ABSTRACT

ObjectiveTo explore effect of Huanglian Jiedutang (HLJDT) on autophagy-related protein expression in septic mice with liver injury induced by cecal ligation and puncture (CLP). MethodSixty eight-week-old C57BL/6 mice were randomly divided into four groups, namely, the sham operation group, model group, and low- (1.44 g∙kg-1) and high-dose (2.88 g∙kg-1) HLJDT groups, with 15 in each group. The septic model was established by CLP after the last administration of HLJDT for three successive days. The survival rate of mice with 24 h was observed. The mice were sacrificed 12 h after operation for collecting the serum and liver tissue. The levels of serum interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α), and interleukin-1β (IL-1β) were detected by enzyme-linked immunosorbent assay (ELISA), and the levels of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) in serum by biochemical method. The pathological changes in liver tissue were observed by hematoxylin-eosin (HE) staining, and the apoptosis index (AI) of hepatocytes by TdT-mediated dUTP-biotin nick end-labeling (TUNEL). The expression levels of protein high-mobility group box 1 protein (HMGB1), Beclin1, and microtubule-associated protein 1 light chain 3 (LC3)-Ⅱ/Ⅰ in the liver tissue were assayed by Western blot. ResultCompared with the sham operation group, the model group showed reduced survival rate at 12 and 24 h, elevated IL-6, TNF-α, and IL-1β levels, enhanced AST and ALT activities (P<0.05), hepatocyte swelling, inflammatory cell infiltration, and apoptosis, and up-regulated HMGB1 (P<0.05), Beclin1, and LC3-Ⅱ/Ⅰ (P<0.05). Compared with the model group, each medication group exhibited increased survival rate at 12 and 24 h, lowered IL-6 and TNF-α levels, weakened AST and ALT activities (P<0.05), alleviated liver injury and apoptosis (P<0.05), down-regulated HMGB1 expression ( P<0.05), and up-regulated Beclin1 and LC3-Ⅱ/Ⅰ (P<0.05). ConclusionHLJDT alleviates the liver injury of septic mice possibly by inducing autophagy and inhibiting apoptosis.

ABSTRACT

AIM To investigate the dose-proportion relation of Tibetan medicine Siwei Jianghuang Prescription (SJP) for protective effects on diabetic nephropathy (DN),and the underlying mechanism.METHODS Diabetes mellitus rats induced by intraperitoneal injection of streptozotocin (STZ) (60 mg/kg) were randomly divided into model group,metformin support group,and eight SJP groups with dose-proportion variation (with reference to the uniform design method) for corresponding drug administration once a day,for four weeks.Measurement of fasting blood glucose (FBG) by a blood glucose meter,the concentrations of blood urea nitrogen (BUN),uric acid (UA),serum creatinine (SCr) and total protein (TP) by chemical methods,serum transforming growth factor-β1 (TGF-β1) and vascular endothelial growth factor (VEGF) by ELISA kits were conducted,the pathological morphology observation and glomerular basement membrane thickness detection by electron microscope were accomplished as well.Principal components analysis (PCA) and multivariate progressive regression analysis (MSRA) were employed to analyze the relationship between the dose-proportion to pharmacodynamics.RESULTS The resultant indexes revealed variant pharmacological improvement in each treatment group.MSRA results showed that the levels of BUN,renal index,FBG,glomerular basement membrane thickness,VEGF,Scr,and UA had correlative relations with a multiple linear or a multiple non-linear in all groups,which regression equation had a statistical significance (P < 0.05);TGF-β1 level and total protein index with the dose-proportion had no linear or non-linear relation,which the regression equation statistical showed non-significance (P > 0.05).In the global optimization comparison around the range of uniform design,the optimal dosage of the rats model was Curcumae Longae Rhizoma ∶ Berberidis dictyophyllae Cortex ∶ Phyllanthi Fructus ∶ Tribuli Fructus =1 ∶ 2 ∶ 1 ∶ 2.CONCLUSION Siwei Jianghuang Prescription shows better therapeutic effects on DN,which may be related to reducing the levels of BUN,renal index,FBG,glomerular basement membrane thickness,VEGF,Scr and UA.

