ABSTRACT

OBJECTIVE To investigate the attenuation and synergism of Hugan buzure recipe (HBR) combined with oxaliplatin on hepatocellular carcinoma tumor bearing nude mice and its mechanism. METHODS Eight nude mice were selected from 40 nude mice as the blank group (normal saline), and the remaining nude mice were inoculated with hepatoma cells Huh7 to establish the tumor-bearing model. The 32 modeled nude mice were randomly allocated to four groups: model group (normal saline, ig), HBR group (0.69 g/kg, ig), oxaliplatin group (10 mg/kg, ip), and combination group (intraperitoneal injection of 0.69 g/kg HBR+intragastric administration of 10 mg/kg oxaliplatin), with 8 mice in each group. Administer drug/normal saline once a day for 32 consecutive days; administer subcutaneous injection once every 7 days for a total of 5 times. During the experiment, the general condition of nude mice in each group was observed, and the tumor volume was measured every 4 days. On the 30th day of administration, the thermal stimulation paw withdrawal latency of nude mice in each group were detected. The tumor inhibition rate, spleen coefficient, the number of red blood cells, white blood cells and platelets in the whole blood of nude mice in each group, and the content of aspartate aminotransferase (AST) and creatinine in serum were detected after the end of administration. HE staining was used to observe the pathological changes in tumor tissues in nude mice in each group. The expression of microtubule-associated protein 1 light chain 3 (LC3),selective autophagy adaptor protein p62, B-cell lymphoma-2 (Bcl-2), Bcl-2 associated X protein (Bax), and Caspase-3 protein in tumor tissues. RESULT Compared with the model group, the tumor volume, tumor weight, white blood cells,red blood cells in the whole blood and spleen coefficients of nude mice in the oxaliplatin group were significantly decreased (P<0.01); the thermal stimulation paw withdrawal latency, AST and creatinine in serum were significantly increased (P<0.05 or P<0.01). Compared with the oxaliplatin group, the tumor volume and tumor weight of nude mice in the combination group were significantly decreased (P<0.01); the white blood cells, red blood cells and platelets in the whole blood and spleen coefficients of nude mice were significantly increased (P<0.05 or P<0.01); the thermal stimulation paw withdrawal latency, AST and creatinine in serum were significantly decreased (P<0.01); the expression levels of LC3, Bax and Caspase-3 proteins in tumor tissues of nude mice were significantly increased (P<0.01), and the expression levels of p62 and Bcl-2 proteins were significantly decreased (P<0.01). CONCLUSIONS HBR enhances the tumor inhibition rate of oxaliplatin by inducing apoptosis and autophagy, and can alleviate the peripheral neurotoxicity, hematological toxicity, hepatorenal toxicity, and immune organ toxicity caused by oxaliplatin in nude mice.

ABSTRACT

Polycystic ovary syndrome (PCOS) is a common reproductive endocrine and metabolic disease, and its pathogenesis is closely related to inflammation, insulin resistance, and metabolic disorders. Bilirubin is the final product of the destruction and degradation of senescent red blood cells in the body. In addition, bilirubin can be not only used to evaluate liver function damage and cytotoxicity, but also can anti-inflammatory, antioxidant, alleviate metabolic disorders, etc. Recently, studies have found a certain correlation between low levels of bilirubin and PCOS: the level of bilirubin in patients with PCOS is low, and the anti-inflammatory and antioxidant properties of bilirubin may play a protective role in the pathogenesis of PCOS.

Then, this paper analyzes the application effects of mobile health in the five aspects of treatment process management, health education, medication management, lifestyle management and psychological care for infertility patients based on the literature. The existing problems are analyzed and prospected on this basis, in order to provide a reference for the application of mobile health to infertility patients′nursing in China.

