ABSTRACT

Objective:To investigate the effects of modified radical prostatectomy via an extraperitoneal approach on urinary control and sexual function in patients with prostate cancer.Methods:Fifty-six patients with stable prostate cancer who received treatment in Deqing People's Hospital between March 2015 and March 2018 were included in this study. They were randomly divided into observation and control groups ( n = 28/group). The observation group was subjected to modified radical prostatectomy via an extraperitoneal approach. The control group underwent standard laparoscopic surgery. Clinical efficacy and the effects of modified radical prostatectomy via an extraperitoneal approach on urinary control and sexual function were compared between the two groups. Results:Amount of blood loss and postoperative drainage were (125.39 ± 11.12) mL and (65.39 ± 10.12) mL in the observation group, and (224.79 ± 14.01) mL and (104.79 ± 15.01) mL in the control group. There were no significant differences in amount of blood loss and postoperative drainage between the two groups ( t = 18.83, 15.67, both P < 0.05). At 1, 3 and 6 months after surgery, the percentage of patients who had urinary control recovery in the observation group was 53.57% (15/28), 78.57% (22/28), 98.21% (27/28), respectively, which were significantly higher than those in the control group [21.43% (6/28), 35.71% (10/28), 67.86% (19/28), χ2 = 4.12, 7.21, 5.01, all P < 0.05]. At 1, 3 and 6 months after surgery, the score of erectile function recovery in the observation group was (15.98 ± 0.28) points, (15.99 ± 0.72) points, and (18.91 ± 0.48) points, which were significantly higher than those in the control group [(17.11 ± 0.34) points, (13.11 ± 0.48) points, (13.41 ± 0.39) points, t = 3.01, 12.89, 15.78, all P < 0.05]. Conclusion:Modified radical prostatectomy via an extraperitoneal approach can improve postoperative urinary control and sexual dysfunction.

ABSTRACT

Background: Intravenous glucocorticoids (GCs) are the first-line therapy of severe ulcerative colitis (SUC), however, part of the SUC patients are steroids-refractory. Aims: To investigate the predictive factors of therapeutic response to GCs and whether prolonging GCs therapy is beneficial for patients with SUC. Methods: Data of patients with SUC admitted in Anhui Provincial Hospital from Jan. 2006 to Dec. 2017 were collected consecutively. Patients were categorized into different groups according to the responsiveness to intensive intravenous GCs therapy on day 7 and day 14 (in prolonged treatment group), and their general conditions, clinical, laboratory, endoscopic, pathological features and outcomes were compared and analyzed for finding the predictive factors. Results: Complete remission and partial remission was achieved on day 7 in 20 (22.7%) and 43 (48.9%) of the 88 SUC patients, respectively, and the other 25 cases (28.4%) were non-responders. Univariate analysis revealed that defecation >10 times on admission, >6 times on day 3, bloody stool on day 3, pancolitis, and abnormal laboratory findings including ESR and albumin (ALB) on admission, and CRP and CRP/ALB ratio on admission and on day 3, were associated with poor response to GCs (P40 mm/h on admission and defecation >6 times on day 3 were the independent risk factors for treatment failure of GCs (P40 mm/h on admission and defecation >6 times on day 3 might be the independent predictors for poor response to GCs in patients with SUC. A small part of SUC patients with partial remission on day 7 can achieve complete remission by prolonged (14-day) GCs therapy.

ABSTRACT

A series of oxazole[5,4-d] pyrimidine derivatives were designed and synthesized to discover novel compounds with antitumor activity.Compounds 8a-8m were synthesized using acetamidine hydrochloride as the start material.The structures of synthesized compounds were confirmed by IR,1H NMR,EI-MS and elemental analysis.The antiangiogenesis activities of the synthesized compounds were determined by MTT in human umbilical vein endothelial cell (HUVEC).The in vitro antitumor activities of the synthesized compounds were determined by MTT assay in A549,HepG2 and U251.Compounds 8c,8d,8g,8i and 8l were found to inhibit the proliferation of all the tested cell lines.Compound 8l exhibited noteworthy activities in A549,HepG2 and U251 cell lines with IC50value lower than the positive reference sunitinib,suggesting that compound 8l might be the promising antitumor agent for further investigation.

