ABSTRACT

Objective@#To investigate parent-child conflict among primary and secondary school students and their parents, and to promote family harmony and mental health of children and adolescents during the COVID-19 pandemic.@*Methods@#An anonymous online survey was completed by 12 711 parents of primary and middle school students during mid-March, 2020. Emotional translation, coping style, parent-child conflict were collected and analyzed by grade.@*Results@#The incidence of parent-child conflict was emotional opposition (62.3%), verbal conflict (52.6%) and physical conflict (20.5%), respectively. About 38.1% of parents and 23.6% of students experienced negative emotions, such as anxiety and anger, and the students used coping styles that were more uniform than those of their parents. About 77.9% of families reported that they experienced different levels of parent-child conflict, which was mainly caused by child learning difficulties(45.6%), daily arrangements(22.1%), and the use of electronic devices(10.2%); compared with the previous year, about 31.4% of families reported an increase in the frequency of parent-child conflicts in the last month.@*Conclusion@#Parent-child conflict is highly prevalent in the families of primary and middle school students, and such conflicts were affected by stress-related factors, which included COVID-19. It s necessary to improve the provision of relevant mental health education and psychological assistance.

ABSTRACT

To evaluate the optimal administration frequency for interferon-α (IFN-α) and the effect of its combined use with inactive virus on chicken flocks, the prokaryotic expression plasmid pET-22b-ChIFN-α was constructed and transferred into Escherichia coli BL21(DE3) host bacteria to induce the expression of chicken IFN-α and to harvest recombinant proteins inclusion bodies. The expression of recombinant chicken IFN-α was confirmed by SDS-PAGE, and the results demonstrated that the chicken IFN-α (20 kDa) was highly expressed using the prokaryotic expression vector with a concentration of 0.2 mg/mL in the medium. Chicken IFN-α was diluted to 2.5×10⁴ U/fowls and administered to immunized specific-pathogen-free chickens orally in combination with inactivated H9N2 subtype influenza virus. Chicken that received chicken IFN-α were safe after three repeated immunizations (96 h). In addition, chicken IFN-α could induce higher levels of antiviral-related inducible genes in peripheral blood, spleen, and thymus of chicken flocks. The results of a challenge assay revealed that the lowest detoxification rates of chicken IFN-α ranged from three to five days, suggesting a higher capacity to resist H9N2 subtype avian influenza virus. The present study obtained the optimal immune frequency and immunization period for chicken IFN-α to provide theoretical support for the optimal clinical application of IFN-α.

ABSTRACT

To evaluate the immunogenicity of HA globular head domain of H5 subtype influenza virus (H5HA), the gene of H5HA was optimized and the recombinant pPICZaA-H5HA expressing vector was constructed and transfected into Pichia pastoris. The expression of the recombinant H5HA was confirmed by SDS-PAGE and Western blotting and the results demonstrated that the recombinant H5HA (37 kDa) was highly expressed in Pichia pastoris with concentration of 0.2 mg/mL in medium. The recombinant H5HA was concentrated and purified using Ni-NTA affinity chromatography. The immunogenicity of H5HA was evaluated by immunizing eight groups of chicken through intranasal or intramuscular injection with different doses of purified H5HA combined with different adjuvants, respectively. The results showed that the recombinant H5HA could induce high level IgG (HI titer was 1:64 and neutralizing antibody titer was 1:218) and the optimal dosage of the recombinant H5HA was 50 μg combined with oil. In addition, intramuscular injection was better than nasal immunization. This study provided a theoretical support for subunit vaccine development.

ABSTRACT

The HA gene of H9N2 influenza virus (A/chicken/Hunan/04.14 (H9N2)) was amplified and sequenced. The RNA was synthesized by in vitro transcription. The RNA transcription solutions were diluted to 10⁹ copies/μL using the RNA storage solution. The aliquoted RNA solutions were used to evaluate the homogeneity and stability. The results were determined by the average value obtained from four independent laboratories. Furthermore, the fluorescence quantitative RT-PCR method was also developed to verify the detection accuracy of clinical samples. The detection limit of this method is approximately 10 copies. Taken together, the RNA transcription solution established in our study can used as positive standard reference for rapid detection of H9N2 influenza virus.

