ABSTRACT

Fundamentos: Idosos que vivem na comunidade são propensos a desenvolver fragilidade, considerada como um estado clinicamente identificável que aumenta a vulnerabilidade a resultados adversos e prediz incapacidade e mortalidade na população idosa. Objetivo: Identificar a prevalência e fatores associados à fragilidade em idosos que vivem em uma comunidade. Método: Trata-se de um inquérito domiciliar transversal e analítico, de abordagem quantitativa realizado com 854 idosos que vivem na comunidade. A fragilidade foi mensurada pela Edmonton Frail Scale (EFS). A associação entre fragilidade e variáveis sociodemográficas e de condições clínicas foi mensurada pela análise múltipla por regressão logística. Resultados: A prevalência de fragilidade encontrada neste estudo foi de 12,3% (IC95%: 10,1 a 14,5). O modelo de regressão logística mostrou que as variáveis estatisticamente associadas à fragilidade foram: queda recorrente, uso de dispositivo para auxílio à marcha, polifarmácia, autopercepção ruim de saúde, dependência nas atividades básicas e instrumentais de vida diária. Conclusão: A prevalência de fragilidade em idosos foi baixa em comparação a outros estudos nacionais que empregaram a EFS. Os resultados indicaram múltiplos fatores associados à fragilidade modificáveis. Assim, a investigação da síndrome da fragilidade bem como dos seus fatores relacionados passíveis de prevenção são ações a serem incluídas na prática clínica. (AU)


Foundations: Elderly people living in the community are prone to developing frailty, considered as a clinically identifiable state that increases vulnerability to adverse events and predicts disability and mortality in the elderly population. Objective: To identify the prevalence and factors associated with frailty in the elderly living in the community. Materials and method: This is a cross-sectional and analytical household survey with a quantitative approach conducted with 854 elderly people living in the community. Frailty was measured by Edmonton Frail Scale (EFS). The association between frailty and sociodemographic and clinical condition variables was measured by multiple analysis using logistic regression. Results: The prevalence of frailty found in this study was 12.3% (95% CI: 10.1 to 14.5). The logistic regression model showed that the variables statistically associated with frailty were: recurrent fall, use of walking aids, polypharmacy, poor self-rated health, dependence on basic and instrumental activities of daily living. Conclusion: The prevalence of frailty in the elderly was low compared to other national studies that employed the SAI. Results indicated potentially modifiable factors associated with frailty. Thus, the investigation of frailty syndrome and its related preventable factors are actions to be included in clinical practice. (AU)

ABSTRACT

The aim of this study was to evaluate the reproductive traits of the non-inbred and inbred AquaAmérica, GIFT and AquaAmérica × GIFTgenetic groups. Six fish from each genetic group were used (2 females:1 male). Females were examined for the presence of eggs in their mouth at every four days, for 12 weeks. Reproduction occurred in all genetic groups (GIFT: 100%; non-inbred AquaAmérica and AquaAmérica ×GIFT: 75%; inbred AquaAmérica: 50%). Female weight, female standard length, total spawning weight, absolute fecundity, relative fecundity, spawn index and hatching rate did not differ significantly between the genetic groups. However, the non-inbred AquaAmérica variety showed lower values (P<0.05) for egg diameter (2.4mm) and egg weight (4.2mg) and higher values (P<0.05) for relative number of eggs (247.6 eggs/g of egg) than GIFT (egg diameter: 2.8mm; egg weight: 5.7mg; relative number of eggs: 175.4 eggs/g of egg) and AquaAmérica ×GIFT (egg diameter: 2.8mm; egg weight: 5.9mg; relative number of eggs: 168.8 eggs/g of egg). In conclusion, the non-inbred AquaAmérica variety produces smaller, lighter eggs but a higher relative number of eggs than the GIFT variety and the AquaAmérica ×GIFT cross; and inbreeding negatively affects spawning rate.(AU)


