ABSTRACT

Objective:To investigate the expression levels of peripheral blood lymphocytes in patients with high-grade squamous intraepithelial lesions (HSIL) of the cervix and early cervical cancer, and to analyze their correlation with the clinicopathological characteristics of cervical cancer.Methods:The clinical data of 65 patients with HSIL and 78 patients with early cervical cancer (2018 International Federation of Gynecology and Obstetrics stage ≤ stage Ⅱ A) treated in Shanxi Province Cancer Hospital from October 2020 to November 2021 were retrospectively analyzed, and 31 healthy people undergoing physical examination during the same period were treated as the healthy control group. The expressions of CD3 + T cells, CD4 + T cells, CD8 + T cells, NK cells, NK/T cells and other immune cells in fasting peripheral blood of the patients were detected by using flow cytometry. Results:The expression levels of CD3 + T cells, CD4 + T cells, CD4 +/CD8 + and NK cells were 71±8, 39±7, 1.5±0.5, 16±7, respectively in HSIL group, and 73±9, 41±9, 1.5±0.6, 16±9, respectively in early cervical cancer group, which were lower than those in the healthy control group (76±9, 45±10, 2.0±1.3, 20±7) (all P < 0.05). The expression levels of CD8 + T cells was 28±7, 29±8, respectively in HSIL group and early cervical cancer group, which were higher than those in the healthy control group (24±7) (all P < 0.05). The expression level of total B cells in early cervical cancer group was lower than that in healthy control group (10±4 vs.12±3, P < 0.05). The expression level of CD3 + T cells in peripheral blood of early cervical cancer patients with tumor diameter >4 cm and nerve/vascular invasion was 71±10 and 72±8, which was lower than that of patients with tumor diameter 2-4 cm, ≤2 cm and without nerve/vascular invasion (72±8, 75±8, 78±7); the expression level of CD8 + T cell was 32±8 and 35±4, which was higher than that of patients with tumor diameter 2-4 cm, ≤2 cm, and without nerve/vascular invasion (28±8, 28±7, 29±8) (all P < 0.05). The levels of CD3 + T cells and total B cells were negatively correlated with the tumor diameter (all P < 0.05), while the level of CD8 + T cells was positively correlated with tumor diameter ( P < 0.05); the levels of CD3 + T cells and NK cells were negatively correlated with nerve/vascular invasion (all P < 0.05). Conclusions:The immune function of the body starts to change in the early progression of cervical cancer, and is related to the tumor diameter and nerve/vascular invasion of cervical cancer.

