ABSTRACT

Abstract Serrapeptase, a proteolytic enzyme, has been used for the adjuvant treatment of many diseases. However, its fibrinolytic activity is still uncertain. Herein, the fibrinolytic activity of serrapeptase and its in vitro thrombolytic effects were investigated. The results showed that the fibrinolytic activity of serrapeptase was 1295 U/mg, and the specific activity was 3867 U/mg of protein when its proteolytic activity toward casein was 2800 U/mg. The optimum temperature and pH for serrapeptase activity were 37-40°C and 9.0, respectively. At 1 mmol/L, Zn2+, Mn2+ and Fe2+ could activate the fibrinolytic activity of serrapeptase, while K+, Cu2+, sodium dodecyl sulfate (SDS) and ethylene diamine tetraacetic acid (EDTA) inhibited it. In vitro tests showed that serrapeptase could completely prevent blood coagulation at 150 U/mL, and the percentage of blood clot lysis reached 96.6% at 37°C after 4 h at 300 U/mL. These results indicate that serrapeptase has excellent fibrinolytic activity, and can be used as a health food or candidate drug for the prevention or treatment of thrombotic diseases.

ABSTRACT

AIM To explore the effects of Shexiang Wulong Pills (Moschus Artifactus,Aconiti Radix Cocta,Pheretima,etc.) drug serum on the expression levels of miR-146a,miR-130 and miR-223 in peripheral blood mononuclear cells (PBMCs) of patients with rheumatoid arthritis (RA).METHODS PBMCs were extracted from blood of 30 cases of RA patients,cells were divided into three groups,blank control group,serum-free groupand Shexiang Wulong Pills drug serum group.After 48 hours,total RNA was extracted,then the expression levels of three miRs were detected by RT-PCR.RESULTS Compared with the serum-free group,the expression levels of miR-146a,miR-130 and miR-223 in the Shexiang Wulong Pills drug serum group were significantly decreased (P < 0.01).CONCLUSION Shexiang Wulong Pills can inhibit inflammation in RA patients by down-regulating the expression levels of miR-223,miR-130 and miR-146a in PBMCs.

ABSTRACT

Objective To determine the expression levels of zinc finger antisense 1 (ZFAS1) in tumor tissues of non-small cell lung cancer (NSCLC) and investigate the biological roles of ZFAS1 in NSCLC.Methods The relative expression levels of ZFAS1 in tumor tissues of NSCLC patients were determined by using real-time fluorescent qRT-PCR.RNA interference was used to knock down ZFAS1 expression in A549 cells.The proliferations of A549 cells with ZFAS1 knockdown were determined by cell counting and cell colony formation assays.Flow cytometric analysis was used to determine the cellular cycle distribution and apoptosis of the A549 cells with ZFAS1 knockdown.Transwell migration and matrigel invasion assays were used to determine the migration and invasion of the A549 cells with ZFAS1 knockdown.The gene expression levels of cyclin D1,Bcl2,N-cadherin,ZEB1,Slug and Twist were determined by real-time fluorescent qRT-PCR.Results The mean expression levels of ZFAS1 in tumor tissues of NSCLC patients [0.01 (0.002 to 0.054)] were significantly higher than those in the adjacent non-cancerous tissues [0.002 (0.001 to 0.012)] (Z =-2.638;P < 0.01).After ZFAS1 knockdown,the proliferation of A549 cells was remarkably retarded (P < 0.01).The percentage of cells at G1 phase was increased while the cells at S phase was decreased (P < 0.01).The rate of apoptotic cells was significantly increased in A549 cells with ZFAS1 knockdown (P < 0.01).A549 cells showed decreased migration and invasion abilities after ZFAS1 knockdown (P <0.01).The expression levels of cyclin D1,Bcl2,N-cadherin,ZEB1,Slug and Twist genes were decreased in ZFAS1 knockdown A549 cells (P < 0.05).Conclusion ZFASI was highly expressed in the tumor tissues of NSCLC patients.ZFAS1 knockdown induced cell cycle arrest,cell apoptosis and suppression of epithelial-mesenchymal transition (EMT),leading to the inhibition of proliferation,migration,and invasion of NSCLC cells.