ABSTRACT

OBJECTIVE To study the effects of Hugan buzure formula (HBF) on intrahepatic cholestatic liver injury in rats and its potential mechanism. METHODS Rats were randomly divided into control group, model group, ursodeoxycholic acid (UDCA) group (positive control, 60 mg/kg ) and HBF low-dose, middle-dose and high-dose groups (HBF-L, HBF-M, HBF-H groups, 0.4, 0.8, 1.6 g/kg ), with 6 rats in each group. The rats in each drug group were given the corresponding drug solution intragastrically, once a day, for 7 consecutive days. The rats in the control group and the model group were given equal volumes of water intragastrically. On the 5th day, except for the control group, the rats in other groups were single intragastrically administered with alpha-naphthyl isothiocyanate olive oil solution (100 mg/kg) to establish the model. After 48 h of modeling, the contents of liver function indexes (aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, total bile acid, total bilirubin, direct bilirubin) and oxidative stress indexes [malondialdehyde (MDA), glutathione (GSH), superoxide dismutase] in serum of rats were detected; the pathological changes of liver tissue were observed. The mRNA expressions of inflammation-related factors [tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β)] and farnesoid X receptor (FXR) signaling pathway-related factors [FXR, small heterodimer partner (SHP), multidrug resistance protein 2 (MRP2), bile salt export pump (BSEP), Na+-taurocholate cotransporting polypeptide (NTCP), organic anion-transporting polypeptide 2 (OATP2) and cholesterol 7α-hydroxylase (CYP7A1)], the expressions of FXR signaling pathway-related proteins (FXR, MRP2, BSEP, NTCP) and nuclear factor- κB p65 (NF- κB p65) in liver tissue were detected.RESULTS Compared with the model group, the contents of liver function indexes and the level of MDA in serum, the mRNA expressions of the above inflammation-related factors and CYP7A1, and the relative expression of NF-κB p65 in liver tissue were significantly decreased; the levels of GSH in serum, the mRNA expressions of FXR, SHP, MRP2, BSEP, NTCP and OATP2, and the relative expressions of FXR, MRP2, BSEP and NTCP in liver tissue were significantly increased (P<0.05 or P<0.01); the pathological changes of liver tissue were significantly improved. Only some indexes in HBF-L group, HBF-M group and UDCA group were significantly reversed (P<0.05 or P<0.01). CONCLUSIONS HBF can prevent intrahepatic cholestatic liver injury in rats, and the effects may be related to the activation of FXR signaling pathway and the reduction of inflammation and oxidative stress.

ABSTRACT

OBJECTIVE To inv estigate the in vitro inhibitory effects of acteoside on cytochrome P 450(CYP)enzymes in liver microsomes of rats. METHODS Using probe substrates method ,acteoside(0.1,0.3,1,3,10,30 μmol/L)was incubated with probe substrates phenacetin ,mephentoin,diclofenac,coumarin,dextromethorphan and testosterone (substrates of CYP 1A2, CYP2C19,CYP2C9,CYP2A6,CYP2D6 and CYP 3A4 enzymes,respectively)in liver microsomes of rats. Another blank control group and positive inhibitor group [ α-naphthoflavone,ticlopidine,sulfabendazole,pilocarpine,quinidine and ketoconazole (inhibitors of CYP 1A2,CYP2C19,CYP2C9,CYP2A6,CYP2D6 and CYP 3A4 enzymes,respectively)] were set up. Using indapamide as the internal standard , the contents of corresponding metabolites (acetaminophen, 4-hydroxymephenytoin, 4-hydroxydiclofenac,7-hydroxycoumarin,dextran,6 β-hydroxytestosterone) were detected by ultra high performance liquid chromatography-tandem mass spectrometry . The IC 50 values were calculated by GraphPad v 8.0 software. By computer molecular docking technology ,acteoside and positive inhibitors were molecularly docked with the CYP enzyme ,and the binding mode and strength of the two molecules were analyzed. RESULTS The IC 50 values of acteoside to CYP 1A2 and CYP 2A6 enzymes were more than 30 μmol/L,and those of acteoside to CYP 2D6,CYP2C19,CYP2C9 and CYP 3A4 enzymes were 24.87,21.52,12.56 and 7.55 μmol/L,respectively. The hydrogen bond and hydrophobic force could form between acteoside and CYP 3A4 enzyme,and the hydrogen bond and electrostatic interaction could form between ketoconazole and CYP 3A4 enzyme. The binding free energy of acteoside and ketoconazole to CYP 3A4 enzyme were - 10.2 and - 12.4 kcal/mol (1 kcal/mol=4.19 kJ),respectively. CONCLUSIONS Acteoside shows moderate inhibitory effect on CYP 3A4 enzyme in liver microsomes of rats ,and its affinity is equivalent to that of positive inhibitor ;the compound shows weak inhibitory effect on other 5 CYP enzymes.

