ABSTRACT

Objective To observe the expression levels of fork head box protein N2 (FOXN2) and fork head box protein F2 (FOXF2) in normal brain tissue, low-grade glioma and high-grade glioma, and to explore the relationship between the two and the grade of glioma, and then deduce their roles in the occurrence and development of glioma, to look for possibilities target of drug therapy. Methods From January 2016 to December 2020, 36 glioma specimens were collected from the Department of Pathology, Yijishan Hospital, Wannan Medical College. The expressions of F0XN2 and F0XF2 were detected by immunohistochemistry and immunofluorescence with the double-blind method, and analyzed by SPSS18. 0 software, with a statistical significance of P<0. 05. Then Western blotting and Real-time PCR experiments were carried out on fresh normal brain tissues, low-grade glioma tissues, high-grade glioma tissues during neurosurgery in Yijishan Hospital of Wannan Medical College from January 2020 to January 2021 (normal brain tissues were all trauma patients). Results The results of immunohistochemistry and immunofluorescence showed that the higher grade of glioma, the lower expressions of FOXN2 and FOXF2 would be (P<0. 05). Western blotting showed that the expressions of FOXN2 and FOXF2 in normal brain tissue and low-grade glioma were higher than those in high-grade glioma (P<0. 05). The result of Real-time PCR showed that the expressions of FOXN2 and FOXF2 in normal brain tissue were higher than those in high-grade glioma (P<0. 01). Conclusion FOXN2 and FOXF2 are expressed in normal brain tissues, and their expression is low in glioma in a grade-dependent manner, suggesting that FOXN2 and FOXF2 are related to the grade and poor prognosis of glioma. Enhancing the expression of F0XN2 and F0XF2 ma)' provide a new idea for target therapy of glioma.

ABSTRACT

Fifteen patients with Crohn′s disease (CD) in remission diagnosed at Shanghai Jing′an District Central Hospital from February 2018 to June 2019, and 26 matched healthy subjects were recruited. All participants underwent resting-state functional magnetic resonance imaging (fMRI) scans of hippocampus. The amplitude of low-frequency fluctuations (ALFF) was calculated to determine differences in the brain. Left hippocampus was selected as seeds for functional connectivity (FC) analysis, and the results were compared between two groups. The relationship between disease duration and ALFF/FC values in abnormal regions were analyzed with Pearson correlation. Compared with the controls, the ALFF of the left hippocampus (voxel size 32) of CD patients decreased [family-wise error correstion(FWE correction), cluster level P<0.05], and the ALFF of the left medial superior frontal gyrus (voxel size 126), left supplementary motor area (voxel size 126) and left anterior cingulate gyrus increased (voxel size 37) (FWE corrected, cluster level P<0.05). Using the left hippocampus as the seed point for the whole brain functional connectivity analysis, CD patients showed increased FC strength with the left superior temporal gyrus, left medial superior frontal gyrus, left inferior frontal gyrus (opercular part), and right supplementary motor area(FEW corrected, cluster level P<0.05). Correlation analysis did not show a significantly differences between ALFF/FC value of altered brain areas and the disease duration. It suggests that there are changes in spontaneous activities and functional connectivity in the left hippocampus in patients with CD.

