ABSTRACT

Plant growth promotion by microorganisms may be a viable alternative to increase lettuce production through pathogens control and nutrients absorption increase. Trichoderma and Pseudomonasgenus are examples of widely studied microorganisms with the capacity to promote plant growth. However, there are still gaps regarding the action of the combined effect of these two microorganisms. Therefore, the objective of this study was to evaluate the combined effect of Pseudomonas sp. UAGF14 and Trichoderma aureoviride URM5158 on the development of lettuce plants. The experimental design was completely randomized, with five treatments: CONT (control), CM (soil with organic fertilization), CMB (soil withorganic fertilization and Pseudomonas sp.), CMF (soil with organic fertilization and T. aureoviride), and CMFB (soil with organic fertilization, Pseudomonas sp. and T. aureoviride), with ten repetitions. At 30, 40 and 60 days after sowing, the following parameters were analyzed: plant and canopy height and number of leaves. At 60 days after emergence, shoot dry matter, leaf area, root dry matter, root length and chlorophyll were analyzed. Catalase, peroxidase and polyphenol oxidase enzymatic activity were determined. The CMFB treatment had the highest means of lettuce growth promotion, confirming the synergistic effect of the combination of the two microorganism types, as it increased height, canopy, shoot and root dry matter, and chlorophyll levels compared to CONT, although did not differ from CM in some variables. Enzymatic activity was also influenced by the action of these microorganisms combined, evidencing by polyphenol oxidase increase. The CMFB or CM were efficient in promoting lettuce growth, showing positive response to the plant morphological and physiological characteristics. However, few responses were observed in lettuce plant growth in the first cycle evaluated after 60 days, compared CM and CMFB treatments, but both treatments showed superiority in lettuce plant growth submitted to CONT treatment. Therefore, further studies are needed to estimate the long-term effects of combined effect of Pseudomonas sp. UAGF14 and T. aureoviride URM5158 on crop productivity in field conditions.


A promoção do crescimento das plantas por micro-organismos pode ser uma alternativa viável para aumentar a produção de alface através de controle de patógenos e aumento da absorção de nutrientes. O gênero fúngico Trichoderma e o gênero bacteriano Pseudomonas são exemplos de micro-organismos amplamente estudados com capacidade para promover o crescimento da planta. No entanto, ainda existem lacunas quanto à ação do efeito combinado desses dois micro-organismos. Portanto, o objetivo deste estudo foi avaliar o efeito combinado de Pseudomonas UAGF14 e Trichoderma aureoviride URM5158 sobre o desenvolvimento de plantas de alface. O delineamento experimental foi completamente casualizados, com cinco tratamentos: CONT (controle, sem fertilização orgânica), CM (solo com fertilização orgânica), CMB (solo com fertilização orgânicae Pseudomonas sp.), CMF (solo com fertilização orgânica e T. aureoviride) e CMFB (solo com fertilização orgânica, Pseudomonas sp. e T. aureoviride), com dez repetições. Aos 30, 40 e 60 dias após a semeadura, foram analisados os seguintes parâmetros: altura da planta e dossel e número de folhas. Aos 60 dias após a emergência, a matéria seca da parte aérea, a área foliar, a massa seca das raízes, o comprimento radicular e a clorofila foram analisados. Catalase, peroxidase e atividade enzimática da polifenol oxidase foram determinadas. O CMFB apresentou o maior crescimento de alface, confirmando o efeito benéfico da combinação dos dois tipos de micro-organismos com a planta, na medida em que aumentou a altura, o dossel, a matéria seca da parte aérea e da raiz, e os níveis de clorofila em relação ao CONT, embora não tenha diferido do CM em algumas variáveis. As atividades enzimáticas também foram influenciadas pela ação desses micro-organismos combinados, evidenciada pelo aumento de polifenol oxidase. O CMFB ou CM foram eficientes na promoção do crescimento da alface, mostrando respostas positivas às características morfológicas e fisiológicas. Entretanto, poucas respostas foram observadas no crescimento da alface noprimeiro ciclo da planta avaliado depois de 60 dias, comparando os tratamentos CM e CMFB, mas ambos tratamentos mostraram superioridade em relação ao crescimento das plantas de alface submetidas ao tratamento controle. Por isso, são necessários futuros estudos para estimar à longo prazo o efeito combinado de Pseudomonas sp. UAGF14 e Trichoderma aureoviride URM5158 na produção de cultura em condições de campo.

