ABSTRACT

The period detection of the biological rhythms is one ofthe hot topics in the research of chronobiology,which will help to understand the mechanism of pathological changes and how the function of the clock genes affect the organism and thus guide clinical drug administration and treatment timely.Some new methods for detecting the periods of the biological rhythms,including Lomb-Scargle periodogram,spectral analysis based on Cosinor method(e.g.Percent Rhythm Spectrum and Reverse EHiptie Spectrum),and the Maximum Entropy Spectral Analysis(MESA),ale introduced in this paper.Advantages and disadvantages of the methods ale reviewed and some suggestions to the research in the future are also proposed.These methods provide promising approaches for detecting the precise periods of the biological rhythms and discovering the rule of the changes of the biological rhythms.

ABSTRACT

Objective To investigate the circadian rhythm of peripheral blood cell counts in Chinese healthy volunteers and nasopharyngeal carcinoma patients. Methods A total of 13 nasopharyngeal carcinoma patients and 14 healthy volunteers were enrolled in this study. Venous blood was sampled every 4 hours during a 24-hour period. EDTA anticoagulated whole-blood was tested for blood cell counts immediately by automated hematology analyzer. The plasma cortisol level was determined by Radioimmunoassay. Data were analyzed by Analysis of Variance for Repeated Measures using SPSS 13.0 software and by Cosinor Analysis using Cosinor Analysis Software. Results Both healthy volunteers and nasopharyngeal carcinoma patients displayed clear circadian rhythms in red blood cells, hemoglobin, leukocytes, lymphocytes, and possibly platelets. Those blood cell counts also showed a significant variation during 24h. Conclusion Peripheral blood cell counts display significant circadian rhythms in both healthy volunteers and advanced nasopharyngeal carcinoma patients, suggesting the possibility of performing the time-selective collection and transfusion of blood.

ABSTRACT

Objective To isolate, clone and identify the acetylcholinesterase (AChE) fragment from the mosquito, Aedes albopictus, in relation to exploring mechanism of insecticide resistance. \ Methods\ The genome DNA extracted from the mosquito was used for degenerate polymerase chain reaction (PCR) and the two pairs of oligonucleotides encoding the highly conserved protein sequences were used as primers. The reaction products were cloned to T\|vector and transfected into E^coli JM 109. The replicative form DNA of recombinant vector extracted from E^coli JM 109 through alkalilysis was identified by the methods of digestion with EcoRⅠand SalⅠ and PCR. \ Results\ The products of degenerate primers polymerase chain reaction were obtained and the identified clone belongs to the AChE fragment of the mosquito.\ Conclusion\ The clone was identified as the AChE fragment of Aedes albopictus.
