ABSTRACT

The exploration of biomarkers predicting response to immune checkpoint inhibitors in microsatellite stability colorectal cancer can enable more patients to benefit from immunotherapy. Tumor mutational burden (TMB), POLE/POLD1 mutation, CMS classifications, MGMT methylation, and other indicators own the potential and value of predicting response to immune checkpoint inhibitors in microsatellite stability colorectal cancer. In this paper, we reviewed the related research on predictive biomarkers of immune checkpoint inhibitors in microsatellite stability colorectal cancer, provide a reference for the best treatment strategy for microsatellite stability colorectal cancer.

ABSTRACT

Objective@#To investigate the sleeping status of the first grade primary students aged 6-8 in Beijing, to investigate the association between maternal parenting stress and children s sleep, and to inform intervention of addressing sleep problems among children in Beijing.@*Methods@#Online survey was conducted based on the child cohort study designed to study puberty, obesity, and cardiovascular risk (PROC). The Children s Sleep Habits Questionnaire (CSHQ) and Parenting Stress Index Short Form (PSI-SF) were completed by parents, and 1 136 school aged children and their mothers were included as study participants. Logistic regression models were used to determine the association between maternal parenting stress and children s sleep.@*Results@#The prevalence of sleep problems among first grade primary students was 78.52%. Among the children who received PSI-SF, the score of children with sleeping problems(86.66±17.31) was higher than that of the children with no sleeping problems(78.59±17.75)( t =6.42, P <0.01). After adjusting sociodemographic characteristics, multivariable logistic regression results showed children of mothers with parenting stress were more likely to have sleep problems ( OR =2.52, 95% CI =1.55-4.11, P <0.01). Taking 3 dimensions in PSI-SF as independent variables, the analysis indicated that compared with the children having mothers free from parental stress and difficult children, the children with mothers with parental stress were more likely to suffer from the sleeping problems( OR = 1.66 ,1.76, P <0.05).@*Conclusion@#Sleep problems are prevalent among first grade primary students, and it underscores providing psychological support to alleviate maternal parenting stress to address children s sleep problems.

ABSTRACT

Objective@#To understand the epidemiological characteristic of the prevalence of aasthma in children in China, and to provide clues for the prevention and treatment of asthma in China.@*Methods@#A systematical literature retrieval on children asthma prevalence from CNKI, WanFang Data, VIP and PubMed databases was performed. Random effects model was used to calculate the combined prevalence of asthma and its 95% confidence interval (CI) with Stata 16.0 software, the subgroup analysis was conducted according to sex, age and region.@*Results@#A total of 26 studies were included, including 213 729 children, 112 169 boys and 101 560 girls. Meta-analysis showed that the combined prevalence of asthma in children aged 14 and under in China was 3.3%(95%CI=2.7%-4.0%). The subgroup analysis showed that the prevalence of asthma in boys was higher than that in girls(Q=11.42, P=0.01). The prevalence of asthma in preschool children aged 4-6 was higher than that in infants aged 0-3 and school children aged 7-14(Q=8.03, P=0.02). And there was no significant discrepancy of the asthma prevalence among children between the north and south(Q=0.72, P>0.05).@*Conclusion@#The prevalence of childhood asthma in China presented an increasing trend. Our findings underscore more efforts to control and prevent asthma among children.

ABSTRACT

The incidence of colorectal cancer is high threatening human health. About 60%~70%cases of CRC are derived from colorectal polyps, which can be treated by endoscopic electrotomy to prevent the possibility of canceration. Therefore, in the prevention and treatment of CRC, the role of screening is of great significance. CRC screening methods include the most commonly used fecal occult blood test ( FOBT ) and the more sensitive fecal immunochemical test (FIT), cost-effective fiber sigmoidoscopy and colonoscopy, CT colonoscopy (CTC), and fecal DNA testing and immature CRC hematology screening. In this paper, the CRC screening technologies were reviewed, including the principles, characteristics and the latest research progress to provide a theoretical basis for the application and development of CRC screening technology.

