ABSTRACT

Purpose@#This study was aimed to evaluate the clinical significance and prognostic value of CRP/albumin ratio (CAR) in patients with gastric cancer. @*Methods@#The data of 205 gastric cancer patients who underwent surgery was analyzed retrospectively. The association of CAR with the clinical features and prognostic value in gastric cancer was analyzed. The data of this study was combined with previous studies to further determine the prognostic value of CAR in patients with gastric cancer using a metaanalysis method. @*Results@#Cox analysis revealed that preoperative CAR was an independent prognosis indicator in patients with gastric cancer. High expression of CAR indicated a shorter survival time than in those with lower expression. CAR has a higher prognostic value in the 1-, 3-, and 5-year overall survival in patients with gastric cancer. CAR showed significant difference regarding the gastric cancer patients’ age, M stage, and clinical stage. The discriminate value of CAR in M stage of gastric cancer was high (area under the curve, 0.809). A meta-analysis combining previous data and our data showed that preoperative CAR demonstrated a significant association with the overall survival of patients with gastric cancer. @*Conclusion@#This study demonstrated that preoperative CAR could serve as an important prognostic indicator in patients with gastric cancer.

ABSTRACT

Seventy-nine injectable herb extractions have been approved by the Chinese Food and Drug Administration (CFDA) and are frequently administered intravenously for various diseases. Unfortunately, herb-drug interactions are under-investigated and sometimes overlooked in the clinic. In the present investigation the in vitro inhibition of 9 drug metabolizing enzymes including CYP1A, CYP2A6, CYP2B6, CYP2C8, CYP2C9, CYP2C19, CYP2D6, CYP2E1, and CYP3A was assessed using an appropriate probe substrate for each enzyme with human liver microsomes. Metabolite formation was quantified using a validated and sensitive high-performance liquid chromatography-tandem mass spectrometry (LC-MS/MS) assay. The IC50 of each herb extract was estimated using a concentration range from 5% to 0.5%, and the time-dependent inhibition of the nine CYP450 isoenzymes was also determined. Of the 79 approved iv herb injectables, 37 inhibited CYP1A, 24 inhibited CYP2A6, 41 inhibited CYP2B6, 36 inhibited CYP2C8, 31 inhibited CYP2C9, 41 inhibited CYP2C19, 13 inhibited CYP2D6, 25 inhibited CYP2E1, and 42 inhibited CYP3A with 50% or greater inhibition at a test concentration of 5% (v/v). IC50 differences were noted between pre-incubation or co-incubation assays with HLM for 30 min, with the time-dependent inhibitory (TDI) effects were observed with 2 injectables on CYP1A, 5 injectables on CYP2A6, 5 injectables on CYP2B6, 6 injectables on CYP2C8, 1 injectable on CYP2D6 and 6 injectables on CYP3A. Collectively, the results demonstrate that potential herb-drug interactions (HDIs) can occur with the concomitant use of herb injectables and prescription drugs that are cleared by CYP450 enzymes, and further investigation is warrant for the clinical relevance of these interactions.

ABSTRACT

@#【Objective】To explore the effects and the possible mechanism of KPT- 8602,a novel selective inhibitor of nuclear export protein (XPO1),on proliferation, cell cycle and apoptosis in human histiocytic lymphoma cell line U937 cells.【Methods】U937 cells were treated with different concentrations of KPT- 8602. Cell viability was assessed by CCK-8 assay. The cell cycle distribution and the apoptosis rate were analyzed by flow cytometry. The proteins expression of XPO1,p-AKT,AKT,Cleaved Caspase-3,p21 were determined by Western blot. Fluorescence microscope was used in observing the intracellular location of XPO1. 【Results】 KPT- 8602 inhibited the growth of U937 cells in a dose- dependent(P<0.001)and time- dependent manner(P<0.001),but normal PBMC were unaffected. 48 h after treatment with KPT-8602,a higher proportion of cells in G1 phase was observed(P<0.001)and the apoptosis rate increased(P=0.016)with drug concentration in U937 cells. XPO1 protein expression of U937 cells was significantly higher than normal PBMC(P=0.003). 48 h after treatment with KPT- 8602,the protein expression of XPO1 decreased(P=0.011),p-AKT decreased(P=0.011),and Cleaved Caspase- 3 increased(P=0.009). In addition, the protein expression of p21,the cargo protein of XPO1,increased in both the nuclei and the cytoplasm(P<0.05)after treatment with KPT- 8602. XPO1 decreased in both the nuclei and the cytoplasm under the fluorescence microscope after treatment with KPT- 8602.【Conclusion】KPT- 8602 can inhibit the proliferation of U937 cells, block the cell cycle at G1 phase, and induce cell apoptosis, which may partially be attributed to the down-regulation of XPO1 and inhibition of PI3K/AKT signaling.

