ABSTRACT

Objective:This study aimed to investigate the change of the position of the tongue before and after combined treatment of maxillary expansion and orofacial myofunctional therapy in children with mouth-breathing and skeletal class Ⅱmalocclusion. Methods:A total of 30 children with skeletal class Ⅱ malocclusion and unobstructed upper airway were selected. The 30 children were divided into mouth-breathing group(n=15) and nasal-breathing group(n=15) and CBCT was taken. The images were measured by Invivo5 software. The measurement results of the tongue position of the two groups were analyzed by independent samples t-test. 15 mouth-breathing children with skeletal class Ⅱ malocclusion were selected for maxillary expansion and orofacial myofunctional therapy. CBCT was taken before and after treatment, the measurements were analyzed by paired sample t test with SPSS 27.0 software package. Results:The measurement of the tongue position of the mouth-breathing and nasal-breathing groups were compared, the differences were statistically significant(P<0.05). The measurement of the tongue position showed significant difference after the combined treatment of maxillary expansion and orofacial myofunctional therapy in children with mouth-breathing and skeletal class Ⅱmalocclusion(P<0.05). Conclusion:Skeletal class Ⅱ malocclusion children with mouth-breathing have low tongue posture. The combined treatment of maxillary expansion and orofacial myofunctional therapy can change the position of the tongue.

ABSTRACT

The pediatric total facial management refers to a series of diagnosis and treatment processes to achieve the healthy development of the face through reasonable medical intervention. The main reason for the poor treatment effect is that the first contact doctor is limited to his own disciplinary analysis and treatment. The importance of multidisciplinary cooperation in the diagnosis and treatment of facial dysplasia in children has become increasingly prominent. it is necessary to comprehensively analyze and find the pathogenic factors of patients and formulate a comprehensive treatment plan to restore normal upper airway structure and nasal breathing, and then reshape the healthy craniomaxillofacial tissue structure, and the monitoring of the results of medical intervention should accompany the whole process of children's growth and development. This paper summarizes the current situation of the treatment of children with facial dysplasia and puts forward the concept of orderly individualized multi-disciplinary diagnosis and treatment of pediatric oral maxillofacial management.

ABSTRACT

Objective To investigate the epidemiological characteristics of major kidney disease deaths and the potential years of life lost among residents in Wuhan from 2014 to 2019, and to provide a scientific basis for the prevention and treatment of kidney diseases. Methods The major kidney diseases deaths among residents in Wuhan during 2014-2019 were collected from the population-based Mortality Surveillance System. The standardized mortality rate and potential years of life lost rate (PYLLR) of major kidney diseases among residents in different ages and genders were calculated, and the epidemiological characteristics and trends were analyzed. Results There were 4 100 deaths (2 380 in male and 1 720 in female) from major kidney diseases among residents in Wuhan between 2014 to 2019, with an age-standardized mortality rate of 6.22/100 000. The mortality rate of major kidney diseases showed an upward trend with the increasing age groups. The age-standardized mortality rate and the age-standardized potential years of life lost rate (SPYLLR) in glomerular disease and tubulo-interstitial diseases were significantly decreased (P<0.05). The age-standardized mortality rate of the kidney failure was significantly increased (P<0.05), especially in the male (APC=25.10% , P<0.05). Conclusion From 2014 to 2019, there was no significant change in the overall mortality rate of major kidney diseases among residents in Wuhan. The death burden and disease burden of glomerular diseases and tubulo-interstitial diseases were significantly decreased, while the mortality rate of male kidney failure was significantly increased, indicating the need for targeted prevention and treatment of kidney diseases.

