ABSTRACT

Objective:To observe the effect of annexin A1 peptidomimetic Ac2-26 on lung injury in rats with cardiopulmonary bypass, and analysis its anti-inflammatory mechanism. Methods:Eighteen male SD rats were divided into 3 groups(n=6) according to the random number table: sham operation group(S group), ischemia reperfusion group(IR group), Ac2-26 group(A group). The sham operation group received femoral arteriovenous puncture and catheterization, only the left lung was opened and the cardiopulmonary bypass tube was not connected. The other two groups were established with cardiopulmonary bypass left lung ischemia-reperfusion injury model. The IR group was given the same volume 10 minutes before cardiopulmonary bypass with the normal saline solution. Group A was injected with Ac2-26(1 mg/kg) through the tail vein 10 minutes before cardiopulmonary bypass. The arterial blood of the three groups of rats was taken for blood gas analysis before CPB(T1), immediately after opening the left lung hilum(T2), and at the end of the experiment(T3) to calculate OI and RI. At T1 and T3, the femoral vein blood was taken to measure the number of PMN. At T3, pulmonary artery blood was taken to measure the number of PMN and VCAM-1 content, left lung tissue was taken to measure NE and MPO content by ELISA, lung tissue neutrophil apoptosis was measured by Tunel method, and lung tissue ICAM-1 and AnxA1 protein expression was measured by Western-Blot.Results:At T3, compared with group S, the number of PMN in peripheral blood and pulmonary artery blood and the content of pulmonary artery VCAM-1 in IR group were significantly higher, the content of MPO and NE in lung tissue increased, the lung tissue pathological damage score increased, OI decreased and RI increased, all P<0.05. Compared with IR group, the lung tissue pathological damage score of rats in group A was significantly reduced, the number of PMN in peripheral blood and pulmonary artery blood and the content of VCAM-1 in pulmonary artery blood were significantly reduced, the expression of AnxA1 and ICAM-1 in the lung tissue of rats decreased; the content of MPO and NE in lung tissue decreased, and the apoptosis rate of PMN increased, all P<0.05. Conclusion:Ac2-26 annexin A1 peptidomimetics can inhibit the aggregation of neutrophils in the lung, weaken the adhesion of neutrophils to the pulmonary vascular endothelium, promote the apoptosis of neutrophils in the lungs, and reduce the content of NE and MPO. It has a protective effect on lung injury after cardiopulmonary bypass in rats.

ABSTRACT

Objective:To develop overgeneral autobiographical memory questionnaire(OGMQ) and test its reliability and validity. Methods:Through literature search, expert opinions and group discussion the initial items of the questionnaire were formed. A random cluster method was adopted to select 1 911 college students for the preliminary test.Totally 1 840 valid questionnaires were obtained, and 920 of which were assessed for exploratory factor analyses and 920 of which were assessed for confirmatory factor analyses and reliability and validity analysis. Thirty subjects with depression and 30 normal subjects were selected to test its criterion validity. Results:The questionnaire consisted of 19 items, two dimensions (construction, elaboration) and four factors (spatial-temporal factor, control factor, context factor, emotion factor). Exploratory analysis extracted 4 factors and 19 items, KMO (Kaiser-Meyer-Olkin) = 0.905, Bartlett test: χ 2=4 728.134, df=171, P<0.001.Confirmatory analysis showed that the factor loads of spatio-temporal factor and control factor in the construction dimension were 0.770 and 0.539 respectively, while the factor loads of context factor and emotional factor in the elaboration dimension were 0.911 and 0.800 respectively. The questionnaire had good internal consistency reliability (Cronbach α=0.870), structural validity (χ 2/ df=3.550, RMSEA=0.053, GFI=0.944, CFI=0.970, NFI=0.958, IFI=0.970) and criterion validity ( r(OGMQ, AMT)=0.870, r(OGMQ, BDI)=0.507), and it was able to effectively distinguish between depressed and normal groups (AUC=0.795). Conclusion:The OGMQ has satisfactory validity and reliability, and meets the needs of psychometrics, which can be used in depression-related research and practice.

ABSTRACT

PURPOSE: Long non-coding RNA taurine upregulated gene 1 (TUG1) is reported to be a vital regulator of the progression of various cancers. This study aimed to explore the exact roles and molecular mechanisms of TUG1 in osteosarcoma (OS) development. MATERIALS AND METHODS: Real-time quantitative PCR was applied to detect the expressions of TUG1 and microRNA-132-3p (miR-132-3p) in OS tissues and cells. Western blot was performed to measure protein levels of sex determining region Y-box 4 (SOX4). Cell viability was assessed using XTT assay. Cell apoptosis was evaluated using flow cytometry and caspase-3 activity detection assays. Bioinformatics analysis and luciferase reporter experiments were employed to confirm relationships among TUG1, miR-132-3p, and SOX4. RESULTS: TUG1 was highly expressed in human OS tissues, OS cell lines, and primary OS cells. TUG1 knockdown hindered proliferation and induced apoptosis in human OS cell lines and primary OS cells. Moreover, TUG1 inhibited miR-132-3p expression by direct interaction, and introduction of miR-132-3p inhibitor partly abrogated the effect of TUG1 knockdown on the proliferation and apoptosis of OS cells. Furthermore, SOX4 was validated as a target of miR-132-3p. Further functional analyses revealed that miR-132-3p inhibited proliferation and induced apoptosis of OS cells, while this effect was greatly abated following SOX4 overexpression. Moreover, TUG1 knockdown suppressed proliferation and promoted apoptosis by upregulating miR-132-3p and downregulating SOX4 in primary OS cells. CONCLUSION: TUG1 facilitated proliferation and suppressed apoptosis by regulating the miR-132-3p/SOX4 axis in human OS cell lines and primary OS cells. This finding provides a potential target for OS therapy.