ABSTRACT

Objective To study the role of salvianolate in the treatment of inflammation of intestinal mucosa by colitis mice model.Methods A total of 60 C57BL/6J mice were divided into acute control group,acute model group,acute interventional group,chronic control group,chronic model group and chronic interventional group with 10 mice in each group.Dextran sodium sulfate (DSS) was used to establish the model.Meanwhile,the mice of interventional group received salvianolate peritoneal injection during modeling.The expression levels of NLR family pyrin domain containing 3 (NLRP3),apoptosis-assoeiated speck-like protein containing CARD (ACS) and cysteinyl aspartate specific proteinase 1 (caspase-1) were detected.And the absorbances of serum superoxide dismutase (SOD) and malonaldehyde (MDA) in mice serum were determined.Kruskal Wallis single factor analysis of variance,single factor analysis of variance,Student-Newman-Keuls (SNK) method and least-significant difference method were performed for statistical analysis.Results The expressions of NLRP3 at protein level of acute control group,acute model group,acute interventional group,chronic control group,chronic model group and chronic interventional group were 9 965.20 (196.81),16 703.38 (291.21),13 423.74 (209.28),10 112.01 (183.55),16 247.90 (505.44) and 12 674.95 (229.32),respectively.The expressions of NLRP3 of acute and chronic interventional group were both lower than those of acute and chronic model group,and the differences were statistically significant (both x2 =15.158,P<0.01).The expressions of ACS at protein level were 9 744.09-±-546.58,16 555.44 ± 407.80,12 568.21±586.49,10066.32±435.63,14 911.73±384.51 and 13 751.37±322.30,respectively.The expressions of caspase-1 at protein level were 5 300.40±843.14,15 478.98± 174.09,10 587.46±821.60,5 517.28±876.16,13 164.01 ±416.47 and 8 856.27 ± 545.01,respectively.The expressions of ACS and caspase-1 of acute and chronic interventional groups were both lower than those of model groups,and the differences were statistically significant (F=260.56,329.36,240.38 and 236.26,all P<0.01).The serum levels of SOD were (282.08±28.24),(221.87±5.81),(231.45±1.84),(267.08±43.63),(237.09±34.09) and (257.78 ± 4.68) U/mL,respectively.There was no significant difference between acute model group and acute interventional group (P>0.05).SOD level of chronic interventional group was higher than that of model group,and the difference was statistically significant (t=-2.65,P =0.021).The serum levels of MDAwere (1.31±0.40),(6.95±1.57),(2.98±1.41),(1.21±0.16),(9.38±3.73)and (3.74±0.81) nmol/L,respectively.MDA levels of acute and chronic interventional groups were lower than those of acute and chronic model groups,and the differences were statistically significant (t=4.88 and 4.02,both P<0.01).Conclusion Salvianolate may alleviate oxidative stress response by adjusting the activity of NLRP3 inflammasome which takes part in reducing intestinal inflammation.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the growth and development of brain derived neurophic factor(BDNF)-positive neurons in the frontal lobe of human fetus.</p><p><b>METHODS</b>The expression of the BDNF-positive neurons in the frontal lobe of human fetus in the 2(nd),3(rd),and 4(th) month of gestation were observed with the streptavidin-biotin-complex/immunoperoxidase(SABC)method.</p><p><b>RESULTS</b>By the second month of gestation,BDNF-positive neurons were seen in the subventricular layer of the frontal lobe of cerebellum.By the third month of gestation,BDNF-positive neurons in the central layer were in various shapes,with big nucleus,less cytoplasm,and small processes.By the fourth month of gestation,BDNF-positive neurons in the central layer grew larger in size,cytoplasm increased,the BDNF-positive expression was enhanced with deeper dyeing,and the nerve fibers and particles were distributed between neurons;also,the BDNF-positive neurons were seen in the marginal layer of the frontal lobe of cerebrum.</p><p><b>CONCLUSION</b>BDNF-positive neurons may participate in the early development of the frontal lobe of cerebrum of human fetus.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the growth and development of nerve growth factor (NGF)-positive neurons in the cerebellum of midanaphase human fetus.</p><p><b>METHODS</b>The expression of the NGF-positive neurons in the cerebrum of human fetus was observed by immunohistochemical methods, and the integral absorbance (IA) was detected.