ABSTRACT

The aim of this study was to evaluate the blood leukocytes and the MMP-2 and -9 expression in the hoof laminar tissue of horses undergoing intraluminal small colon obstruction. Laparotomy and the small colon obstruction was performed in eight healthy horses, inserting a ball in the intestinal lumen. The ball was inflated to 80 mmHg pressure and the occlusion was maintained for 4 hours. The blood was collectedBlood samples were taken before the obstruction (M0), immediately after intestinal clearance (M4), and every 12 hours until completeuntil 72 hours (M12, M24, M36, M48, M60 and M72). The hoof biopsies were performed at M0, M4, and M72 and the samples were subjected to zymography analysis. There was an increase in leukocytes in M12 and M24, due to the increase in segmented neutrophils and band neutrophils, which decreased as of M36. According to zymography technique not observed changes were not not observed in MMP-2 and -9, possibly due to the low intensity of the small colon lesions. Wherefore, it is concludedIn conclusion, that the inflammatory changes resulting from small colon obstruction were not enough to cause changes in the expression of MMP-2 and -9 in the hoof laminar tissue.(AU)

ABSTRACT

ABSTRACT

The aim of this study was to estimate the energy requirements of crossbreed ewe lambs in a feedlot fed with increasing levels of concentrate in the diet. 36 ewe lambs were evaluated with initial body weight of 23.7±3.67kg. Six animals were slaughtered at the beginning of the experiment to obtain the reference group values. Animals (30) were distributed in five treatments: maintenance (feed restricted with Tifton grass hay) and those supplemented with increasing dietary levels of concentrate (20, 40, 60 and 80%) on a dry matter basis (six animals per treatment). The ewe lambs were slaughtered when they reached 37.70±9.89kg. The concentrate levels influenced the utilization efficiency of metabolizable energy for maintenance (km) and consequently the daily requirements of metabolizable energy for maintenance. The net energy requirements for maintenance of crossbred lambs in growth is 57kcal/FCW 0.75/day, where FCW is body weight on fasting. The ewe lambs with 20kg presented net energy requirement for 100g daily weight gain of 465kcal/day. The ewe lambs with 40kg of body weight required 930 kcal/day of net energy for the same gain. The net energy requirement for maintenance of ewe lambs can be estimated by the equation: NEm (Mcal/day) = 0.057 * FCW0.75. The net energy requirement for crossbred ewe lambs gain (Mcal/day) can be estimated from the equation: NEg (Mcal/day) = 0.524*FCW 0.75*GEBW1.21, where GEBW represented gain empty body weight. Diets affect the efficiencies of uses of metabolizable energy for maintenance (km) and gain (kg).(AU)

ABSTRACT

Dye exclusion tests are used to determine the number of live and dead cells. These assays are based on the principle that intact plasma membranes in live cells exclude specific dyes, whereas dead cells do not. Although widely used, the trypan blue (TB) exclusion assay has limitations. The dye can be incorporated by live cells after a short exposure time, and personal reliability, related to the expertise of the analyst, can affect the results. We propose an alternative assay for evaluating cell viability that combines the TB exclusion test and the high sensitivity of the flow cytometry technique. Previous studies have demonstrated the ability of TB to emit fluorescence when complexed with proteins. According to our results, TB/bovine serum albumin and TB/cytoplasmic protein complexes emit fluorescence at 660 nm, which is detectable by flow cytometry using a 650-nm low-pass band filter. TB at 0.002% (w/v) was defined as the optimum concentration for distinguishing unstained living cells from fluorescent dead cells, and fluorescence emission was stable for 30 min after cell treatment. Although previous studies have shown that TB promotes green fluorescence quenching, TB at 0.002% did not interfere with green fluorescence in human live T-cells stained with anti-CD3/fluorescein isothiocyanate (FITC) monoclonal antibody. We observed a high correlation between the percentage of propidium iodide+CD3/FITC+ and TB+CD3/FITC+ cells, as well as similar double-stained cell profiles in flow cytometry dot-plot graphs. Taken together, the results indicate that a TB exclusion assay by flow cytometry can be employed as an alternative tool for quick and reliable cell viability analysis.

ABSTRACT

The effects of replacing monensin with sodium bicarbonate in diets for confined heifers on weight gain (WG), dry matter intake (DMI), feed:gain ratio (FGR) and rumen fermentation parameters were evaluated. Fifity six heifers were utilized (Santa Gertrudis x Braunvieh). Diets were formulated with two levels of concentrate (50% and 80% concentrate) with sodium monensin or sodium bicarbonate. The DMI was higest in heifers with an intake of 80% concentrate. There was not difference when the same concentrate levels were evaluated in relation to the DMI. The heifers at 80% concentrate showed highest WG. There was no significant difference in relation to WG between monensin and bicarbonate treatments and levels concentrate proportions. There were significant effects regarding FGR between treatments. The heifers on 80% concentrate showed lowest FGR. In the evaluation of N-NH3, the highest values were observed, on the whole, at 80:20 diets in relation to 50:50 diets. In the additive use, the monensin on 80:20 diets promoted the highest values of N-NH3 at 1 for 9 hours after feed intake. In the values of ruminal pH, the bicarbonate increased the values on 50:50 at 6 for18 hours after feed intake, in relation to the 80:20 diet. In this case, it was concluded that the use of sodium bicarbonate in diets for confined finishing heifers is a viable option for the replacement of monensin sodium.

