ABSTRACT

O presente trabalho objetivou avaliar os efeitos da administração em dose única de progestágenos em fêmeas caninas hígidas, as quais nunca haviam recebido tais fármacos. Foram selecionadas 20 cadelas, que foram examinadas clinicamente e por meio de exames complementares. Nessas cadelas, foi aplicado medroxiprogesterona por via subcutânea. Noventa dias após, as fêmeas foram esterilizadas cirurgicamente, sendo os tecidos reprodutivos encaminhados para histopatologia. Foi possível verificar que, aos 30 dias, 12 animais (60%) apresentaram hiperplasia mamária. Aos 90 dias, 18 animais (90%) apresentavam sinais de hiperplasia endometrial cística, tendo cinco (27,77%) destes animais apresentado conteúdo purulento no lúmen uterino. No exame microscópico, apenas uma fêmea não demonstrou alterações patológicas, sendo a única que recebeu o contraceptivo na fase correta (anestro). As demais fêmeas apresentaram alterações que variaram entre alterações circulatórias a hiperplasia endometrial cística grave. Assim, foi possível concluir que uma única aplicação de anticoncepcional em fêmeas hígidas pode causar complicações leves a graves.(AU)

ABSTRACT

Abstract This study aimed to inventory the herbivore insects associated with Enterolobium contortisiliquum (Vell.) Morong (Fabaceae) fruits and seeds and their primary and secondary parasitoids. Six samples collected between May and October 2013 yielded 210 fruits, from which 326 insects of six orders emerged: Coleoptera, Hymenoptera, Lepidoptera, Diptera, Thysanoptera and Psocoptera. Coleoptera (five families) was represented by the seed consumers Merobruchus bicoloripes Pic, Stator sp. Bridwell (Chrysomelidae, Bruchinae), two species of Silvanidae, one species of Scolytinae (Curculionidae), one species of Nitidulidae and one species of Cerambycidae. The cerambycid was also observed forming galleries on fruit mesocarp. Immature individuals of Lepidoptera were observed consuming the fruits and seeds. From the seven Hymenoptera families, only two species were associated with Coleoptera, being Horismenus Walker sp. (Eulophidae) as parasitoid of M. bicoloripes, and Neoheterospilus falcatus (Marsh) (Braconidae) as parasitoid of Scolytinae. The Lepidoptera parasitoids represented four genera: Pseudophanerotoma Zetel, Chelonus Panzer (Braconidae), Orgilus Nees (Braconidae) and Goniozus Forster (Bethylidae). The host associations for the reared parasitoids Bracon Fabricius (Braconidae), Pimplinae sp. (Ichneumonidae) and Perilampus Forster (Perilampidae) were not confirmed. We obtained a single representative of Diptera (Tachinidae) associated with Lepidoptera hosts in this food web.


Resumo Este estudo teve como objetivo inventariar os insetos herbívoros associados a frutos e sementes de Enterolobium contortisiliquum (Vell.) Morong (Fabaceae) e seus parasitoides primários e secundários. De seis amostras coletadas entre maio e outubro de 2013, obteve-se 210 frutos, dos quais emergiram 326 insetos pertencentes a seis ordens: Coleoptera, Hymenoptera, Lepidoptera, Diptera, Thysanoptera e Psocoptera. Coleoptera (cinco famílias) foi representada pelos consumidores de sementes: Merobruchus bicoloripes Pic, Stator sp. Bridwell (Chrysomelidae, Bruchinae), Silvanidae sp. 1 e sp. 2, Scolytinae sp. (Curculionidae), Nitidulidae sp. e Cerambycidae sp. A última espécie também foi observada formando galerias no mesocarpo do fruto. Indivíduos imaturos de Lepidoptera também foram observados consumindo os frutos e sementes. Dos Hymenoptera (sete famílias), duas espécies foram associadas a Coleoptera, sendo Horismenus Walker sp. (Eulophidae) parasitoide de M. bicoloripes e Neoheterospilus falcatus (Marsh) (Braconidae) parasitoide de Scolytinae. Os parasitoides de Lepidoptera foram totalizados em quatro gêneros: Pseudophanerotoma Zetel e Chelonus Panzer (Braconidae), Orgilus Nees (Braconidae) e Goniozus Forster (Bethylidae). As interações para os parasitoides Bracon Fabricius (Braconidae), Pimplinae sp. (Ichneumonidae) e Perilampus Forster (Perilampidae) não foram confirmadas. ABSTRACT

