ABSTRACT

ObjectiveTo observe the influence of basic fibroblast growth factor (bFGF),vascular endothelial growth factor (VEGF),epidermal growth factor (EGF),insulin and self-corium granule on the fat granule under cultivating and after transplantation.MethodsThe adipocytes of mouse were treated with bFGF,VEGF,EGF,insulin and self-corium granule in different concentration gradient post inoculation,respectively,to determine their different cytoactivity with MTT method,and then draw their activity curve with time.The various grafts were implanted into 32 SD rats,which contained bFGF,VEGF,EGF,insulin,self-corium granule or physiologic saline,and then the general condition of animals,quality determination,pathological changes and immunohistochemical staining were observed.At last,the effects of the different implants on the fat granule were compared in different methods.ResultsThe best cytoactive saturation of every experiental group was the same at different time points,except self-corium granule group,the cytoactivity of which was higher than that of control group (P<0.05).In animal experiment,focal necrosis and fat cistern were seen in comparatively larger area in the control group.The adipocyte of the experiental groups differed in size,and considerable small adipocytes were present.The capillary vessel density of all the experiental groups except the insuline group were higher than those of the control group (P<0.05).But the difference between the experiental groups and the control group became illegible gradually with time.ConclusionsThe activity of adipocyte being cultivated is differently increased by bFGF,VEGF,EGF and Insulin.The quantity of fat graft could be significantly raised by bFGF,VEGF,EGF and self-corium granule,which improve the revascularization of the fat graft after transplantation.

ABSTRACT

Objective To investigate a medicine which can inhibit the expression of connective tissue growth factor (CTGF) in hypertrophic scar of rabbit ears. Methods 24 bigears white rabbits were used to establish a model of hypertrophic scar of rabbit ears, which was randomly divided into four groups. The hypertrophic scar was injected intralesionally with antisense CTGF (group A), betamethason (group B), triamcinolone acetonide (group C) and physiological saline (group D). Some scar tissue samples were sectioned in every group when the scar was treated after 7, 14, 30, and 60 days, respectively. The expression of CTGF mRNA in the scar was assessed by in situ hybridization and hematoxylin and eosin stain (HE stain) in every samples. Results The expression of CTGF mRNA and the counting of fibroblasts decreased in group A, which showed statistical difference as compared with groups B, C and D. Conclusions The results suggesz that antisense CTGF is able to inhibit the proliferation process of hypertrophic scar in rabbit ears and to remarkably decrease the degree of fiborsis in the scar.

ABSTRACT

Objective To seek for a new method in the treatment of devided nevus of the eyelid. Methods On the outside (or the inside) of the lesion, using the level of canthi externa (or inner) as the center, a hatchet skin flap was designed on the upper and lower eyelids, respectively, which had the same pedicle and proportion each other. "The cutting edge" of the skin flap nearby the side of diseased region should be extended to the 1/2 transverse diameter of the lesion so as to make it to be a figure of hatchet. The skin flap should be anatomized from the fat level, which revolved the defect region to cover the wound surface. Results This method was performed in 9 cases of devided nevus of eyelid. The size of their defected areas (eyes being closed) ranged from 1.0 cm ? 1.0 cm to 2.5 cm ? 2.0 cm. All of the skin flaps were completely survival and achieved aesthetic contouring results. Conclusions The method of hatchet skin flap is safe and simple, with satisfactory effect, similar external appearance in color and texture compared with adjacent normal skin. Both the patients and their families are satisfied with the results. Therefore, it is an efficient approach in treating devided nevus of eyelid.