ABSTRACT

Objective@#To identify the epidemiological characteristics of the common diarrhea-related virusesof children under 5 years of age in Chengdu area, and provide the objective evidences for prevention and control of diarrhea.@*Methods@#Fecal specimens collected from children with acute gastroenteritis between March 2006 and June 2015 were sent to Center for Disease Control and Prevention(CDC) of Sichuan province for detection of viral RNA. Clinical data were also documented. Enzyme-linked immunosorbent assay(ELISA) and/or Reverse transcriptase-polymerase chain reaction(RT-PCR) were used to detect and classify rotavirus, human calicivirus, adenovirus and astrovirus.@*Results@#A total of 2 331 fecal specimens from children (1 446 male and 885 female) under 5 were collected. 1 351 were identified as having viral gastroenteritis with the overall positive rate of 58.0%. Children at the age from 7 to 12 months were the susceptible population. Rotavirus was detected in 659 specimens (28.3%) with epidemic time from November to December. Human calicivirus was detected in 542 specimens (23.3%) and September was its epidemic time. Norovirus GII was the main strain of the virus, but no outbreak was observed in our study. Prevalence of rotavirus declined after 2007, while the detection rate of calicivirus was increasing, which led it to be one of the primary pathogens related to viral gastroenteritis in children under 5. Astrovirus was detected in only 35 patients (1.5%) mainly identified from January to March. Adenovirus was detected in 118 patients (5.1%) mainly from May to August with limited epidemic in 2011. Most patients had acute progress(91.2%), none have chronic progress. Mild dehydration was the most common symptom among all the children, followed by moderate dehydration, while none of the patient had severe symptom. Digestive symptoms are usually accompanied by extra-intestinal symptoms in both virus infection. However, extra-intestinal symptoms had higher incidences in rotavirus infection than in calicivirus infection, while the patients with these symptoms recovered during the follow-up period.@*Conclusions@#Virus infection is the common cause of acute gastroenteritis in children under 5. Rotavirus and human calicivirus were the leading pathogens in Chengdu area.

ABSTRACT

Objective To investigate the expression and possible mechanism of miR-21 and Ski-related novel protein N( SnoN) in the renal fibrosis diabetic process.Methods The animal model was established by tail-vein injection of Streptozotocin,and the other group were normal control ( NC) group.After 10 weeks, the rats were sacrificed to measure biochemical parameters and renal index , and to observe the changes of pathomorphology by HE staining as well.Meanwhile, immunohistochemistry and Western blot were employed to examine protein ex-pression of E-cadherin,α-smooth muscle actin(α-SMA), fibronectin(FN), collagen-Ⅰ(Col-Ⅰ), collagen-Ⅲ(Col-Ⅲ), transforming growth factor-β1(TGF-β1), Smad3, p-Smad3(Ser423/425) and SnoN in the renal tissue. In addition, the expression of SonN mRNA and miR-21 were detected by qPCR.Results In DM group,the ex-pressions of Col-Ⅰ, Col-Ⅲ and FN in renal interstitium were increased ( P <0.05 ) , TGF-β1 increased (P<0.05),while E-cadherin decreased(P<0.05).Compared with NC group, the expression of α-SMA,p-Smad3 (Ser423/425) protein increased in DM group(P<0.05),while the protein level of SnoN decreased but the level of SnoN mRNA increased ( P <0.05 ) .Moreover, the level of miR-21 markedly increased in DM group ( P <0.05 ) .Conclusions TGF-β1 may up-regulate the expression of miR-21 but restrain the translational expression of SnoN, aggravating fibrosis.

ABSTRACT

Objective To investigate the actual cost and benefit of stoma nursing at present as a reference for the improvement of stoma care cost management program. Methods A cross sectional study was carried out in gastrointestinal surgery of a tertiary- level hospital in southwestern China from April 2014 to December 2014. The consumption of intestinal stoma care work, the average monthly income/average monthly working hours ratio and other indicators of stoma care nurse specialist were measured by the step cost allocation method, to investigate the actually cost of a single stoma care service which colostomy patients had received and analyze the gap between actually costs and charge standard of Sichuan Province. Results The directly human cost of stoma care expenses were (22.33 ± 3.66) yuan; working fee expenses were (4.18 ± 0.68) yuan;administrative expenses were (1.87 ± 1.32 ) yuan; research and education expenses were (1.87 ± 1.32 ) yuan; the total cost of investment was (29.14 ± 4.76) yuan, which was much higher than the current charge standard of Chengdu Bureau of Commodity Prices (t=119.50, P <0.05). Conclusions There is a difference between the actual cost and charge standard of stoma care. The situation of low pay to labor is not in line with economic laws, and not conducive to the future development of stoma care speciality.