Um caso de abiotrofia cerebelar em um gato com 45 dias de idade foi diagnosticado no Laboratório de Patologia Animal, Hospital Veterinário da Universidade Federal de Campina Grande. O animal apresentava, havia 15 dias, apatia, anorexia, desidratação, ataxia, hipermetria, espasticidade dos membros torácicos e pélvicos, tremores de intenção, nistagmo, opistótono, déficit proprioceptivo e ausência de resposta de ameaça. Clinicamente, havia a suspeita de hipoplasia cerebelar, e, devido ao prognóstico desfavorável, o animal foi eutanasiado. Na necropsia, não foram observadas alterações macroscópicas. Microscopicamente, as lesões estavam restritas ao cerebelo e caracterizavam-se por alterações neurodegenerativas e necróticas, com desaparecimento segmentar dos neurônios de Purkinje. Nessas áreas, também se verificaram espaços em branco, denominado aspecto de cesto vazio, resultantes da perda dos neurônios de Purkinje, além de raros esferoides axonais e proliferação dos astrócitos de Bergmann. Em algumas áreas, a camada granular estava hipocelular e havia moderada gliose multifocal na camada molecular. O diagnóstico de abiotrofia cerebelar foi realizado com base nos dados epidemiológicos, clínicos e, principalmente, pelas alterações histopatológicas dos neurônios de Purkinje características da doença.(AU)


The aim of this report was to describe a case of cerebellar abiotrophy in cat with 45-year-old diagnosed at the Animal Pathology Laboratory, Veterinary Hospital of the Federal University of Campina Grande. The animal had presented 15-day apathy, anorexia, dehydration and neurological signs, characterized by ataxia, hypermetria, spasticity of fore and hindlimbs, intention tremor, nystagmus, opisthotonos, proprioceptive deficits, and absence of threat response. Clinically, cerebellar hypoplasia was suspected and the animal was euthanized due to poor prognosis. During necropsy, gross lesions were not observed. Microscopically the lesions were restricted to the cerebellum and were characterized by neurodegenerative and necrotic damage with segmental disappearance of the Purkinje cells. In these areas, there were also empty spaces, called the empty basket aspect, resulting from the loss of Purkinje cells, as well as rare axonal spheroids and proliferation of Bergmann's astrocytes. In some areas, the granular layer was hypocellular and there was moderate multifocal gliosis in the molecular layer. The diagnosis of cerebellar abiotrophy was based on epidemiological, clinical and mainly on histopathological changes in neurons of Purkinje disease characteristics.(AU)

ABSTRACT

Relata-se um caso de lesão cutânea secundária à infecção por Corynebacterium pseudotuberculosis em bovino. Abscessos e múltiplas lesões cutâneas nodulares, variando de 8x10 para 20x25cm de diâmetro, firmes, sensíveis ao toque e sem mobilidade, algumas com superfície ulcerada, circundada por halo avermelhado e drenando secreção piossanguinolenta, e outras com superfícies alopécicas, foram identificados na região torácica lateral do animal. Exames histopatológicos e o isolamento do agente de fragmentos obtidos após remoção cirúrgica das lesões confirmaram tratar-se da forma cutânea da infecção pelo C. pseudotuberculosis. As lesões microscópicas caracterizaram-se por dermatite nodular piogranulomatosa e ulcerativa. A avaliação da susceptibilidade in vitro do C. pseudotuberculosis a antimicrobianos demonstrou que o agente era resistente à amicacina, kanamicina, neomicina e penicilina G, apresentando sensibilidade à ampicilina adicionada de subactam, amoxicilia com ácido clavulônico, cefalexina, cefalotina, cefotaxima, enrofloxacina, gentamicina e tetraciclina. A retirada cirúrgica das lesões e o tratamento com enrofloxacina resultaram na cura do animal. Lesões de pele observadas em casos de ptiose, carcinoma de células escamosas e na forma atípica da actinobacilose devem ser consideradas no diagnóstico diferencial da forma cutânea da infecção por C. pseudotuberculosis em bovinos. Os dados apresentados demonstram que a infecção pelo C. pseudotuberculosis deve ser considerada no diagnóstico diferencial das lesões de pele em bovinos no Brasil.(AU)


