ABSTRACT

Abstract One of the texts in the "Biodiversity in the State of São Paulo" series, within the FAPESP-Biota Program, was dedicated to the Infrastructure for Biodiversity Conservation, with a focus on Biological Collections and Conservation Units. From the early 1960s, when FAPESP was established, to the present day, financial resources have been invested in the preservation of the biodiversity of the national genetic heritage, besides other fields. History of years of advances in scientific knowledge was built, which can be portrayed through the projects that resulted in high-quality data of national and international impact. Microbiological collections are centers that generate technology and specialized human resources, and act (among other things) as living repositories preserving reference material and as witnesses to the history of microbial biodiversity because they preserve what may no longer exist. They have enormous potential to promote the global bioeconomy and address problems that have resulted from the misuse of natural resources. This reading brings everyone the history, advances, and future perspectives of culture collections, within the efforts of 60-year scientific activities in Brazil.


Resumo Um dos textos da série "Biodiversidade do Estado de São Paulo", dentro do Programa FAPESP-Biota, foi dedicado à Infraestrutura para Conservação da Biodiversidade, com foco nas coleções biológicas e nas unidades de conservação. Do início dos anos 60, quando a FAPESP foi criada, até os dias atuais muito foi investido em pesquisa nas mais diversas áreas, incluindo a preservação da biodiversidade do patrimônio genético nacional. Uma história de longos anos de avanços no conhecimento científico foi construída, a qual pode ser retratada através dos projetos que resultaram em dados de alta qualidade com impacto nacional e internacional. As coleções microbiológicas são centros geradores de tecnologia e recursos humanos especializados, que atuam (dentre outros) como repositórios vivos, preservando material de referência, e como testemunho da história da biodiversidade microbiana, preservando o que pode não mais existir. Possuem enorme potencial para alavancar a bioeconomia global e tratar de problemas que resultaram do mau uso dos recursos naturais. Essa leitura traz a todos o histórico, os avanços e as perspectivas futuras das coleções de microrganismos, dentro dos esforços de 60 anos de atividades científicas no Brasil.

ABSTRACT Purpose: This study aims to evaluate the link between preoperative parameters and oxidative stress (OS) markers in the bladder wall of men undergoing open prostatectomy. Materials and Methods: From July 2014 to August 2016, men aged ≥ 50 years and presenting with LUTS were prospectively enrolled. Preoperative assessment included validated questionnaires (IPSS and OAB - V8), lower urinary tract ultrasound and urodynamics. Bladder biopsies were taken during open prostatectomy for determination of OS markers. Increased OS was defined by increased concentration of malondialdehyde (MDA) and / or decreased concentration of antioxidant enzymes (superoxide dismutase and / or catalase). P<0.05 was regarded as statistically significant. Results: Thirty - eight consecutive patients were included. Mean age was 66.36 ± 6.44 years, mean prostate volume was 77.7 ± 20.63 cm3, and mean IPSS was 11.05 ± 8.72 points. MDA concentration was increased in men with severe bladder outlet obstruction (BOO grade V - VI according to the Schaefer's nomogram) in comparison with BOO grade III - IV (p = 0.022). Patients with severe LUTS also had higher MDA concentration when compared to those with mild LUTS (p = 0.031). There was a statistically significant association between increased post - void residual urine (cut off ≥ 50 mL) and not only higher levels of MDA, but also reduced activity of SOD and catalase (p < 0.05). Conclusions: This pilot study showed that severity of LUTS and BOO were associated with increased MDA concentration in the bladder wall of men undergoing open prostatectomy. Further studies are still needed to assess the role of non - invasive biomarkers of OS in predicting bladder dysfunction in men with LUTS.

Microbial culture collections underpin biotechnology applications and are important resources for clinical microbiology by supplying reference strains and/or performing microbial identifications as a service. Proteomic profiles by MALDI-TOF MS have been used for Candida spp. identification in clinical laboratories and demonstrated to be a fast and reliable technique for the routine identification of pathogenic yeasts. The main aim of this study was to apply MALDI-TOF MS combined with classical phenotypic and molecular approaches to identify Candida clinical isolates preserved from 1 up to 52 years in a Brazilian culture collection and assess its value for the identification of yeasts preserved in this type of collections. Forty Candida spp. clinical isolates were identified by morphological and biochemical analyses. Identifications were also performed by the new proteomic approach based on MALDI-TOF MS. Results demonstrated 15% discordance when compared with morphological and biochemical analyses. Discordant isolates were analysed by ITS sequencing, which confirmed the MALDI-TOF MS identifications and these strains were renamed in the culture collection catalogue. In conclusion, proteomic profiles by MALDI-TOF MS represents a rapid and reliable method for identifying clinical Candida species preserved in culture collections and may present clear benefits when compared with the performance of existing daily routine methods applied at health centres and hospitals.

ABSTRACT

OBJECTIVE: To evaluate the concordance between the Gleason scores of prostate biopsies and radical prostatectomy specimens, thereby highlighting the importance of the prostate-specific antigen (PSA) level as a predictive factor of concordance. METHODS: We retrospectively analyzed 253 radical prostatectomy cases performed between 2006 and 2011. The patients were divided into 4 groups for the data analysis and dichotomized according to the preoperative PSA, <10 ng/mL and ≥10 ng/mL. A p-score <0.05 was considered significant. RESULTS: The average patient age was 63.3±7.8 years. The median PSA level was 9.3±4.9 ng/mL. The overall concordance between the Gleason scores was 52%. Patients presented preoperative PSA levels <10 ng/mL in 153 of 235 cases (65%) and ≥10 ng/mL in 82 of 235 cases (35%). The Gleason scores were identical in 86 of 153 cases (56%) in the <10 ng/mL group and 36 of 82 (44%) cases in the ≥10 ng/mL group (p = 0.017). The biopsy underestimated the Gleason score in 45 (30%) patients in the <10 ng/mL group and 38 (46%) patients in the ≥10 ng/mL (p = 0.243). Specifically, the patients with Gleason 3 + 3 scores according to the biopsies demonstrated global concordance in 56 of 110 cases (51%). In this group, the patients with preoperative PSA levels <10 ng/dL had higher concordance than those with preoperative PSA levels ≥10 ng/dL (61% x 23%, p = 0.023), which resulted in 77% upgrading after surgery in those patients with PSA levels ≥10 ng/dl. CONCLUSION: The Gleason scores of needle prostate biopsies and those of the surgical specimens were concordant in approximately half of the global sample. ABSTRACT