ABSTRACT

Objective To explore the impact of temperature and environmental factors on the risk of deaths from cardiovascular and cerebrovascular diseases (CVD) in Wuhan during the course of high temperature. Methods The daily CVD death data and meteorological and atmospheric pollutant concentration data during the high temperature process in Wuhan from 2014 to 2019 were collected. The generalized additive models (GAM) were used to obtain the characteristics of meteorological factors, atmospheric pollutant concentrations, high impact factors and thresholds affecting the relative risk of CVD death. Results The analysis results showed that: (1) When the maximum temperature reached 36.7°C and the lowest temperature reached 25.3°C, the relative risk of CVD death increased significantly; (2) The risk of CVD death during the first high temperature process was the largest, and the average CVD excess mortality rate during the first high temperature process from 2014 to 2019 reached 21.7%; and (3) The average temperature, maximum temperature and PM10 during the course of high temperature were important environmental factors that increased the risk of CVD deaths, and the relative risks were 1.14 (95% confidence interval (95% CI): 1.11-1.17), 1.11 (95% confidence interval (95% CI): 1.08-1.15, and 1.06 (95% confidence interval (95% CI): 1.02-1.09), respectively. Conclusion Temperature and PM10 are important environmental factors that increase the risk of death from CVD during the high temperature process in Wuhan from 2014 to 2019, with the first annual high temperature process having the greatest impact on the risk of CVD death.

ABSTRACT

ObjectiveTo determine the association between air pollutants (PM2.5, PM10, SO2, NO2) and death from respiratory diseases in Wuhan. MethodsDaily air pollutants, meteorological data and mortality from respiratory disease between 2014 and 2019 were collected for a descriptive analysis. A time series semi-parametric generalized additive model (GAM) was used to determine the exposure-effect relationship between atmospheric pollutants and daily mortality from respiratory diseases,and the excess risk (ER) was used to quantify the effects of air pollutants on death from respiratory diseases. ResultsThere was significant effect of PM2.5, PM10, SO2 and NO2 on respiratory diseases mortality. In the period with strongest effect, the ER of death from respiratory diseases were 2.803%(95%CI:2.151%‒3.460%), 1.878%(95%CI:1.477%‒2.281%), 10.210%(95%CI:7.922%‒12.549%), 4.564%(95%CI:3.530%‒5.608%), along with an incremental 10 μg·m-3 of PM2.5,PM10,SO2 and NO2, respectively. Furthermore, females were more sensitive to PM2.5, SO2 and NO2, while males were more sensitive to PM10. Residents aged less than 65 years were more sensitive to PM2.5 and NO2, and those older than 65 years were more sensitive to PM10 and SO2. ConclusionAir pollutants (PM2.5, PM10, SO2, and NO2) in Wuhan are associated with the death from respiratory diseases. Therefore, at-risk groups should be considered for formulating local policies against air pollution.

ABSTRACT

Schistosomiasis is one of the major communicable diseases, which seriously impacts human health, Oncomelania hupensis snail is unique intermediate host of Schistosoma japonicum as well as posts significant influence on schistosomiasis transmission. The long-term serial data of oncomelania snail area in the Middle and Lower Reaches of the Yangtze River in the Schistosomiasis situation in People's Republic of China ,were collected from 1999 to 2018. The spatial and temporal variation characteristics of oncomelania hupensis area in five provinces were analyzed by Manner-Kendall test. In the spatial change, the area composition ratio of lake-and-marshland regions in Hunan increased, while that in Hubei and Jiangxi showed a slight decline. The lake-and-marshland snails in Anhui showed a significant downward trend, while Jiangsu showed no stable trend. In the spatial change, the area of lake-and-marshland snails in Jiangsu, Anhui, and Hubei showed a downward trend, among which the downward trend in Hubei was not obvious, while the upward trend in Jiangxi and Hunan had an upward trend but the trend was not obvious. the year of the sudden change in the area of the lake and marshes in in Jiangsu and Hubei were 2010 and 2017 ; There was no significant mutation in Anhui Province; the mutation point of the lake-and-marshland snails in Hunan was between 2001 and 2002; the mutation year of the lake-and-marshland snails in Jiangxi was 2002.This paper studies the temporal and spatial changes of the snail breeding area in the middle and lower Yangtze River, and the results can provide a basis for the elimination of snails in different types of distribution areas.