</p><p><b>RESULTS</b>By the 3rd to 4th month of gestation, neurons was seen in the ependymal, central, and marginal plate of cerebellum; the nucleus was oval and the neurons had short and small processes. By the 5th to 7th month of gestation, the number of NGF-positive neurons increased, the expressions enhanced, the nucleus was round-, oval-, or fusiform-shaped, the neurons grew larger in size, and the Purkinje cells showed NGF-positive expression. By the 8th to 10th month of gestation, the NGF-positive expression was enhanced with deeper dying, the body of Purkinje cells grew larger gradually, and the number of NGF-positive neurons in the granular cell layer and molecular layer increased. IA of the cerebellar cortical neurons of the 3rd, 4th, 5th, 6th, 7th, and 8th month of gestation showed an increasing trend, and significant difference was observed (P < 0. 05).</p><p><b>CONCLUSION</b>NGF-positive neurons in the cerebellum play an important role for differentiation, proliferation, migration, and growth of neurons in the cerebellum.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the influential factors for the prognosis in patients with paraquat poisoning.</p><p><b>METHODS</b>Retrospective analysis was performed on 40 patients with acute paraquat poisoning to evaluate serum urea nitrogen, creatinine, alanine aminotransferase, and aspartate aminotransferase levels and organ injuries.</p><p><b>RESULTS</b>Among the patients, 21 had lung injury, 26 had kidney injury, 13 had multiple organ dysfunction syndrome, and 12 died. The factors associated with lung injury were kidney injury and serum urea nitrogen and creatinine levels within 24 hours after paraquat poisoning.</p><p><b>CONCLUSION</b>The renal function in early stage of paraquat poisoning is related to lung injury and thus can be used asa predictor for the incidence of lung injury.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To assess the effect of electroacupuncture in different frequencies by electromyography and walking function measure in post-stroke patients with lower-extremity (OLE) spasticity and hemiparesis.</p><p><b>METHODS</b>Fifty cases of post-stroke whose motor deficit was classified into Brunnstrom stage II - IV were randomly divided into a 100 Hz group, a 50 Hz group and a 2 Hz group. They were accepted 100 Hz, 50 Hz or 2 Hz of electroacupuncture (EA) therapy combined with standard rehabilitation program. Main outcome measures included integrated electromyography (IEMG) score during maximum isometric voluntary contraction (MIVC) of the knee flexors and extensors, ankle dorsiflexors and planterflexors in the affected LE recorded by surface EMG, Co-contraction ratio calculated by IEMG score of the antagonist over that of the agonist plus antagonist, Composite Spasticity Scale (CSS), Fugl-Meyer Motor Scale (FMS) and Functional Ambulation Categories (FAC) on LE. All outcomes were assessed at the baseline and after treatment by the professional practitioners who blinded to the treatment.</p><p><b>RESULTS</b>After EA treatment, IEMG of rectus femoris were decreased in 100 Hz and 50 Hz groups (P < 0.01, P < 0.05), and better than that in 2 Hz group (both P < 0.05); gastrocnemius IEMG were decreased in 100 Hz and 50 Hz groups (P < 0.05, P < 0.01); but IEMG of tibialis anterior muscle was increased only in 50 Hz group (P < 0.05). During knee flexion, EMG co-contraction ratio in MIVC declined in 100 Hz and 50 Hz groups were decreased significantly (P < 0.05, P < 0.01), and the co-contraction ratio between the non-affected and affected side were increased significantly in all the 3 groups after treatment (P < 0.01, P < 0.05). During ankle dorsiflexion, co-contraction ratio were decreased significantly in all the 3 groups (P < 0.05, P < 0.01), and cocontraction ratio between the non-affected and affected side was increased significantly only in 100 Hz after treatment (P < 0.01). FMS score, CSS and FAC were improved in all the 3 groups after treatment (all P < 0.01), but only FAC in 100 Hz group showed better effect than that in 50 Hz group or 2 Hz group (both P < 0.05).</p><p><b>CONCLUSION</b>Electroacupuncture therapy combined with rehabilitation program is effective for the spasticity status of lower-extremity in post-stroke. The therapeutic effect of EA in the frequencies of 100 Hz or 50 Hz is superior to that of 2 Hz stimulation and parameter of 100 Hz may be optimal.