To observe the effects of Guanxinkang (GXK) decoction, a compound traditional Chinese herbal medicine, on serum lipids and apolipoprotein A I (ApoA I), apolipoprotein B (ApoB), apolipoprotein E (ApoE), C-reactive protein (CRP), serum amyloid A protein (SAA) and fibrinogen (Fbg) concentrations of ApoE-knockout mice with atherosclerosis, and to explore the mechanism of GXK decoction in anti-atherosclerosis.

ABSTRACT

<p><b>OBJECTIVE</b>To compare the antitussive activity of three kinds of Stemonae Radix specified in the Chinese Pharmacopoeia, including roots of Stemona sessilifolia, S. japonica and S. tuberosa.</p><p><b>METHOD</b>The antitussive activity was determined in mouse after cough induction by ammonia aerosol stimulation and the number of cough in 2 min were detected with codeine as positive control.</p><p><b>RESULT</b>All the decoctions, the total alkaloid fractions and non-alkaloid fractions of S. sessilifolia, S. japonica and three chemical types of S. tuberosa showed significant antitussive effect and exhibited a dose-dependent inhibition of coughing. The ED50 values showed that the antitussive activity strength for both total alkaloid fractions and the decoctions are: S. tuberosa (Type I) > S. sessilifolia > S. japonica. The total alkaloid fractions had more potent atitussive activity than the decoctions and non-alkaloid fractions. The antitussive activity strength for the three chemical types of S. tuberosa is: Type I > Type III > Type II. The samples from different producing areas for the same species of Stemonae Radix had no significant differences in antitussive activity. The result also showed that the honey-processed slice had much stronger antitussive activity than raw slice.</p><p><b>CONCLUSION</b>The antitussive efficacies of Stemonae Radix were influenced by chemical diversity both in same species and among different species, different fractions and processed method.</p>

ABSTRACT

Objective To develop a method for the determination of acrylamide in flocculant by gas chromatography-mass spectrometry.Methods Acrylamide was derived through bromination and extracted by organic solvent.GC-MS method was used to identify the target substance and quantify acrylamide.Results The recoveries obtained from samples spiked with standards were in the range of 70.2%-109.2% and relative standard deviations (n=5) was less than 5.0%.Linearity of peak area was obtained over a wide range of concentration (4.3-2 000 ?g/ml),and the correlation coefficients was 0.999 4.Conclusion The method is reliable,selective and reproducible and it is applicable to the determination of the acrylamide in flocculant.

ABSTRACT

Objective To investigate the influence of passive smoking to pregnant woman's and fetus's health by the detection of biomarker. Methods A questionnaires about passive smoking made by ourselves were answered by 236 pregnant women including quantity and the number of year of smoking in the family and working places. At the same time pregnant woman's saliva, venous blood, urine and umbilical cord blood were collected. The sulphocyanate in their saliva was determined with pyridine-barbiturate method; the cotinine in venous blood, urine and umbilical cord blood were determined with barbiturate method. Results The content of sulphocyanate in saliva was different upon the quantity of passive smoking of pregnant women; there were significant differences had been seen among groups of passive smoking in cotinine in venous blood, urine and umbilical cord blood. Conclusion The contents of cotinine in venous blood, urine and umbilical cord blood are biomarkers of pregnant woman subjected to passive smoking, and passive smoking of pregnant woman will influence the health of pregnant woman and fetus.
