.
