ABSTRACT

Objective A lentivirus-mediated colon cancer cell line stably overexpressing human SAMHD1 gene was constructed to observe the effect of this gene on the ability of the cells to resist herpes simplex virus type 1 (HSV-1) infection. Methods p CDH-EF1-MCS-SAMHD1-T2 A-GFP recombinant plasmid was constructed using pCDH-EF1-MCS-T2 A-copGFP lentiviral vector. After confirming successful synthesis with sequencing, the recombinant plasmid and lentivirus packaging plasmids were co-transfected into 293 T cells. The pseudovirus solution was collected and concentrated, then human colon cancer cells were infected with the concentrated solution. The cell line overexpressing SAMHD1 gene was infected with HSV-1 in contrast to the control group, and the virus infection efficiency was assessed by Western blotting and immunofluorescence analyses. Results The pCDH-EF1-MCS-SAMHD1-T2 A-GFP recombinant plasmid was successfully constructed and verified by gene sequencing. Western blotting confirmed the overexpression of the SAMHD1 gene with higher levels of the gene in the transfected cells in contrast to control human colon cancer cell line. Western blotting and immunofluorescence showed that the cell line overexpressing human SAMHD1 gene could effectively inhibit the infection of HSV-1. Conclusion Lentivirus-mediated stable cell line overexpressing human SAMHD1 gene was effective in inhibiting HSV-1 replication and could help us to further investigate the function of SAMHD1.

ABSTRACT

Objective: To comprehensively compare and evaluate the yield and quality of Pinellia ternate from Sichuan province and provide a basis for breeding and high-yield cultivation of it. Methods: Principal component analysis (PCA) on 14 main agronomic characters and three quality characters of 22 species of P. ternata came from Sichuan were analyzed, and the comprehensive evaluation and cluster analysis were carried out. Results: The yield increase rate, leaf weight per plant, and proliferation rate had greater coefficients of variation among species, while the contents of uridine, guanosine, uridine, and guanosine had smaller coefficients of variation. PCA showed that the 17 main traits might be represented by six principal components, and the cumulative contribution rate was 89.829%, and induced that tuber morphology factor, plant type, quality factor, proliferation rate factor, large grain rate factor, high yield increase rate, low reproduction factor, and yield increase rate factor, respectively. The comprehensive score of 6 was the highest, which was the higher yield and inferior quality material, and the comprehensive score of 13 was the lowest, which was the lower yield and higher quality material. The test materials could be divided into four types by cluster analysis. Conclusion: The comprehensive evaluation method is reliable by PCA and cluster analysis. The wild resources of P. ternate in Sichuan basin is rich. We can choose some high quality resources among them to provide the basis for the selection of new varieties of Sichuan.

ABSTRACT

<p><b>OBJECTIVE</b>To analyze the characteristics of autopsies in medical dispute.</p><p><b>METHODS</b>The data of 202 autopsy cases in medical disputes performed by the Department of Pathology of Jiangxi Medical College from January 2001 to December 2010 were retrospectively analyzed.</p><p><b>RESULTS</b>The number of autopsy in medical disputes increased year by year. Neonatal, infant and 30 ≊ 60y were more common age groups in dispute; the gender ratio of male to female was 2:1 and more cases were from hospitals in rural areas. Most medical disputes came from in departments of pediatric, obstetrics and gynecology and general surgery. Death caused by cardiovascular diseases ranked at the first place.</p><p><b>CONCLUSION</b>Autopsy is important in medical disputes to define the cause of death and to preserve evidence.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To compare the contents of tetrahydropalmatine and dehydrocorydaline in corydalis, broken cell wall corydalis and its different processed products.</p><p><b>METHOD</b>The broken cell wall technique was used to corydalis, and then both the corydalis and broken cell wall corydalis were processed. The method of chromatography which was used to determine the contents of tetrahydropalmatine and dehydrocorydaline in corydalis broken cell wall corydalis and its different processed products was performed by RP-HPLC with Kromasil ODS-C18 (4.6 mm x 250 mm, 5 microm) column was used at 35 degrees C, acetonitrile-acetate buffer solution (pH 6.0) (30:70) as mobile phase of 1 mL x min(-1) flow rate, detection wavelength was set at 280 nm.</p><p><b>RESULT</b>The contents of the two active components in broken cell wall corydalis were higher than that in corydalis, while that in broken cell wall and vinegar-fried corydalis was highest in the products of breaked cell wall corydalis.</p><p><b>CONCLUSION</b>Breaking the cell wall of corydalis can help to dissolute alkaloids.</p>

ABSTRACT

Objective To explore the effect of different processes on the contents of active ingredients in Rhizoma Corydalis. Methods Processes of vinegar-boiling, vinegar-frying and alcohol-frying were used to fresh corydalis, boiled corydalis and purchased corydalis. Then the method of extraction by ultrasound with water and by reflux with methanol were used to extract the sample. The contents of Protopine, Tetrahydropalmatine and Dehydrocorydaline in corydalis was determined by RP-HPLC. Agilent TC-C18 (25 mm?4.6 mm, 5 ?m) column was used at 35 ℃, with acetonitrile-0.1% phosphonic acid (pH 5.3 with triethylamine) (28∶72) as mobile phase. The flow rate was 1 mL/min and detection wavelength was set at 280 nm. Results In the sample extracted by water, the contents of active ingredients in processed products were higher than in corydalis. And in the sample extracted by methanol, it also has the tendency that the contents of active ingredients in processed products were higher than in corydalis. The content of Dehydrocorydaline in fresh corydalis which was processed by vinegar-boiling, vinegar-frying and alcohol- frying was high. The content of Tetrahydropalmatine in purchased corydalis which was processed by water-boiling at production place was high. Conclusion The method of determining active ingredients in Rhizoma Corydalis was established. It was convenient and the result was accurate. The process technology that fresh corydalis was boiled by water and then processed or fresh corydalis was processed in production place can be used.
