ABSTRACT

Objective:To investigate the relationship between SLC2A2 rs5393, rs5400 loci gene polymorphism and metformin efficacy in patients with type 2 diabetes mellitus (T2DM) .Methods:From Jan. 2016 to Apr. 2019, 90 T2DM patients and 90 healthy persons were selected as the subjects. The frequencies of SLC2A2 rs5393 and rs5400 loci gene polymorphisms were detected by PCR restriction fragment length polymorphism (PCR-RELP) method. Patients with T2DM were treated with metformin and followed up for 90 days. The relationship between metformin efficacy and genotypes was analyzed.Results:The genotypes of SLC2A2 rs5393 and rs5400 in the control group and T2DM group were in accordance with Hardy Weinberg equilibrium law. Compared with the control group, the ratios of AC type and CC type at rs5393 locus in T2DM group was significantly higher ( P<0.05) , and the frequencies of CT type and TT type at rs5400 locus were significantly higher ( P<0.05) . There was no significant difference in the levels of FPG, 2 h PBG and HbA1c of AA type, AC type and CC type at rs5393 locus before treatment ( P>0.05) , after metformin treatment, FPG, 2hPBG and HbA1c of patients with AC type and CC type were lower than those with AA type ( P<0.05) , and the decreases of FPG and HbA1c in CC type were lower than those in AC type ( P<0.05) . There was no significant difference in the levels of FPG, 2hPBG and HbA1c of CC type, CT type and TT type at rs5400 locus before treatment ( P>0.05) . After metformin treatment, FPG, 2 h PBG and HbA1c of patients with CT type and TT type were lower than those with CC type ( P<0.05) , and the decreases of FPG and HbA1c in TT type were lower than those in CT type ( P<0.05) . Conclusion:T2DM patients with AA type at rs5393 locus and CC type at rs5400 locus of SLC2A2 gene polymorphism loci are sensitive to metformin and has good hypoglycemic effect.

ABSTRACT

Stromal preadipocytes from human omental adipose tissues were isolated with enzyme-digesting method,and cultured for inducing differentiation.Preadipocyte factor-1 was abundant during preadipoeyte stage,whereas lipoprotein lipase and PPARγ2 mRNA were rapidly up-regulated at early stage of differentiation and CCAAT/enhancer binding protein expression was increased rather late.In addition,resistin expression and secretion was primarily detectable in preadipocytes,leptin was gradually increased during adipogenesis,whereas adiponectin appeared specifically in mature adipocytes.Insulin-like growth factor-Ⅰ secretion was remarkably detected in both preadipocytes and mature adipocytes.

ABSTRACT

Objective To determine the content of indirubin in Kangbingan Oral Liquid by UV spectrophotometer methods.Methods The content of indirubin was determined by Heltos Gamma ultravioletvisible spectrophotometer with chloroformic solution of 2~8 ?g/mL confected by standard sample of indigotin and indirubin.The sample of negative blank solution was confected by the prescription without Radix Isatidis.The standard sample and the blank sample were determined separately with the wavelength at 500~700 nm.Results Indirubin had the maximum absorption at wavelength of 540.0 nm and indigotin had the maximum absorption at wavelength of 634.5 nm.The linear correlation was available at the range of 0.20~1.40 ?g/mL,and the correct coefficient was 0.999 4.The average recovery rate was 99.05% and RSD was 0.87%(n=6).Conclusion This method is simple and accurate,and can be used to control the quality of Kangbingan Oral Liquid.