Abstract Protein hydrolysates originating from egg white have already been reported to be bioactive and, among their biological activities, possess the antioxidant property that protects the body from early ageing and diseases linked to oxidation. Therefore, the objective of this work was to evaluate the antioxidant activity of hydrolysates obtained by the hydrolysis of egg white from hen poultry. The protease produced by Aspergillus avenaceus URM 6706 was purified and subsequently applied to hydrolysate the egg white, and the degree of hydrolysis was verified during the protease exposure time (4-24 h). The hydrolysis was intensified over time of exposure to the protease. It was possible to detect the antioxidant activities of eliminating the 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) radical (ABTS•+) from 97% to 99% and 2,2-diphenyl-1-picrylhydrazyl (DPPH•) up to 27%, as well as the chelation of Cu2+ metal ions up to 62% and Fe2+ up to 54%. The elimination of ABTS•+ radical had a positive correlation with the degree of hydrolysis; however, all the other activities tested showed a negative correlation with the degree of hydrolysis. The results obtained suggest that the egg white of hen chicken represents a food source of animal origin with potential application in the functional food industry.

Abstract Aspergillus fumigatus is an opportunistic saprobe fungus that accounts for 90% of cases of pulmonary aspergillosis in immunosuppressed patients and is known for its angiotropism. When it reaches the respiratory tract, A. fumigatus interacts with structural components and blood vessels of the lungs, such as elastin. To understand the effect of this structural component, we examined the effect of elastin on the production and development of the biofilm of A. fumigatus. In RPMI containing 10 mg/mL of elastin, a significant increase (absorbance p < 0.0001; dry weight p < 0.0001) in the production of biofilm was observed in comparison to when RPMI was used alone, reaching a maximum growth of 18.8 mg (dry weight) of biofilm in 72 h. In addition, elastin stimulates the production (p = 0.0042) of extracellular matrix (ECM) and decreases (p = 0.005) the hydrophobicity during the development of the biofilm. These results suggest that elastin plays an important role in the growth of A. fumigatus and that it participates in the formation of thick biofilm.

ABSTRACT

Os dermatófitos são fungos que podem causar infecções superficiais da pele, cabelo e unhas em humanos e animais. As espécies de dermatófitos mais frequentemente isoladas dos cães e gatos afetados por micoses são Microsporum gypseum e principalmente Microsporum canis. O papel crucial durante o processo de infecção é a produção de enzimas extracelulares essenciais para a invasão e estabelecimento do agente patogênico no tecido do hospedeiro. O objetivo deste trabalho foi isolar dermatófitos de cães e gatos e avaliar o perfil enzimático dos isolados obtidos. Amostras de pelos e escamas epidérmicas foram coletadas de cães e gatos em instalações veterinárias em Recife/PE, e os isolados foram identificados com base nas características macroscópicas e microscópicas. A análise qualitativa das enzimas urease, protease, lipase, colagenase e fosfolipase foi avaliada a partir dos dermatófitos isolados. Durante 10 meses, um total de 106 animais, que compreendeu de 99 cães e sete gatos com sinais clínicos, independentemente do sexo e raça foram avaliados. Apenas oito animais foram confirmados com dermatofitose, principalmente cães (n=7), sendo seis afetados por M. canis e um por M. gypseum, a raça mais afetada foi Yorkshire (n=3). No entanto, apenas um gato foi confirmado com M. canis. Não foi observada predisposição relacionada ao sexo quanto à ocorrência de dermatofitose nos cães e gatos avaliados. Os dermatófitos isolados apresentaram perfis semelhantes para as enzimas urease, lipase, protease, fosfolipase e colagenase, característica importante em infecções patogênicas. O diagnóstico clínico destas zoonoses com base na confirmação microbiológica e uma compreensão dos mecanismos subjacentes é de grande importância para o tratamento e prevenção de doenças fúngicas em animais.(AU)


