ABSTRACT

BACKGROUND: Because of the non-homology of protein and gene between human and animals, to promote osteogenesis or spinal fusion of animals by construction of tissue-engineered bone with the human gene has influenced the experimental validation.OBJECTIVE: To construct the seed cell line for bone tissue engineering with stable expression of human bone morphogenetic protein 2 (hBMP2).METHODS: The full-length hBMP2 gene was cloned from human muscle tissues by nested RT-PCR and transfected to human bone marrow mesenchymal stem cells (hBMSCs) with lipidosome. The transfected hBMSCs were cultured with G418 in vitro to screen and purify the cells. A series of analyses such as RT-PCR, dot-ELISA, immunohistochemstry and alkaline phosphatase activity analysis were performed to evaluate the situation of hBMP2 expression and secretion at 48 hours and 3 weeks after the transduction. hBMSCs transduced with empty plasmid and the normal hBMSCs served as positive control and blank control groups, respectively, which were used for observation of cell growth, proliferation and biological characteristics of transfected cells. RESULTS AND CONCLUSION: The transfected hBMSCs appeared in small groups or clusters, and had a good proliferation after subculture in vitro. Some G418-resistance cell clones and calcium nodules were found when cultured with G418 in vitro. No significant difference was noted in the cell proliferation between the hBMP2 transfection group and two control groups. The ALP activity in the hBMP2 transfection group remained significantly higher than that in the two control groups (P < 0.01). At 48 hours and 3 weeks after transduction, hBMSCs could express actively hBMP2 by RT-PCR monitoring, and had a positive reaction of dot-ELISA and immunohistochemical analysis. The expression of hBMP2 gene in the experiment group at 48 hours was significantly higher than that at 3 weeks after transduction while there was no expression of hBMP2 gene in the two control groups. The above results show that the hBMSCs transfected by hBMP2 gene not only have potentials of normal proliferation and osteogenic differentiation, but also can stably express hBMP2.

ABSTRACT

Objective:To investigate the centralized management mode of cleaning disinfection and for ventilator, monitor, injection pump and infusion pump in our hospital so as to achieve scientific management and further development for hospital.Methods: The qualified rates of ventilator, monitor, injection pump and infusion pump, and satisfied degree of various department for them were analyzed after the centralized management was implemented.Results:After the centralized management was implemented, the cleaning qualified rates of ventilator, monitor, injection pump and infusion pump were, 99.25%, 99.78%, 99.47% and 98.94%, respectively. And the satisfaction scores of various departments for the application of equipment, equipment allocation and equipment recovery were significantly higher than those in 2013 (t=4.18,t=4.62,t=3.86,P<0.01).Conclusion: After the centralized management for routine emergency equipment is carried out by the cleaning disinfection center, the cleaning qualified rates of equipment and satisfaction of various departments has been significantly increased. Therefore, it is worthy to be applied and promoted.

ABSTRACT

BACKGROUND:In clinical application, the structure of crista lambdoidalis of L5 was unclear. It needs to expose more tissue to define L5 entry point through transverse process or superior and inferior articular process. This increased the risk of trauma and iatrogenic superior intervertebral degeneration. Therefore, it is necessary to expose L5 entry point with a minimaly invasive way. OBJECTIVE:To investigate the accuracy of L5 pedicle screw insertion with the entry point of mastoid process slope by imaging. METHODS:Mastoid process was located on the base of L5 superior articular process. A cant was formed when the highest point of L5 mastoid process backward protuberance extended inwards and downwards. The cant was defined as mastoid process slope; it was lateral to pedicle medial superior side internaly, medial to transverse process root and superior to the top of crista lambdoidalis. The slope was first easily touched and exposed in lumbar posterior surgery through paraspinal muscle space approach. Fifty patients of lumbar spine disorders were treated by L5 pedicle screws fixation through the entry point of mastoid process slope. According to preoperative radiographic and CT images, pedicle screw insertion direction of the sagittal and transverse sections was calculated. The diameter of pedicle screw was 6.5 mm. The condition of intraoperative successful rate of screws placement at one time was analyzed. The accuracy of screw placement was evaluated by postoperative radiographic and CT images. RESULTS AND CONCLUSION:With the method of the mastoid process slope, the successful rate of screw placement at one time was 96% (96/100). Totaly 100 screws were inserted into L5. According to the criterion by Gertzbein, 95 screws (95%) totaly located in pedicles and 5 screws (5%) encroached on the pedicle from medial wal. Three (3%) out of 5 inaccurately placed screws cut out less than 2 mm of the inner wal, while 2 (2%) between 2 mm and 4 mm, without neurologic deficits. The method of mastoid process slope had a high successful rate of screw placement. Combined with preoperative X-ray films and CT images could obtain a high accuracy rate of screw insertion.

ABSTRACT

Objective:To observe the clinical effect of self-prescribed Zuoyu No. 2 (ZY2) in the treatment of anal eczema after complex anal fistula operation. Methods: Totally 100 patients were randomly divided into two groups, the patients in the treatment group were given ZY2 combined with the conventional wound dressing treatment after defecation everyday, while the patients in the con-trol group were treated with washing infection area with warm water followed by applying triamcinolone acetonide and econazole creams on the lesion or itching skin combined with the conventional wound dressing after defecation everyday. Results:The efficiency rate in the treatment group was higher than that in the control group (82. 0% vs 60. 0%, P<0. 05), the total effective rate in the treatment group was also significantly higher than that in the control group (100% vs 90. 0%, P<0. 05). The anal pruritus disappearance time and the skin lesion recovery time in the treatment group were better than those in the control group(P<0. 05). Conclusion:ZY2 has a good clinical curative efficacy in the treatment of anal eczema after complex anal fistula operation with improvement of clinical symp-toms, which is worthy of promoted application.