ABSTRACT

Objective To investigate the mechanism of MDM2-p53 signaling pathway in the development of colorectal cancer and correlation between p53 with clinicopathological parameters, so as to further analyze the effect of p53 on prognosis. Methods The colorectal cancer tissues and the adjacent normal tissues from 86 cases of patients with colorectal cancer were collected . The expression of p53 and murine double minute 2 (MDM2) in colorectal cancer and adjacent normal tissues were detected by immunohistochemistry, Western Blot and real-time fluorescence quantitative polymerase chain reaction (RT-PCR). The prognosis of the patients was analyzed by the Kaplan-Meier survival curves. Results The protein expression and the mRNA expression of p53 and MDM2 in colorectal cancer tissues were significantly higher than that in the adjacent non-cancerous tissues (all P<0.01). A positive correlation was observed between the expression of p53 and MDM2 (r=0.785). The expression of p53 in colorectal cancer tissues were correlated well with the degree of tumor differentiation, TNM stage, lymph node metastasis and infiltration depth (all P<0.05). Survival analysis demonstrated that the mean overall survival time in p53 high expression group was (53.92±1.56) months which was significantly lower than that in p53 low expression group of (69.16±3.72) months, and the difference was statistically significant (χ2=14.78, P<0.01). Conclusions The risk and prognosis of colorectal cancer are closely related to the MDM2-p53 signaling pathway. p53 can be used as a potential target for the prognosis and treatment of colorectal cancer.

ABSTRACT

2,4,6-Trinitrotoluene (TNT) and its by-products dinitrotoluene (DNT) pose a significant threat to human health and other living organisms.However, the conventional analytical methods involved in bulky and expensive instruments are complicated and time-consuming, impeding quick and on-line determination.In this work, a facile yet effective strategy of utilizing UV-vis spectroscopy coupled with partial least squares (PLS) was proposed, through which TNT and two isomers of DNT (2,4-DNT and 2,6-DNT) in nature water could be rapidly and simultaneously determined without any pre-separation.Variable combination population analysis (VCPA) was utilized to select important feather variables and significantly improved the predictive performance of the PLS model.The calibration set contained 25 samples constructed by orthogonal array design (OAD).The predictive ability of the models was validated by an independent prediction set including 15 samples, achieving up to 0.99 of the determination coefficients (R2) for each of the analytes.The optimized models were successfully applied to determine the 3 ingredients in 8 environmental samples involving in tap, lake and two kinds of river water with the recovery values of great than 97%.Finally, the proposed method was further validated by high performance liquid chromatography method.UV-vis spectroscopy coupled with chemometrics may be used as simple and effective strategy with high potential in environmental monitoring.

ABSTRACT

To fill the urgent need of researches on the structure and function of proteins to obtain conformation information, we combined targeted molecular dynamics ( TMD) simulation with protein network methods to analyze conformation variations in the activation process of β2 adrenergic receptor. First, targeted MD was used to obtain the conformation resembles in the activation process, and then the protein network method was applied to identify the key residues and pathways in the activation process. The results indicate that the activa-tion process of β2 adrenergic receptor involves in the cooperation of all regions and the connector region of transmembrane helices is signaling hubs. In addition, the helix ends, including intracellular and extracellular loops, are the core areas. The pathway analysis reveals that there is more than one signaling pathway. All the pathways start from Ser204 of the ligand pocket and finally transmit to NPxxY or ICL2 region, which are depended on the different pathways. While the helix TMIII, TMV, TMVI are the important areas in all the pathways. The observations from the work provide valuable information at molecular level for unraveling the signal transduction mechanism associated with the activation process.

ABSTRACT

In the framework of ensemble, a partial least squares(PLS) regression ensemble algorithm in subspace(MIESPLS), which is the combination of bootstrap and variable selection based on mutual information(MI), was proposed. The key of the proposed algorithm is to introduce the diversity of member models by bootstrap re-sampling on the training set and the subsequent MI calculation. Each time, those variables whose MI are lower than a defined threshold are first eliminated;then, a member model can be trained on a smaller subspace of original spectral variables. Two kinds of model fusion strategies, i.e., simple average fusion(SAF) and weighted average fusion(WAF), were adopted and compared. By two experiments concerning quantitative application of near-infrared(NIR) spectroscopy, MISEPLS is confirmed to be superior to the full-spectrum PLS and MIPLS method, i.e., PLS combined with MI-induced variable selection. The proposed MISEPLS can produce a more accurate and robust calibration model, but without increasing the complexity.