We report a case of secondary skin lesionby infection with Corynebacterium pseudotuberculosis in bovine. Abscesses and multiple nodular lesions, ranging from 8x10 to 20x25 cm in diameter, firm, sensitive to touch, and without mobility, some with ulcerated surface, surrounded by reddish halo and draining piosanguinolenta secretion, and other surfaces with alopecia, were identified in the skin of the animal`s thoracic area. Histopathology and isolation of the agent from fragments obtained after surgical removal of the lesions confirmed the cutaneous infection by C. pseudotuberculosis. Microscopic lesions were characterized by lumpy skin disease and ulcerative pyogranulomatous. Evaluation of in vitro susceptibility to antimicrobial demonstrated that the agent was resistant to amikacin, kanamycin, neomycin and penicillin G, and sensitive to ampicillin + subactam, amoxicilia with clavulonic acid, cephalexin, cephalothin, cefotaxime, enrofloxacin, gentamicin, and tetracycline. The surgical removal of the lesions, and treatment with enrofloxacin resulted in animal cure. Skin lesions observed in case of ptiose, squamous cell carcinoma and atypical form of Actinobacillosis should be considered in the differential diagnosis of cutaneous form of C. pseudotuberculosis infection in cattle. The data presented demonstrate that infection with C. pseudotuberculosis should be considered in the differential diagnosis of skin lesions in cattle in Brazil.(AU)

ABSTRACT

We report a case of intestinal intussusception secondary to enteritis caused by Pythium insidiosum infection in a 1-year-old mixed breed bitch that died after a 30-day history of bloody diarrhea, anorexia, weight loss, listlessness, dehydration, and pale mucous membranes. Necropsy revealed two areas of intussusception, one jejunum-jejunal and one ileum-cecum-colic. The first intussusception showed slight congestion of the intestinal loop, which was resolved by manual traction, while the second intussusception exhibited thickening, irregular yellow spots, and transmural congestion. Histologically, the jejunum-jejunal segment had ischemic lesions secondary and ileum-cecum-colic intussusception there was also necrotizing, pyogranulomatous enteritis associated with negative images of intralesional fungal hyphae that were well visualized by silver impregnation. Immunohistochemistry identified the hyphae as Pythium insidiosum. The diagnosis was made based on the histological changes and confirmed by immunohistochemistry. We can conclude that enteritis by P. insidiosum in dogs can occasionally cause intestinal intussusception and result in acute death.(AU)


Relata-se um caso de intussuscepção intestinal secundária à enterite causada por Pythium insidiosusm em uma cadela, sem raça definida, com um ano de idade, que morreu após apresentar 30 dias de diarreia sanguinolenta, anorexia, perda de peso, apatia, desidratação e mucosas pálidas. Na necropsia verificaram-se duas áreas de intussuscepções, sendo uma jejuno-jejunal e outra íleo-ceco-cólica. Na primeira havia discreta congestão, que foi desfeita pela tração manual, e na segunda observou-se espessamento segmentar, focos amarelados irregulares e congestão transmural do intestino. Microscopicamente, no segmento jejuno-jejunal observaram-se moderadas lesões isquêmicas secundárias à intussuscepção, e no segmento íleo-ceco-cólica verificou-se enterite piogranulomatosa e necrosante, segmentar, associada a imagens negativas de hifas intralesionais, impregnadas pela prata. Pela imuno-histoquímica, as hifas foram imunomarcadas para Pythium insidiosum. O diagnóstico foi realizado com base nas características microscópicas das lesões e morfotintoriais das hifas e foi confirmado pela imuno-histoquímica. De acordo com esses dados, é possível concluir que a infecção intestinal por P. insidiosum em cães pode causar intussuscepção e morte.(AU)