Objective To investigate and analyze the clinical analysis and nursing intervention of intravenous thrombolysis with urokinase in the treatment of acute myocardial infarction. Methods 40 patients with acute myocardial infarction treated in Hangzhou Hospital of Zhejiang Medical and Health Group from February 2015 to August 2016 were selected and randomly divided into the control group and the experimental group, with 20 patients in each group. The control group received routine treatment, intravenous drip of nitroglycerin and subcutaneous injection of low molecular weight heparin. On this basis, the experimental group was given urokinase intravenous thrombolysis treatment. Two groups of patients were given reasonable nursing intervention measures to improve the quality of care. The therapeutic effects of the experimental group and the control group were compared and analyzed. Results After the corresponding treatment, the effective number of patients in the experimental group was 19 cases (95.0%). In the control group, the effective number of treatment was 13 cases (65.0%). The effective rate of treatment in the experimental group was significantly higher than that in the control group, with statistical difference (P<0.05). The remission time of the chest pain in the control group was (15.32±7.21) h, and the remission time of the chest pain in the experimental group was (2.72±0.67) h. The improvement time of chest pain in the experimental group was significantly shorter than that in the control group, with statistical difference (P<0.05). In addition, the probability of complications in the experimental group was 10%, which was significantly lower than that in the control group (40.0%), and there was a statistical difference (P<0.05). Conclusion Intravenous thrombolysis with urokinase in treatment of acute myocardial infarction treatment effect is ideal, can improve clinical symptoms in a large extent, improve the efficiency of treatment, the treatment of high safety, with the further promotion of the clinical significance.

ABSTRACT

This study was designed to investigate the effect of Xiao-Ai-Ping injection on cancer angiogenesis. CCK8 assay and BrdU incorporation immunofluorescence assay were used to detect the effect of Xiao-Ai-Ping injection on HUVECs proliferation; wound healing assay and transwell assay were employed to test the effect of Xiao-Ai-Ping injection on HUVECs migration. The anti-angiogenic effect of Xiao-Ai-Ping injection was examined by tube formation assay, rat aortic ring assay and chicken chorioallantoic membrane (CAM) assay. ELISA assay was used to measure the secretion of vascular endothelial growth factor (VEGF); and the activation of vascular endothelial growth factor receptor 2 (VEGFR2) protein and its downstream signaling pathways were examined by Western blot. Our data demonstrated that Xiao-Ai-Ping injection inhibited HU-VECs proliferation in a time-and dose-dependent manner, and the IC50 (mg·mL-1) values for 24, 48 and 72 h were 48.7±7.14, 29.1±2.25 and 22.0±4.53, individually. Xiao-Ai-Ping injection inhibited HUVECs DNA synthesis and migration. Xiao-Ai-Ping injection suppressed HUVECs tube formation, and reduced microvessel sprouting from rat aortic rings and vessel growth in CAMs. Furthermore, Xiao-Ai-Ping injection attenuated the secretion of VEGF, and inhibited the expression of p-VEGFR2 and phosphorylation of protein kinase B (p-AKT). We conclude that Xiao-Ai-Ping injection inhibits angiogenesis by down-regulation of VEGF signaling and AKT pathway.

ABSTRACT

β-arrestins, important soluble proteins mediating receptor desensitization, have diverse biological functions, such as regulating cell proliferation, cell survival, apoptosis and gene transcription. β-arrestins regulate inflammatory and immune reactions by inhibiting the basal activity of pro-inflammatory transcription factor NF-kB and participating in Toll-like receptors (TLR)/NF-kB signal pathway-mediated NF-kB activation. β-arrestins are involved in the pathogenesis of various inflammatory diseases, such as inflammatory bowel disease, rheumatoid arthritis, ankylosing spondylitis, asthma and so on. The study on β-arrestins will reveal the mechanisms of inflammatory diseases and provide new strategy for clinical treatment. Here we reviewed the recent progress on the role of β-arrestins in regulating the inflammatory diseases.

ABSTRACT

Introduction: Tuberculosis (TB) in internal migrants is one of three threats for TB control in China. To address this threat, a project was launched in eight of the 19 districts of Shanghai in 2007 to provide transportation subsidies and living allowances for all migrant TB cases. This study aims to determine if this project contributed to improved TB control outcomes among migrants in urban Shanghai. Methods: This was a community intervention study. The data were derived from the TB Management Information System in three project districts and three non-project districts in Shanghai between 2006 and 2010. The impact of the project was estimated in a difference-in–difference (DID) analysis framework, and a multivariable binary logistic regression analysis. Results: A total of 1872 pulmonary TB (PTB) cases in internal migrants were included in the study. The treatment success rate (TSR) for migrant smear-positive cases in project districts increased from 59.9% in 2006 to 87.6% in 2010 (P < 0.001). The crude DID improvement of TSR was 18.9%. There was an increased probability of TSR in the project group before and after the project intervention period (coefficient = 1.156, odds ratio = 3.178, 95% confidence interval: 1.305–7.736, P = 0.011). Conclusion: The study showed the project could improve treatment success in migrant PTB cases. This was a short-term programme using special financial subsidies for all migrant PTB cases. It is recommended that project funds be continuously invested by governments with particular focus on the more vulnerable PTB cases among migrants.