ABSTRACT

AIM:To identify the serum proteins that might serve as biomarkers for predicting mucosal healing ( MH) in the patients with Crohn’ s disease ( CD) treated with infliximab ( IFX) .METHODS:We collected serum sam-ples before treatment (0 week, group A) and 14 weeks after treatment (group B) from 7 CD patients with IFX treatment who had achieved MH, as well as the serum samples from 7 CD patients who had not achieved MH (0 week, group C;14 weeks, group D) .Two-dimensional fluorescence difference gel electrophoresis was applied to analyze and compare the re-sults of serum profiles between groups A and B, C and D, A and C, B and D.Matrix-assisted laser desorption/ionization time-of-flight tandem mass spectrometry and bioinformatics tools were utilized to preliminarily identify and figure out the dif-ferentially expressed proteins.RESULTS:(1) In total, there were 44 differentially expressed spots, 36, 3, 10 and 31 differentially expressed spots were detected while comparing A with B, C with D, A with C and B with D, respectively. (2) Among those spots, 17, 2, 2 and 15 proteins were identified, respectively.In total, there were 19 differentially ex-pressed proteins, including apolipoprotein E, apolipoprotein A-I, complement factor H, and so on.(3) Protein functional association networks were carried out based on STRING database.CONCLUSION: The serum protein profiles obviously change after IFX treatment in MH CD patients, and the serum protein profiles of MH patients are different from that of non-MH patients after IFX treatment.The 19 proteins we identified may serve as potential biomarkers for predicting MH in CD patients with IFX treatment.

ABSTRACT

Objective To investigate the effect of dexmedetomidine on the outcome of rats with acute lung injury following blunt chest trauma.Method Forty male SD rats weighing 250~300 g were randomly (random number)divided into 5 groups(n=8 each),namely,group C(normal),group D(normal plus dexmedetomidine),group T(trauma),group TD(trauma plus dexmedetomidine),group TDY(trauma plus dexmedetomidine plus yohimbine).Au rats were sacrificed by using exsanguination from arteria femoralis 6 hours later.The TNF-α and IL-1β levels in plasma were examined by using ELISA.Lung wet/dry(W/D)weight and the percentage of neutrophil in leucocytes in bronehoalveolar lavage fluid(BALF)of rats were detected.HE staining was performed to observe the injury of lung tissue under microscope.Results There was significant lung injury after blunt chest trauma.The serum concentrations of TNF-α and IL-1β,PMN%and lung wet/dry(W/D)weight were significantly higher in traumatic group(P<0.05,P<0.01).chest trauma,but this protective effect of dexmedetomidine could be blocked by yohimbine,at least in part,via the inhibition of α2-adrenergie receptor.Conclusions Dexmedetomidine has a certain protective effect on acute traumatic acute lung injury in rats.

ABSTRACT

Aim To study the effect of chromomycin on MPP~+-induced dopaminergic neuronal apoptosis.Methods An apoptosis model induced by 1-methyl-4-phenylpridnium ion(MPP~+)was established in cultured fetal mesencephalic neurons in vitro.Dopaminergic neuronal apoptosis was detected by Hoechst 33258 staining and phospho-Tau levels were detected by immunofluorescence.Results 10 μmol·L~(-1) MPP~+ hyperphosphorylated Tau at Ser396 and induced dopaminergic neuronal apoptosis.Chromomycin increased the number of TH-positive cells via inhibiting Tau phosphorylation.Conclusion These results indicate thatchromomycin inhibits Tau phosphorylation at Ser396 and therefore protects dopaminergic mesencephalic neurons from apoptosis induced by MPP+,which suggests that Chromomycin may be used to cure neurodegenerative disorders such as Parkinson's disease in clinic.

ABSTRACT

[Objective]To apply an acetonitrile-pretreatment method for proteomic analysis of serum of patient with hepatocellular carcinoma(HCC),searching for the proteins that might be related to occurrence and development of HCC.[Methods]Serum samples were recruited from 12 patients with primary HCC and 12 health adult.Firsdy,the sera were pretreated with acetonitrile (ACN),and the high-abundance proteins were removed,then two-dimensional electrophoreaia(2-DE)was performed to screen the differentially expressed proteins between the patient with HCC and health control.Finally,the differentially expressed proteins in the 2-DE gel were identified by a MALDI-TOF/TOF mass spectrometry.[Results]After the sera were pretreated with 0%,20% and 30% ACN,2-DE analysis showed that the number of the protein spots in the gels were 532±96,623±102,and 674±123,respectively.By the proteomic analysis of the ACN-pretreatment sera of patient with HCC and health control,we found that tetraiodo-L-thyronine and proapolipoproteinin were up-regulated,vitamin D-binding protein precurser and transferrin were downregulated in the HCC serum.[Conclusion]Proteomic analysis of serum using the ACN-pretreatment method can improve the detection of proteins which were non-specifically binding to the albumin.Abnormal expression of tetraiodo-L-thyronine,proapolipoproteinin,vitamin D-binding protein precursor and transferrin in the serum are related to HCC.