ABSTRACT

Este trabalho teve por objetivo avaliar, clínica e macroscopicamente, o tratamento adjuvante com plasma rico em plaquetas na forma de colírio ou tampão, em úlceras de córnea de cães atendidos no Serviço de Oftalmologia Veterinária. Foram analisados 20 olhos com diagnóstico de ceratite ulcerativa, distribuídos em dois grupos experimentais. O grupo colírio (GC) foi constituído por olhos tratados topicamente com colírio autólogo de plasma rico em plaquetas (PRP), e o grupo tampão (GT) por olhos submetidos ao tratamento à base de tampão sólido de PRP, associado ao recobrimento com terceira pálpebra para retenção deste. Os grupos foram avaliados, por meio de avaliação clínica, macroscópica e análise da redução do defeito epitelial, em diferentes momentos, aos três, cinco, dez, 15 e 30 dias, com exceção do terceiro dia no GT. O recobrimento da terceira pálpebra foi removido no quinto dia no GT. Em ambos os grupos, houve redução dos sinais de inflamação, melhora na sensibilidade ocular e adequada reparação do defeito epitelial. Todos os olhos do GT apresentaram completa cicatrização no quinto dia e 70% no GC, atingindo a totalidade no 10º dia. O PRP na forma de colírio ou tampão é uma excelente terapia adjuvante a ser instituída no tratamento clínico da úlcera de córnea em cães, pois atua na diminuição dos sinais inflamatórios, da dor ocular e auxilia potencialmente na cicatrização do defeito epitelial...


This study aims to clinically and macroscopically evaluate the adjuvant therapy with platelet-rich plasma in the form of eyedrops or clot, for corneal ulcers in dogs treated at the Veterinary Ophthalmology Service. We analyzed 20 eyes diagnosed with ulcerative keratitis, divided into two experimental groups. The eyedrop group (GC) was composed of eyes treated topically with eyedrops of autologous platelet-rich plasma (PRP), and the clot group (GT) was composed of eyes treated with a platelet-rich clot and covered with a third eyelid for retention of the clot. The groups were evaluated by clinical and macroscopic analysis and by the analysis of epithelial defect reduction, at different times, at three, five, ten, 15 and 30 days, except for the third day in GT. The coverage of the third eyelid was removed on the fifth day. In both groups the inflammation signs reduced, there was an improvement in ocular sensibility and proper repair of epithelial defect. All GT eyes and 70% GC eyes showed complete healing on the fifth day, the remainder of GC completed healing on the tenth day. PRP in the form of eyedrops and clot is an excellent adjuvant therapy to be instituted in the clinical treatment for corneal ulcer in dogs, because it decreases the inflammatory signs and the ocular pain and it potentially assists in healing epithelial defects...

ABSTRACT

A terapia celular vem sendo utilizada com resultados promissores no tratamento da tendinite equina, entretanto ainda existem dúvidas quanto à persistência e ao comportamento dessas células quando implantadas no local da lesão, e quanto à sua migração para outros focos inflamatórios. O objetivo deste estudo foi avaliar a marcação das células-tronco mesenquimais (CTMs) com nanocristal antes e após o implante em lesões tendíneas experimentais do tendão flexor digital superficial (TFDS) de equinos, bem como observar a possibilidade de migração das CTMs marcadas para outro foco de lesão, o membro contralateral do mesmo animal. Para isso, foi realizada a indução de lesão experimental no TFDS em ambos os membros torácicos de cinco equinos e, após sete dias, foram implantadas as CTMs autólogas marcadas com o nanocristal Qtracker 655 em um dos membros dos animais. Após sete dias do implante, foi realizada a biópsia tendínea para posterior avaliação histopatológica, utilizando-se microscopia com fluorescência. Também foi realizado o teste de viabilidade celular antes e após a incubação com o nanocristal. As CTMs marcadas e injetadas no tecido tendíneo mantiveram sua fluorescência sete dias após seu implante, e não ocorreu migração para o membro contralateral. O uso do nanocristal para a marcação das CTMs derivadas da medula óssea equina mostrou-se efetivo pelo fato de essa nanopartícula não ter alterado a viabilidade celular e por ela ter permanecido ativa durante o período implantado...


