ABSTRACT

Antibody against Pseudomonas aeruginosa exotoxin A can be used in immunotherapy together with antibiotics to treat acute burn patients. Exotoxin A is one of the virulence factors in Pseudomonas aeruginosa that comprises of three domains, binding domain, translocation and catalytic domain. The purpose of this study was to construct the recombinant domain of the catalytic part of this microorganism in order to produce antibody against it. Pseudomonas aeruginosa samples were isolated from burn patients hospitalized in Mousavi Hospital, Zanjan, Iran and its species was identified by Biochemical tests. Bacteria genomic DNA and also the catalyhc domain of exotoxin A was amplified by PCR. PCR Products and plasmid extracts was digested by restriction enzymes. Subsequently PCR products and plasmids transformed into E. coli BL21 [DE3]. Clones containing gene of interest was determined by restriction enzyme digestion and sequencing. The sequence homology of the catalytic domain of exotoxin A was compared with that of the published gene data bank. The results showed a complete homology between our gene species and the published genome in data banks. The results of this study showed that about 90% of the isolated bacteria contained exotoxin A and there was a sequence homology between our species and published gene data banks

ABSTRACT

Positioning is an important part of perioperative care that optimal respiratory function is a main purpose. Thus oxygen saturation measurement in routine surgical positions could be a proper index of respiratory function. To determine effects of supine, prone and lithotomy positions on arterial oxygen saturation, 31 healthy volunteers were positioned in each three position for ten minutes periods oxygen saturation was measured by pulse oximeter. There was no significant difference between 3 positions in oxygen saturation. Absent of prone and lithotomy positions effect on oxygenation in this study may be attributed to the short time that the subjects were in each position [ten minutes], we suggest that evaluation of position effect on oxygenation require further investigation

ABSTRACT

Continues monitoring of oxygen saturation by pulse oximetery is known as a routine technique in critical care of patients. But previous researches showed some differences in it's measurement according to probe placement.In this cross sectional analytical study 36 admitted neonates below one months age were selected. Sampling method was convenience and sample size was estimated according to alpha = 5% and beta = 20%. Samples'1 oxygen saturation was measured three times by hand and foot pulse oximetery and direct method. Data was analyzed using paired t test.Analysis of results showed significantly difference between both two pulse oximetery methods with arterial blood gas. Comparison of oximetery methods with direct one showed more accuracy of foot probe [p<0/05].We concluded that pulse oximetery do not reliably predict changes in SaO[2] and also probe placement causes some differences in oximetery results

ABSTRACT

Introduction and Objective: there are many noninvasive methods to measure systolic blood pressure. A new noninvasive method by pulse oximetery has recently been reported. The purpose of this study was the comparison of systolic blood pressure measurement by pulse oximetery and with auscultatory method


Material and Methods: systolic blood pressure measurement using pulse oximeter was compared with current auscultatory method. Thirty one healthy volunteers enrolled and were positioned in supine, prone and lithotomy for three 10 minutes periods. Blood pressure was measured at the end of each position by two methods


Results: there was a significant correlation between two method in all positions; supine [r= 0.72], prone [r= 0.88] and lithotomy [r= 0.78]


Conclusion: pulse oximeter seems to be a useful non-invasive method of systolic blood pressure measurement. In order to increase the accuracy of this method, low speed of cuff inflation and deflation is strongly recommended