ABSTRACT

The aim of this study was to investigate the effect of adding whole-body vibration (WBV; frequency = 35 to 40 Hz; amplitude = 4 mm) to squat training on the T-cell proliferative response of elderly patients with osteoarthritis (OA) of the knee. This study was a randomized controlled trial in which the selected variables were assessed before and after 12 weeks of training. Twenty-six subjects (72 ± 5 years of age) were divided into three groups: 1) squat training with WBV (WBV, N = 8); 2) squat training without WBV (N = 10), and 3) a control group (N = 8). Women who were ≥60 years of age and had been diagnosed with OA in at least one knee were eligible. The intervention consisted of 12 uninterrupted weeks of squatting exercise training performed 3 times/week. Peripheral blood mononuclear cells were obtained from peripheral blood collected before and after training. The proliferation of TCD4+ and TCD8+ cells was evaluated by flow cytometry measuring the carboxyfluorescein succinimidyl ester fluorescence decay before and after the intervention (∆). The proliferative response of TCD4+ cells (P = 0.02, effect size = 1.0) showed a significant decrease (23%) in the WBV group compared to the control group, while there was no difference between groups regarding the proliferative response of TCD8+ cells (P = 0.12, effect size = 2.23). The data suggest that the addition of WBV to squat exercise training might modulate T-cell-mediated immunity, minimizing or slowing disease progression in elderly patients with OA of the knee.

ABSTRACT

Leprosy is caused by Mycobacterium leprae, which induces chronic granulomatous infection of the skin and peripheral nerves. The disease ranges from the tuberculoid to the lepromatous forms, depending on the cellular immune response of the host. Chemokines are thought to be involved in the immunopathogenesis of leprosy, but few studies have investigated the expression of chemokine receptors on leukocytes of leprosy patients. In the present study, we evaluated 21 leprosy patients (M/F: 16/5) with a new diagnosis from the Dermatology Outpatient Clinic of the University Hospital, Federal University of Minas Gerais. The control group was composed of 20 healthy members (M/F: 15/5) of the community recruited by means of announcements. The expression of CCR2, CCR3, CCR5, and CXCR4 was investigated by flow cytometry on the surface of peripheral blood lymphocytes. There was a decrease in percentage of CD3+CXCR4+ and CD4+CXCR4+ lymphocytes in the peripheral blood of leprosy patients (median [range], 17.6 [2.7-41.9] and 65.3 [3.9-91.9], respectively) compared to the control group (median [range], 43.0 [3.7-61.3] and 77.2 [43.6-93.5], respectively). The percentage of CD4+CXCR4+ was significantly lower in patients with the tuberculoid form (median [range], 45.7 [0.0-83.1]) of the disease, but not in lepromatous patients (median [range], 81.5 [44.9-91.9]). The CXCR4 chemokine receptor may play a role in leprosy immunopathogenesis, probably directing cell migration to tissue lesions in tuberculoid leprosy patients.

ABSTRACT

The objective of the present study was to develop a quantitative method to evaluate laser-induced choroidal neovascularization (CNV) in a rat model using Heidelberg Retina Angiograph 2 (HRA2) imaging. The expression of two heparan sulfate proteoglycans (HSPG) related to inflammation and angiogenesis was also investigated. CNV lesions were induced with argon laser in 21 heterozygous Zucker rats and after three weeks a fluorescein angiogram and autofluorescence exams were performed using HRA2. The area and greatest linear dimension were measured by two observers not aware of the protocol. Bland-Altman plots showed agreement between the observers, suggesting that the technique was reproducible. After fluorescein angiogram, HSPG (perlecan and syndecan-4) were analyzed by real-time RT-PCR and immunohistochemistry. There was a significant increase in the expression of perlecan and syndecan-4 (P < 0.0001) in retinas bearing CNV lesions compared to control retinas. The expression of these two HSPG increased with increasing CNV area. Immunohistochemistry demonstrated that the rat retina damaged with laser shots presented increased expression of perlecan and syndecan-4. Moreover, we observed that the overexpression occurred in the outer layer of the retina, which is related to choroidal damage. It was possible to develop a standardized quantitative method to evaluate CNV in a rat model using HRA2. In addition, we presented data indicating that the expression of HSPG parallels the area of CNV lesion. The understanding of these events offers opportunities for studies of new therapeutic interventions targeting these HSPG.