Cell therapy has been used with promising results in the treatment of equine tendinitis. However, there are still doubts about the persistence and behavior of these cells implanted in the injured tissue and their migration to other inflamed sites. The aim of this study was to evaluate the labeling of mesenchymal stem cells (MSCs) with nanocrystals before and after implantation in experimental tendinitis of the superficial digital flexor tendon (SDFT) of horses, observing the migration possibility of MSCs marked to another lesion, performed on the contralateral limb of the same animal. An experimental lesion was induced in SDFT in both forelimbs of five horses, and after seven days autologous MSCs labeled with Qtracker(r) 655 were implanted in one member of the animals. Tendon biopsy was performed for subsequent histopathological evaluation using fluorescence microscopy seven days after the implant. Cell viability test was also performed before and after incubation with the cell labeling kit. MSCs labeled and injected into the tendon tissue maintained their fluorescence seven days after their implantation and there was no migration to the contralateral limb. The use of nanocrystals for labeling MSCs was effective because it does not alter cell viability and remains active during the experimental period...

ABSTRACT

A Brazilian isolate of Beauveria bassiana (CG425) that shows high virulence against the coffee berry borer (CBB) was examined for the production of subtilisin-like (Pr1) and trypsin-like (Pr2) cuticle-degrading proteases. Fungal growth was either in nitrate-medium or in CBB cuticle-containing medium under both buffered and unbuffered conditions. In unbuffered medium supplemented with cuticle, the pH of cultures dropped and Pr1 and Pr2 activities were detected in high amounts only at a pH of 5.5 or higher. In buffered cultures, Pr1 and Pr2 activities were higher in medium supplemented with cuticle compared to activities with nitrate-medium. The Pr1 and Pr2 activities detected were mostly in the culture supernatant. These data suggest that Pr1 and Pr2 proteases produced by strain CG425 are induced by components of CBB cuticle, and that the culture pH influences the expression of these proteases, indicating the occurrence of an efficient mechanism of protein secretion in this fungus. The results obtained in this study extend the knowledge about protease production in B. bassiana CG425, opening new avenues for studying the role of secreted proteases in virulence against the coffee berry borer during the infection process.


O isolado brasileiro de Beauveria bassiana (CG425) que apresenta alta virulência contra a broca do café (CBB) foi analisado quanto à produção de proteases degradadoras de cutícula, tipo-subtilisina (Pr1) e tipo-tripsina (Pr2). O crescimento fúngico foi realizado em meio contendo nitrato e em meio contendo cutícula da broca em condições de pH tamponado e não tamponado. Em meio não tamponado, suplementado com cutícula, o pH da cultura caiu e as atividades de Pr1 e Pr2 foram detectadas somente em valores de pH igual ou superior a 5,5. Em culturas tamponadas, as atividades Pr1 e Pr2 foram superiores em meio suplementado com cutícula, comparativamente as atividades em meio contendo nitrato. As atividades Pr1 e Pr2 ocorreram predominantemente no sobrenadante de cultivo. Os dados obtidos sugerem que Pr1 e Pr2 produzidas pelo isolado CG425 são induzidas por componentes da cutícula da broca do café (CBB), e que o pH da cultura influencia a expressão destas proteases, indicando a ocorrência de um mecanismo eficiente de secreção por este fungo. Os resultados obtidos neste estudo aumentam o conhecimento a respeito da produção de proteases por B. bassiana CG425, abrindo novos caminhos para o estudo do papel de proteases na virulência contra a broca do café durante o processo de infecção.

