ABSTRACT

A method was established for simultaneous determination of 21 active constituents including flavanols, isoflavones, flavonols, dihydroflavones, dihydroflavonols, chalcones, pterocarpan, anthocyanidins and phenolic acids in Spatholobi Caulis by ultra fast liquid chromatography with triple quadrupole linear ion trap mass spectrometry(UFLC-QTRAP-MS/MS). Then, it was employed to analyze and evaluate the dynamic accumulation of multiple bioactive constituents in Spatholobi Caulis. The chromatographic separation was performed on a XBridge®C_(18)(4.6 mm×100 mm, 3.5 μm) at 30 ℃ with a gradient elution of 0.3% formic acid aqueous solution-methanol, and the flow rate was 0.8 mL·min~(-1), using multiple-reaction monitoring(MRM) mode. A comprehensive evaluation of the multiple bioactive constituents was carried out by gray correlation analysis(GRA). The 21 target components showed good linearity(r>0.999 0) in the range of the tested concentrations. The average recovery rates of the 21 components were from 97.46% to 103.6% with relative standard deviations less than 5.0%. There were differences in the contents of 21 components in Spatholobi Caulis at diffe-rent harvest periods. Spatholobi Caulis had high quality from early November to early December, which is consistent with the local tradi-tional harvest period. This study reveals the rule of the dynamic accumulation of 21 components in Spatholobi Caulis and provides basic information for the suitable harvest time. At the same time, it provides a new method reference for the comprehensive evaluation of the internal quality of Spatholobi Caulis.

ABSTRACT

<p><b>OBJECTIVE</b>To observe the maintenance effect of polyglycosides of Tripterygium wilfordii (GTW) on remission in postoperative Crohn disease (CD).</p><p><b>METHODS</b>From 2005 to 2007, 45 adult cases of postoperative Crohn disease were randomly divided into two groups, GTW group and mesalazine group, which received GTW and mesalazine treatment respectively. CD activity index (CDAI) and clinical markers were collected at 0, 3, 6, 12 months or at the onset of symptoms. Ileocolonoscopy was performed at the end of the trial (1 year after operation) or at the onset of symptoms, and recurrence score were recorded.</p><p><b>RESULTS</b>No clinical recurrence was ascertained in both groups at 3 months. Four patients (18.2%) in GTW group relapsed and 5 (21.7%) in mesalazine group relapsed at 6 months (P=0.530). Seven patients (31.8%) in GTW group and 9 (39.1%) in mesalazine group relapsed at one year (P=0.421). Ten patients (45.5%) in GTW group had endoscopic recurrence compared with 14 (60.9%) in mesalazine group at one year(P=0.231). There were no significant differences between two groups.</p><p><b>CONCLUSION</b>GTW is similar to mesalazine in maintenance of remission of postoperative Crohn disease.</p>

ABSTRACT

Aim To observe homoharringtonine(HHT) distribution in eyeball tissue of rabbit. Methods Harringtonine(HT) was used as internal standard. The chromatography consisted of Kromasil C18 column with methanol 0.01mol?L-1 (60∶40,pH 2.5) as the mobile phase at the flow rate of 0.8ml?min-1. Results The chromatography retention time of HHT and HT were 6.35min and 4.70min. The standard curve was linear in the concentration range of 5~200 ?g?L-1 in serum and the relation coefficient was 0.994. The recovery of spiked sample was(93.17?4.31)%. After injection of 2mg of HHT in side of rabbit eyeball, the contents in sclera, cornea, retina, vitreous body and crystalline lens were reduced progressively, but the eliminate rate were reduced in a proper order. Conclusion HHT can distribute to every eyeball tissue when it was injected in side of rabbit eyeball.
