ABSTRACT

Alcantarea nahoumii(Leme) J. R. Grant is a species native to theAtlantic Forest that stands out for ornamental purposes. The objective of this work was to evaluate the in vitrogerminationof A. nahoumiiseeds and establish a micropropagation protocol for production of seedlings so as to minimize the effects of predatory extractivism and develop an in vitroconservation system. Mature seeds were disinfested, established in three culture media (MS, MS½ and MS⅓) and incubated at four temperatures (20, 25, 30 and 35ºC)in a germination chamber. In the micropropagation experiment, stem segments were introduced in MS medium supplemented with 0.5 μM of 1-naphthaleneacetic acid (NAA) and 0.0, 2.2, 4.4 and 6.6 μM of 6-benzylaminopurine (BAP). For the in vitroconservation, plantlets were established in MS or MS½ medium supplemented with 15 g L-1or 30 g L-1of sucrose. The plants were acclimated with commercial substrate. The highest seed germination percentages were promoted by temperature conditions of 20 and 25ºC, with MS culture medium. The highest multiplication rate of shoots was obtained from the treatment without addition of the growth regulator or when combined with 2.2 μM of BAP + 0.5 μM of NAA. The acclimation of the plants occurred with high survival rate. The species can be conserved in vitrounder slow growth condition for 24 months when incubated in MS medium supplemented with 30 g L-1of sucrose.

ABSTRACT

Bauhinia cheilantha, conhecida como "pata-de-vaca", possui grande relevância econômica e etnofarmacológica no semiárido brasileiro. Nas suas sementes constatou-se dormência, o que dificulta a obtenção de plantas uniformes e em curto período de tempo, diante disso, o presente trabalho objetivou estabelecer um protocolo de micropropagação para a espécie. Os segmentos cotiledonar e nodal de plântulas emergidas in vitro foram inoculados em meio de cultura WPM, suplementado com diferentes concentrações de 6-benzilaminopurina (BAP), thidiazuron (TDZ) ou cinetina (KIN) para induzir a regeneração de brotos adventícios. Na fase de enraizamento foram testadas diferentes concentrações de ácido indol-3-butírico (AIB) e de carvão ativado. Nos segmentos nodais houve maior capacidade organogênica do que no segmento cotiledonar. O maior número de brotos (4,3 e 2,1) foi obtido com 1,0mg L-1 de TDZ e 1,5mg L-1 de BAP, respectivamente. No entanto, na presença de TDZ foram observadas as menores brotações. A presença de 2,0mg L-1 de AIB com carvão ativado (CA) promoveu a maior porcentagem de enraizamento (cerca de 60,0 por cento) e maior número de raízes adventícias (2,5). As brotações enraizadas foram transferidas para casa de vegetação e aclimatizadas com sucesso.


Bauhinia cheilantha, known as "pata-de-vaca", is of great economic and ethnopharmacological importance in the semiarid Brazilian. Seeds are dormant which makes it difficult to obtain uniform plants in a short time before this, our study aimed to establish a micropropagation protocol for the species. Cotyledonary and nodal segments of seedlings grown in vitro were inoculated on Woody Plant Medium (WPM) supplemented with various concentrations of 6-benzylaminopurine (BAP), thidiazuron (TDZ) or kinetin (KN) to induce adventitious shoot regeneration. In the rooting phase, different concentrations of indole-3-butyric acid (IBA) and the activated charcoal were tested. The nodal segments showed organogenic capacity greater than the cotyledonary segment. The highest number of shoots (4.3 and 2.1) was obtained at 1.0mg L-1 TDZ and 1.5mg L-1 BAP, respectively. However, in the presence of TDZ was observed the smaller shoots.The use of 2.0mg L-1 IBA within activated charcoal (AC) promoted the highest percentage of rooting (about 60.0 percent) and number of adventitious roots (2.5). The rooted shoots were transferred to greenhouse and successfully acclimatized.

ABSTRACT

Erythrina velutina (mulungu) é uma planta nativa do semiárido brasileiro que pode ser propagada via cultura de tecidos, o que poderá contribuir com a rápida multiplicação dessa espécie, além da redução de possíveis impactos ambientais. O objetivo deste trabalho foi estabelecer um protocolo de micropropagação para o mulungu utilizando explantes de plântulas assépticas obtidas in vitro. Para a indução de brotos, foram utilizados como explantes hipocótilos segmentos nodais e nós cotiledonares inoculados em meio de cultura WPM acrescido de diferentes concentrações de BAP e ANA. O enraizamento dos brotos foi realizado em meio WPM suplementado com 0,0; 2,46; 4,92; e 9,88µM de AIB. A melhor resposta para a multiplicação foi obtida em meio suplementado com 17,76µM de BAP, sendo utilizado segmento nodal ou nó cotiledonar como explante. Os brotos enraizaram em todas as condições testadas, inclusive no meio de cultura sem auxina.

The objective of this study was to establish a protocol for micropropagation of mulungu using aseptic explants seedlings grown in vitro. For the induction of shoots, , nodal segments and cotyledons were used as explants hypocontyl and were inoculated in a WPM culture medium supplemented with different concentrations of BAP and NAA. The rooting of shoots was achieved in a WPM medium supplemented with 0.0; 2.46; 4.92; 9.88µM IBA. The best answer to the multiplication was obtained in medium supplemented with 17.76µM BAP using nodal segment or cotyledonary node explants. The shoots rooted in all conditions tested, including the culture medium without auxin.