Dermatophytes are fungi that can cause superficial infections of the skin, hair and nails in man and animals. The most frequent dermatophyte species isolated from dogs and cats are Microsporum gypseum, most notably Microsporum canis. The crucial role during the infection process is the production of extracellular enzymes essential for the invasion and establishment of the pathogen in the host tissue. The objective of this research was to isolate dermatophytes from dogs and cats and evaluate the enzymatic profile of the isolates obtained. Hair samples and epidermal scales were collected from dogs and cats in veterinary facilities in Recife-PE, and the isolates were identified based on macroscopic and microscopic characteristics. The qualitative analysis of the enzymes urease, protease, lipase, collagenase and phospholipase was evaluated from the isolated dermatophytes. During 10 months, a total of 106 animals, comprising of 99 dogs and seven cats with clinical signs, regardless of sex and race were evaluated. Only eight animals were confirmed with dermatophytosis, mostly dogs (n=7), being six affected by M. canis and one by M. gypseum, the race most affected was Yorkshire (n=3). However, only one cat was confirmed with M. canis. No sex-related predisposition was observed regarding the occurrence of dermatophytosis in dogs and cats evaluated. Isolated dermatophytes showed similar profiles for the enzymes urease, lipase, protease, phospholipase and collagenase, important characteristic for pathogenic infections. The diagnosis of this zoonosis based on microbiological confirmation and a better understanding of the underlying mechanisms is of great importance for the treatment and prevention of fungal diseases in animals.(AU)

ResumenEl suelo es un sistema biológico complejo, que desempeña un papel fundamental en las plantas y los animales, especialmente en los bosques secos como la Caatinga. Los hongos del suelo, tales como Aspergillus y Penicillium, pueden ser utilizados como bioindicadores para la conservación de la biodiversidad. El objetivo de este estudio fue aislar e identificar las especies de Aspergillus y Penicillium del suelo, en los municipios de Ibimirim y Tupanatinga en el Parque Nacional Catimbau. Cinco colecciones se llevaron a cabo en cada área durante la estación seca de 2012, un total de 25 muestras de suelos por área. Los hongos fueron aislados mediante la suspensión en agua destilada estéril y se sembraron en medio de cultivo Agar Sabouraud más Cloranfenicol y Rosa de Bengala, y también en el medio Agar Dicloran Glicerol. Los aislamientos fueron identificados en el Laboratorio de Colección de Hongos y se confirmaron por secuenciación del espaciador transcrito interno de ADN. Un total de 42 especies fueron identificadas, 22 de ellas pertenecientes al género Aspergillus y 20 al género Penicillium. Los aislamientos de Penicillium mostraron una distribución uniforme en Tupanatinga con índices de uniformidad entre 0.92 y 0.88 en Ibimirim. Entre los aislamientos de Aspergillus el valor encontrado en Tupanatinga (0.85) fue muy similar al encontrado en Ibimirim (0.86). Se observó una gran diversidad y bajo predominio de hongos en las muestras de suelo. Estos resultados contribuyen a la estimación de la diversidad de hongos en ambientes secos, especialmente en la Caatinga, donde la diversidad es decreciente en los suelos que han sufrido alteraciones.


Abstract Soil is a complex biological system that plays a key role for plants and animals, especially in dry forests such as the Caatinga.Fungi from soils, such as Aspergillus and Penicillium, can be used as bioindicators for biodiversity conservation. The aim of this study was to isolate and identify species of Aspergillus and Penicillium in soil, from the municipalities of Tupanatinga and Ibimirim, with dry forests, in the Catimbau National Park. Five collections were performed in each area during the drought season of 2012, totaling 25 soil samples per area. Fungi were isolated by suspending soil samples in sterile distilled water and plating on Sabouraud Agar media plus Chloramphenicol and Rose Bengal, and Glycerol Dicloran Agar. Isolates were identified by morphological taxonomy in the Culture Collection Laboratory and confirmed by sequencing of the Internal Transcribed Spacer of rDNA. A total of 42 species were identified, of which 22 belong to the genus Aspergillus and 20 to Penicillium. Penicillium isolates showed uniform distribution from the collecting area in Tupanatinga, and the evenness indices found were 0.92 and 0.88 in Tupanatinga and Ibimirim, respectively. Among isolates of Aspergillus evenness, the value found in Tupanatinga (0.85) was very close to that found in Ibimirim (0.86). High diversity and low dominance of fungi in soil samples was observed. These results contributed to the estimation of fungal diversity in dry environments of the Caatinga, where diversity is decreasing in soils that have undergone disturbance. Rev. Biol. ABSTRACT

