ABSTRACT

The mitochondrion is a particularly important organelle in eukaryotic cells. It contains its own genetic material and is coined as “the powerhouse of cells”. Mitochondria are involved in many cellular progresses such as cell signaling and metabolic homeostasis. Its dysfunction is linked to various human diseases, including cancer, neurodegenerative diseases, and diabetes. Mitochondrion has a unique DNA, a small size with 16 569 bp circular genome, encoding only 37 genes, which are key components of the electron transport chain (ETC) and translational machinery. Furthermore, the mutations of mitochondrion DNA correlate with some inherited disease such as Leber’ s hereditary optic neuropathy (LHON) and mitochondrial encephalopathy, lactic acidosis and stroke-like episodes (MELAS). There are very few treatments to fully cure these diseases. As a result, researchers are interested in developing a wide range of methods to understand mitochondrial functions. In this review, we mainly focus on works in targeting mitochondrial DNA, including drug modification, material delivery and gene editing.

ABSTRACT

Classical Representative Famous Prescription is the valuable cultural heritage of Chinese medicine. In November 2018,the State Council issued the " Intensive Implementation of the National Intellectual Property Strategy in 2018 to Accelerate the Construction of IP Strong Country",explicitly proposing to strengthen the intellectual property protection of Classical Representative Famous Prescription. How about the current situation of intellectual property protection of lassical Representative Famous Prescription in China? We selected Liuwei Dihuang Pills,Shengmai Powder and Guizhi Fuling Pills( three representative drugs on market) from Chinese Pharmacopoeia2015 Volume I issued by Chinese Pharmacopoeia Commission to analyze their patent layout, reflecting its status quo of patent protection as follows: first,in recent years, the number of related patent applications for Classical Representative Famous Prescriptions has declined, which was positively correlated with the drug registration and approval policies in recent years, but the policy dividend has not been reflected in the patent application, which may be related to the long period of pharmaceutical R&D; secondly, the patent applicant in the field of Chinese medicine is mainly based on individuals, but the applicant of Classical Representative Famous Prescription is mainly of enterprises, and in addition, the company applicants have the highest authorization rate; thirdly, the main technologies are to improve preparation method and the dosage form in the research and development of Classical Representative Famous Prescription, but these two types of authorized patents have much difficulty in further application on the market. Therefore, the innovative entities shall look for a new breakthrough in secondary development and utilization of Classical Representative Famous Prescriptions.

ABSTRACT

Objectives To observe the effect of low intensity pulsed ultrasound (LIPUS) on osteocyte injuries induced by the tricalcium phosphate(TCP) wear particles in the calvaria of mice. Methods Thirty ICR male mice of 6 to 8 weeks were randomly divided into a normal control group(n=10),a model group (n=10) and a LIPUS-treated group(n=10).A murine calvarial model of osteolysis was established in the model and LIPUS-treated groups through injecting TCP particles onto the surface of bilateral parietal bones at week 1,3,5,7 and 11.Mice in the normal group received negative ultrasound probe pressing, while those in the LIPUS-treated received LIPUS radiation.Three months later, the calvarias were obtained. The micro-CT,HE staining, flow cytometry and Western blotting were performed to estimate the calvarial osteolysis, osteocyte death, apoptosis and proteins expression of the dentin matrix protein 1 (DMP-1),sclerosis protein (SOST),glucose-regulated protein78 (GRP78),inositol-requiring enzyme(IRE 1 α),spliced X-box binding protein 1 (XBP1 s),c-Jun N-terminal kinase (JNK) and phosphorylated c-Jun N-terminal kinase(p-JNK) respectively. Results Compared with the normal control group,in the model group the viability of prosthetic osteocytes decreased significantly, and cell apoptosis was more obvious(P<0.05);the osteocytic marker protein DMP-1 down-regulated significantly, but another marker protein SOST up-regulated significantly, which caused the decline in DMP-1/SOST(P<0.05).Moreover,the expression levels of GRP78,IRE1,XBPls and p-JNK of the model group increased significantly(P<0.05) in the calvaria osteocytes compared to the control group. However,in the LIPUS treatment group, osteocyte injuries and endoplasmic reticulum(ER) stress both decreased significantly, shown by a significant increase in the number and activity of osteocytes,DMP-1/SOST,and significant inhibition of the IRE1α-XBP1-JNK activation(P<0.05).Conclusion LIPUS prevents osteocyte injuries induced by TCP wear particles in the calvaria of mice, which may be due to the inhibition of IRE1α-XBP1-JNK pathway activation through ER stress reaction.

ABSTRACT

OBJECTIVETo describe the incidence trends and to access the histological changing patterns of thyroid cancer in urban areas of Beijing.