ABSTRACT

BACKGROUND: Umbilical cord mesenchymal stem cells (UC-MSCs) are a group of cells that have self-renewal, highly proliferative and multidrug differentiation potential. The properties of UC-MSCs and their tumor tropism make them an ideal tool for glioma cell therapy. These cells can act by paracrine or as a delivery system for genes and drugs. It has been demonstrated that UC-MSCs can inhibit the growth of glioma and improve the survival after transplantation into the brain. OBJECTIVE: To summarize the molecular mechanisms and safety of UC-MSCs in the treatment of glioma and to provide a useful reference for further research. METHODS: We searched the PubMed and CNKI databases from 2000 to 2017 with the English terms of "glioma; umbilical cord mesenchymal stem cells" and the Chinese terms of "glioma; umbilical cord mesenchymal stem cells; safety; molecular mechanism". Based on the inclusion and exclusion criteria, 55 articles were finally reserved for review. RESULTS AND CONCLUSION: UC-MSCs have obvious effect on treating glioma. These cells can treat glioma through homing mechanism and paracrine mechanism as gene carrier and co-culture. Moreover, UC-MSCs have certain safety in the treatment of glioma.

ABSTRACT

Objective To detect hs‐cTnT and cTnI for exploring their application value in the early diagnosis of acute myocardial infarction(AMI) .Methods From October 2015 to March 2016 ,100 patients with AMI in the cardiology department of our hospital were selected as the observation group(AMI group) and contemporaneous 100 adults undergoing physical examination as the con‐trol group .The chemiluminescence immunoassay was used to detect the levels of hs‐cTnT and cTnI .The positive rate ,sensitivity and specificity of the hs‐cTnT and cTnI in the AMI group for early diagnosis of AMI were compared .Results The level of hs‐cT‐nT in the AMI group was (4 .89 ± 1 .83)ng/mL ,which was higher than that in the healthy population by 99 percentile value 0 .014 ng/mL ;the cTnI level in the AMI group was (28 .82 ± 12 .32)ng/mL ,which was higher than that in the healthy population by the upper limit of normal reference value 0 .4 ng/mL ,both of them were significantly higher than those in the control group ( P<0 .05) ,the positive rate and sensitivity of hs‐cTnT was both 92 .0% ,which was significantly higher than 79 .0% of cTnI both(P<0 .05) ,the specificity of cTnI was 96 .0% ,which was significantly higher than 76 .0% of hs‐cTnT(P<0 .05) .Conclusion The pos‐itive rate and the sensitivity of hs‐cTnT are high ,the specificity of cTnI is high ,their joint detection has an important significance for the early diagnosis of AMI .

ABSTRACT

Objective To explore the application value of serum procalcitonin (PCT ) and amyloid protein A (SAA ) in the early diagnosis and curative evaluation of bacterial infectious diseases among preschool children in order to improve the clinical diagnosis level of early bacterial infection .Methods A total of 120 cases of pediatric infectious diseases in our hospital from January to De‐cember 2015 were selected and divided into the bacterial infection group (60 cases) and viral infection group (60 cases) ,meanwhile 50 children undergoing the physical examination were selected as the control group .The serum levels of PCT and SAA were ob‐served and compared among the three groups ,and the statistical analysis was performed by using the SPSS 19 .0 software .Results The levels of PCT and SAA in the bacterial infection group were significantly increased compared with the viral infection group and the healthy control group(P<0 .05) ,both PCT and SAA levels after effective antibiotic treatment were significantly decreased (P<0 .05);the positive rates of PCT and SAA in the bacterial infection group were 91 .7% and 95 .0% respectively ,which were signifi‐cantly higher than those in the viral infection group and the healthy control group (P<0 .05) ,the sensitivity of PCT in the diagno‐sis of bacterial infection was lower than that of SAA ,but the specificity ,positive predictive value and negative predictive value were all higher than those of SAA ,the correlation analysis showed that PCT level in the bacterial infection group was positively correla‐ted with and the SAA level(r=0 .782 ,P<0 .01) .Conclusion Detecting the PCT and SAA levels is conducive to early diagnosis , judgment of the disease condition and guidance of rational medication in bacterial infection among preschool children ,in which the efficiency of PCT for diagnosing bacterial infection is superior to SAA .