Bauhinia forficata is native to South America and used with relative success in the folk medicine in Brazil. The diversity, antibacterial activity, and extracellular hydrolytic enzymes of endophytic fungi associated with this plant were studied. Plant samples, which included leaves, sepals, stems, and seeds, were used. Ninety-five endophytic fungal were isolated (18 from leaves, 22 from sepals, 46 from stems, and nine from seeds), comprising 28 species. The most frequently isolated species were Acremonium curvulum (9.5%), Aspergillus ochraceus (7.37%), Gibberella fujikuroi (10.53%), Myrothecium verrucaria (10.53%) and Trichoderma piluliferum (7.37%). Diversity and species richness were higher in stem tissues, and Sorensen’s index of similarity between the tissues was low. Eleven fungi showed antibacterial activity. Aspergillus ochraceus, Gibberella baccata, Penicillium commune, and P. glabrum were those with the greatest antibacterial activity against Staphylococcus aureus and/or Streptococcus pyogenes. Thirteen species showed proteolytic activity, particularly Phoma putaminum. Fourteen species were cellulase positive, particularly the Penicillium species and Myrmecridium schulzeri. All isolates tested were xylanase positive and 10 showed lipolytic activity, especially Penicillium glabrum. It is clear that the endophytic fungi from B. forficata have potential for the production of bioactive compounds and may be a source of new therapeutic agents for the effective treatment of diseases in humans, other animals, and plants. To our knowledge, this is the first study of endophytic fungi from different tissues of B. forficata and their biotechnological potential.

ABSTRACT

The current study evaluated the proteases production from 11 fungal species belonging to the genera Mucor, Rhizomucor and Absidia. The species were obtained from the Collection of Cultures URM at the Mycology Department-UFPE, Brazil. The best producing species was Mucor hiemalis URM 3773 (1.689 U mL-1). Plackett-Burman design methodology was employed to select the most effective parameter for protease production out of 11 medium components, including: concentration of filtrate soybean, glucose, incubation period, yeast extract, tryptone, pH, aeration, rotation, NH4Cl, MgSO4 and K2HPO4. Filtrated soybean concentration was the significant variable over the response variable, which was the specific protease activity. The crude enzyme extract showed optimal activity in pH 7.5 and at 50ºC. The enzyme was stable within a wide pH range from 5.8 to 8.0, in the phosphate buffer 0.1M and in stable temperature variation of 40-70ºC, for 180 minutes. The ions FeSO4, NaCl, MnCl2, MgCl2 and KCl stimulated the protease activity, whereas ZnCl2 ion inhibited the activity in 2.27%. Iodoacetic acid at 1mM was the proteases inhibitor that presented greater action.The results indicate that the studied enzyme have great potential for industrial application.


Foi avaliada a produção de proteases por 11 espécies fúngicas pertencentes aos gêneros Mucor, Rhizomucor e Absidia, obtidas da Coleção de Culturas URM do Departamento de Micologia- UFPE, Brasil. A melhor espécie produtora foi Mucor hiemalis URM3773 (1,689 U mL-1). A metodologia de planejamento Plackett-Burman foi empregada para selecionar o parâmetro mais efetivo para a produção de proteases através de 11 componentes do meio, incluindo: concentração do filtrado de soja, glicose, período de incubação, extrato de levedura, triptona, pH, aeração, rotação, NH4Cl, MgSO4 e K2HPO4. A variável significante sobre a variável- resposta, atividade proteásica específica, foi a concentração do filtrado de soja. O extrato enzimático bruto apresentou atividade ótima ao pH 7,5 a 50ºC. A enzima foi estável em uma ampla variação de pH de 5,8­8,0 em tampão fosfato 0,1M e termicamente estável a uma variação de 40-70°C, durante 180 minutos. Os íons FeSO4, NaCl, MnCl2, MgCl2 e KCl estimularam a atividade proteásica, enquanto que o íon ZnCl 2 inibiu 2,27% da atividade. O inibidor de proteases que teve maior ação foi o ácido iodoacético a 1mM. Os resultados obtidos indicam que a enzima estudada tem grande potencial de aplicação industrial.

