ABSTRACT

Objective:To customize the individualized 3D printed head film for patients with head tumors undergoing radiotherapy, and to evaluate the physical properties of the material and the precision of this technology compared with the thermoplastic head film.Methods:The 3D printed head film and thermoplastic head film were placed on the solid water surface, and the depth and surface dose were measured at 5 cm by ionization chamber and film, respectively. Thirty patients with head tumors receiving radiotherapy were randomly divided into the control and experimental groups. The patients were fixed with thermoplastic head film and 3D printed head film. The translational and rotational errors in the x, y and z direction were obtained by CBCT.Results:The radiation attenuation rate of two materials at the depth of 5 cm was less than 1%. The dose of thermoplastic head film in the surface position was increased by 27%, and increased by 18% in the 3D printed head film. In two groups, 116 sets of setup errors were collected. The average translational setup errors in the control and experimental groups were 1.29 mm and 1.16 mm, 1.42 mm and 1.24 mm, 1.38 mm and 1.16 mm, respectively, and the average rotational setup errors were 1.29°and 1.08°, 1.02°and 0.96°, 1.01°and 1.00°, respectively. The translational setup errors in the y and z direction and rotational setup errors in the x direction significantly differed between the control and experimental groups (all P<0.05), but no statistical significance was found in the other direction (all P>0.05). Conclusion:The 3D printed head film fixation meets the precise setup requirements of modern radiotherapy, which deserves further application in clinical trials.

ABSTRACT

OBJECTIVE: To investigate the mechanism of calycosin (CA) inhibiting the proliferation and migration of lung adenocarcinoma cells by regulating miR-21/PTEN signaling pathway. METHODS: Using lung adenocarcinoma SPC-A1 cells as objects, cell proliferation was detected by MTT method after treated with different doses of CA (5, 15, 25, 50, 75, 100   μg/mL) for 12, 24, 48, 72 h. Cell survival rate, 30% cell growth inhibition concentration (IC30) and half inhibition concentration (IC50) were calculated. Transwell migration test was used to detect the migration of cells after treated with low-dose, medium-dose and high-dose of CA (50, 75, 100 μg/mL) for 24 h. The number of stained cells was recorded and inhibition rate of cell migration were calculated. Western blotting assay and real-time PCR were used to detect the expression of miR-21 as well as the proteins and their mRNAs expression of PTEN, VEGF, MMP-9 after treated with low-dose, medium-dose and high-dose of CA (50, 75, 100 μg/mL) for 24 h. After transfected with miR-21 mimics and miR-21 inhibitor, the effects of CA (75 μg/mL) on the expression of miR-21 and the protein expression of PETN, VEGF and MMP-9 were detected. RESULTS: After treated with 50, 75, 100 μg/mL CA for 12, 24, 48 h, 25, 50, 75, 100 μg/mL CA for 72 h, cell survival rate was decreased significantly (P<0.05 or P<0.01). IC30 of CA were 82.24, 50.45, 46.34, 31.81 μg/mL ; IC50 of CA were 108.06, 73.35, 70.08, 49.89 μg/mL during 12-72 h. Compared with normal control group, the number of stained cells in CA groups, protein expression of VEGF in CA low-dose group, expression of miR-21 as well as proteins and their mRNAs expression of VEGF, MMP-9 in CA medium-dose and high-dose groups were decreased significantly; the medium-dose and high-dose groups were significantly less or lower than low-dose group; the high-dose group was significantly less or lower than medium-dose group (P<0.05 or P<0.01). Cell migration rate of CA groups as well as protein and its mRNA expression of PTEN in CA medium-dose and high-dose groups were increased significantly; the medium-dose and high-dose groups were significantly higher than the low-dose group; the high-dose group was significantly higher than the medium-dose groups (P<0.05 or P<0.01). After transfected with miR-21 mimics, expression of miR-21 as well as protein expression of VEGF and MMP-9 were increased significantly in miR-21 mimic group, compared with normal control group; protein expression of PTEN was decreased significantly (P<0.01). After intervened by CA, expression of miR-21 as well as protein expression of VEGF and MMP-9 in cells were decreased significantly, compared with miR-21 mimic group; protein expression of PTEN was increased significantly (P<0.05 or P<0.01). After transfected with miR-21 inhibitor, expression of miR-21 as well as  protein expression of VEGF and MMP-9 were decreased significantly in miR-21 inhibitor group, compared with normal control group; protein expression of PTEN was increased significantly (P<0.05 or P<0.01). After intervened by CA, the expression of miR-21 and above protein had no significant change in cells, compared with miR-21 inhibitor group (P>0.05). CONCLUSIONS: CA can inhibit the proliferation and migration of lung adenocarcinoma SPC-A1 cells in a dose-dependent manner, which may be associated with the regulation of miR-21/PTEN signaling pathway.