ABSTRACT

Objective: To observe the effects of acupoint thread-embedding therapy on serum apelin and glucagon-like peptide-1 (GLP-1) in type 2 diabetes mellitus patients with obesity due to dampness-heat encumbering spleen.Methods: Sixty-six patients were randomly divided into a control group and an observation group according to the random number table method, with 33 cases in each group. Patients in the control group were treated with exenatide and metformin, while patients in the observation group were treated with additional acupoint thread-embedding. After 12-week treatment, the obesity-related indicators, including body mass index (BMI), waist circumference and body fat rate, the glycometabolism indicators, including fasting blood glucose, 2 h postprandial blood glucose and glycosylated hemoglobin, and the lipid metabolism indicators, including total cholesterol (TC), triglyceride (TG) and low-density lipoprotein cholesterol (LDL-C), as well as serum apelin and GLP-1 levels were observed in patients of the two groups. Results: After treatment, the BMI, waist circumference and body fat rate of patients in the two groups were all reduced (all P<0.05), and were lower in the observation group than in the control group (all P<0.05); the fasting blood glucose, 2 h postprandial blood glucose and glycosylated hemoglobin levels of patients in both groups were all decreased (all P<0.05), and were significantly lower in the observation group than in the control group (all P<0.05); the TC level was decreased (P<0.05), while the TG and LDL-C levels did not change significantly in the control group (both P>0.05); the TC, TG and LDL-C levels were all significantly reduced in the observation group (all P<0.05), lower than those in the control group (all P<0.05); the serum apelin level was decreased (P<0.05) and the serum GLP-1 level was increased (P<0.05) in the observation group, statistically different from those in the control group (both P<0.05). Conclusion: Combined with the conventional medication, acupoint thread-embedding therapy can significantly improve the obesity-related indicators, glycometabolism and lipid metabolism in type 2 diabetes mellitus patients with obesity due to dampness-heat encumbering spleen. This may be achieved by regulating the serum apelin and GLP-1 levels.

The use of non-invasive blood glucose detection techniques can help diabetic patients to alleviate the pain of intrusive detection, reduce the cost of detection, and achieve real-time monitoring and effective control of blood glucose. Given the existing limitations of the minimally invasive or invasive blood glucose detection methods, such as low detection accuracy, high cost and complex operation, and the laser source's wavelength and cost, this paper, based on the non-invasive blood glucose detector developed by the research group, designs a non-invasive blood glucose detection method. It is founded on dual-wavelength near-infrared light diffuse reflection by using the 1 550 nm near-infrared light as measuring light to collect blood glucose information and the 1 310 nm near-infrared light as reference light to remove the effects of water molecules in the blood. ABSTRACT