ABSTRACT

Objective To investigate the diagnostic value of combined detection of neutrophil CD64 index and C reactive protein (CRP) levels in newborn bacterial infectious diseases .Methods The retrospective analysis was performed on 139 neonates admitted to the neonatal department of our hospital from January to December 2015 ,including 72 cases in the bacterial infection group and 67 cases in the viral infection group ,and 50 healthy neonates were selected as the control group .The levels of CD64 and CRP and the positive rates were compared among 3 groups and analyzed .The sensitivity ,specificity ,positive predictive value and negative predictive value of CD64 and CRP in the diagnosis of bacterial infection were analyzed .Results The CD64 index and CRP levels had statistical difference between the bacterial infection with the viral infection group and normal control group (P<0 .05) ,the CD64 index and CRP levels after treatment in the bacterial infection group were significantly decreased ,the differences were statistieally significant(P<0 .05) .The sensitivity ,specificity ,positive predictive value and negative predictive value of CD64 index in the diag‐nosis of bacterial infection were 91 .7% ,92 .5% ,93 .0% and 91 .2% respectively ,which were significantly higher than those of CRP ,the differences were statistically significant (P< 0 .05).CD64 index was positively correlated with CRP (r= 0 .781 ,P<0 .01) .Conclusion The combined detection of CD64 index and CRP level helps to early diagnosis ,differential diagnosis and the therapeutic effect evaluation of bacterial infection in neonates ,moreover the diagnostic efficiency of CD64 is obviously better than that of CRP .