O objetivo deste estudo foi avaliar as características reprodutivas dos grupos genéticos AquaAmérica não endogâmicos e endogâmicos, GIFT e AquaAmérica × GIFT. Foram utilizados seis peixes de cada grupo genético (duas fêmeas:um macho). As fêmeas foram examinadas quanto à presença de ovos na boca a cada quatro dias, durante 12 semanas. A reprodução ocorreu em todos os grupos genéticos (GIFT: 100%; AquaAmérica não endogâmica e AquaAmérica × GIFT: 75%; AquaAmérica endogâmica: 50%). Peso e comprimento padrão de fêmea, peso total de desova, fecundidade absoluta, fecundidade relativa, índice de desova e taxa de eclosão não diferiram significativamente entre os grupos genéticos. Entretanto, a variedade não endogâmica da AquaAmérica apresentou valores mais baixos (P<0,05) para diâmetro do ovo (2,4mm) e peso do ovo (4,2mg) e maiores valores (P<0,05) para número relativo de ovos (247,6 ovos/g de ovo ) que GIFT (diâmetro do ovo: 2,8mm; peso do ovo: 5,7mg; número relativo de ovos: 175,4 ovos/g de ovo) e AquaAmérica × GIFT (diâmetro do ovo: 2,8mm; peso do ovo: 5,9mg; número relativo de ovos: 168,8 ovos/g de ovo). Em conclusão, a variedade AquaAmérica não endogâmica produz ovos menores e mais leves, mas um número relativo maior de ovos que a variedade GIFT e o cruzamento AquaAmérica × GIFT; a consanguinidade afeta negativamente a taxa de desova.(AU)

ABSTRACT

Objective: To evaluate the antinociceptive and anti-inflammatory activities and the toxic effects of Rhipicephalus microplus saliva for elucidating the modulation mechanism between arthropod saliva and host. Methods: For saliva collection, engorged ticks were obtained from a controlled bovine infestation and collected by natural fall. The ticks were fixed and injected pilocarpine 0.2%for induction of salivation. Saliva was collected, lyophilized and stored at - 80 ℃. Cytotoxic activity was assessed by the hemolysis method (25, 50, 100, 200 and 300 μg/mL) and MTT cell viability assay (2.5, 5, 10, 20 and 40 μg/mL) for 24, 48 and 72 h. Anti-inflammatory activity was evaluated using the method of neutrophil migration to the peritoneal cavity of mice at doses of 10, 15 and 20 mg/kg; antinociceptive activity was assessed using the acetic acid-inducedwrithing test, and formalin-induced paw-licking in mice at dose of 15 mg/kg. Results: Saliva did not cause erythrocytes hemolysis at any concentration tested, as well as did not decrease cell viability in the MTT assay. Saliva inhibited neutrophil migration by 87% and 73% at doses of 15 and 20 mg/kg, respectively. In the nociceptive tests, saliva presented analgesic activity of 69.96% in the abdominal writhing test, and of 84.41% in the formalin test. Conclusions: The study proves that Rhipicephalus microplus saliva has significant in vivo anti-inflammatory and antinociceptive activities. The data presented herein support the development of further studies to elucidate the active principles of Rhipicephalus microplus saliva and its mechanism of action and,in future, to develop novel anti-inflammatory and analgesic drugs.