ABSTRACT

Abstract: Bifid or trifid condyle is a rare morphological alteration of the temporomandibular joint (TMJ), of unknown etiology. The aim of this paper is to describe the imaging findings of two patients with bifid and trifid mandibular condyle associated with functional disorders. Material and Method: Two female patients, who over the last six months had presented with noise, click and unilateral TMJ pain radiated to the ear in addition to chronic progressive joint hypomobility underwent CTand MR I examinations. Results: A severe asymmetry of mandibular condyles regarding size and shape, along with irregular contours and presence of cracks or channels in condylar apex, were observed. The bone structure of cortical and trabecular bone of the condyles, as well as the condylar bone marrow, had a normal appearance. The morphology of both glenoid fossa and transverse apophysis of the temporal bone was normal. The articular disc presented a deformation adapted to the condylar shape, showing the biggest disc volume as related to the condylar groove; a case with an accumulation of fluid in the superior articular recess, and signs ofsynovitis in the anterior supradiscal recess. Both joint surfaces were covered with fibrocartilage of homogeneous appearance and regular thickness. Conclusions: Changes in condylar morphology deserve special attention in differential diagnosis of bone tumors. It is important for the radiologist to be aware of the extreme anatomic variability of the TMJ. Bifid condyles are affected by TMJ dysfunctions.

ABSTRACT

Methicillin-resistant Staphylococcus aureus (MRSA) and Methicillin-resistant Coagulase-negative Staphylococci (MRCoNS) are among the most frequent hospital infections around the world, and are associated with a significant rise in the use of vancomycin. In order to assess the presence of staphylococci resistant to glycopeptides in patients taking this antibiotic in Uberlândia Federal University Hospital, Uberlândia (MG, Brazil), a prospective longitudinal study of 41 patients (21 adults and 20 children) was performed between December 2000 and March 2002. Microbiological monitoring was carried out by means of swabs collected from the oral cavity and rectum, screened by culturing in Salt Mannitol Agar plus 6 ?g/mL oxacillin. Selected samples were tested for susceptibility to glycopeptides, by the techniques of gel diffusion and dilution in agar, and for heteroresistance, by seeding with an inoculum at a density of 0.5 on the McFarland scale (108 CFU/mL) and performing a population analysis profile (PAP). A single nephropathy patient was colonized with a strain of vancomycin intermediate S. aureus (VISA) phenotype (MIC = 8 miug/mL). In twelve patients, heteroresistant staphylococci were isolated, corresponding to eight hVISA and four hVICoNS. The PAP study confirmed the presence of six hVISA and two hVICoNS. The presence of heteroresistant vancomycin samples may pose a potential risk in the future.

ABSTRACT

The hospital environment, especially surfaces, represents a secondary reservoir for pathogens such as Staphylococcus aureus. The present study was carried out at the Hospital de Clínicas da Universidade Federal de Uberlândia (HC-UFU) in Brazil, from January to August 2004, with the aim of qualitatively and quantitatively assessing the presence of S. aureus on surfaces touched by hands (bed rail, bedside table and door handle), on the floor and in the air, in wards where the patients were infected or not infected with this organism. Twenty-six wards with at least one infected patient and 26 wards with uninfected patients were investigated. Surfaces and air were sampled during bedmaking, by means of sterile 10cm2 adhesive tapes and 90mm-diameter exposed culture plates, respectively. Cultures were carried out on Egg Yolk Salt Agar, containing 7.5% NaCl and 1% egg yolk, and colonies identified by lecithinase and coagulase tests and Gramstain. About 50% of the wards were S. aureus contaminated, without significant differences between the groups (46.1% in the infected patient wards versus 53.8% in controls); the correlation between counts of the pathogen on the surfaces and in the air was 66.7%. Inspite of the fact that these bacteria were frequently present in the assessed wards, the microbial density waslow (less than 1 CFU/cm2). The association between environmental contamination and the epidemiology of S. aureus nosocomial infections is complex and thus further investigations are needed to reach a better understanding of this relationship.

ABSTRACT

Chondroid lesions present some radiologic features which are common to benign chondroid lesions and chondrosarcomas of low-grade. It is not always possible to differentiate them, and a careful analysis must be looked. We revised the main characteristics of the continuum Enchondroma-Conventional Chondrosarcoma from the perspective of plain X-ray films, Computed Tomography and Magnetic Resonance imaging.

ABSTRACT

ABSTRACT

The use of medicinal plants has strongly increased by the world population. The objetive of this study is to assess the effects of the aqueous extract of Hyptis pectinata leaves, popularly known as "sambacaitá" or "canudinho", on liver regeneration after 70% partial hepatectomy. Twenty four rats were divided into 4 groups: group OS, submitted to sham operation and oral administration of distilled water during 4 days; group OSD200, also submitted to sham operation and ingestion of 200 mg of extract/Kg of animal for the same period of time; group HP, which underwent 70% hepatectomy after 4 days of distilled water administration; and group HPD200, which underwent 70% hepatectomy after 4 days of oral administration of 200 mg of extract/Kg of animal. Alkaline phosphatase, total bilirubin and the serum level of aminotransferases were measured and state III of mitochondrial respiratory activity was assessed. Group OSD200, when compared to OS, presented significant decrease of alcaline fosfatase. Group HPD200, when compared to HP, showed statistically significant decrease of AST level and state III respiratory process.