The effects of mycorrhizal inoculation and increasing soil P levels on the expression of total proteins and peroxidase activity on passion fruit roots were evaluated. The experimental design was entirely at random, with four treatments of inoculation (a - control; b - Gigaspora albida; c - Scutellospora heterogama; d - mixture of G. albida, G. margarita, S. heterogama, and Glomus clarum) × three levels of soil P (4, 11, and 30 mg/dm³ of soil), each with three replicates. Plants were harvested 70 days after inoculation, when root colonization, shoot P level, protein content, and enzymatic activity of peroxidase (PAGE - 7%) on root extract were evaluated. Regarding protein, there was no significant difference among the treatments, except between those roots receiving mixed inoculum and 11 mg P/dm³ of soil. Effect of P on protein concentration, when compared with the inoculation effect was observed. For peroxidase, there was an eletrophoretic band common to all treatments (rf: 0.43) and another that was absent only in noncolonized plants, grown in soil with lower P (rf: 0.46). Mycorrhizal specific bands were not present but a small decrease of intensity of bands in noncolonized plants was observed. Conversely, the control roots presented a single band (rf: 0.33) not observed in the other extracts, that may demonstrate an inhibitory effect of AMF on some host activities. The data showed the influence of P level in soil on the protein expression of roots, suggesting the influence of this nutrient on root genetic expression as well as on the mechanisms of symbiotic control/recognition.


O efeito da inoculação micorrízica e de crescentes níveis de P no solo sobre a expressão de proteínas e a atividade da peroxidase foi avaliado em raízes de maracujazeiro-amarelo (Passiflora edulis f. flavicarpa Deg.). O arranjo experimental foi feito totalmente ao acaso com quatro tratamentos de inoculação com fungos micorrízicos arbusculares, FMA (a - controle, b - Gigaspora albida, c - Scutellospora heterogama e d - mistura de G. albida, S. heterogama, G. margarita e Glomus clarum) × três níveis de P no solo (4, 11 e 30 mg/dm³ de solo) com três repetições. As plantas foram colhidas aos 70 dias de inoculação, para avaliação da colonização micorrízica, determinação do P na parte aérea, estimativa do conteúdo protéico e avaliação da atividade enzimática da peroxidase (em gel de poliacrilamida - PAGE - 7%) nos extratos radiculares. Nos valores de proteína, não houve diferença significativa, exceto entre as raízes tratadas com inóculo misto e 11 mg de P/dm³ de solo e as plantas crescidas com G. albida em solo com 30 mg de P/dm³ de solo. Observou-se influência destacada do P sobre a concentração protéica, quando comparado com o efeito fúngico. Para peroxidase, evidenciou-se a formação de uma banda eletroforética comum a todos os tratamentos (rf: 0,43) e outra que foi ausente apenas nas plantas não colonizadas, crescidas em solo com menores concentrações de P (rf: 0,46). Não foram notadas bandas micorrízicas específicas nos extratos, mas uma pequena diminuição na intensidade em bandas de plantas não colonizadas. Porém, a raiz controle apresentou uma banda única (rf: 0,33), não presente nos demais extratos, o que demonstra, talvez, um efeito inibitório dos FMA sobre algumas atividades do hospedeiro. Os dados demonstram a influência do nível de P no solo sobre a expressão protéica radicular, sugerindo a influência desse elemento na expressão gênica radicular, bem como nos mecanismos de controle/reconhecimento simbiótico.