ABSTRACT

O objetivo deste estudo foi avaliar a morfometria intestinal por meio da inclusão de níveis de treonina digestível na dieta de codornas para corte em crescimento. O delineamento experimental adotado foi o inteiramente ao acaso, em esquema fatorial: quatro níveis nutricionais e três segmentos do intestino com dez repetições. Foram coletadas amostras dos três segmentos do intestino delgado. As variáveis estudadas foram área e altura das vilosidades intestinais, profundidade de cripta e relação vilo e cripta. No estudo da altura da vilosidade, foi obtida interação significativa para todas as idades estudadas. A profundidade da cripta em codornas de corte aos 21 dias de idade mostrou-se significativa para as variáveis inclusão de treonina na dieta e para a porção do intestino estudada. Na profundidade da cripta aos 42 dias, ficou evidenciada uma interação significativa entre níveis de inclusão de treonina na dieta e porção do intestino delgado. No estudo da relação altura de vilosidade e profundidade de cripta em codornas aos 21 dias de idade, foram encontrados resultados significativos para a interação tratamento e porção do intestino. Aos 21 e aos 42 dias de idade, a suplementação de 1,04 e 0,78%, respectivamente, de treonina digestível na dieta promove melhores resultados para morfometria intestinal.(AU)


The aim of the study was to evaluate intestinal morphology data by including different levels of threonine in the diet of meat quails in the growing phase. The experimental design was completely randomized in a factorial arrangement: four nutritional levels and three intestinal segments with ten repetitions. Samples were collected to obtain morphometry data (villi and crypt) of the three segments of the small intestine (duodenum, jejunum, and ileum). In the study of the intestinal villus height a highly significant interaction was obtained for all ages studied. A crypt depth was evidenced at 42 days with a significant interaction between levels of inclusion of threonine in the diet and small intestine. In the study of the relationship villus height and depth crypt quails at 21 days of age, the results were significant for the interaction treatment (T) and part of the intestine. At 21 and 42 days of age, supplementation of 1.04 and 0.78% respectively of threonine in the diet promotes better outcomes for intestinal morphology.(AU)

ABSTRACT

The aim of this study was to determine the nutritional requirements of energy and protein for maintenance and weight gain of crossbred cattle, as well as their efficiencies. Fifty 3/4 Zebu × 1/4 Holstein crossbred bulls with initial weights of 214±4kg and aged 11±0.2 months on average were used in this experiment. Four animals were used in the reference group; ten bulls were fed at the maintenance level; and the remaining 36 bulls were fed ad libitum and distributed in a completely randomized design in a 3×3 factorial arrangement, which had three feedlot periods (56, 112 or 168 days) and three calcium and phosphorus levels (low, medium and normal) in the diet. Four of the maintenance animals had their heat production measured by respirometry at the Laboratory of Metabolism and Calorimetry of UFMG. After slaughter, composite samples, referred to as carcass and noncarcass samples were obtained from each animal. The net energy requirements for maintenance (NEm) and metabolizable energy for maintenance (MEm) were 68.9 and 90.1 kcal/EBW0.75/day, respectively. The efficiency (km) was 76.41%. The NEm requirement determined in the respirometry chamber was 85.5 kcal/kg0.75. The following equations were obtained for net energy for gain (NEg) and net protein for gain (NPg): NEg (Mcal/day) = 0.0505±0.000986 × EBW0.75× EBWG1.095 and NPg (g/day) = 162.79±18.2546 × EBWG - 1.30±5.3010 × RE. The efficiencies of fat and protein deposition were 70.04 and 15.12%, respectively. In conclusion, the requirements of NEm for growing and finishing non-castrated 3/4 Zebu × 1/4 Holstein crossbred cattle are 68.9 kcal/EBW0.75/day. Requirements of NEg and NPg can be obtained by the following equations: NEg (Mcal/day) = 0.05050.000986 × EBW0.75 × EBWG1.095 and NPg (g/day) = 162.79±18.2546 × EBWG - 1.30±5.3010× RE.(AU)