ABSTRACT

Objective:To investigate the predictive value of MRI radiomics model in assessing the early response to concurrent chemoradiotherapy for locally advanced cervical squamous cell carcinoma.Methods:A total of 367 patients with pathologically proven locally advanced cervical squamous cell carcinoma (International Federation of Gynecology and Obstetrics stage ⅡB-ⅣA) in Liaoning Cancer Hospital & Institute from January 2013 to June 2019 were retrospectively collected. The patients were unable to undergo surgery and received complete concurrent chemoradiotherapy. Pelvic plain MRI, DWI and dynamic contrast-enhanced MRI were performed within 2 weeks before treatment and at the end of the 4th week of treatment. Patients were divided into complete response (CR) group ( n=247) and non-CR group ( n=120) according to response evaluation criteria in solid tumors 1.1. The patients were divided into a training set ( n=256) and a validation set (n=111) via a randomized split method at a ratio of 7∶3. Two radiologists drew the region of interest on the DWI, T 2WI and contrast-enhanced T 1WI (delayed phase) images before treatment to form the volume of interest finally. Totally 1 906 radiomics features were extracted from 3 single sequence images, respectively. Feature correlation analysis and tree model were used for feature selection. Three classifier learning algorithms, namely logistic regression (LR), support vector machine and random forest, were used for machine learning and the best classifier was selected. Based on the best classifier, 3 single sequence radiomics models were built, and a multi-sequence combined model was obtained by multivariate LR analysis. The differences in the area under the receiver operating characteristic curve (AUC) of the 3 single sequence models and the multi-sequence combined model were compared by DeLong test. The clinical application value of the multi-sequence combined model was evaluated by decision analysis curve. Results:In the training set and validation set, the LR classifier model had the best performance. Based on the LR classifier, AUCs of DWI, T 2WI, contrast-enhanced T 1WI and combined sequences in the training set were 0.77, 0.74, 0.79 and 0.86, respectively, and AUCs in the validation set were 0.71, 0.66, 0.75 and 0.77, respectively. In the training set, the AUC of multi-sequence combined model was higher than those of DWI, T 2WI and contrast-enhanced T 1WI sequence models, and the differences were statistically significant ( Z=3.01, 3.56, 2.83; P=0.003, 0.001, 0.005). In the validation set, the AUC of multi-sequence combined model and T 2WI model had significant difference ( Z=2.46, P=0.015). The decision analysis curve showed that when the threshold probability was in the range of 0.44 to 0.88, the multi-sequence combined model yielded a net benefit. Conclusion:Based on the LR classifier, the combined model built by radiomics features of multi-sequence MRI images has predictive value for assessing the early response of concurrent chemoradiotherapy for locally advanced cervical squamous cell carcinoma.

ABSTRACT

Plant biology is a basic course for students majoring in biological science. Under the guidance of developmental biology, we have constructed a comprehensive, systematic and dynamic new curriculum system for plant biology. There are still some problems existing in the current curriculum teaching mode, e.g. the students do not listen carefully to the lecture due to single classroom teaching mode, the mismatch between theoretical and experimental courses in terms of content and timing, the incomplete understanding on plants and the difficulty of integrating learning and practicing due to insufficient field practices. In view of these problems, a series of new teaching strategies for bioscience major of Harbin Institute of Technology were proposed and implemented. The combination of lectures and high quality massive open online courses (MOOC) were used for theoretical study, to which the flipped classroom was added. In addition, the theoretical study and the experiment work were combined. Moreover, the plant cognitive practices were carried out in the campus and the botanical garden in early spring, late spring and summer, respectively. Satisfactory results were achieved after two rounds of teaching practice, which fulfilled the education requirements and laid foundations for students to continue follow-up basic and professional courses.

ABSTRACT

Objective:To explore the effect of problem-based learning (PBL) combined with video teaching mode on the standardized residency training of digestive operating room.Methods:The study selected 52 digestive operating room residents who participated in the standardized residency training in The First Affiliated Hospital of Air Force Medical University from December, 2019 to March, 2020 as research subjects and they were randomized into the routine group and the observation group, with 26 residents in each group. The routine group only issued the standardized training materials for learning, and the observation group introduced PBL combined with video teaching mode on this basis, with 1 month of learning time for both group. The scores of theory and practice, the scores of core competence before and after learning and the satisfaction with teaching mode were compared between the two groups. SPSS 24.0 was used for t test, rank sum test and chi-square test. Results:The results of theory and practice, excellent and good rate of observation group were higher than those of routine group ( P<0.05). After learning, the scores of basic knowledge reserve, emergency identification ability, event handling ability, field investigation ability and safety protection ability of the two groups were higher than those before learning, of which the observation group were significantly higher than the routine group, with statistical differences ( P<0.05). The satisfaction of the observation group was higher than that of the routine group. Conclusion:PBL combined with video teaching mode can improve the performance and core competence of digestive operating room residents, thus reaching their satisfaction.