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the growth and development of nitric oxide synthase (NOS)-positive neurons in the cerebellum of human fetus in the midanaphase.</p><p><b>METHOD</b>The positive expression of the NOS-positive neurons in the cerebellum of midanaphase human fetus was observed by immunohistochemistry.</p><p><b>RESULTS</b>By the sixth to seventh month of gestation, NOS-positive neurons were seen in the ependymal layer of the cerebellum. The nucleus was oval-shaped and the neurons had short and small processes. By the eighth to ninth month, NOS-positive neurons were found in the central layer of the cerebellum and the nucleus was round-, oval-, or fusiform-shaped; meanwhile, the neurons grew larger in size with richer cytoplast and heavier staining. The beaded nerve fibers reached the marginal layer and the layer became thickened on the tenth month, which generally was composed of 5 to 6 layers of NOS-positive neurons that were tightly aligned. Some NOS-positive neurons were in smaller size with the cell body and the nerve fibers grew well.</p><p><b>CONCLUSION</b>Nitric oxide generated by NOS of the NOS-positive neurons in the cerebellum plays an important role in the differentiation, proliferation, and migration of neurons and gliacytes.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the development of nitric oxide synthase (NOS)-positive neurons in the frontal lobe of the cerebrum of human fetus in midanaphase.</p><p><b>METHODS</b>The positive expression of the NOS-positive neurons in the frontal lobe of cerebrum of human fetus was observed by immunohistochemistry.</p><p><b>RESULTS</b>By the 7th to 8th month of gestation, NOS-positive neurons in the cortical plate of frontal lobe demonstrated themselves inequality of sizes and morphological difference in the deeper layers with interspersed distribution and increased NOS response, and the distribution of beaded nerve fiber was observed between neurons of cerebral tissues. By the 9th to 10th month of gestation, NOS-positive neurons in the deeper layers of cortical plate of frontal lobe developed slightly in size of the cell body with richer cytoplast, full shape and deeper dyeing and extrusive beaded nerve fibers, and the NOS-positive neurons scattered in the shallow layer of cortical plate presented with round or oval shape. The nucleus developed bigger but with sparse cytoplasm and clear nerve process.</p><p><b>CONCLUSION</b>NOS-positive neurons in the deeper layer of cortical plate of lobus frontal consist of largely network of neural system and produce micro-environment with higher concentration of NO, which favors the differentiation, proliferation, migration, and development of various neurons.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore the factors associated with retention in a community-based methadone maintenance treatment(MMT) among drug users in Urumqi.</p><p><b>METHODS</b>With national MMT guideline( drift) for heroine addicted drug users, local heroine-dependent people were admitted to community-based MMT program affiliated to Xinjiang Uigur Autonomous Region Center for Mental Health. Data on outpatients' social-demo characteristics, baseline behaviors on drug use and daily stabilized dose of drugs were entered to MMT database.</p><p><b>RESULTS</b>Up to 10, Feb. , 2006,353 persons withdrew MMT treatment among 709 heroin dependant drug users. Between 11, Aug., 2005 and 10, Feb., 2006, with median duration as 77 days, cumulative drop-out rate of 90 days and 180 days after first dose of MMT were 73.8% among 455 and 99.4% among 355 drug users, respectively. The incidence of drop-outs was 29.8 per 100 person-month. The median length of stay (days) in MMT was 68.0 (95% CI: 59.0- 78.0). Correlates of retention were found as: Being Uigur(HR = 1.35;95% CI :1.09-1.67), duration of drug use (HR =0.74; 95% CI:0.55-0.99) and stabilized dose(HR = 0.60;95% CI: 0.48-0.74) was found in multiple Cox proportional hazard regression model.</p><p><b>CONCLUSION</b>Retention of MMT among drug users in Urumqi was low. Uigur people should be given individual counseling to help them increase the compliance rate. Within the ranges of clinic dosage, adjustment of the methadone dose on an individual base might serve as an appropriate approach to increase the effectiveness of the program.</p>