ABSTRACT

OBJECTIVE@#To evaluate the effectiveness of IL-10 in suppressing 18F-FDG uptake in the inflammatory granuloma of SD rats.@*METHODS@#Eight SD rats were killed, and their blood was collected sterily. After centrifugion, the white blood cells were incubated in PRMI 1640 for 3 days. Then each culture flask of white blood cells was divided into two equal parts. To one group was added 0.2 mL IL-10 solution (0.1 mg/mL); to the control was added with 0.2 mL of 0.9% sodium chloride solution. All cells were then incubated for 120 minutes at 37 degree, after which 18F-FDG (1.85 MBq) was added. Sixty minutes later, the cells were washed twice with PBS and the extent of uptake 18F-FDG determined. In vivo, an inflammatory granuloma was produced by hypodermic injection of rats with a mixture of Freund's complete adjuvant, bovine serum albumin and talcum powder. Each rat was maintained for 8 weeks. Imaging of the inflammatory granulomas was performed using the 18F-FDG signal. IL-10 was injected into SD rats at 10 μg/kg of body weight. Sixty minutes later, 7.4 MBq of 18F-FDG were injected, and, after a further 60 minutes, the rats underwent a PET-CT scan. The region of interest (ROI) of the inflammatory granuloma was delineated and the standard uptake value (SUV) calculated. A second PET-CT scan was done without IL-10 on the next day. The granulomatous tissue underwent pathological examination.@*RESULTS@#In the intro test, the with blood cell uptaking ratio of 18F-FDG was (50.3±6.7)% without IL-10, and (34.6±3.5)% with IL-10(t=8.9, P<0.01). IL-10 suppressed the rat white blood cell uptaking 18F-FDG. In the PET-CT scan, the SUV of ROI on inflammatory granuloma was 1.7±0.4 with IL-10 and 2.1±0.3 without IL-10 (t=20.6, P<0.01). IL-10 suppressed the inflammatory granuloma uptaking 18F-FDG.@*CONCLUSION@#IL-10 can suppress the inflammatory granuloma of SD rats uptaking 18F-FDG.

ABSTRACT

To evaluate the sensitivity, specificity, and accuracy of the serum tumor markers cytokeratin 19 fragment [CYFRA 21-1], carcinoembryonic antigen [CEA] and cancer antigen 125 [CA125] in the diagnosis of bone metastases in patients with lung cancer. A total of 134 patients with lung cancer, diagnosed by pathological examination or bronchoscopic biopsy, as well as 105 healthy subjects, were enrolled in the study. The cancer patients were divided into a metastasis-negative group, a localized metastasis group [1 to 2 metastases], and an extensive metastasis group [3 or more metastases]. Serum levels of CYFRA 21-1, CEA, and CA125 were measured in each subject. The diagnostic efficiency of three tumor markers, used alone or in combination, was assessed using ROC analysis and compared with that of X-ray, CT, and emission computed tomography [ECT]. The levels of CYFRA 21-1, CEA, and CA125 were highest in patients with extensive metastasis, and lowest in patients without metastasis. The optimal cutoff value of CYFRA 21-1, CEA, and CA125 for diagnosing bone metastasis of lung cancer was 42.59 ng/ml [AUC: 0.908], 36.35 ng/ml [AUC: 0.854] and 36.21 U/ml [AUC: 0.786], respectively. The combined use of CYFRA 21-1, CEA, and CA125 was more sensitive than X-ray and more specific than CT. Combined determination of CYFRA 21-1, CEA, and CA125 can significantly improve accuracy and positive rates of diagnosis of bone metastasis in patients with lung cancer. Compared with imaging methods, serological detection is simple, rapid, and highly cost-effective