OBJECTIVE@#To observe the effect of acupoint application therapy at different timing points on the gastrointestinal function recovery and heart rate variability (HRV) after laparoscopic resection of colorectal cancer under the instruction of enhanced recovery after surgery (ERAS).@*METHODS@#A total of 105 patients for the selective laparoscopic resection of colorectal cancer were selected and randomized into a preoperative acupoint application group (35 cases, 3 cases dropped off), a postoperative acupoint application group (35 cases, 1 case dropped out) and a control group (35 cases, 2 cases dropped off). In the control group, ERAS interventions were provided, such as health education, fluid supplementation and multi-mode analgesia. On the base of the treatment as the control group, in the preoperative acupoint application group and the postoperative acupoint application group, 3 days before operation and 6 h after operation, the acupoint application therapy was given respectively. The acupoints were Zusanli (ST 36), Shangjuxu (ST 37), Sanyinjiao (SP 6), Neiguan (PC 6) and Xiajuxu (ST 39). The acupoint application was exerted for 6 h each time, once daily till the first postoperative exhaust and defecation presented. It was to observe the time of the first postoperative exhaust, defecation and food intake, the score of visual analogue scale (VAS) 1 to 3 days after operation, the total score of gastrointestinal symptom rating scale (GSRS) before and 1 week after operation, as well as the related indicators of HRV [standard deviation of NN intervals (SDNN) and the ratio of low-frequency power and high frequency power (LF/HF)] in the three groups successively. Besides, the adverse reactions were recorded during intervention in the three groups.@*RESULTS@#Compared with the control group, the time of the first postoperative exhaust and the time of the first postoperative defecation were all earlier in the preoperative acupoint application group and the postoperative acupoint application group respectively (<0.05), and VAS scores 1 to 3 days after operation and total GSRS scores 1 week after operation were all reduced (<0.05); the time of first food intake was earlier after operation (<0.05), and SDNN and LF/LF were increased 1 day and 3 days after operation in the preoperative acupoint application group (<0.05). Compared with the postoperative acupoint application group, in the preoperative acupoint application group, the time of the first postoperative exhaust and the time of the first postoperative defecation were all earlier (<0.05), VAS scores were reduced in 1 to 3 days after operation (<0.05), and SDNN 1 day and 3 days after operation and LF/HF 1 day after operation were all increased (<0.05). No adverse reaction was detected in patients of the three groups.@*CONCLUSION@#Under the instruction of ERAS, the preoperative acupoint application effectively promotes the postoperative gastrointestinal function recovery, improves HRV and autonomous nerve function in the patients after laparoscopic resection of colorectal cancer. The therapeutic effect of this therapy is better than the postoperative acupoint application.

ABSTRACT

Objective To identify specific peptides binding to hepatitis B virus ( HBV) DNA pol-ymerase TP region ( HBP-TP) and to study their effects on HBV replication. -ethods The phage polypep-tide display library was screened using the HBP-TP recombinant protein expressed in Escherichia coli as a substrate to obtain the specific phage, the polypeptide region of which was then sequenced. Polypeptide se-quences with a repetition rate greater than 10% were counted and commercially synthesized. Changes in HBV replication were detected after treatment of HBV-expressing cells with synthetic peptides as drugs. Re-sults Eight peptides targeting HBP-TP recombinant protein were screened out from the phage display librar-y. One of the peptides was found to have a significant inhibitory effect on HBV replication at cellular level. Conclusions A HBP-TP protein-targeting polypeptide with an inhibitory effect on HBV replication was ob-tained.

AIM:To investigate influence of demethylation/acetylation by 5-Aza-2'-deoxycytidine/trichostatin A(5-Aza/TSA)treatment on B-cell specific phenotype of non-Hodgkin lymphoma cells.METHODS:CD19 promoter-driven reporter with NEO cassette was constructed to realize transfection and stable selection of Hodgkin and non -Hodgkin lymphoma cells.The exogenous CD19 promoter activity in both cell line clusters with and without 5-Aza/TSA treatment was detected and compared.The B-cell specific expression profiling in Eμ-myc transgenic mouse model developed lymphoma was isolated and identified.The effects of 5-Aza/TSA treatment on B-cell specific phenotype were analyzed.RESULTS:Epigenetic modification via 5-Aza/TSA repressed B-cell specific phenotype in B-cell-derived non-Hodgkin lymphoma cells. CONCLUSION:Epigenetic modification of pivotal master repressor genes plays an essential role in B -cell phenotype of both human and murine developed B-cell non-Hodgkin lymphoma cells.