ABSTRACT

AIM:To investigate the effects of proteasome inhibitor MG 132 on the expression of SnoN in renal tubule epithelial cells incubated in high glucose , and to explore the possible mechanism and function that MG 132 reduces or slows down renal tubular interstitial injury after incubated in high glucose .METHODS:The NRK-52E cells were divid-ed into normal control group (NG), high glucose group (HG) and high glucose plus pretreatment with different doses of MG132 group (HG+MG132).The immunofluorescence staining was used to detect the protein expression of E-cadherin and α-smooth muscle actin (α-SMA) in NRK-52E cells under different conditions .The relative protein expression levels of SnoN, Smad ubiquitination regulatory factor 2 (Smurf2), Arkadia, E-cadherin, α-SMA and collagen type Ⅰ(Col-Ⅰ) were detected by Western blotting .RESULTS:Compared with NG group , the expression of E-cadherin and SnoN was de-creased (P<0.05), while the expression of α-SMA, Col-Ⅰ, Smurf2 and Arkadia was increased (P<0.05).Compared with HG group, the protein expression of SnoN and E-cadherin was significantly up-regulated in HG+MG132 group ( P<0.05 ) , and the protein expression of α-SMA and Col-Ⅰwas significantly down-regulated in a dose-depended manner ( P<0.05).However, no effect on the protein expression of Smurf2 and Arkadia was observed.CONCLUSION: MG132 in-hibits the degradation of SnoN protein induced by high glucose , thus reducing the renal fibrosis .

Objective: To get a general estimation of the pneumatic system in ventilators and supply technical assistance for screening and removing the faults of pneumatic component. Methods: Pneumatic system of three kinds of ventilators, Sevro-i/s, Rapheal XTC and PB840 were analyzed and the differences of the three pneumatic systems were compared. Results:Known the construct and differences of pneumatic system. Conclusion: Pneumatic system is the major component of ventilator, and the analysis of the pneumatic component is benefited for the common troubleshooting of ventilators.

ABSTRACT

An amphiphilic LDL adsorbent was synthesized with dietary fiber as the carrier, laurylamine as hydrophobic group and 3-Chloro-2-hydroxylpropysodium sulfonate as sulphonation reagent. The effects of reaction time, reaction temperature and amount of 3-Chloro-2-hydroxylpropysodium sulfonate on SO4(2-) content were studied, and the required preparations were made. The condition for adsorption was obtained by investigating the influence of adsorbent amount and adsorption time. The results show that the adsorption percentages for the removal of total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C) and high-density lipoprotein cholesterol (HDL-C) are 49.6%, 60.0% and 18.2%, respectively. This amphiphilic adsorbent possesses a better selectivity in removing cholesterol.

ABSTRACT

The preparation of a novel amphiphilic dietary fiber LDL adsorbent with sulfonic group and laurylamine group was studied. The effects of reaction time and reaction temperature on the adsorption rate were studied. The results show that the adsorption rates for the removal of Total cholesterol (TC), Low-density lipoprotein cholesterol (LDL-C) and High-density lipoprotein cholesterol (HDL-C) are 40.8%, 50.8% and 23.6%, respectively. The amphiphilic adsorbent has better selectivity in removing LDL-C.

ABSTRACT

OBJECTIVE To investigate the nosocomial infection and risk factors of extended spectrum beta-lactamases (ESBLs),and the concomitant infection with other bacteria and double infection in patients of malignant hematopathy.METHODS Forty six malignant hematopathy patients with nosocomial infection by ESBLs positive bacteria were studied retrospectively.RESULTS ESBLs positive bacteria of nosocomial infection in patients of malignant hematopathy were Escherichia coli and Klebsiella.67.4% Were pulmonary infection and 30.4% with septicemia.Major risk factors were the usage of the third generation cephalosporin,the usage of chemotherapy and adrenal cortical hormone,and agranulocytosis.41.3% Cases had infection with other bacteria and 13.0% cases had double infection at the same time.All cases were sensitive to carbopenems,35.7% cases were sensitive to amikacin.CONCLUSIONS The prognosis of patients with nosocomial infection by ESBLs positive bacteria is bad.It is important to select appropriate chemotherapy,measure granulocyte amount in time,control the use of third generation cephalosporin and intensify support therapy.

ABSTRACT

A PC-based software was developed and programmed with VC++6 for reconstructing MR images from the data acquired on an irregular k-space trajectory. It can read clinical MRI raw data and image data, create numerical phantoms, design k-space trajectories, generate k-space data from numerical phantom, calculate weighting functions, reconstruct images, and carry out error analysis for the reconstructed images. It is helpful to the investigations of new k-space trajectories and new reconstruction algorithms.
