ABSTRACT

Objective To observe the disease type and the changes of symptom load during treatment of patients with Ph chromosome/ BCR-ABL fusion gene negative myeloproliferative neoplasm (MPN). Methods A total of 84 patients with MPN diagnosed from May 2017 to January 2019 in People's Hospital of Longhua Dis-trict of Shenzhen were selected,and were divided into polycythemia vera (PV)group,essential thrombocy-hemia (ET)group,and myelofibrosis (PMF)group according to their subtypes,with 28 cases in each group. The scores of MPN-SAF-TSS were compared among the three groups. Besides,the scores of the scale (myelo-proliferative neoplasm symptom assessment form total symptom score,MPN-SAF-TSS)in different treatment periods (at the time of the visit,when the disease progressed,when the disease was stable,when the clinical improvement was made,when the partial remission was completed,at the time of remission and recurrence) were also compared. Results At the time of initial diagnosis,there were significant differences in the inci-dences of symptom burdens among the three groups of MPN patients with abdominal fullness (χ2 = 6. 095,P =0. 047),abdominal discomfort (χ2 = 7. 342,P = 0. 025),poor mobility (χ2 = 13. 029,P = 0. 001),inatten-tion (χ2 = 6. 099,P = 0. 047),pruritus (χ2 = 6. 956,P = 0. 031),bone pain (χ2 = 7. 807,P = 0. 020),fever (χ2 = 8. 000,P = 0. 018)and weight loss (χ2 = 27. 340,P < 0. 001). The incidences of poor mobility (85. 71%,24 / 28),inattention (67. 86%,19 / 28)and weight loss (82. 14%,23 / 28)in PMF group were significantly higher than those in PV group [42. 86% (12 / 28),39. 29% (11 / 28),35. 71% (10 / 28)]and ET group [46. 43% (13 / 28),39. 29% (11 / 28),14. 29% (4 / 28)](all P < 0. 05). The incidences of abdominal discomfort (75. 00%,21 / 28)and bone pain (60. 71%,17 / 28)in PMF group were higher than those in PV group [39. 29% (11 / 28),25. 00% (7 / 28)](both P < 0. 05). ABSTRACT

Objective The dynamic accumulation trend of secondary metabolite in cultivated Spatholobus suberectus Dunn at different growth time was studied to provide scientific basis for confirming its reasonable harvesting time. Methods Ethanol-soluble extractives were measured by the method recorded in Chinese Pharmacopoeia ( A) Appendix XA ( 2010 edition). The content of total flavonoids was tested by ultraviolet spectrophotometer. The contents of protocatechuic acid, catechin, and epicatechin in Spatholobus suberectus Dunn were determined by high performance liquid chromatography ( HPLC). Results The contents of ethanol-soluble extractives, total flavonoids and the major components such as protocatechui cacid, catechin and epicatechin in Spatholobus suberectus Dunn showed increasing trend with the prolongation of growth time. The changes of contents of alcohol-soluble extract, total flavonoids and epicatechin in samples collecting at different months in one year showed a parabolic trend, and the contents reached the peak values from July to September. Catechin content kept increasing continually. The content of ethanol -soluble extractives of Spatholobus suberectus Dunn aged 4-6 years met the requirements of the current edition of Chinese Pharmacopoeia when harvested from July to September. Conclusion It is preliminarily recommended that Spatholobus suberectus Dunn cultivated for more than 7 years is suitable for harvesting, but the optimal harvest period and the harvest month still need to be further studied.

ABSTRACT

Objective To explore the seed protein extraction method for Amomum villosum Lour., so as to lay the foundation for the study of differential proteomics of Amomum villosum Lour. during seed dormancy and germination. Methods We used the four kinds of commonly-used protein extraction methods for the experiment, including Tris-HCl extraction method, trichloroacetic acid (TCA) /acetone precipitation method, direct lysis buffer method and improved Tris-saturated phenol method. The obtained proteins were quantified, and then were analyzed by sodium dodecyl sulphate- polyacrylamide gel electrophoresis (SDS-PAGE) . Results The protein yield was the highest when using direct lysis buffer method, which was mainly composed of small molecule protein. The protein yield was the lowest using Tris/HCl method, characterized by the diffused stripes in SDS-PAGE spectra. The protein yield was moderate using TCA/acetone method and improved Tris-saturated phenol method, and the obtained protein by the TCA/acetone method had less stripes in SDS-PAGE spectra than that by Tris-saturated phenol method, and was characterized by macromolecular protein. Conclusion The combination of direct lysis buffer extraction method and improved Tris-saturated phenol method may provide reference for seed protein extraction method of two-dimensional electrophoresis for Amomum villosum Lour..

