ABSTRACT

Objective@#To investigate the genetic characteristics of VP1 coding region of enterovirus Coxsackievirus A16(CV-A16) and etiological features of hand, foot and mouth disease (HFMD) in 2017 in Xining city.@*Methods@#The pharyngeal swab specimens were collected from HFMD patients, and detected by real-time reverse transcription-polymerase chain reaction (RT-PCR). For CV-A16 positive samples, virus isolation was performed. Then RNA was extracted, and then VP1 coding region was amplified by RT-PCR. The phylogenetic tree was constructed by comparing with other genotypes and sub-genotypes strains of EV-A71.@*Results@#It was shown that 70 strains of CV-A16 were isolated from 2017 to 2018 in Xining city. In 2017, 10 strains were isolated and divided into two different lineages by phylogenetic analysis, 3 strains of B1a and 7 stains of B1b. In 2018, 60 stains were isolated, which were all belong to B1b.@*Conclusions@#B1a and B1b of CV-A16 are prevalent in Xining city from 2017 to 2018, in which B1b is the prominent isolates.

ABSTRACT

Objective@#To evaluate 2017 poliovirus surveillance in Qinghai Province.@*Methods@#According to the World Health Organization (WHO) 4 th edition of the polio laboratory manual procedure for virus isolation, the isolated L20B positive strain was identified as intratypic differentiation (ITD) by the China Center for Disease Control and Prevention, CDC). The National Polio Laboratory performed the nucleotide sequence determination of the capsid protein VP1 coding region of poliovirus (PV) and analyzed the poliovirus surveillance and the result of analysis of the cases with acute flaccid paralysis (AFP) reported in Qinghai Province in 2017 and stool samples of healthy children.@*Results@#In 2017, Qinghai CDC Polio Laboratory received specimens of 211 AFP cases and healthy stool samples. PV2 strains were isolated with a separation rate of 0.95%. Non-polio enterovirus (NPEV) strains were isolated from 25 strains with the isolation rate of 11.85%. Two PVs were used for ITD. All of them were vaccine-associated strains.@*Conclusions@#In 2017, the Qinghai CDC Polio Laboratory did not find any poliovirus and vaccine-derived poliovirus in the AFP cases and stool samples from healthy persons, maintaining the polio-free status.

Objective@#To investigate the genetic characteristics of enterovirus A 71 (EV-A71) and etiological features of hand, foot and mouth disease (HFMD) in Qinghai province from 2016 to 2017.@*Methods@#Specimens were collected from HFMD patients in Qinghai province, and detected by real-time reverse transcription-polymerase chain reaction (RT-PCR). For EV-A71 positive samples, virus was isolated and RNA was extracted, and then VP1 coding region was amplified by RT-PCR. The phylogenetic tree was constructed by comparing with other genotypes and sub-genotypes strains of EV-A71.@*Results@#It was shown that 114 strains of EV-A71 were isolated in Qinghai province from 2016 to 2017, which all belonged to genotype C4a, and could be divided to two different lineages by phylogenetic analysis. From 2016 to 2017, the epidemic strains of EV-A71 in the different transmission chains of Qinghai province was closely related to other provinces of China.@*Conclusions@#C4a was the dominant genotype of EV-A71 in Qinghai province from 2016 to 2017, and no other genotype was detected. In addition, EV-A71 isolated from Qinghai province co-evolved with EV-A71 in other provinces of China.

ABSTRACT

Objective@#To evaluate the running status of Qinghai provincial hand foot and mouth disease (HFMD) laboratory network in 2017.@*Methods@#The surveillance database developed in Qinghai provincial HFMD laboratory network in 2017 were analyzed, and the indicators for the running status of HFMD laboratory network in Qinghai province were evaluated.@*Results@#It was shown that 574 samples of suspected HFMD cases were detected by real time RT-PCR in 2017, and 368 were positive, the positive rate was 64.11%. Then 121 virus strains were isolated, the results of sequencing and analysis showed that 100 strains were EV71 with C4a genotype, 16 strains were CA16 with B1b genotype, and 5 strains belonged to the other enterovirus. In addition, Qinghai provincial HFMD network labs passed all the confirmatory test and proficiency test, and on-site review held by national HFMD laboratory in 2017, respectively.@*Conclusions@#Qinghai provincial HFMD laboratory network has been established and running well. It provided important scientific basis for HFMD surveillance in Qinghai province.