ABSTRACT

Objective To compare the difference of dose distribution between inverse planning simulated annealing (IPSA) and hybrid inverse treatment planning and optimization (HIPO) in 3D brachytherapy plan of cervical cancer,and to provide evidence for selection of reverse planning optimization method for cervical cancer brachytherapy.Methods From Dec 2016 to May 2017,totally 43 cases of patients with cervical cancer radical surgery were selected.Original IPSA brachytherapy treatment plan optimization was applied to all cases.Based on the information of original image,IPSA and HIPO plans were established according to the same initial conditions.Parameters of Dg0,D100,V100%,Homogeneity Index (HI),and conformal index (CI) of the bladder,rectum and sigmoid D2 cm3 data for High-Risk Clinical Target Volume (HR-CTV) were assessed.Results There was no statistically significant difference in D90,D100 and CI for HR-CTV between the two groups.But the V100% of HR-CTV in HIPO group was significantly higher than that in IPSA group [(87.72 ±0.49)% vs.(85.01 ± 0.55)%,t =2.54,P <0.05].Furthermore,HI in HIPO group was (0.51 ±0.08),which was higher than that in IPSA group (0.42 ± 0.06),and the difference was statistically significant (t =3.02,P < 0.05).Compared with IPSA,bladder D2 cm3 and rectum D2 cm3 [(3.04 ± 0.37) Gy] for HIPO plan were lower [(3.42 ± 0.17) Gy vs.(3.57 ± 0.28) Gy,(3.04 ± 0.37) Gy vs.(3.57 ± 0.28) Gy],which had reached statistical significance (t =0.27,0.19,P < 0.05).There was no statistical significance in the D2 cm3 dose of sigmoid.Conclusions In the treatment of cervical cancer,better target area HI and less irradiated dose of bladder and rectum can be obtained by HIPO optimization than IPSA optimization.

ABSTRACT

Purpose To investigate the diagnostic value of prenatal MRI in fetal intracranial hemorrhage (FICH). Materials and Methods The imaging and clinical data of 41 cases of FICH accepting MRI diagnosis were retrospectively analyzed. The hemorrhage location, signal characteristics and the associated intracranial abnormalities were observed. The pregnancy outcomes and clinical data after birth were followed up. The correlation between periventricular hemorrhage/intraventricular hemorrhage (PVH/IVH) classification and clinical outcomes was analyzed by combining prenatal risk factors. Results Forty-one cases of FICH were evaluated. There were 23 cases of multifocal cerebral hemorrhage and 18 cases of single focal hemorrhage. According to the bleeding site, the 41 cases were classified into PVH/IVH (33 cases), cerebral hemispheres near cortex hemorrhage (3 cases), cerebellar hemorrhage (2 cases), subdural hemorrhage (2 cases) and subarachnoid hemorrhage (1 case). Most of the FICH cases were in subacute period (36/41) and a few were in chronic period or forming encephalomalacia (5/41). The associated changes included ventriculomegaly, vascular malformation, agenesis of corpus callosum, agenesis of vermis, etc. Follow-up results showed that there were 25 cases of labor induction (autopsy after labor induction was performed in 3 cases), 16 cases were followed-up after birth. Among the 16 newborn, there was 1 case of PVH/IVH grade II fetus showing left ear hearing loss after birth, 1 case of grade II fetus showed dyskinesia within one year after birth, and 1 case of grade IV fetus showed unilateral limb movement disorder. The other 13 cases had no obvious neurological sequelae. Spearman correlation test of ranked data indicated that PVH/IVH classification was moderately correlated with birth outcome (r=0.689, P<0.05). Conclusion Prenatal MRI can evaluate the type and severity of fetal intracranial hemorrhage, and provide references for clinical diagnosis and treatment.