ABSTRACT

<p><b>BACKGROUND</b>It has not been solved what kind of needle sensation might influence outcomes of acupuncture treatment. Effects of personality factors on the therapeutic effect of acupuncture have not been investigated. This study aimed to find the effects of the traits of personality on the objective outcome when different acupuncture techniques were used in treating patients with Bell's palsy.</p><p><b>METHODS</b>We performed a secondary analysis of a prospective multicenter randomized controlled trial of acupuncture for Bell's palsy. Patients were randomly assigned to the de qi and control groups, respectively. The primary outcome was facial nerve function at month 6. The intensity of each needle sensation was rated by a visual analog scale. Psychosocial factors were assessed by the pretreatment mediator questionnaire; 16 Personality Factor Questionnaire (16PF) was used for assessing personality factors and digit cancellation test for assessing attention.</p><p><b>RESULTS</b>After 6 months, patients in the de qi group had better facial function (adjusted odds ratio [OR]: 4.16, 95% confidence interval [CI]: 2.23-7.78). Path analysis showed that intensity of needle sensation of fullness had direct effect on House-Brackmann (HB) score at month 6. In de qi group, the low HB score on day 1 (OR: 0.13, 95% CI: 0.03-0.45) and the low Social Boldness score (OR: 0.63, 95% CI: 0.41-0.97) in 16PF were associated with better facial function. In control group, low HB score on day 1 (OR: 0.25, 95% CI: 0.13-0.50), low Vigilance score (OR: 0.66, 95% CI: 0.50-0.88), and high Tension score (OR: 1.41, 95% CI: 1.12-1.77) in 16PF were related to better facial function.</p><p><b>CONCLUSIONS</b>The needle sensation of fullness could predict better facial function and personality traits might influence outcomes of acupuncture treatment. Both of them should be considered seriously in acupuncture treatment and research.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the internal fixation methods in the two different surgical approach to the unstable distal radius fractures.</p><p><b>METHODS</b>From August 2006 to October 2010,47 patients with distal radius fractures were treated with open reduction and plate internal fixation. Among the patients, 21 patients were male and 26 patients were female,ranging in age from 39 to 73 years old. All the patients were divided into two groups: 32 patients were treated with plate fixation at the palm side with palm side incision (group A),including locking plate used in 27 patients and ordinary T shape plate used in 5 patients; 15 patients were treated with dorsal plate fixation (group B), including locking plate used in 7 patients and ordinpry T shape plate used in 8 patients. The postoperative wrist function, X-ray score,complications were compared between the two groups.</p><p><b>RESULTS</b>Compare to preoperative, postoperative volar tilt,ulnar deviation and radial styloid height of both groups improved; and comparison between 1 week after operation and 6 months after operation,volar tilt,ulnar deviation and radial styloid height of both groups had no obvious loss. In group A, 1 patient had tension injury of median nerve, 1 patient had incision infection, 2 patients had adhesion of tendon; and in group B,1 patient had incision swelling and infection, 1 patient had adhesion of tendon,3 patients had stimulation of extensor pollicis longus muscle tendon. The complication occurrence rate of group A is lower than that of group B. The wrist functional assessment: 17 patients got an excellent results, 11 good, 3 poor and 1 bad in group A; and 8 excellent, 4 good, 2 poor and 1 bad in group B. There was no statistically significant difference of wrist function between the two groups.</p><p><b>CONCLUSION</b>Plate fixation at the palm side for the treatment of unstable distal radius fractures can also achieve a good reduction,and has less complication such as tendon damage than that of dorsal plate fixation.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To analyze the relation between recurrent miscarriage and routine semen parameters.</p><p><b>METHODS</b>We compared the semen parameters of normal healthy men with those of the spouses of recurrent miscarriage women through 1: 1 age-matched case-control study.</p><p><b>RESULTS</b>Compared with the healthy controls, the men of the case group showed a significantly lower mean semen volume ([1.95 +/- 1.11] ml vs [2.74 +/- 1.43] ml), sperm concentration ([48.68 +/- 20.07] x 10(6)/ml vs [59.26 +/- 25.35] x 10(6)/ml), percentage of grade b sperm ([12.07 +/- 3.34] % vs [16.18 +/- 6.74] %), fruit-sugar content ([1.73 +/- 0.64] g/L vs [2.21 +/- 0.75] g/L), acrosomal enzyme activity ([84.34 +/- 26.69] U/mg prot vs [94.20 +/- 26.35] U/mg prot), alpha-glucuronidase (alpha-GLU) content ([36.28 +/- 15.98] U/ml vs [44.45 +/- 12. 54] U/ml), and acid phosphatase (ACP) content ([68.55 +/- 35.45] U/ml vs [84.78 +/- 51. 10] U/ml) (P < 0.05), but remarkably higher percentages of head teratospermia ([47.36 +/- 4.59] % vs [46.50 +/- 6.32] %) and tail teratospermia ([7.56 +/- 2.27] % vs [7.28 +/- 3.10] %), and elastase content ([885.64 +/- 1 272.30] ng/ml vs [661.08 +/- 764.64] ng/ml) (P < 0.05). Based on the results of discriminant analysis, the semen volume, percentages of grade b sperm and combined teratospermia, and contents of fruit-sugar, alpha-GLU and ACP could be used to evaluate the semen and sperm quality of the spouses of recurrent miscarriage women.</p><p><b>CONCLUSION</b>Routine semen and sperm tests might help evaluate the seminal factors of recurrent miscarriage, but they lack specificity and need comprehensive analysis. Poorer semen quality is associated with higher incidence of recurrent miscarriage.</p>