ABSTRACT

The aim of this study was to evaluate peptidase production by Aspergillus terreus in solid-state bioprocess and evaluate its parameters. The best conditions were 5.0 g of wheat bran as substrate, incubation temperature 30°C, inoculum 2.0x105 spores/g and 75% saline volume, with production reaching 677 U/mL (5400 U/g culture medium) after 72 h of fermentation. Biochemical characterization of the crude enzymatic extract showed the optimum pH and temperature of 6.5 and 55°C, respectively. The stability at different temperatures and pH values showed that the extract could endure different pH. The evaluation of the ions influence and inhibitors proved that the enzyme required an ion for better activity, which was corroborated with the inhibition of EDTA and PMSF, characterizing serine and/or metallo peptidase. The extract was also tested for specific activities and showed promising results for keratinolytic and collagenolytic activities (0.252 and 0.165 OD/mL, respectively).

ABSTRACT

Endophytic microorganisms from the base and top leaves of Spermacoce verticillata were isolated and the antimicrobial potential was evaluated. A total of 56 strains were isolated in pure culture, 44 fungi and 12 actinobacteria. The isolation frequency was higher in the base leaves (12.5%), when compared to the top leaves (3.05%). Among all fungi and actionobacteria identified, the majority belonged to the genus Guignardia (25%) and Microbispora (41.66%), respectively. The antimicrobial screening was firstly evaluated by agar plug assay and showed that 28.57% of the isolates presented activity mainly against gram positive bacteria Staphylococus aureus (ATCC-6538) and Bacillus subtilis (UFPEDA-16). The microorganisms that presented the best activities were then selected, evaluated in different culture media broth, and tested by disk diffusion assay using their fermented broth. The microorganisms selected for this assay exhibited antimicrobial activity mainly for Bacillus subtilis (UFPEDA-16). Since many isolates showed inhibitory activity against pathogenic microorganisms, it is suggestive that endophytic microorganisms from S. verticillata could be an interesting source to explore for bioactive metabolites and new tools need to be employed to explore the real potential of these microorganisms.

ABSTRACT

Enzymes obtained by fermentation processes offer a number of advantages and have been widely researched and used throughout the world. This study aimed to partially characterise an inulinase produced from palm and cassava peel. The enzyme was produced via the solid-state fermentation of Aspergillus japonicus URM5633. The optimal temperatures were 50ºC and 55ºC, and the optimal pH values were 5.2 and 3.4 for inulinase fermentatively produced from palm and cassava peel, respectively. The thermostability measurements for inulinase produced in palm showed that the relative activity remained below 100% until 30 minutes of stability for all temperatures, but reached 106.8% at a temperature of 50ºC after 60 minutes. Inulinase from the crude extract of cassava peel was pH stable and only decreased to 55% of the maximal activity over the course of the assay, suggesting that this enzyme can be used in inulinase production and can be utilized in food industries.

ABSTRACT

Lectins are carbohydrate-binding proteins of non-imune origin. This group of proteins is distributed widely in nature and they have been found in viruses, microorganisms, plants and animals. Lectins of plants have been isolated and characterized according to their chemical, physical-chemical, structural and biological properties. Among their biological activities, we can stress its fungicidal action. It has been previously described the effect of the lectins Dviol, DRL, ConBr and LSL obtained from the seeds of leguminous plants on the growth of yeasts isolated from vaginal secretions. In the present work the experiments were carried out in microtiter plates and the results interpreted by both methods: visual observations and a microplate reader at 530nm. The lectin concentrations varied from 0.5 to 256µg/mL, and the inoculum was established between 65-70% of trammitance. All yeast samples isolated from vaginal secretion were evaluated taxonomically, where were observed macroscopic and microscopic characteristics to each species. The LSL lectin did not demonstrate any antifungal activity to any isolate studied. The other lectins DRL, ConBr and DvioL, showed antifungal potential against yeast isolated from vaginal secretion. These findings offering offer a promising field of investigation to develop new therapeutic strategies against vaginal yeast infections, collaborating to improve women's health.