ABSTRACT

Objective To observe the effect of calycosin on cisplatin?induced inhibition of human gastric BGC823 cells. Methods BGC823 cells were treated with different drugs:saline,cisplatin,calycosin and cisplatin combined with calycosin. MTT assays were used to detect the prolif?eration rate of BGC823 cell. Then the protein level and RNA level of cyclin D1,CDK4 and CDK6 were detected by Western blotting and Real?time PCR. Results The proliferation inhibition rates of BGC823 cells treated with 20μg/mL cisplatin,10μg/mL meperoflavone,and combina?tion of the 2 drugs were 56.44%± 2.08%,9.52%± 2.77%and 74.44%± 0.82%,respectively. The inhibition rate of the combination of drugs was significantly higher than that of the single drug treatment group(P<0.05). In addition,we found that calycosin can significantly enhance the inhi?bition of Cyclin D1,CDK4 and CDK6 by cisplatin in protein level and RNA level. Conclusion Calycosin can significantly increase the inhibitory rate of cisplatin on BGC823 cell proliferation,and the combination of the two drugs can reduce the side effects of cisplatin.

ABSTRACT

Objective To analyze the mechanism of pigment epithelium?derived factor(PEDF)regulation in lung cancer cell proliferation and migration by vascular endothelial growth factor(VEGF). Methods VEGF expression was observed after overexpression or silencing of PEDF. Proliferation and migration were analyzed by MTT and transwell assays. Real?time PCR and Western blotting were performed to investigate the mechanism underlying PEDF regulation of the VEGF/SRC/FAK pathways. Results PEDF could inhibit the proliferation and migration of A549 cells by VEGF. Conclusion PEDF can be considered as a potential therapeutic target for lung cancer.

ABSTRACT

Objective To study the clinical effect of laparoscopy-assisted improved Soave operation for treating neonatal congeni-tal megacolon .Methods 68 cases of neonatal congenital megacolon treated in our hospital from March 2010 to July 2013 were di-vided into 2 groups .The treatment group(34 cases) was performed the laparoscopy-assisted improved Soave operation after the sys-tematic examinations ,while the control group(34 cases) was given the conventional operation therapy .The postoperative curative effects were observed and compared between the two groups .Results The effects of intraoperative bleeding amount ,operative time and the pain score in the treatment group were superior to those in the control group with statistical difference between the two group(P<0 .05);the postoperative hospital stay ,postoperative intestinal peristalsis recovery time and fasting time in the treatment group were superior to those in the control group with statistical difference between the two group (P<0 .05) .Conclusion Laparo-scope-assisted improved Soave operation has better effect for treating neonatal congenital megacolon and should be widely used in clinic .

ABSTRACT

Objective To observe the results of multifocal visual evoked potential (mfVEP) examination in patients with anterior ischemic optic neuropathy (AION) before and ater treatment,and to probe its clinical significance.Methods A total of 90 patients (90 eyes) with AION were examined by mfVEP;the second-order reaction of mfVEP was analyzed.The reaction was divided into upper and lower hemi-field of visual field,or 1 / 4 quadrant visual field (superior nasal,inferior nasal,superior temporal,and inferior temporal).The sum of waves of each response was analyzed and the results in various regions were compared.The features of wave configuration was compared between the AION eyes and the contralateral eye,and between the AION eyes before and after treatment.Results The amplitude and latency of P-wave of mfVEP was 0.198±0.033 and 100.197±7.354 respectively in AION eyes before treatment,and was 0.271±0.024 and 98.567±6.794 in the contralateral eyes;the difference was significant (t=16.556,18.330;P<0.01).The amplitude and latency of P-wave of mfVEP was 0.229±0.016 and 100.104±10.603 respectively in AION eyes after treatment,which differed much from that before the treatment (t=13.649,8.858;P<0.01) and also from that of the contralateral eyes (t=13.649,8.858;P<0.01).Conclusions The amplitude and latency of P-wave of mfVEP may accurately reflect the recovery of local optic nerve damage in AION eyes before and after treatment with good repeatability.AION can be used as a new method for AION diagnosis and detection of the prognosis.

ABSTRACT

AIM:To choose the best formulation of marzulene-S grranuls oral membrane and to evaluate the clinical effects.METHODS: Orthogonal method was used for studying.Take the appearance,viscidity and dissolubility of the oral membrane as the indexes of scoring 108 cases of patients were chosen and divided into two groups randomly.58 case were treated with marzulene-S grranuls oral membrane and 50 cases were treated with gargarisma chlorhexidine as the control group.RESULTS: The best prescription is sodium carboxymethyl cellulose 4%,polyvinyl alcohol_(1750) 1% and polyvinyl alcohol_(17-88) 3%.CONCLUSION: The design of the formulation is reasonable and the clinical effect was satisfactory.