ABSTRACT

Objective:To observe the clinical effects of Zuoyu üo. 1(ZY1)in the prevention of concomitant concurrent complica-tions after operation for anorectal diseases. Methods:164 patients who had received operation for anorectal diseases were randomly di-vided into 2 groups. Patients in the treatment group were treated with sitz baths in ZY1 and the conventional wound dressing method. Patients in the control group were treated with sitz baths in warm water and the conventional wound dressing method. Results:The effi-ciency rate of the treatment group was significantly higher than that of the control group(53. 57% vs. 27. 5%,P<0. 05),as well as the total effectiveness(95. 23% vs. 88. 75%,P<0. 05). The comparison showed that better curative effects existed in the treatment group on distress,bleeding,and edema ratings and wound healing time. Conclusion:Sitz bath in ZY1 after operation for anorectal dis-eases reduces complications and shortens the wound healing time without any side effect. It is thus extensively promising in clinical ap-plications.

ABSTRACT

PURPOSE: Synuclein-gamma (SNCG), which was initially identified as breast cancer specific gene 1, is highly expressed in advanced breast cancers, but not in normal or benign breast tissue. This study aimed to evaluate the effects of SNCG siRNA-treatment on breast cancer cells and elucidate the associated mechanisms. METHODS: Vectors containing SNCG and negative control (NC) siRNAs were transfected into MDA-MB-231 cells; mRNA levels were determined by real-time polymerase chain reaction. Cell proliferation was evaluated using the MTT assay, cell migration was assessed by the Transwell assay, apoptosis and cell cycle analyses were conducted with the flow cytometer, and Western blot analysis was performed to determine the relative levels of AKT, ERK, p-AKT, and p-ERK expression. RESULTS: SNCG mRNA levels were significantly reduced in MDA-MB-231 cells transfected with SNCG siRNA. Our results indicate that in SNCG siRNA-treated cells, cell migration and proliferation decreased significantly, apoptosis was induced, and the cell cycle was arrested. Western blot analysis indicated that the protein levels of p-AKT and p-ERK were much lower in the SNCG siRNA-treated groups, than in the control and NC groups. CONCLUSION: SNCG siRNA could decrease the migration and proliferation of breast cancer cells by downregulating the phosphorylation of AKT and ERK.

BACKGROUND: Fracture of humeral ectocondyle is a kind of articular epiphyseal fracture. It is one of the most prevalent disorder of cubital articulation. The occurrence of fracture and the mechanical mechanism of treatment is not very well cognized in clinic.OBJECTIVE: To analyze the stress distribution of the humeral ectocondyle in different postures so as to investigate the treatment mechanism of the humeral ectocondyle fracture.DESIGN: Computer simulation study on stress.SETTING: Department of Experimental Mechanics, Fuzhou University, and Department of Orthopedics, Third Affiliated Hospital of Peking University.MATERIALS: This experiment was conducted in the Fujian Experimental Mechanic Committee of Fuzhou University between January 2000 and December 2002. Humerus from a 6-year-old. corpse (Provided by the Department of Anatomy of Fujian College of Traditional Chinese Medicine). Instruments were computer, SUPER-SAP93 software, material sickle and measuring instrument and so on.METHODS: 9 kinds of loading conditions were simulated and added on three dimensional finite elemental model of humerus in SUPER-SAP93software, and then the stress distribution was filed out and analyzed.MAIN OUTCOME MEASURES: The average Mises stress of lateral condylar area and humeral trochlea area.RESULTS: At flection posture of elbow, the average Mises stress of lateral condylar area was the largest while at extension posture the stresses of nodes affected by extensor tendon were larger than those of lateral condyle.CONCLUSION: At the flection posture of elbow, the fracture of lateral condyle of humerus was resulted from c9llision occurring when radius and ulna struck humerus while at extension posture, it was chiefly due to extensor tractive force. We can use extensor tractive force to reduce the fracture of lateral condyle and avoid extensor tractive force to fix it.

ABSTRACT

AIM: To establish an HPLC-MS for determining seven bioactive components,including vanillin ferulaic acid,ligustrazine,senkyunolid I,H,and A,and ligustilide in Ligusticum chuanxiong Hort. . METHODS: The bioactive components were extracted by ultrasonic method,and then were separated by reverse HPLC,and finally were determined by the multiple reaction mode of MS. RESULTS: The detected limits of the seven bioactive components such as vanillin were 40,20,80,20,10,60 and 50 ng/mL,respectively. Their linear range were 1450,4-490,1-625,2-360,2-700,1-244 and 21-210 ?g/mL,respectively. The average contents of the above components in Ligusticum chuanxiong Hort. were 0. 12,0. 25,0. 41,0. 99,0. 30,8. 96 and 11. 25 mg/g,respectively. CONCLUSION: The proposed method has the characteristic of simplicity,rapidty and high sensitivity. It could be used in analyzing the whole components of Ligusticum chuanxiong Hort. .