ABSTRACT

Objective To analyze the risk factors and pathogen distribution of ventilator-associated pneumonia (VAP) in patients following coronary artery bypass grafting (CABG).Methods The clinical data of 1414 patients undergoing CABG in Teda International Cardiovascular Hospital from June 2014 to June 2016 were retrospectively analyzed .The VAP developed following CABG in 42 patients ( VAP group ) and not occurred in 1372 patients ( non-VAP group) .Multivariate logistic regression was used to analyze the risk factors of VAP in patients after CABG , and the microbial culture and drug sensitivity test were performed in VAP patients.Results The incidence rate of VAP after CABG was 2.97% (42/1414).Multivariate logistic regression analysis showed that history of smoking ( OR =2.216, 95% CI 1.018 -4.825, P <0.05), mechanical ventilation time >48 h (OR=7.457, 95% CI 3.443-16.161, P<0.01), LVEF<40%(OR=3.524 , 95% CI 1.203-10.325, P<0.05) and postoperative acute kidney injury (OR=16.239, 95% CI 7.551 -34.924, P <0.01) were independent risk factors for VAP in patients after CABG.A total of 42 pathogen strains were detected in 42 patients with VAP, including 37 strains of Gram-negative bacteria, 2 strains of Gram-positive bacteria, and 3 strains of fungus.Gram-negative bacteria mainly were Klebsiella pneumoniae subspecies ( n =23, 54.76%) and Burkholderia cepacia ( n =6, 14.27%);the Gram-positive bacteria were Staphylococcus aureus ( n =2, 4.76%);the fungus was Candida albicans ( n =3, 7.14%).Klebsiella pneumoniae was sensitive to many antibiotics;and the resistance rate to amikacin , aztreonam , meropenem , and levofloxacin was <10%, the resistance rate to ceftazidime and piperacillin was <25%. Burkholderia cepacia was naturally resistant to amikacin , ampicillin, aztreonam, cefazolin, gentamicin and sulfamethoxazole .Conclusion The incidence of VAP was higher in patients after CABG , and the involved pathogens were mainly Gram-negative bacteria .Clinically , it is necessary to take necessary measures to prevent and treat VAP in order to improve the prognosis of patients undergoing CABG .

ABSTRACT

Aim To investigate the effect of cotreat-ment norcantharidin (NCTD) and cisplatin (DDP) on the cisplatin sensitivity and proliferation in A549 cells, and whether YAP molecule involved in this process. Methods A549 cells were used as a model for inves-tigating the function of NCTD and DDP in this study. The expression of caspase-3,Annexin V,YAP,CTGF and Cyr61 were detected using RT-PCR and Western blot assay. The cell growth and proliferation were as-sessed by MTT and CCK-8 assay. Moreover, the tran-scriptional activity of YAP was detected by luciferase reporter gene assays. Results YAP was required for the cell growth and proliferation. NCTD,DDP and co-treatment of NCTD and DDP depressed cell viability, inhibited cell proliferation and promoted the sensitivity of cisplatin in A549 cells. Our results showed that the higher expressed oncogene YAP activated and promoted cell proliferation in lung cancer cells. Moreover, the high concentration of NCTD or DDP significantly re-duced cell proliferation, but cotreatment of NCTD and DDP in low concentration could significantly increase the cisplatin sensitivity via YAP pathway in A549 cells. Conclusions The cotreatment of NCTD and DDP in low concentration significantly reduces the transcriptional activity and protein level of YAP, then inhibits cell proliferation and thus increases the sensi-tivity of DDP in A549 cells.