ABSTRACT

Objective To examine the adaptability of Carver and White' s behavioral activation/in-hibition scales(BIS/BAS Scales)in a large sample of Chinese military-man,and to revise the instrument and validate its factor structure. Methods A total of 2 921 Chinese military-men completed four subscales:one for the Behavioral Inhibition System, and three for the Behavioral Approach System(drive, reward re-sponsiveness,fun seeking).Data were analyzed using exploratory and confirmatory factor analysis. The internal consistency of the subscales and the complete scale were examined. Results The findings did not support Carver and White's 4-factor structure which was validated in a sample of Chinese college students.A 3-factor structure(behavioral inhibition, reward responsiveness, drive)in Chinese military-man were extracted and validated using confirmatory factor analysis(CFA)(KMO=0.87,CFI=0.94,RMSEA=0.06).The Cronbach α of behavioral inhibition, drive and reward responsiveness were 0.78,0.71 and 0.67 respectively. The internal consistencies of behavioral inhibition and drive were higher than four-factor model of Chinese college students and the internal consistencies of drive and reward responsiveness were less than American young adults.Be-havioral inhibition and drive positively predicted depressive symptoms(βBIS=0.332,βDrive=0.076,P<0.001),and reward responsiveness negatively predicted depressive symptoms(βRRS=-0.140,P<0.001). Conclusion The BIS/BAS Scales(revised Chinese military version)has acceptable reliability and validi-ty,and can be utilized as a measurement tool for assessing the approach/avoidance motivation of Chinese ser-vicemen.

ABSTRACT

Objective To explore the relationship between WT1 and prognosis of patients with acute myeloid leukemia (AML) or acute lymphoblastic leukemia (ALL), and to evaluate the possibility of WT1 as a potential marker for monitoring the minimal residual disease (MRD). Methods Bone marrow mononuclear cells from 58 patients with primary AML, 32 patients with primary ALL, 40 patients with AML-complete remission (CR), 28 patients with ALL-CR and 31 patients with trilineage hyperplasia (control group) were collected. Real-time fluorescent quantitative PCR method was used to detect the expression of WT1 in all patients. The expression threshold of WT1 in each group was established. WT1 copy number/ABL copy number ratio×100%denotes the relative expression level of WT1 gene. Results Median relative expression level of WT1 in the control patients was much lower than that in primary AML patients [0.026%(0-0.240%) vs. 20.880 % (3.550 %-48.500 %), Z=-7.74, P20.880 %) was 60.7 % (17/28), while the CR rate was 76.7 % (23/30) in those with lower WT1 expression. WT1 expression was increased dramatically in recurrent AML patients. Relative expression level of WT1 was significantly higher in primary ALL patients [0.350 % (0.021 %-10.780 %)] compared with that in the control group Z=-2.58, P<0.05. There was no significant difference in relative expression level of WT1 between ALL and ALL-CR patients [0.038 % (0-2.800 %), P=0.065]. Conclusion WT1 expression level in AML patients is relatively high, which could be used as an effective index of prognosis evaluation and MRD monitoring for AML patients, but not for ALL patients.

ABSTRACT

Objective:To translate the Attention to Positive and Negative Inventory(APNI)and analyze the validity and reliability in Chinese undergraduates sample,to offer a convenient and reliable tool of measuring the cognitive bias for national researchers. Methods:The English-version APNI went through translation into Chinese, retroversion into English, translation into Chinese again, and revision several stages. Two parts of samples (1450 Chinese college students)were surveyed. Sample one (n=1000)was used for item analysis, exploratory factor a-nalysis (EFA),concurrent validity and reliability analysis, while sample 2 (n=450)was used for confirmatory fac-tor analysis (CFA). Totally 68 subjects of sample 1 were randomly chosen and resurveyed with an interval of one week. Beck depression inventory (BDI-II)and patient health questionnaire (PHQ-9)was used for concurrent validi-ty. Results:Item analysis indicated that the 22 items of Chinese APNI had good discriminability. EFA focused onattention to positive information(API)and attention to negative information(ANI)two factors. CFA showed good model fit (χ2 =1376,RMESA=0. 09,CFI=0. 94). Concurrent validity result showed that the total scores of BDI-II and PHQ-9 was negatively correlated with total scores of API (r=-0. 24,-0. 29,Ps<0. 01 ),and posi-tively correlated with total scores of ANI (r=0. 36,0. 31,Ps<0. 01). The Cronbach'αcoefficients of API and ANI sub-scale were 0. 86 and 0. 82,while the retest reliability coefficients were 0. 79 and 0. 62. Conclusion:It suggests that the Chinese APNI has good validity and reliability in a sample of college students, which could be used to eval-uate the cognitive bias of Chinese college students.