ABSTRACT

Objective To investigate the protein and mRNA expressions of Toll-like receptor 2 (TLR2) and Toll-like receptor 4 (TLR4) on colons of rats with trinitrobenzene sulfonic acid (TNBS)- induced colitis,and to evaluate the effects of probiotics.Methods Thirty male Wistar rats were randomly divided into normal control group (NC group),model control group(UC group) and probiotics-treated group(PC group).The experimental colitis was induced by TNBS/ethanol enema.Rats in PC group were fed with Bifico [live probiotics of combined hifidobacterium(Bif),lactobaeillus(Lac) and enteroeoccus] by 2.2?10~9 CFU/d for 4 weeks.Inflammatory scores were studied.Expressions of protein and mRNA of TLR2 and TLR4 were measured by Western blot and real-time quantitative polymerase chain reaction (PCR),respectively.Results Inflammatory scores in NC group,UC group and PC group were 4.35?0.88,10.25?1.36 and 7.94?0.85,respectively.The inflammatory scores in PC group were decreased compared with that in UC group (P

ABSTRACT

Enteric microorganisms in cecum were fostered with standard methods.Results Inflammatory scores in PC group decreased compared with those in UC group(7.94?0.85 vs 10.25?1.36,P<0.05),but still higher than those in NC group(7.94?0.85 vs 4.35?0.88,P<0.01).There was a significant alteration in the enteric microbial flora,Bif and Lac were decreased significantly in UC group,Bifico supplement ameliorated this imbalance.Plasma endotoxin and mucosal TNF-?levels in PC were lower than those in UC group[endotoxin:(93.33?21.22)pg/ml vs (121.25?39.07)pg/ml;TNF-?:67.51?14.63 vs 85.99?18.17,P<0.05],but higher than those in NC group[endotoxin:(93.33?21.22) pg/ml vs(35.20?15.12)pg/ml;TNF+?:67.51?14.63 vs 43.28 4?19.98,P<0.01].There was no statistically significant difference in mucosal IL-6 levels between PC group and UC group,though its level in PC group was lower than that in UC group(155.22?34.01 vs 184.09?29.11,P>0.05),but higher than that in NC group(155.22?34.01 vs 108.73?37.35,P<0.01).Conclusions Probiotics can attenuate inflammation of experimental colitis of rats through reinstituting gut flora bal- anee.Improving gut flora imbalance,reducing endotoxin and pro-inflammatory cytokines may be the effects of probiotics.

ABSTRACT

Objective To study the effect of millimeter wave radiation on human hepatoma cell. Methods BEL7404 hepatoma cells were cultured in vitro and randomly divided into 4 groups: the control group, the group radiated by millimeter wave for 30 min, the group treated with Fluorouracil(5 FU), and the group radiated by millimeter wave and treated with 5 FU simultaneously at same time. The ability of 35.8 GHz millimeter wave to induce the apoptosis of hepatoma cell was evaluated by analyses of fluorescence microscopy, DNA fragmentation assay and flow cytometry assay. Results BEL7404 cells radiated by the millimeter wave had the typical characteristics of apoptosis. Comparaed with the control group [(3.21? 1.06)%], the apoptosis rates were higher in 30 min radiating or/and 5 FU groups[ (14.33? 2.66)%, (18.58? 2.57)%, (27.91? 3.66)%]. Poly adp ribose polymerase(PARP) was found to be cleavaged in all the cells in millmeter wave radiation or/and 5 FU groups. Conclusion Radiation of 35.8 GHz could induce apoptosis of BEL7404 cell in vitro, and could act synergistcally with 5 FU treatment.

ABSTRACT

Objective To study the expression of proliferating cell nuclear antigen (PCNA), CDK4 and P16 on rat hepatocellular carcinoma by millimeter wave radiation. Methods Fourty male Wistar rats were randomly divided into four groups. Group one to three were feeded by diethylnitrosamine (DEN). Group one was a tumor control group. In group two and three the liver was directly radiated by 35.8 GHz, 100 mW/cm 2 millimeter wave for 20 min, twice a week for 10 or 5 weeks. Group four was a normal control radiated group. Fourteen weeks later all rats were sacrificed to undergo serological test and immunohistochemical stain of liver. Results The serum levels of ? glutamyltransferase in group two and three were lower than that in group one. Adenocarcinoma was only existed in group one by histological examination of liver tissue. Other groups of DEN exposure only had basophilic and eosinophilic nodules. Liver tissue expression of PCNA and CDK4 in group two and three were significantly lower than in group one, but the expression of P16 in group two and three was higher than that in group one. Conclusions Radiation with millimeter wave can partially inhibit cell proliferation and suppress the DEN induced hepatocellular carcinoma in rats.