ABSTRACT

Litopenaeus vannamei, which is the most common shrimp species cultivated in the northeast of Brazil, is very susceptible to microbial diseases, and this consequently affects productivity. There are reports of bacteria, viruses and protozoa in these shrimp, but not fungi. This study aims to isolate and identify fungi present in shrimp Litopenaeus vannamei, and in their nursery waters, at two breeding farms in Brazil. The pathogenic potential of the isolates was assessed through the qualitative detection of proteases and aflatoxin B production. The 146 isolated fungi comprised 46 species. Aspergillus, Penicillium and Furarium were the three most relevant genera and Aspergillus flavus was the predominant species with a total of 33 isolates. Most of the isolated species are known as potentially pathogenic to humans and other animals. Eighteen isolates of A. flavus and two of A. parasiticus were able to produce aflatoxin B and 33 out of the 46 species produced protease, indicating that these fungi may also become pathogenic to shrimp and their consumers.

INTRODUÇÃO: Onicomicoses são comuns em pacientes imunocomprometidos embora espécies emergentes tenham sido verificadas, modificado o perfil epidemiológico desta micose. Assim, o objetivo desta pesquisa é avaliar o perfil clínico e micológico da onicomicose em pacientes com infecção pelo HIV/AIDS. MÉTODOS: Amostras clínicas foram coletadas, processados para exame direto e a cultura mantida a temperatura de 30°C e 37ºC durante 15 dias. RESULTADOS: Dos 100 pacientes, 32 apresentavam onicomicose. Os agentes isolados foram Candida albicans, C. parapsilosis, C. tropicalis, C. guilliermondii, Trichophyton rubrum, T. mentagrophytes, Fusarium solani, Scytalidium hialinum, S. japonicum, Aspergillus niger, Cylindrocarpon destructans e Phialophora reptans. CONCLUSÕES: Onicomicoses em HIV/AIDS apresentam variadas manifestações clínicas e podem ser causadas por fungos emergentes. As peculiaridades apresentadas pelos diferentes agentes de origem fúngica justificam a necessidade de identificação ao nível da espécie, com a finalidade de orientar uma melhor abordagem terapêutica e minimizar a exposição desses pacientes a condições de risco de uma infecção disseminada.


INTRODUCTION: Onychomycosis is common in immunocompromised patients, but emerging species have been verified, thereby modifying the epidemiological profile of this mycosis. Thus, the aim of this study was to evaluate clinical and mycological profile of onychomycosis among HIV/AIDS patients. METHODS: Clinical samples were collected and processed for direct examination, and cultures were maintained at a temperature of 30°C and 37°C for 15 days. RESULTS: Out of 100 patients, 32 had onychomycosis. The etiological agents isolated were Candida albicans, C. parapsilosis, C. tropicalis, C. guilliermondii, Trichophyton rubrum, T. mentagrophytes, Fusarium solani, Scytalidium hialinum, S. japonicum, Aspergillus niger, Cylindrocarpon destructans and Phialophora reptans. CONCLUSIONS: Onychomycosis in HIV/AIDS patients presents various clinical manifestations and may be caused by emerging fungi. The peculiarities presented by different fungal agents justify the need for identification to species level, with the purpose of guiding better therapeutic approaches and minimizing these patients' exposure to conditions presenting a risk of disseminated infection.

No presente trabalho foi investigada a ocorrência de fungos de importância médica na água, torneira e pia dos bebedouros do Centro de Ciências Biológicas da Universidade Federal de Pernambuco, Recife, Brasil. De cada bebedouro foram coletados 100 mL de água, e das superfícies da torneira e pia as amostras foram obtidas com swab. As amostras foram semeadas em meio ágar Sabouraud adicionado de 50mg/L de cloranfenicol, em placas de Petri. Das amostras de água, 10 culturas de fungos filamentosos foram isoladas e estas foram identificadas como Aspergillus flavus (5), A. niger (3) e Cladosporium cladosporioides (2). Das torneiras foram obtidos 21 isolados de fungos filamentosos e 164 das amostras das pias. O número de unidades formadoras de colônias (UFCs) (48h) variou de 12 a 56 em amostras de água, de 115 a 228 em amostras coletadas das torneiras e de 204 a 690 das pias. A presença de fungos com capacidade de produzir micotoxinas foi detectada nas amostras de água, bem como das torneiras. Com a finalidade de preservar a saúde dos consumidores, o monitoramento e a manutenção de equipamentos de purificação da água são práticas essenciais para garantir a qualidade microbiana e química da água.