ABSTRACT

Objective To compare the impacts of subtotal nephrectomy and ischemiareperfusion injury on renal stem cells and progenitor cells of rats, and to explore the significance of renal stem cells and progenitor cells for renal repair and the possible mechanisms of prognosis in rats with acute renal failure (ARF) or chronic renal failure (CRF).Methods Rats of CRF or ARF model underwent 5/6 nephrectomy or renal artery ligation and repedusion respectively, and rars in control group underwent sham operation.Scr,BUN and 24 hour urine protein were regularly measured.Kidney specimens were obtained at the set time for HE staining and fluorescence staining.Expressions of CD24,CD133 and podocin were detected by immunofluorescence.RT-PCR was performed to quantify the expression of transforming growth factor β1 (TGF-β1),Notch2,hepatocyte growth factor (HGF),bone morphogenetic protein 7 (BMP7) and Pax-2 mRNA in renal tissue and the expression of podocin mRNA in renal cortex.Correlation among the expressions of Pax-2 mRNA,podocin mRNA and glomemlosclerosis index were analyzed. Results The rats of two models presented typical ARF or CRF in renal pathology and function.Glomerulosclerosis index in CRF group increased gradually with time, which were (2.34±0.28)%,(25.12±5.67)%,(89.42±12.28)% and (171.23±32.28)% at day 14,day 30,day 60 and day 90 respectively.Compared with sham group, the CD24+CD133+ cells of the ARF rats showed no significant change in quantity and distribution, while the CRF rats showed gradual reduction of CD24 +CD133+ cells. The expression of podocin in glomerulus decreased temporarily and recovered finally after ischemiareperfusion injury, but decreased gradually after 5/6 nephrectomy.Compared with sham group, expression of TGF-β1,Notch2 mRNA in renal tissue was increased in CRF group, while the expression of HGF,BMP7 mRNA in renal tissue of ARF group were increased.Between the expression of Pax-2 mRNA in renal tissue and the expression of podocin mRNA in renal cortex, there was positive correlation in CRF group, while they both were negatively correlated with glomerulosclerosis index. Conclusions Ischemia-reperfusion injury makes no obvious impairment to renal progenitor cells.Having progressively injured the living environment of renal progenitor cells, subtotal nephrectomy reduces renal progenitor cells, and causes podocytes to repairing incompetently, which may be the main pathogenesis of CRF with poor prognosis.

ABSTRACT

To study the expression of beta-human chorionic gonadotropin (beta hCG) genes in renal cell carcinomas (RCC) and benign renal disease tissues, nested reverse transcription-polymerase chain reaction (RT-PCR) and restriction endonuclease analysis were employed to detect the expression of beta hCG genes in 44 cases of RCC tissues and 24 cases of benign renal disease tissues. It was found that 52% RCC samples revealed positive for beta hCG mRNA expression. Positive rate in advanced stage and poorly differentiated RCC was higher, but there was no significant difference. The positive rate of beta hCG mRNA expression was 54% in 24 cases of benign renal tissues, including 3 cases out of 6 polycystic kidneys, 7 cases out of 13 renal atrophies, 2 cases out of 2 oncocytomas and 1 case out of 2 pyonephrotic kidneys. beta 7 was most frequently transcribed subtype gene independent on the histology. These findings suggested beta hCG gene transcription is not only involved in RCC but also in benign renal diseases.

Gilg.decreased with the prolongation of storage time and with the decrease of water content. The seeds with water content being 9.34% and 7.35% had a higher germination capacity when stored at 4℃ for 35 days than that stored at room temperature. The fresh seeds stored in wet sand sprouted and the seeds stored in drying apparatus and dry sand had lower germination capacity and went mouldy.【Conclusion】Low temperature and moderate water content are benificial to the storage of the seeds of Aquilaria sinensis(Lour.) Gilg..

ABSTRACT

[Objective] To observe the effect of biological fertilizers on the yield and quality of Amomum villosum (AV). [Methods] Field experiment method was used to observe the effect of different kinds and different amount of biological fertilizers on AV. Biological fertilizers were: A (mainly composed of fermented chicken manure), B (mainly composed of fermented filter of sugarcane) and C (mixed with fertilizer A and biological fertilizer at certain proportion). The fertilizer amount was 150, 112 and 75 g****m-2 per time. Content of bornyl acetate in AV was detected by gas chromatography. [Results] The three kinds of biological fertilizers increased the yield of AV but had no obvious effect on its quality; high- and moderate-amount fertilizer increased the yield of AV, in particular fertilizer A (112 g?m-2 per time). The three kinds of fertilizers had no effect on the content of bornyl acetate in AV. [Conclusion] Reasonable application of fertilizers is one of the important ways to raise the yield of AV.

ABSTRACT

Different varieties of Terminalia chebula Retz. were identified by DNA finger prints, andat the same time, some problems encountered during the application of RAPD for the identification ofTCM were studied. The polymorphic features of DNA finger prints of different species were compared. Results of the study showed that they were differently related in heredity which can provide a basis for thedifferentiation of one species from the other. Means for the solution of problems arised during RAPD ap-plication were suggested. It was concluded that RAPD is an effective method to identify T. chebula from itsconfusable varieties.