ABSTRACT

Objective To study on the application value of flipped classroom and PBL in the teaching process of sanitary chemistry experiment. Methods 112 students studied in School of Public Health in Nanchang University were considered as research subjects, including Preventive Medicine grade 2014 class 1 (observation group, n=57) and class 2 (control group, n=55). The students were taught with flipped classroom model in the control group , while flipped classroom combined with PBL was applied in the observation group. The learning effect of the two groups was evaluated by using the form of "Richter score scale"in the self-made form. The data was input using Epidata 3.0 and processed by SPSS 18.0, and it was analyzed by t test and 2 test. Results The evaluation index scores (learning attitude, ability improvement, learning cooperation, professional quality improvement) in the observation group were significantly higher than that of thecontrol group (P<0.05). Besides, the final test scores were also significantly higher in the observation group than those in control group [(83.56 ±10.81) vs. (69.37 ±10.45), t=7.059, P=0.000]. Conclusion Combining flipped classroom with PBL in the teaching process of sanitary chemistry experiment for the students majored in Preventive Medicine not only improves students' exam results, but also helps students improve their learning attitude, learning ability, professional quality and so on. Therefore, the teaching method is worthy of wider application.

ABSTRACT

Ovarian cancer is one of the three major malignant tumors in gynecology, which is characterized with occult onset and rapid progression. Most diagnosed patients are in the advanced stage, therefore, it is important to seek new and effective early diagnosis methods to improve survival rate and theprognosis. miRNA is a key molecule involved in the regulation of post-transcriptional gene expression and almost all biological processes, including cell cycle, proliferation and differentiation, and tumor formation. More and more studies have found that miRNA is closely related to human malignancies and is overexpressed in ovarian cancer tissues compared with the healthy people. The expressions of miRNA have differences in different clinical stage and the histological differentiation, chemotherapy resistance and sensitivity of ovarian cancer. At the same time, miRNA is also involved in the regulation of the development of ovarian cancer, affecting the prognosis of ovarian cancer. This paper reviews the progress of miRNA in ovarian cancer, providing new ideas for the detection, diagnosis and treatment of ovarian cancer. This paper reviews the progress of miRNA in ovarian carcinoma.

ABSTRACT

Objective To evaluate the effectiveness on informational support of the parenting sense of competence and parenting stress in primiparas. Methods Ninety-five primiparas in the First Affiliated Hospital of Zhengzhou University were recruited from November 2017 to April 2018, mothers of premature infants discharged from hospital from November 2017 to January 2018 were divided into the control group,and which discharged from February 2018 to April 2018 were divided into the experimental group conveniently.Seven cases in the control group and 5 cases in the experimental group dropped out. The control group received routine nursing care, 4 times of informational support were implemented to the mothers of premature infants at the first 4 months after discharged in the experimental group,included team teaching, live demonstration, set up a wechat group, et al. All primiparas were investigated using the Chinese version of Parenting Sense of Competence Scale(C-PSOC) and the Parenting Stress Index-Short Form(PSI-SF) at the 1th,3th,6th month postpartum. Results At the 6th month postpartum, total scores of the Chinese version of Parenting Sense of Competence Scale and subscale scores of self-efficacy, satisfaction of the mothers of premature infants were 82.60 ± 4.45, 39.19 ± 3.25, 43.40 ± 3.47, respectively in the experimental group,and the scores were 76.83 ± 7.88, 36.90 ± 4.82, 39.93 ± 5.16, respectively in the control group, the difference between the two groups was statistically significant(t=4.117, 2.541, 3.613, P<0.05). At the 6th month postpartum,total scores of the Parenting Stress Index-Short Form and subscale scores of parenting distress,parent-child dysfunctional interaction,difficult child of the mothers of premature infants were 80.31 ± 9.50, 28.62 ± 4.54, 23.98 ± 4.91, 26.14 ± 5.15, respectively in the experimental group,and the scores were 87.36 ± 8.58, 32.55 ± 4.88, 25.95 ± 4.15, 28.84 ± 4.88, respectively in the control group,the difference between the two groups was statistically significant(t=-3.568-3.823, P<0.05). Conclusions Informational support can effectively improve the parenting sense of competence and relieve the parenting stress in primiparas.