ABSTRACT

To observe the expression of cyclin D1, hTERT, and telomerase activity in MNC, HL-60, HL-60A and to explore their effects on leukemogenesis and drug-resistance, normal human peripheral blood mononuclear cells, HL-60 cells sensitive to adriamycin and HL-60A cells resistant to adriamycin were investigated. The cell cycle was analyzed by flow cytometry, and the apoptosis was analyzed by Annexin V-FITC(+) PI staining. Expressions of cyclin D1 and hTERT were determined by real-time PCR and Western blot. Telomerase activity was detected by TRAP-ELISA. The results indicated that the percentage of MNC, HL-60 and HL-60A in S phase was (10.21 + 2.11)%, (44.93 + 3.00)%, and (51.38 + 1.10)% respectively; the percentage of apoptosis cells was (16.14 + 2.13)%, (7.53 + 0.92)%, (4.15 + 0.96)% respectively; the expression of mRNA and protein for cyclin D1 and hTERT increased; the telomerase activities of HL-60 and HL-60A were higher (p = 0.000), whereas the difference between HL-60 and HL-60A was no statistically significant (p = 0.232); positive correlation between cyclin D1, hTERT and telomerase activity had been found (p < 0.01). It is concluded that the cells of S phase increased while the apoptotic cells decreased in HL-60 and HL-60A, especially in HL-60A, which may be due to the up-regulation of cyclin D1, hTERT and telomerase activity.

ABSTRACT

Objective To evaluate the value of MSCT perfusion imaging in assessment of single renal function of hydronephrotic kidney. Methods 64-slice spiral CT perfusion was performed in 36 obstructive nephrohydrosis patients whose split renal glomerular filtration rate(GFR) was measured by SPECT renal dynamic imaging. ①The perfusion parameters of the renal cortex and renal medulla of the hydronephrotic kidney were compared with the normal kidney studied by contrast group. ②The 72 kidneys were divided into normal renal function, mild and severe renal impairment groups according to renal function. Differences between the groups respect to all the mean perfusion parameters of the renal cortex and renal medulla were assesses by ANOVA. ③Using Pearsons correlation test, the correlations between all the mean perfusion parameters of the renal cortex and renal medulla and renal GFR were examined. Results ① The time-density curves of bilateral normal renal cortex and medulla were not symmetric. The mean BF, BV, PS, PBV of renal cortex were (203. 2±44.9)ml·100 ml~(-1)·min~(-1), (27.6±3.9)ml/100 ml, (30.7±6.5)ml·100 ml~(-1)· min~(-1), (46.5±10. 9)ml/100 ml; and the mean BF, BV, PS, PBV of renal medulla were (99.9±24.1)ml·100 ml~(-1)·min~(-1) ,(18. 3±4.3)ml/100 ml, (51.8±12.1)ml · 100 ml~(-1)· min~(-1) , (21.3±3.0)ml/100 ml. The mean perfusion parameters of the cortex and medulla of obstructed kidney were lower compared to that of normal kidney. ②There were significant differences of all the perfusion parameters of the renal cortex and me-dulla between 3 groups (P<0. 05). ③The perfusion parameters of the renal cortex and medulla had positive linear correlation with GFR. The best correlation was the blood flow of the cortex of kidney. The correlation coefficient r=0.852.Conclusions64-slice spiral CT perfusion imaging can quantita-tively evaluate the haemodynamic condition and functional lesion of the kidney, classify the impaired kidney function. The perfusion parameters of the renal cortex and medulla had positive linear correlation with GFR.