Six strains of white-rot fungi isolated from southern Chile were evaluated for their ergosterol/biomass correlation and ligninolytic potential in solid medium to formulate pellets for Reactive Orange 165 (RO165) decolourization. The fungus Anthracophyllum discolor was selected to formulate complex pellets (fungal mycelium, sawdust, and activated carbon), coated pellets (complex pellet + alginate) and simple pellets (fungal mycelium). The activity of ligninolytic enzymes (laccase, manganese peroxidase, manganese-independent peroxidase, and lignin peroxidase) was evaluated in both the complex and coated pellets in modified Kirk medium, and the morphology of the pellets was studied using scanning electron microscopy (SEM). Complex pellets of A. discolor showed a higher enzymatic production mainly MnP (38 U L-1 at day 15) compared to coated and simple pellets. Examinations using SEM showed that both pellets produced a black core that was entrapped by a layer of fungal mycelium. Decolourization of RO165 was demonstrated with all the pellets formulated. However, the highest and fastest decolourization was obtained with complex pellets (100 percent at day 8). Therefore, complex pellets of A. discolor can be used for the biological treatment of wastewater contaminated with RO165.

ABSTRACT

The antifungal activity of essential oils of fennel (Foeniculum vulgare Mill., Apiaceae), ginger (Zingiber officinale Roscoe, Zingiberaceae), mint (Mentha piperita L., Lamiaceae) and thyme (Thymus vulgaris L., Lamiaceae) was evaluated against mycotoxin producers Aspergillus flavus and A. parasiticus. High Resolution Gas Chromatography was applied to analyze chemical constituents of essential oils. The effect of different concentrations of essential oils was determined by solid medium diffusion assay. Mycelial growth and sporulation were determined for each essential oil at the concentrations established by solid medium diffusion assay. At the fifth, seventh and ninth days the mycelial diameter (Ø mm) and spore production were also determined. FUN-1 staining was performed to assess cell viability after broth macrodilution assay. Trans-anethole, zingiberene, menthol and thymol are the major component of essential oils of fennel, ginger, mint and thyme, respectively. The effective concentrations for fennel, ginger, mint and thyme were 50, 80, 50 and 50% (oil/DMSO; v/v), respectively. The four essential oils analysed in this study showed antifungal effect. Additionally, FUN-1 staining showed to be a suitable method to evaluate cell viability of potential mycotoxigenic fungi A. flavus and A. parasiticus after treatment with essential oils.

ABSTRACT

A hiperplasia benigna da próstata (HBP) é uma condição muito prevalente em homens a partir dos 40 anos, atingindo mais da metade da população masculina na sétima década e a quase totalidade na oitava década. Os sintomas a ela relacionados podem causar grande prejuízo à qualidade de vida desses pacientes. O objetivo da presente revisão é resumir a literatura atual sobre a patologia, focando na sua epidemiologia, diagnóstico e manejo. A avaliação inicial do paciente com HBP deve compreender a anamnese com aplicação do escore de sintomas prostáticos (IPSS), exame físico com toque retal, avaliação laboratorial (PSA sérico, exame de urina e função renal), além de métodos de imagem e urodinâmica para casos selecionados. O tratamento inicial para casos leves e moderados é medicamentoso, com o uso de alfa-bloqueadores ou inibidores da 5-alfaredutase, ou ainda a combinação desses. Pacientes que não respondem ao tratamento medicamentoso, com sintomas graves, ou que desenvolvem complicações da HBP devem ser considerados para tratamento cirúrgico. Este consiste em cirurgia aberta para próstatas de grande volume, ressecção transuretral para próstatas menores, além de outros tratamentos invasivos pouco disponíveis ou indisponíveis no nosso meio. O tratamento expectante para casos leves deve ser lembrado. Em conclusão, a HBP é doença com alta prevalência, cujo tratamento deve ser individualizado e instituído antes do surgimento de complicações maiores


Benign prostatic hyperplasia (BPH) is a very prevalent condition in men after 40 years of age, affecting more than half the male population in the seventh decade, and almost all in the eighth decade of life. The related symptoms can cause great damage to the quality of life of these patients. The purpose of this review is to summarize the current literature on the disease, focusing on epidemiology, diagnosis, and management. The initial evaluation of patients with BPH must include anamnesis with scoring of prostatic symptoms (IPSS), physical examination with digital rectal examination, laboratory evaluation (serum PSA, urinalysis and renal function), and imaging and urodynamics for selected cases. The initial treatment for mild to moderate cases is drug-based, using alpha blockers or 5-alpha-reductase inhibitors, or a combination of these. Patients who do respond to drug treatment, with severe symptoms, or who develop complications of BPH should be considered for surgical treatment. This consists in open surgery for large-volume prostates, transurethral resection for smaller prostates, and other invasive treatments little or unavailable in our community. The expectant treatment for mild cases should be considered. In conclusion, BPH is a highly prevalent disease whose treatment should be individualized and started before the onset of major complications