ABSTRACT

<p><b>OBJECTIVE</b>To isolate, culture, and identify bone marrow mesenchymal stem cells (BMSCs) from enhanced green fluorescent protein (EGFP)-transgenic rats in vitro.</p><p><b>METHODS</b>Bone marrows were isolated from tibia and femur of healthy EGFP-transgenic rats of specific pathogen free (SPF) grade. Then,the whole bone marrow adherent method was used for isolation,culture,and purification of BMSCs. The morphological change was noted by continuous observation under inverted fluorescence microscope. The growth curve of cells was drawn through the method of CCK-8 and the proliferation compared with wild type BMSCs. The surface markers of BMSCs were detected by flow cytometry. The BMSCs were induced to differentiate into osteoblasts, adipocytes, and chondrocytes lineages. The EGFP-BMSCs were transplanted into the rats intravenously, and the expression of GFP was detected.</p><p><b>RESULTS</b>BMSCs stably expressing EGFP gene were obtained successfully, with the fusiform-shaped appearance and the forming of circinate cell colonies. The growth curve of EGFP-MSCs showed the characteristic of active proliferation, showing no significant difference compared with the wild-type BMSCs. The expression rates of the surface markers of BMSCs CD29, CD90, CD34, CD49d, and CD45 were 99.4%, 96.4%, 0.171%, 0.049%, and 0.038%. The GFP were detected in lung 3 days after transplantation. After osteogenic, adipogenic, and chondrogenic induction, oil red-O and alizarin red positive signals and toluidine blue positive cells were detected.</p><p><b>CONCLUSIONS</b>High-purity BMSCs stably expressing green fluorescent protein gene can be cultured using the whole bone marrow adherent method. EGFP does not affect the stem cell properties and expresses stably after transplantation. The cells can be used as seed cells for subsequent research.</p>

ABSTRACT

Objective To reveal the self-management status of patients with hypertension in community, and to analyze the influencing factors of self-management behavior so as to provide theoretical basis for the formation and maintenance of self-management. Methods With the method of stratified random cluster sampling, 559 patients with hypertension were selected from twelve communities in six urban districts of Wuhan completed the questionnaire survey to analyze the demographic characteristics, executive condition of self-management behavior and related psychological behavior with hypertension patients. Results The average age was (61.2 ± 8.4) years. The rate of measuring blood pressure regularly was 38.39%, rate of regular medication was 63.83%, rate of low salt diet was 37.72%, and the rate of regular exercise was 50.45%. The score of self-management was 0.60 ± 0.21. Self efficacy was the influencing factor of self-management score (β=0.042 1,P<0.01). Conclusion Self assessed efficacy is closely related to self-management. Intervention should be guided by psychological behavior theory, and emphasized on psychological aspect.

Results Before implementation health defects rate was 43.3%(303/700), after the implementation health education defect rate was 11.4%(223/1 950). The health defects rate before and after the implementation showed significant difference (χ2=8.387, P=0.004). The incidence of anxiety before and after the implementation was 11.8%(23/195) and 4.1%(8/195), the difference was significantly different (χ2=7.885, P =0.005). Conclusions To carry out the quality circle activity, can effectively reduce the perioperative health education defect rate and improve the quality of care and patient satisfaction, method is simple and easy to operate, it is worth clinical promotion.

ABSTRACT

Background:The incidence of inflammatory bowel disease(IBD)is increasing recently. However,the pathogenesis has not been fully clarified. Aims:To investigate the expressions and significance of interleukin-22( IL-22),matrix metalloproteinase-9(MMP-9)and macrophage migration inhibitory factor(MIF)in peripheral blood of patients with IBD. Methods:A total of 80 patients with IBD admitted from May 2011 to Nov. 2014 at the First Affiliated Hospital of Soochow University were enrolled,in which 43 cases were Crohn’s disease(CD),37 cases were ulcerative colitis(UC). Forty healthy subjects were served as normal controls. Peripheral levels of IL-22,MMP-9 and MIF were detected by ELISA. Multivariate Logistic regression model was used to analyze IL-22,MMP-9 and MIF in active CD and UC and ROC curve was used to evaluate the diagnostic performance of these markers for screening of active CD and UC. Results:Compared with normal control group,peripheral levels of IL-22,MMP-9 and MIF increased significantly in CD and UC groups(P 0. 05). Peripheral levels of IL-22, MMP-9 and MIF in active CD and UC were significantly higher than those in remission stage(P < 0. 05). For screening of active IBD,the area under ROC curve(AUC)of combined detection of IL-22 and MMP-9(0. 853 for CD,0. 867 for UC) was superior to that of IL-22,MMP-9 or MIF only(0. 747,0. 770 and 0. 699 for CD,0. 774,0. 815 and 0. 761 for UC). Conclusions:Peripheral levels of IL-22,MMP-9 and MIF increase markedly in IBD patients,which are correlated closely with the activity of IBD. Combined detection of IL-22 and MMP-9 might greatly increase the accuracy for screening of active IBD.