ABSTRACT

Objective To explore disease-associated proteins in the serum of patients with inflammatory bowel disease by serum proteomic analysis combined with mixed sampling strategy. Methods The serum proteins from 8 healthy adults and 8 patients with inflammatory bowel disease who had been admitted to the Sixth Affiliated Hospital of Sun Yat-sen University from March 2007 to June 2008 were collected. Two-dimensional differential in-gel electrophoresis (2D-DIGE) was used to define patterns of protein expression after the serum proteins were cross-labeled with cariant CyDye. Proteins that showed differential expressions were analyzed by matrix-assisted laser dcsorption/ionization time of flight mass spectrometry. The 2D-DIGE images were analyzed using DeCyder V6.0 software, and the differences between the groups were analyzed by t-test. Results Maps of 2D-DIGE of patients with inflammatory bowel disease and healthy adults were obtained. Fifty-six spots of proteins with abnormal expression were detected in patients with inflammatory bowel disease, and 30 proteins were identified using mass spectrometry and database retrieval. The 30 proteins included haptoglobin, complement factor B, apelipoprotein A- Ⅱ precursor and GTPase K-ras. Conclusions Serum proteomic analysis combined with mixed sampling strategy can clearly detect the difference in the expression of serum proteins between patients with inflammatory bowel disease and healthy adults. The differentially expressed proteins may provide new biornarkers for investigating the biological behavior of inflammatory bowel disease.

ABSTRACT

Objective To explore the safety and invasion of endoscopic thyroidectomy(ET).Methods A total of 156 cases of benign thyroid diseases were surgically treated from January 2003 to December 2005,including 64 cases of endoscopic thyroidectomy(Endoscopic Group) and 92 cases of traditional thyroidectomy(Traditional Group).The operation time,hemorrhage volume,length of incision,Visual Analogue Scale(VAS) scores for pain severity and cosmetic appearance,analgesic requirements,postoperative hospital stay,hospitalization costs,wound infection rate,and recurrent rate were compared between the two groups,respectively.Results The operation was successfully completed in both of groups.No hemorrhage,hoarseness,or hypocalcemia was encountered.Wound hydrops occurred in 4 cases in the Traditional Group.As compared with the Traditional Group,the Endoscopic Group had less blood loss(15?10 ml vs 20?15 ml;t=-2.330,P=0.021),lower VAS pain scores(2.5?1.0 vs(5.0?)1.6;t=-11.079,P=0.000),higher VAS cosmetic scores(8.0?1.5 vs 2.0?1.2;t=27.696,P=0.000),and less analgesic requirements(10 cases vs 28 cases;?~2=4.493,P=0.034),but longer operation time(80?20 min vs 70?30 min;t=2.330,P=0.021) and higher hospitalization costs(5 352.7?978 yuan vs 4 738.5?672 yuan;t=4.651,P=0.000).Follow-up observations for 2~30 months(mean,13 months) revealed recurrence in 2 cases in the Endoscopic Group and 3 cases in the Traditional Group.Conclusions As compared with traditional operation,endoscopic thyroidectomy has advantages of minimal invasion,early recovery,short hospitalization time,and high safety,being an ideal option for benign thyroid diseases.

ABSTRACT

【Objective】To study the effect of the specific p38 mitogen-activated protein kinase(p38 MAPK) inhibitor SB203580 on apoptosis of cerebellar granule neurons induced by low potassium.【Methods】Apoptosis was induced by switching the cultured cerebellar granule neurons from a culture medium containing K+ 25 mmol*L-1 to a medium containing K+ 5 mmol*L-1 (cLK).Fragmentation of DNA was analyzed using agarose gel eletrophoresis.SAPK/JNK activity was measured by SAPK/JNK assay kit.【Results】Low potassium resulted in apoptosis as characterized by morphological and biochemical features,but the specific p38 MAPK inhibitor SB203580 improved the survival of cerebellar granule neurons cultured in cLK medium by blocking apoptosis in a concentration-dependent manner.The expression and phosphorylation of c-Jun increased and the activity of c-Jun N-terminal protein kinase (JNK) elevated when cerebellar granule neurons were cultured in cLK medium.But when the cerebellar granule neurons cultured in cLK medium were exposed to 25 μmol*L-1 SB203580,the expression and phosphorylation of c-Jun and the activity of JNK were both decreased evidently.【Conclusions】These results indicate that SB203580 inhibits the activation of JNK and phosphorylation of c-Jun,and therefore protects granule neurons from apoptosis induced by low potassium.

ABSTRACT

Objective To explore the feasibility and reliability of laparoscopic assisted thyroidectomy. Methods A transversal 2.0 cm incision was made at the site 1.0 cm above the sternal notch. By using a harmonic scalpel under a 5 mm laparoscope, the thyroid glands and involving blood vessels were dealt with by either “coagulation-dissection-suction” technique or “dissection-coagulation-disconnection” technique to complete thyroid enucleation or lobectomy. Results There were 20 cases of adenoma (nodule) excision and 6 cases of unilateral lobectomy. The operation time was 80~130 min (mean, 100 min). The intraoperative blood loss was 10~40 ml (mean, 20 ml). No surgical complications occurred. Follow-up checkups in 23 cases for 1~)20 months (mean, 9 months) found a satisfactory cosmetic result and no recurrence. Conclusions Thyroidectomy under laparoscope is safe and reliable. This procedure offers a shorter incision, less invasion and better cosmetic results as compared with conventional thyroidectomy.