ABSTRACT

En la central de abastecimiento y en tres ferias libres de la ciudad de Recife, PE(Brasil), se analizaron cada tres meses, durante un año, frutos de aguacate (palta), limón, maracuyá-amarillo, pimentón y tomate para detectar la presencia de enfermedades fúngicas. En cada muestra fueron evaluadas 20 unidades de cada tipo de fruto, en cinco puntos diferentes por sitio de colecta, totalizándo 100 frutos/punto de comercialización, la incidencia de enfermedades fúngicas en los frutos de aguacate fue de 38,52 porciento, en maracuyá de 15,48 porciento, en limón de 17,55 porciento, en pimentón de 1,58 porciento y en tomate de 6,5 porciento. El hongo detectado con mayor frecuencia en los frutos de aguacate fue sphaceloma perseae (20,0 porciento), en limón sphaceloma fawcetti (6,2 porciento), en maracuja cladosporium herbarum (10,3 porciento) en pimentón alternaria alternata (0,7 porciento) y en tomate geotrichum candidum (3,2 porciento). Los altos valores de incidencia de enfermedades fúngicas detectados, indican la necesidad de adoptar procedimientos antes y después de la cosecha, para reducir los riesgos de infección, lo que promovería la reducción significativa de pérdidas de postcosecha

ABSTRACT

The present study was carried out to evaluate the effect of anesthetics on reperfusion arrhythmias. Male Wistar rats (200-300 g) were injected ip with heparin (200 IU), followed by anesthesia with thiopental (40 mg/kg), pentobarbital (30 mg/kg), urethane (1.2 g/kg), either, or halothane and sacrificed by decapitation. The isolated heart (5 to 8 per group) was perfused with Locke solution by the Langendorff method and the left coronary artery was ligated for 10 min. The incidence of reperfusion arrhythmias (100 per cent ) was similar in hearts of control and previously anesthetized rats, but the duration of the arrhythmias was significantly increased by anesthesia (5-fold with thiopental, 15-fold with pentobarbital, ether and halothane, and 30-fold with urethane). In hearts taken from unanesthetized rats and perfused with Locke solution containing anesthetics (5-7 per group), the duration of reperfusion arrhythmias decreased with thiopental (0.23 +/- 0.15 min), did not change with pentobarbital (1.14 +/- 0.26 min) and increased with urethane (16.10 +/- 5.60 min). Our results show that anesthetics alter the duration of reperfusion arrhythmias in the isolated rat heart

ABSTRACT

1. The effects of acetylcholine (ACh) on cardiac rate (electrocardiogram), contractile force and coronary flow recorded simultaneously were investigated in isolated guinea pig hearts perfused with Locke solution by the method of Langendorff. 2. Bolus injections of 0.5-550 nmol ACh induced oscillations of cardiac rate. These changes were not directly related to the doses of ACh injected (X**2 test, P > 0.05). 3. The presence of 10 micronM physostigmine in the Locke solution increased the number of heart rate oscillations elicited by ACh. 4. The electrocardiogram showed that the heart rate oscillations were due to wandering pacemakers, such as slow or fast junctional rhythm, and slow or fast indioventricular rhythmy, which were intermingled with sinus rhythym, A - V block or sinus bradycardia. 5. In most experiments, the increase in ventricular rate was associated with an increase in ventricular contractile force ("Bowditch Effect") and a simultaneous reduction of coronary flow. 6. The heart rate oscillations were not prevented bu reserpine or blockade of nicotinic receptors (hexamethonium plus gallamine) but were prevented blockade of muscarinic receptors with atropine. 7. We conclude that the heart rate oscillations induced by ACh are due to several electrophysiological mechanisms (automatism and/or conduction disturbances_ related to activation of muscarinic receptors

ABSTRACT

The consequences of acute Trypanosoma cruzi infection for acetylcholine and histamine levels in gastric wall and for mast cells of the stomach were studied in rats. Intraperitoneal infection with 4,000 trypomastigotes/g of a Y strain of Trypanosoma cruzi led to a 4-fold decrease in gastric acetylcholine level and to a 57 - and 15-fold increase in histamine content in the membranous and glandular regions of the rat stomach, respectively. Infection of rats with Trypanosoma cruzi also induced a 2- and 4-fold increase in mast cell numbers in the membranous and glandular regions of the muscle layer of the gastric wall, respectively, and a ganglionic inflammatory reaction with predominance of mononuclear cells. We conclude that in acutely Trypanosoma cruzi-infected rats, the reduction of acetylcholine content is due to gastric denervation and that the histamine increase might be secondary to gastric denervation and/or to an increase in the number of mast cells of the gastric wall