This study aimed to assess the occurrence of fungi in water sample, tap and sink from drinking fountains atthe university center, Recife-PE, Brazil. From each fountain, 100mL of water were collected, and the samplesfrom the tap and sink surfaces were obtained by using swabs. The samples were cultivated on Sabouraud agarmedium containing 50mg/L of chloramphenicol in Petri dishes. Of water samples, 10 isolates of filamentousfungi were found and identified as Aspergillus flavus (5), A. niger (3) and Cladosporium cladosporioides (2).Twenty-one strains of filamentous fungi were isolated from tap samples and 164 from sink samples. Thenumber of colony forming units (CFU) (48h) ranged from 12 to 56 in water samples, from 115 to 228 insamples collected from taps, and 204 to 690 in sink samples. Fungi capable of producing mycotoxins weredetected in both water and taps samples. Monitoring the microbial and chemical quality of water and themaintenance of water purification equipment are crucial to guarantee the consumers safety.Key-words. filamentous fungi, water, drinking fountain, microbial water quality, mycotoxins.

ABSTRACT

Este trabalho teve como objetivo isolar e identificar os fungos queratinofílicos do solo e correlacioná-los com os agentes de dermatomicose. De acordo com nossos resultados, o predomínio de Trichophyton tonsurans como agente de dermatomicose em crianças na cidade do Recife, PE deve-se provavelmente ao maior contato destas com o solo.


This study had the aim of isolating and identifying keratinophilic fungi from soil and correlating them with dermatomycosis agents. According to our results, the predominance of Trichophyton tonsurans as a dermatomycosis agent among children in the city of Recife, PE, is probably due to children's greater contact with soil.

ABSTRACT

This work aimed at the assessment of fermentative capacity of original diploid, monocellular haploid and recuperated diploid cultures of S. cerevisiae in sterilized sugar-cane wort. Twenty eight cultures were analyzed, four being original diploids (URM-4420, Itaiquara Ferment FIT, Lallemand Ferment FLA and Wild Ferment SEL); 12 monocellular haploids from original ones and 12 recuperated diploids from the monocells. The ethanol percentage ranged from 1.7 to 6.2 percent and the unfermentable reducing sugars from 0.45 to 0.50g/100mL. The highest ethanol percentages were produced by the monocellular cultures URM-MH3 (4.8 percent) in 12 h and SEL-MH1 (6.2 percent) in 24 h, corresponding to the productivity values of 3.15 and 2.03 g.L-1.h-1, respectively. The recuperated diploids did not present detectable ethanol content by the method used. The results showed that depending on the physiological state of the yeasts, being in the diploid or monocellular haploid form, different percentages of ethanol could be produced.


Este trabalho teve como objetivo avaliar culturas de S. cerevisiae diplóides originais, haplóides monocelulares e diplóides recuperados, quanto à capacidade de fermentar em mosto de cana-deaçúcar esterilizado. Vinte e oito culturas foram analisadas, sendo 4 diplóides originais (URM-4420, Fermento Itaiquara FIT, Fermento Lallemand FLA e Fermento Selvagem SEL); 12 haplóides monocelulares provenientes das originais e 12 diplóides recuperados a partir das monocelulares. O percentual de etanol produzido variou de 1,70 por cento a 6,20 por cento e os açúcares redutores infermentescíveis de 0,45 g/100mL a 0,50 g/100mL. Os maiores percentuais de etanol foram produzidos pelas culturas haplóides monocelulares URM-MH3 (4,80 por cento), em 12 horas e SEL-MH1 (6,20 por cento), em 24 horas, correspondendo a valores de produtividade 3,15g.L-1.h-1 e 2,03 g.L-1.h-1 , respectivamente. Os diplóides recuperados não apresentaram teores de etanol detectáveis pelo método utilizado. Os resultados mostram que, dependendo do estado fisiológico das leveduras, seja na forma diplóide ou haplóide monocelular, podem ser produzidos diferentes percentuais de etanol.

ABSTRACT

Aspergillus is a ubiquitous fungus which can cause a variety ofclinical syndromes. This fungus has emerged as agent of systemic infections and has therefore gained considerable public health importance. This paper describes two cases of invasive aspergillosis caused by A. fumigatus in immuno-suppressed patients and underscores the importance of early identification of Aspergillus infection associated with systemic lupus erythematosus and cardiac postoperative complications.