ABSTRACT

Objective@#To evaluate the running status of Qinghai provincial measles laboratory network during 2012—2017.@*Methods@#To analyze serological and virological surveillance database developed in Qinghai provincial measles laboratory network in 2012—2017, and evaluate the indicators for the running status of measles laboratory network in Qinghai Province.@*Results@#It was shown that 5 763 suspected measles cases were reported in Qinghai Province during 2012—2017, and 4 167 serum samples were collected from suspected measles cases, the collection rate is 72.31%; among them, 3 697 were IgM positive for measles and the positive rate was 88.72%; 68 were IgM positive for rubella and the positive rate was 1.63%. And 515 throat swab specimens were collected from suspected measles cases, 82 measles virus isolates were obtained and the positive rate was 15.92%. The result of sequencing and analysis showed that all the measles viruses belonged to genotype H1 and subgenotype H1a, which were predominant genotype circulated in China in recent years. In addition, Qinghai provincial measles network labs passed all the serological confirmatory test and proficiency test, and on-site review held by national or provincial measles laboratory and WHO during 2012—2017, respectively.@*Conclusions@#Qinghai provincial measles laboratory network has been established and running well. It provided important scientific basis for measles elimination in Qinghai province.

ABSTRACT

Objective To investigate the role of microRNA-155 (miR-155) on post-transcription regulation of SH2 domain-containing inositol 5'-phosphatase 1 (SHIP1) gene expression in the pathogenesis of acute myeloid leukemia (AML).Methods Quantitative real-time polymerase chain reaction (RT-PCR) was performed to detect the expression of miR-155 and SHIP1 mRNA in the AML patients and controls.miR-155 mimics was transfected into U937cells (U937m) by using X-treme GENE siRNA transfection reagent.Cells without transfection (U937c) and cells with negative transfection (U937mc) were used as controls.RT-PCR was performed to detect the expression of miR-155 and SHIP1 mRNA in these cells.The expression of SHIP1,TAKT and pAKT were detected by Western blot in U937 cells.Apoptosis was studied by flow cytometry (FCM).Results The average level of SHIP1 protein content in 15 samples of patients with AML-M4 or AML-M5 from 30 AML patients was significantly lower compared with that of patients with the other types of AML,and the levels of miR-155 were significantly higher in the same group of patients (P < 0.05).Significantly decreased levels of SHIP1 protein were found in U937m cells compared that of U937c and U937mc (P < 0.05).Significantly decreased rate of apoptosis was observed in U937m cells compared with that of U937mc and U937c.U937m cells also exhibited no alteration in total AKT content,while increased p-AKT levels were found (P< 0.05).Conclusion SHIP1 was also a primary target of miR-155 in AML.Overexpression of miR-155 could activate PI3K-AKT pathway to depressing SHIP1 and decrease the apoptosis rate of AML cells.

ABSTRACT

Objective To establish a diarrhea rat model using multiple-stimulating factors and choosing the best indexes to assess whether the model is consistent with the disease characteristics of liver -QI stagnation with spleen deficien-cy in traditional Chinese medicine .Methods Newborn SD rats were randomly divided into model group ( n=20 ) and control group (n=10).The rats of model group were stimulated by maternal separation , restraint stress and rectum acetic acid irritation, while the rats in control group were fed as normal .Weight changes, rectal sensitivity, Bristol scores and wa-ter content of feces and histology of the colon tissues were used as evaluation indexes to assess whether the model meets the demands for further studies .Results The rats in the model group showed loss of appetite , increase of water intake and u-rine reduction .Some rats showed increased activity , and even mania .Bristol scores and water content of feces were signifi-cantly higher than that of the control group , and the rectal sensitivity was significantly increased .The colon mucosa showed slightly thickened submucosal layer and mild inflammatory cell infiltration in the model rats .Conclusions The rat model established in this study is better to simulate the clinical manifestation of liver -QI stagnation with spleen deficiency in Chi-nese medicine , and may meet the demands of related researches of this disease .