ABSTRACT

Objective To analyze the application of the HR -HPV,TCT combined with digital colposcopy cervical biopsy in diagnosis of CIN and cervical carcinoma.Methods From 2014 to 2016,250 cases diagnosed as ASCUS and above the level of disease were selected in Shanxi Cancer Hospital ,including 148 cases of ASCUS,68 cases of LSIL,27 cases of HSIL and 7 cases of SCC.The HR-HPV copy number was detected by using PCR fluorescence measurement,the Reid colposcopy scores were measured by using digital colposcopy ,and the cervical histopathological diagnosis was regarded as a measurement rule.Results The diagnostic rate of simple use of TCT for the diagnosis of CIN and above lesions was 41.20%(103/250), HR -HPV & TCT in diagnosis of CIN and above lesions was 58.78%(77/131),TCT & digital colposcopy for the diagnosis of CIN and above lesions was 67.42%(89/132), which were significantly increased (χ2=35.87,P=0.00;χ2=59.01,P=0.00).The diagnostic rate of HR-HPV&TCT combined with electronic colposcopy in diagnosis of lesions above CIN was 79.82%( 91/114 ), which was significantly higher than that of HR -HPV & TCT[58.78%(77/131)],and the diagnostic rate of TCT & digital colposcopy was 67.42%(89/132)(χ2=15.54,P=0.01;χ2=8.05,P=0.02).The incidence of CIN and cervical cancer were closely related with HR -HPV infection and the rate of HR-HPV infection showed an increasing trend with the aggravating degree of disease (the HR-HPV infection rate of ASCUS was 39.86%,the HR-HPV infection rate of LSIL was 61.76%,the HR-HPV infection rate of HSIL was 88.89%,while the HR-HPV infection rate of SCC was 100.00%).Conclusion HR-HPV,TCT combined with digital colposcopy can effectively improve the diagnosis rate of CIN and above the level of disease.At the same time,with the deterioration of the disease ,HR-HPV infection rate increases significantly ,which can provide diagnostic information and significance in the clinical diagnosis of CIN and cervical cancer.

ABSTRACT

Objective To understand the experiences and feelings of major caregivers of children with nocturnal enuresis. Methods A semi-structured interview was conducted with major caregivers of children with nocturnal enuresis. Data were analyzed based on Colaizzi phenomenological research method. Results The major caregivers′experiences and feelings were classified into 4 themes, including overload care tasks, worry about the children with nocturnal enuresis,uncertain feeling of the prognosis, lack of family and medical support system. Conclusions Understanding the experiences and feelings of major caregivers of children with nocturnal enuresis can help medical workers realize their attending demands, provide professional services and promote the recovery of children.