<p><b>OBJECTIVE</b>To investigate the association of activated coagulation factor VII(F7a) and its gene Msp I polymorphism with coronary heart disease in elderly patients.</p><p><b>METHODS</b>This was a case-control study, and the method of candidate gene was adopted. F7 genotypes were identified with polymerase chain reaction amplified genomic deoxyribonulieic acid (DNA) and Msp I restriction fragment length polymorphism analysis, and the level of plasma F7a was detected with recombinant tissue factor method for 108 elderly patients with coronary heart disease and 120 sex- and age-matched healthy control subjects.</p><p><b>RESULTS</b>(1) Plasma F7a levels was significantly higher in elderly patients with coronary heart disease than in healthy control subjects (2.88 +/- 0.62 vs 2.58 +/- 0.60 microg/L, P < 0.05), and was significantly higher in old myocardial infarction than in stable angina pectoris (3.12 +/- 0.62 vs 2.76 +/- 0.60, P < 0.05). F7a was shown to be a risk factor for coronary heart disease in elderly patients by Logistic regression analysis (OR=1.21 P < 0.05). (2) The allelic frequencies were in accordance with Hardy-Weinberg equilibrium. The results suggested that the distribution of genotype and allelic frequencies in the groups displayed no significant difference, and there was no difference between the subgroups of coronary heart disease in elderly patients, either (P > 0.05). (3) F7a level was significantly higher in RR genotype than in Q allele carriers (2.72 +/- 0.60 vs 1.98 +/- 0.59 microg/L, P < 0.05) and was associated with F7 gene polymorphism.</p><p><b>CONCLUSION</b>Plasma F7a level may be an independent risk factor of coronary heart disease in elderly patients, and it may be influenced by the Msp I polymorphism of F7 gene.</p>

ABSTRACT

BACKGROUND: Reading disorder is the main obstacle in children, but its etiology and pathogenesis are still uncovered.OBJECTIVE: To explore the relationship between brain blood flow (BBF)and scores for children reading skill detecting test (CRSDT), in order to provide theoretical references for earlier intervention and functional monitoring for children with reading disorder(RD).DESIGN: Comparative observation study with RD children as subjects and normal children as controlSETTING: Nuclear medicine and psychological-health institute of a university.PARTICIPANTS: This study was carried out in Nuclear Medicine and Psychological-health Institute of Second Xiangya Hospital, Central South University. Between August 1998 and August 1999, 25 children with RD were screened out from the students from grade 3 to grade 6 in two Changsha civic elementary schools, including 15 males and 10 females aged(10±1)teacher lasted for more than one year and began from the earlier stage of school age(before grade three), with their achievement ranked last or often failed in examinations, even stay in the same grade due to learning disorder;and teacher or investigation of their homework accorded to the ICD-10 didiseases. Meanwhile 20 healthy children with normal intelligence were randomly selected as control group from the same class of RD children, including 12 males and 8 females with age of (10 ± 1 )years.METHODS: Non blood sampling-SPECT images was used for detecting cerebral blood flow(CBF), as well as right and left CBF and regional CBF (rCBF) of both RD children and normal controls. Rough scores for CRSDT were obtained for analyzing the relationship between it and CBF.group .RESULTS: CBF was(388.7 ± 37.7) μL/g per minute in RD group obviously lower than(436.5 ± 26.4) μL/g per minute in control gruop( t = 2. 820, P < 0.01 ) ;The distribution frequency of regions with obviously decreased rCBF ranked as follows: frontal lobe, occipital lobe > parietal-occipital boundary > temporal lobe > parietal lobe > thalamus > other regions(cerebellum, brain stem and basal ganglion) in RD children; moreover rough scores for reading skill was found positively correlated with CBF in RD group( r = 0.651,P <0.05).CONCLUSION: CBF was proved decreased in children with RD, and CBF obtained by SPECT image and CRSDT can be used for reflecting the severity of disease and brain function, expecting to improve their long-term life quality of RD children by earlier intervention.