ABSTRACT

<p><b>OBJECTIVE</b>This study further explores the stromal cell-derived factor-1 (SDF-1)/chemokine receptor 4 (CXCR4) signaling axis mechanism in temporomandibular joint osteoarthritis (OA) by detecting the changes in CXCR4, interleukin (IL)-6, and collagen X expression in the ATDC5 cell line stimulated by the cyclic tensile strain and SDF-1.</p><p><b>METHODS</b>Insulin-transferrin-selenium (ITS) was used to induce ATDC5 cells to differentiate into chondrocyte-like cells. After three weeks, the cells were divided into two groups: those with and without cyclic tensile strain. These groups were further divided into the negative control and SDF-1 groups. Strain force of 20% was applied. After 12 h, the total proteins were extracted from cells of the four groups, and Western blot analysis was used to detect the changes in CXCR4, IL-6, and collagen X expression.</p><p><b>RESULTS</b>SDF-1 could enhance CXCR4, IL-6, and collagen X expressions in the chondrocytes, and 20% tensile strain force could further upregulate the three factors.</p><p><b>CONCLUSION</b>Under abnormal tensile force, SDF-1 can upregulate its specific receptor CXCR4, thus increasing its-binding efficiency and resulting in the activation of the SDF-1/CXCR4 axis. This condition enhances the expressions of IL-6 and other inflammatory factors and directly damages to cartilage tissue. Such damage directly promotes chondrocyte hypertrophy, which enhances collagen X expression.</p>

ABSTRACT

Objective To explore the immunomodulatory effects of 1,25-dihydroxyvitamin D3 (1,25 (OH)2D3) in the treatment of experimental colitis induced by dextran sulfate sodium (DSS) in mice.Methods According to random number table,thirty BALB/c mice were randomly divided into control group,model group,low dose,moderate dose,and high dose intervention group.Mice of model group,low dose,moderate dose and high dose intervention group drank 5% DSS solution for seven days to create colitis model.On the 1st,3rd,5th,7th day,the mice of low dose,moderate dose and high dose intervention group were intraperitoneal injected with low,moderate,and high dose of 1,25(OH)2D3 (50,100 and 200 ng/each mouse,respectively).Mice of control group and model group were intraperitoneal injected with sterile soybean oil as control.The observed indicators included disease activity index (DAI) and colonic histopathological score (HPS).On the 8th day,all mice were sacrificed.The expression of interferon (IFN)-γ,interleukin (IL-17) and IL-21 in mice colon tissues and spleens at mRNA and protein level were measured by reverse transcription-polymerose chain reaction (RT-PCR) and flow cytometry,respectively.The data were analyzed by one way ANOVA.LSD-test or Tamhane test were performed for comparison in groups.Results Compared with the control group,the DAI and colitis HPS of mice in the model group significantly increased (0.33±0.52 vs 7.33±1.03,0.17±0.41 vs 12.00±0.63).Compared with the model group,the DAI and colonic HPS of intervention groups treated with 1,25 (OH)2 D3 declined with varying degrees (2.83 ± 0.40,2.83±0.75,2.33±0.52 and 10.83±0.98,7.50±0.84,6.67±0.52,LSD-t=0.39 and 0.41,all P<0.01).The expression of IFN-γ,IL-17 and IL-21 of the model group were significantly higher than those of the control group.The expressions of IFN-γ,IL-17 and IL-21 of intervention group were signifiantly lower than that of the model group (mRNA:LSD-t =0.12,0.13,0.09; protein:F =20.61,22.46,4.80,all P<0.01).Conclusion 1,25 (OH)2D3 might have a direct role on T-cell phenotype,down-regulate effective cytokines IFN-γ,IL-17 and IL-21 and then play an interventional role.
