ABSTRACT

ABSTRACT The karyotypes of three armored catfish species (Loricariidae) from the Iguaçu river, southern of the Brazil, were compared using different techniques: C-banding, Ag-NOR and fluorescence in situ hybridization (FISH), which used 5S and 18S rDNAs and total Cot-1 fraction as probes. Hypostomus commersoni and Hypostomus derbyi presented 2n = 68 chromosomes, with karyotype formulae 12m+12sm+14st+30a and 12m+12sm+10st+34a, respectively; whereas Hypostomus myersi presented 2n = 74 chromosomes and 12m+16sm+12st+34a. The chromosomal localization of the Ag-NORs, 5S and 18S rDNAs differed in number of sites and chromosomal localization among the studied species. The total Cot-1 probe permitted the visualization of the repetitive DNA fraction in karyotypes of each species. Cross-hybridizations using total Cot-1 probe revealed that these species have repetitive DNAs in common. However, this does not occur in H. commersoni in relation to the other species. The apparent karyotype similarity suggests a close relationship between the sympatric H. commersoni and H. derbyi species, but the small differences detected in the examined chromosomal markers indicate evolutionary divergence due to gene flow restriction among them. Hence, the present findings indicate different composition of repetitive sequences among studied species, which permit to infer its role in chromosomal differentiation of Hypostomus.

ABSTRACT

B chromosomes are extra chromosomes from the normal chromosomal set, found in different organisms, highlighting their presence on the group of fishes. Callichthys callichthys from the upper Paraná River has a diploid number of 56 chromosomes (26 m-sm + 30 st-a) for both sexes, with the presence of a sporadically acrocentric B chromosome. Moreover, one individual presented a diploid number of 57 chromosomes, with the presence of a morphologically ill-defined acrocentric B chromosome in all analyzed cells. The physical mapping of 5S and 18S rDNA shows multiple 5S rDNA sites and only one pair of chromosomes with 18S sites in C. callichthys, except for two individuals. These two individuals presented a third chromosome bearing NORs (Ag-staining and 18S rDNA) where 5S and 18S rDNA genes are syntenic, differing only in position. The dispersion of the 18S rDNA genes from the main st-a chromosome pair 25 to one of the chromosomes from the m-sm pair 4 would have originated two variant individuals, one of which with the ill-defined acrocentric B chromosome. Cromossomos B são cromossomos extras ao conjunto cromossômico normal, encontrado em diferentes organismos, com destaque para sua presença no grupo de peixes. Callichthys callichthys do alto rio Paraná tem um número diploide de 56 cromossomos (26 m-sm + 30 st-a) para ambos os sexos, com a presença esporádica de um cromossomo B acrocêntrico. Além do mais, um indivíduo apresentou número diploide de 57 cromossomos, com a presença de um cromossomo B acrocêntrico morfologicamente mal definido em todas as células analisadas. O mapeamento físico do DNAr 5S e 18S mostrou múltiplos sítios de DNAr 5S e apenas um par de cromossomos com sítio para o DNAr 18S em C. callichthys, com exceção para dois indivíduos. Estes dois indivíduos apresentaram um terceiro cromossomo portador das RONs (Ag-RONs e 18S rDNA), onde os genes DNAr 5S e 18S são sintênicos, diferindo apenas na posição. A dispersão dos genes DNAr 18S do par de cromossomos principal st-a 25 para um dos cromossomos do par m-sm 4 teria originado dois indivíduos variantes, um dos quais com cromossomo B acrocêntrico mal definido. ABSTRACT

The genus Astyanax is prominent among Characiformes, due to the large number of species found and its wide geographic distribution. In this work, Astyanax laticeps specimens from the laguna dos Patos system were cytogenetically analyzed. A diploid number of 2n = 50 chromosomes distributed into 6m+16sm+16st+12a (FN = 88) was found, without differences between males and females. A few small heterochromatin blocks were observed, besides three more conspicuous C-bands, corresponding to NORs, as confirmed by silver nitrate and CMA3 staining, FISH, and DAPI negative staining. These regions were located in a medium-sized subtelocentric and in a large subtelocentric chromosomal pair, probably because of a deletion of this region in one homologous chromosome, or due to a transposition event between them.(AU)


O gênero Astyanax é destacado entre os Characiformes, pelo grande número de espécies encontradas e a ampla distribuição geográfica. Neste trabalho, foram analisados citogeneticamente espécimes de Astyanax laticeps do sistema da laguna do Patos. O número diplóide observado foi de 2n = 50 cromossomos distribuídos em 6m+16sm+16st+12a (NF= 88), sem diferenças entre machos e fêmeas. Foram observados poucos blocos de heterocromatina, além de três bandas-C mais conspícuas, correspondentes às NORs, confirmado pela coloração com nitrado de prata,CMA3, FISH, e coloração negativa ao DAPI. Estas regiões foram localizadas em um cromossomo subtelocêntrico de tamanho médio e em um par subtelocêntrico grande, provavelmente devido a deleção desta região em um dos cromossomos homólogos, ou por eventos de transposição entre eles.(AU)

ABSTRACT

Fluorescence in situ hybridization (FISH) was undertaken in order to determinate the chromosomal distribution pattern of 18S and 5S ribosomal DNAs (rDNA) in four populations of the characid fish Astyanax altiparanae from the upper Paraná river basin, Brazil. The 18S rDNA probe FISH revealed numerical and positional variations among specimens from the Keçaba stream compared to specimens of the other populations studied. In contrast to the variable 18S rDNA distribution pattern, highly stable chromosomal positioning of the 5S rDNA sites was observed in the four A. altiparanae populations. Divergence in the distribution pattern of 18S and 5S rDNA sites is also discussed.