ABSTRACT

BACKGROUND:Oligo(poly(ethylene glycol)fumarate) (OPF) hydrogel is a kind of biomaterial with good biocompatibility, injectability, and biodegradability. It is known that there are differences in the characteristics of hydrogels with different molecular weights. Under osteogenic induction, bone marrow mesenchymal stem cels encapsulated in the suitable molecular weight hydrogel have better proliferation and differentiation. Therefore, the use of OPF hydrogel provides new options for bone tissue-engineered scaffold. OBJECTIVE:To investigate the effect of OPF hydrogel with different molecular weights on the proliferation and differentiation of encapsulated rat bone marrow mesenchymal stem celsin vitro. METHODS: Four kinds of OPF hydrogels with molecular weights of 1 000, 3 000, 10 000 and 35 000 were developed by OPF crosslinking with a redox radical initiation system. Hydrogel sweling and degradation properties were detected. Afterwards, bone marrow mesenchymal stem cels encapsulated in different hydrogels were cultured in osteogenic medium for 1-3 weeks. The effects of hydrogels on morphplogy of bone marrow mesenchymal stem cels and osteogenic differentiation were detectedvia histological staining (hematoxylin-eosin staining and alizarin red staining) and immunofluorescence staining. RESULTS AND CONCLUSION: With the increase of molecular weight of hydrogels, the gelation time was shortened, the sweling ratio significantly increased, and the hydrogel degradation rate was proportional to the molecular weight. In addition, the number of mineralized nodules formed in 3 000 and 10 000 molecular weight hydrogels was more than that in the other hydrogels, indicating that hydrogels with appropriate sweling and degradation properties are beneficial to cel proliferation and differentiation. In conclusion, OPF hydrogels have good biocompatibility and hydrogels with 3 000 and 10 000 molecular weight play active regulatory roles in osteogenic differentiation of mesenchymal stem cels.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the effect of bone marrow mesenchymal stem cells (BMSCs) on secretion of inflammatory cytokines in macrophages stimulated by lipopolysacharide (LPS).</p><p><b>METHODS</b>Rat BMSCs and macrophages were isolated, cultured, and identified. The BMSCs and macrophages, cultured alone or in co-culture, were treated with LPS or PBS or without treatment and tested for interleukin-10 (IL-10) and tumor necrosis factor-α (TNF-α) concentrations in the supernatants at 1, 3, 5, 7, 12, and 24 h after the treatment using enzyme-linked immunosorbent assay.</p><p><b>RESULTS</b>Exposure to LPS caused significantly increased IL-10 and TNF-α concentrations in the supernatant of cultured macrophages but not in BMSC culture. Macrophages co-cultured with BMSCs showed significantly lowered IL-10 and TNF-α secretions in response to LPS exposure as compared with the macrophages cultured alone.</p><p><b>CONCLUSION</b>BMSCs can reduce LPS-induced secretion of inflammatory cytokines by the macrophages to ameliorate inflammatory reactions.</p>

ABSTRACT

Objective To detect and analyze the expression of ER and PR in endometrial benign and malignant tumor,and to study its correlation of ER and PR with the establishment and development of endometrial carcinoma.Methods The expressions of ER and PR were examined by immunohistochemical method in 58 eases of endometrial carcinoma,37 cases of atypism endometrial hyperplasia,25 cases of simple endometrial carcinoma.and 25 eases of normal endometrium.Results The expressions of ER and PR were higher in normal endometrium,atypism endometrial hyperplasia and endometrial carcinoma.They were lower in endometrial carcinoma than in atypism endometrial hyperplasia,both were higher than that in normal endometrium(P<0.05).The rates of expression in ER and PR increased gradually from histological grade Ⅰ,Ⅱto Ⅲ.The expressions of ER and PR in histological grade Ⅰ were significantly different from that in histologic grade Ⅲ(P<0.05).There were no correlation between expressions and ages,which expression rates less than 50 years were higher than those above 50 years.Its expression W88 not related to different clinic grade(P>0.10).Conclusion The expressions of ER and PR increased gradually from normal endometrium,atypism endometrial hyperplasia to endometrial carcinoma.ER and PR expressions were obviously related to histologic degree.It maybe related to establishment and development of endometrial carcinoma.

ABSTRACT

Objective To assess the factors which affect the activity of isolated islets,and to construct a steady and effective isolation method of rat pancreatic islets.Methods Pancreatic islets were isolated by collagenase method.The glucose-stimulation of insulin secretion(GSIS) in different conditions was performed by batch incubation method and measured by RIA.Results GSIS was improved by BSA;There was a evident decrease of insulin secretion in cultured islets of 7 days,however,no difference was observed between the freshly isolated islets group and the 1~5days cultured islets groups.Compared to the groups of(5.5 mmol/L) and(25 mmol/L) glucose in the culture medium,(11.1 mmol/L) glucose group stimulated higher insulin release.Conclusion BSA,glucose concentration of RPMI1640 medium and the culture period are related to the activity of isolated islets.
