ABSTRACT

Objective To explore brain cortex proteomics changes during ischemic cerebral stroke (ICS),and to seek the biomarkers associated with ICS.Methods A total of 42 male SD rats were randomly divided into sham group (n =12) and ICS group (n =30).ICS model was induced by distal middle cerebral artery occlusion.Brain cortex tissues were collected at 1,6,24 and 48 h after ICS,respectively,after cerebral ischemia.Samples from different groups were subjected to tandem mass tag (TMT)-based quantitative proteomics analysis for identification of differentially expressed proteins.Gene Ontology enrichment analysis was utilized to classify the differentially expressed proteins.Western blot was used to verify the quantitative proteomics results.Results Compared with sham group,25 proteins were considered to be differentially expressed in ICS group.Gene Ontology enrichment analysis revealed that the differentially expressed proteins were related to acute-phase response,inflammatory response,lipoprotein metabolic process,complement activation,classical pathway and innate immune response.The expression change of heat shock protein 27 (HSP27) was basically consistent with the quantitative proteomics results.Conclusions These 25 differentially expressed proteins may serve as potential biomarkers for ICS.

ABSTRACT

<p><b>OBJECTIVE</b>To explore the biological function of BMAL1 in human acute myeloid leukemia by means of the HL-60 cell line in whica circadian gene BMAL1 was konocked-out by the CRISPR/Cas9 technology.</p><p><b>METHODS</b>Two sgRNAs for BMAL1 were designed and the PX459 knockout vectors containing the sgRNA were constructed. The activity of 2 sgRNAs was detected by T7 endonuclease I. the BMAL1 knocked out HL-60 cells were prepared by transient transfection of the target vectors into the cells. Western blot was used to detect the expression of BMAL1 protein. The apoptosis of the targeted cells was detected by flow cytometry. The proliferation status of the cells was assessed by the CCK-8 assay.</p><p><b>RESULTS</b>The PX459-sgRNA vectors were successfully constructed and screened to assure the activity of the targeting vector. It was found that the expression of BMAL1 protein was not detected in BMAL1-knocked out HL- 60 cells. Further, it was shown that BMAL1 knockdout could promote the apoptosis of HL-60 cells and inhibit the cell proliferation ability.</p><p><b>CONCLUSION</b>BMAL1 knocked out HL-60 cells have bean successfully established using the CRISPR/Cas9 gene editing technique, and BMAL1 knockout can promote the HL-60 cell apoptosis and inhibit its proliferation.These result reveal the biological role of the BMAL1 circadian gene in acute myeloid leukemia.</p>

ABSTRACT

Phytochemical investigation on Forsythia suspensa (Thunb.) Vahl afforded ten compounds, including five lignan glycosides and five phenylethanoid glycosides. The compounds were isolated by using HP-20 macroporous resin, silica gel, octadecyl silica gel (ODS), size exclusion chromatography resin HW-40 chromatography, and preparative HPLC. The structures were established through application of extensive spectroscopic methods, including ESI-MS, 1D-and 2D-NMR spectroscopy. They were identified as forsythialanside E (1), 8'-hydroxypinoresinol-4'-O-β-D-glucoside (2), 8'-hydroxypinoresinol (3), lariciresinol-4'-O-β-D-glucoside (4), lariciresinol-4-O-β-D-glucoside (5), forsythoside H (6), forsythoside I (7), forsythoside F (8), plantainoside B (9), and plantainoside A (10). Compound 1 was a new lignan glycoside.

ABSTRACT

<p><b>OBJECTIVE</b>To explore the computer aided design (CAD) method of veneer restoration, and to assess if the solution can help prosthesis meet morphology esthetics standard.</p><p><b>METHODS</b>A volunteer's upper right central incisor needed to be restored with veneer. Super hard stone models of patient's dentition (before and after tooth preparation) were scanned with the three-dimensional laser scanner. The veneer margin was designed as butt-to-butt type. The veneer was constructed using reverse engineering (RE) software.</p><p><b>RESULTS</b>The technique guideline of veneers CAD was explore based on RE software, and the veneers was smooth, continuous and symmetrical, which met esthetics construction needs.</p><p><b>CONCLUSIONS</b>It was a feasible method to reconstruct veneer restoration based on RE technology.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To establish a three-dimensional (3-D) descartes coordinates of artificial teeth based on concepts of dental anatomy and to obtain their geometric parameters.</p><p><b>METHODS</b>Using the 3-D reverse engineering software, artificial-tooth coordinates were defined based on their anatomic concepts which included long axis, mesiodistal and buccolingual directions, and occlusal plane. Some geometric parameters, such as mesiodistal and buccolingual dimensions, height and obliquity of dental cusp, were measured.</p><p><b>RESULTS</b>The 3-D coordinates of 28 artificial teeth were established and geometric parameters in coordinates were obtained.</p><p><b>CONCLUSIONS</b>The definition of artificial-tooth coordinates can be made using mathematic language based on dental anatomic concepts, which is the basal work of complete denture computer-aided design (CAD).</p>

ABSTRACT

<p><b>OBJECTIVE</b>This study was to explore a CAD method for constructing occlusal and proximal surfaces of inlays using reverse engineering (RE) software and to develop a CAD software for the inlay fabrication.</p><p><b>METHODS</b>The dentition model of a volunteer with individual normal occlusion was scanned with a 3D mechanical scanner. The scanned objects were prepared teeth with G. V. Black class I, class II and class VI (MOD) cavities, neighbor teeth, intercuspal position (ICP) record, and functional bite record (or functional generate path, FGP). Inlays were constructed based on RE technology using the database of Chinese normal teeth.</p><p><b>RESULTS</b>The technique guideline of inlay CAD was developed based on RE softwares. Inlays designed with CAD technique showed smooth and continuous contours. The occlusal and proximal contacts of inlay met physiologic demands.</p><p><b>CONCLUSIONS</b>It is a feasible method to develop a CAD software for inlay fabrication based on RE software.</p>