METHODSTotal of 4883 new cases diagnosed as thyroid cancer in 1995-2010 in urban areas of Beijing were extracted from the population-based data base of Beijing cancer registry, among which 1185 were males and 3698 were females with a coverage population of 56 143 454 person-years and 54 430 655 person-years, respectively. Incidence, age adjusted incidence, annual percentage change (APC) and gender specific incidence change of different histology patterns during last 16 years were calculate.

RESULTSThere were 4883 cases diagnosed as thyroid cancer during 1995 - 2010 with the sex ratio of 1:3 (1185 males and 3698 females). Incidence rate of thyroid cancer in urban areas of Beijing increased dramatically from 1.55/100 000 (97/6 245 016) in 1995 to 9.90/100 000 (768/7 758 759) in 2010 with an increase rate of 538.71%. After adjusted by world population, the standardized incidence rate increased from 1.27/100 000 in 1995 to 6.96/100 000 in 2010 with a 12.12% annual percentage change (APC = 12.12%, P < 0.05). Among males, incidence rate increased from 0.85/100 000 (27/3 185 669) to 4.58/100 000 (179/3 912 458) with an increase rate of 438.82%. After adjusted by world population, the standardized incidence rate of males increased from 0.68/100 000 in 1995 to 3.19/100 000 in 2010 with an 11.09% annual percentage change (APC = 11.09%, P < 0.05). Among females, the incidence rate increased from 2.29/100 000 (70/3 059 347) to 15.31/100 000 (589/3 846 301) with an increase rate of 568.56%. After adjusted by world population, the standardized incidence rate of females increased from 1.89/100 000 in 1995 to 10.82/100 000 in 2010 with a 12.48% annual percentage change (APC = 12.48%, P < 0.05). Incidence rate ranked 18th and 12th in 1995 among males and females, respectively; while it ranked 14th and 5th in 2010 respectively. Among 4883 cases, 4594 cases were confirmed histologically as primary tumor accounting for 94.08% of total cases. Papillary carcinoma was the most common subtype accounting for 77.31% (3775/4883) of total cases. Among all the pathological types of thyroid cancer, the proportion of papillary carcinoma has risen from 51.55% (50/97) in 1995 to 87.63% (673/768) in 2010. However, the proportion of the follicular carcinoma decreased from 5.15% (5/97) to 1.69% (13/768).

CONCLUSIONIncidence of thyroid cancer in urban areas of Beijing keeps increasing which is mainly due to the rising of papillary carcinoma. Risk factors of papillary thyroid cancer should be focused on when strategies for thyroid cancer prevention and control are conducted.

ABSTRACT

We reported a 23-year-old man who was involved in a high-speed motorcycle accident. He sustained a closed fracture at the right distal femur. The primary fracture happened on February 2008. He underwent open reduction and internal fixation with cloverleaf plate. And one hundred days after the surgery, the proximal screws were pulled-out, but the bone union was not achieved. Treatment consisted of exchanging the cloverleaf plate with a locking compression plate and using an auto-iliac bone graft to fill the nonunion gap. In July 2009, the patient had a sharp pain in the right lower limb. The X-ray revealed that the plate implanted last year was broken, causing a nonunion at the fracture site. Immediately the plate and screws were removed and an intramedullary nail was inserted reversely from the distal femur as well as a 7 cm long bone from the right fibula was extracted and longitudinally split into two pieces to construct cortical bone plates. Then we placed them laterally and medially to fracture site, drilled two holes respectively, and fastened them with suture. We carried on auto-iliac bone grafting with the nonunion bone grafts. The follow-up at 15 months after operation showed that the treatment was successful, X-ray confirmed that there was no rotation and no angular or short deformity. We briefly reviewed the literature regarding such an unusual presentation and discussed in details the possible etiology and the advantages of autologous double-strut fibular grafts to cope with such an intractable situation.

ABSTRACT

The effects of tree ages and geographic area on quality of bark of Magnolia officinalis (MO) and M. officinalis var. biloba (MOB) were studied. The volatile oils of samples extracted by a water stream distillation method were analyzed using a GC/MS method. The non-volatile ingredients of samples were determined by an HPLC method. Seven and twenty characteristic peaks were detected in the HPLC and GC fingerprint of MO, respectively, and the similar HPLC and GC fingerprint were showed in MOB. For samples collected in the same geographic area with different ages, the concentrations of active ingredients varied barely, whereas for those samples collected from different geographic area, differences on the concentrations of active ingredients were observed, for example, the concentrations of the amount of magnolol and honokiol in samples collected in Hubei province were higher than those collected in other areas. Big differences on the concentrations of active ingredients were found between MO and MOB, for MO, the ratio of honokiol/ magnolol was more than 0.7, whereas the ratio-was less than 0.7 in the case of MOB, and the concentrations of honokiol in MO were higher than those in MOB.