ABSTRACT

Objective To investigate the status of endemic fluorosis in Boxing County in Shandong Province at present,and to provide the scientific evidence for making strategies in prevention and control.Methods Children aged 8-12 years old and adults above 30 years old were selected from 8 endemic fluorosis villages in 2 fluorosis of children aged 8-12 years old were diagnosed by Dean method and skeletal fluomsis diagnosed by clinic and X-Rays.Results Eight villages in 2 towns were chosen underwent epidemiological investigation.Eight villages had water fluoride content>4.50 mg/L.the highest water fluoride content was 5.78 mg/L.The total rate of dental fluorosis of children aged 8-12 years old WaS 90.70%(195/215),the index of dental fluorosis was 2.15 and the rate of dental damage was 24.65%(53/215).The rate of skeletal fluorosis detected by clinic and X-rays in adults older than 30 years old were 30.71%(78/254)and 16.54%(42/254),respectively.The averaged fuoride level in urine wa8 over 1.50 mg/L in 98.95%(189/191)of children aged 8-12 years old and in 97.92%(235/240)adults older than 30 years old,with the highest respectively being 14.50 mg/L and 17.99 mg/L.Conclusions In Boxing County in Shandong Province,endemic fluorosis is not effectively controlled.So endemic fluorosis control mfist be strengthened.

ABSTRACT

Objective To study the epidemiological characteristics of measles in Fuyong Street in order to predict its occurrence and reduce its incidence by establishing preventive and control measures.Methods Epidemiological data in Fuyong Street between 2003 and 2007 were analyzed.Results The result showed that 393 measles cases were reported in the whole street during 2003 and 2007,329 were confirmed by laboratory,and 60 were local cases.The measles annual average incidence was 18.29/100 000.The high incidence period was from March to August and the most measles cases were young adults and infants.30.79% cases had no immunization history and 24.68% cases had immunization history.Conclusions The incidence of measles is kept at a high level.It will go on possibly for half a year or a year.A comprehensive control measures should be thaken to strengthen vaccination program especially for foreign people and to reinforce the surveillance and analysis of epidemic situation in order to avoid the outbreak of the disease.

ABSTRACT

<p><b>OBJECTIVE</b>To study the infection of human cytomegalovirus (HCMV) and herpes simplex virus type II (HSV-I) and the morphological characteristics of the infected spermatogenic cells in the semen of infertile men.</p><p><b>METHODS</b>We washed and concentrated the spermatogenic cells obtained from 83 semen samples of infertile men, extracted DNA and then screened HCMV and HSV-II by polymerase chain reaction (PCR). Immunocytochemistry (ICC) was used to detect the expression of correlative virus antigens of the positive semen cells, and the cytology smear was employed to observe the morphological changes of the spermatogenic cells under the microscope after cytology staining.</p><p><b>RESULTS</b>Of all the semen samples, 8 were HCMV positive, 4 HSV-II positive, but none were both HCMV and HSV-II positive. HCMV late antigens were positively and HCMV early antigens negatively expressed in the spermatogenic cells of the 8 HCMV positive cases. In the 4 HSV-II positive cases, 3 were positively and 1 weakly positively expressed. In the semen of the 12 positive cases were found large numbers of immature spermatogenic cells, with different manifestations of apoptosis, such as chromatin pycnosis, vacuoles, damaged nuclear membrane, and apoptotic bodies, but without virus infection-induced specific morphological alteration. Sperm concentration of the positive group was significantly lower than that of the negative (P < 0. 05).</p><p><b>CONCLUSION</b>Spermatogenic cells infected by HCMV and HSV-II may cause pathologic lesions and affect spermatogenesis. Morphologically, the infected spermatogenic cells may undergo some pathologic alteration, such as apoptosis. The rate of HCMV infection is higher among infertile males with pathologic cells in the semen.</p>

ABSTRACT

The results of isolation,screening and identification of Bacillus subtilis strains which produced fibronolysion were reported in this paper.20 Bacillus subtilis strains were screened from different withered straw samples.Among them,FM-S1,FM-S2,FM-S6,FM-S8 and FM-S11 produce fibronolysin with higher activity.According to morphological,physiological and biochemical characters,the screened strains were confirmed to be Bacillus subtilis. For FM-S2,the production of fibronolysin type in solid fermentation was considered to be synchronous with growth.