ABSTRACT

Aim To explore the effects and mecha-nisms of choro-oxime derivatives on spatial learning and memory impairment in Kunming mice and SD rats induced by scopolamine and Aβ1-42 , respectively. Methods 40 Kunming mice were randomly divided into 5 groups: control group, model group, donepezil treatment group, arimoclomol treatment group and TCO-2 treatment group. There were 8 mice in each group. Mice of control group were established by intra-peritoneal injection of saline, and mice of other groups were injected with scopolamine and caused memory im-pairment. Both control group and model group were treated with solvent by intraperitoneal administration;donepezil treatment group received donepezil by intra-gastric administration; arimoclomol treatment group and TCO-2 treatment group were given the correspond-ing drugs by abdominal injection, respectively. The solvent and drugs were given at the same time every morning for 8 days. Spatial learning and memory abili-ty were tested by Morris water maze from the fifth day of the drugs administration. 40 SD rats were divided into 5 groups the same as the dementia model men-tioned above. Mice of control group were established by intracerebroventricular injection of saline, and mice of other groups were injected with insoluble Aβ1-42 to be induced of memory impairment. Solvent and drugs were also delivered as mentioned above. Morris water maze was carried out from the fifth day of the drug de-livery. After that, acetyl cholinesterase activity of hip-pocampus was tested with acetyl cholinesterase reagent kit; the content of Aβ1-42 in hippocampus was meas-ured by ELISA assay kit;the expression of phosphoryl-ated tau proteins was detected by Western Blot. Re-sults In both two dementia models, choro-oxime de-rivatives could improve the spatial learning and memory ability, shorten the escape latency and increase the times of crossing the former platform. Choro-oxime de-rivatives could also inhibit the acetyl cholinesterase ac-tivity in animal brain, decrease the concentration of Aβ1-42 and the expression of phosphorylated tau pro-teins in the dementia rats’ hippocampus. Conclusions Spatial learning and memory deficits induced by sco-polamine and Aβ1-42 could be reversed by choro-oxime derivatives. It may be concerned with enhancement of the cholinergic system functions and reduction of the levels of Aβ1-42 and phosphorylated tau proteins in the brain.

ABSTRACT

Femur fracture from falls is considered one of the most serious types of the elderly. FEA has proved to be an extremely useful tool in the structure analysis of the proximal femur. In this paper, a FEA model of proximal femur is introduced, and Hoffman failure criteria are built based on the experimental strength data for both cortical bone and trabecular bone of the femur from some references. The FEA model and the failure criteria are verified using other researcher's experimental results. The predicted trabecular failure load was only 0.5% lower than the experimental data and cortical failure load was 4.2% higher than the experimental result. This result shows that our FEA model, combined with the Hoffman criterion for both cortical bone and trabecular bone, can effectively predict the structural capacity of the femur during falling.

ABSTRACT

The effects of loading conditions on the structural capacity of the proximal femur were investigated parametrically by Finite Element Analysis (FEA) combined with Hoffman failure criterion. The loading conditions included the fall configuration angles, load locations and the friction resistance in hip joint. The results of this parametric study revealed that the load locations are the keys to determining the structural capacity of the proximal femur. There are two low peaks of the structural capacity when the loads are applied to femoral head. If the impact load were applied in this area, the fracture risk would be great. The frictional resistance of the hip joint can severely affect the failure load, which has far reaching implications in terms of osteoarthritis.

ABSTRACT

Objectives: To study the differences between sevoflurane and isoflurane in abdomen surgery anesthetized with the target-controlled close-loop anesthesia system.Methods: Forty patients (ASA I-II) were divided into sevoflurane group (n=25) and isoflurane group (n= 15).BIS value of 55 was the control variable index.The target-controlled infusion system of sevoflurane was administered for the maintenance of anesthesia in both groups after intravenous nduction. The BIS values,blood pressures,heart rates and doses of assistant drugs were recorded and analyzed.Results: No statistical difference was observed in BIS、HR and pulse pressure in two groups. ABSTRACT

Objective To study the apheresis procedure of collecting mononuclear cells from bone marrow for clinical treatment.Methods Bone marrow mononuclear cells were separated from bone marrow using COBE Spectra apheresis system.Cell morphology,cell counts,positive expression of CD34+,CD133+,CD271+ were detected,and the recovery rate of nucleated cell and mononuclear cell were also calculated.And the main parameters of the procedure with upper efficiency were set via data analysis.Results The efficiency of collection line with colourimetric card of 3% was higher than that of 5% and of 7%.The recovery rate of mononuclear cell is 75% when the cycle times ranged from 7 to 9.The positive expression of CD34+,CD133+ and CD271+ were 1.17%,0.63%,and 0.53%,respectively.Conclution This mononuclear apheresis procedure is safety,pollution-free and efficient,and remaming the bone marrow could be retransfused to the donor.