Aspergillus é um fungo ubíquito que pode causar uma variedade de síndromes clínicas. Este fungo tem emergido como agente de infecções sistêmicas e tem conquistado considerável importância para saúde pública. Este trabalho descreve dois casos de aspergilose invasiva causada por A. fumigatus em pacientes imunossuprimidos e relata a importância da identificação precoce em infecção por Aspergillus associada ao lúpus eritematoso sistêmico e a complicações cardíacas pós-operatórias.

ABSTRACT

Enzimas hidrolíticas secretadas por fungos têm um papel importante na patogenicidade das infecções. Objetivando avaliar a atividade enzimática foram testados 31 isolados de Acremonium mantidos na Coleção de Culturas University Recife Mycology. Fragmentos das culturas foram transferidos para caldo glicosado para reativação e posterior crescimento em meio ágar batata dextrose, para verificar viabilidade, pureza e confirmação taxonômica pela observação das características macroscópicas e microscópicas. Para detecção enzimática foram utilizados substratos de caseína do leite e gelatina para protease, amido para amilase e lecitina de soja para fosfolipase. Das 31 culturas, 26 (83,9 por cento) mantiveram-se viáveis e 24 (92,3 por cento) foram confirmadas taxonomicamente. Das 24 culturas, 12 (50 por cento) apresentaram atividade proteásica, duas (16,7 por cento) em caseína do leite, uma (8,3 por cento) em gelatina e nove (75 por cento) em ambos os substratos; 16 (66,7 por cento) degradaram amido. Nenhuma cultura apresentou atividade fosfolipásica. Conclui-se que espécies de Acremonium são capazes de produzir enzimas envolvidas na patogenicidade das infecções fúngicas.


Hydrolytic enzymes secreted by fungi play an important role in the pathogenesis of infection. With the aim of evaluating the enzymatic activity, 31 isolates of Acremonium stored in the University of Recife Mycology (URM) Culture Collection were tested. Culture fragments were transferred to glycoside broth for reactivation and further growth in potato dextrose agar medium in order to investigate viability and purity and to confirm the taxonomy through observing the macroscopic and microscopic characteristics. To detect enzymes, milk casein and gelatin were used as substrates for proteinase, starch for amylase and soy lecithin for phospholipase. Among the 31 cultures, 26 (83.9 percent) remained viable and 24 (92.3 percent) were confirmed taxonomically. Out of these 24 cultures, 12 (50 percent) presented proteinase activity, of which two (16.7 percent) were on milk casein, one (8.3 percent) on gelatin and nine (75 percent) on both substrates; 16 (66.7 percent) degraded starch. None of the cultures presented phospholipase activity. It was concluded that Acremonium species are able to produce enzymes that are involved in the pathogenicity of fungal infections.

Mucoraleswere isolated from maize flour, corn meal and cooked cornflakes using surface and depth plate methods. Rhizopus oryzae, Circinella muscae, Mucor subtilissimus,Mucor hiemalis f. hiemalis, Syncephalastrum racemosum, Rhizopus microsporus var. chinensis and Absidia cylindrospora showed protease activity.


Mucorales foram isolados da farinha de milho, fubá e flocos de milho pré-cozidos pelos métodos de plaqueamento em superfície e em profundidade. Rhizopus oryzae, Circinella muscae, Mucor subtilissimus,Mucor hiemalis f. hiemalis, Syncephalastrum racemosum, Rhizopus microsporus var. chinensis e Absidia cylindrospora exibiram atividade proteásica.

ABSTRACT

Opportunistic mycoses have been increasingly observed among immunocompromised patients. We describe a case in which Engyodontium album was isolated and cultured from the blood of a patient with the Acquired Immunodeficiency Syndrome. E. album grew at 37°C and showed proteinase activity, both indicators of pathogenicity. This is the first time that this organism has been reported as agent of fungaemia.


Micoses oportunistas têm sido progressivamente observadas entre pacientes imunocomprometidos. Nós descrevemos um caso no qual Engyodontium album foi isolado e crescido do sangue de um paciente com a Síndrome da Imunodeficiência Adquirida. E. album cresceu a 37°C e exibiu atividade proteásica, ambos indicadores de patogenicidade. Esta é a primeira vez que este organismo foi reportado como agente de fungemia.