Objetivou-se determinar as exigências nutricionais de energia e proteína para mantença e ganho de peso de bovinos cruzados, bem como suas eficiências. Foram utilizados 50 bovinos cruzados 3/4 Zebu × 1/4 Holandês, não castrados, com peso inicial de 214±4kg e idade média de 11±0,2 meses. Quatro animais foram usados para referência, dez para o grupo mantença e os 36 animais alimentados à vontade foram distribuídos em delineamento inteiramente ao acaso em esquema fatorial 3×3, sendo três períodos de confinamento (56, 112 ou 168 dias) e três níveis de cálcio e fósforo (baixo, médio e normal) na dieta. Quatro dos animais mantença foram encaminhados ao laboratório de metabolismo e calorimetria da UFMG para medição da produção de calor através da respirometria. Após os abates, foram obtidas duas amostras compostas para cada animal, denominadas carcaça e não carcaça. As exigências de energia líquida para mantença (ELm) e energia metabolizável para mantença (EMm) foram, respectivamente, de 68,9 e 90,1kcal/PCVZ0,75/dia. A eficiência (km) foi de 76,41%. A exigência de ELm obtida na câmara respirométrica foi de 85,5kcal/kg0,75. As equações obtidas para energia líquida de ganho (ELg) e proteína líquida de ganho (PLg) foram: ELg (Mcal/dia) = 0,0505±0,000986 × PCVZ 0,75 × GPCVZ 1,095 e PLg (g/dia) = 162,79±18,2546 × GPCVZ - 1,30±5,3010 × ER. As eficiências para deposição de gordura e de proteína foram de 70,04 e 15,12%, respectivamente. Conclui-se que as exigências de ELm para bovinos cruzados 3/4 Zebu × 1/4 Holandês não castrados em crescimento e terminação são de 68,9kcal/PCVZ0,75/dia; e que as exigências de ELg e PLg podem ser obtidas pelas respectivas equações: ELg (Mcal/dia) = 0,0505±0,000986 × PCVZ 0,75 × GPCVZ 1,095 e PLg (g/dia) = 162,79±18,2546 × GPCVZ - 1,30±5,3010 × ER.(AU)

ABSTRACT

The kinetics of gas production and in vitro ruminal degradation of diets contained replacement of soybean meal (SBM) by detoxicated castorbean meal (DCBM) (0, 15, 30 and 45% of substitution) were evaluated in this study. Values of gas production of non-fibrous carbohydrates (Vf1), fibrous carbohydrates (Vf2) and total carbohydrates (V) showed quadratic behavior, and by observation of equations can be obtained inclusions of 25% the V and Vf1 obtained maximum values of 174.7 and 237.8 ml gas / g DM, respectively. The potential degradability (PD) and effective degradability (ED) of dry matter showed quadratic behavior and by observation of equations can be perceive that up to 18.0% there was an increase in the values of PD and ED that obtained maximum values of 93.7% and 55.7%, respectively. The replacement of SBM by DCBM from 15 to 30% increased the kinetics of gas production and rumen degradation.(AU)

ABSTRACT

Lactic acid bacteria species were molecularly identified in milk from Lacaune, Santa Inês and crossbred sheep breeds and their in vitro probiotic potential was evaluated. The species identified were Enterococcus faecium (56.25%), E. durans (31.25%) and E. casseliflavus (12.5%). No other lactic acid bacteria species, such as lactobacilli, was identified. Most of the isolated enterococci were resistant to gastric pH (2.0) and to 0.3% oxgall. All tested enterococci were resistant to ceftazidime, oxacillin and streptomycin and sensible to clindamycin, erythromycin and penicillin. The resistance to ciprofloxacin, gentamicin, tetracycline and vancomycin varied among tested species. All tested enterococci strongly inhibited (P<0.05) Escherichia coli and Listeria monocytogenes, moderately inhibited E. faecalis and Staphylococcus aureus and did not inhibit Pseudomonas aeruginosa, Salmonella enterica var. Typhimurium and also one E. durans sample isolated from sheep milk. Four samples of E. faecium, one of E. durans and one of E. casseliflavus presented the best probiotic potential...


Espécies de bactérias ácido-lácticas foram identificadas em nível molecular em leite das raças ovinas Lacaune, Santa Inês e suas mestiças, e o seu potencial probiótico in vitro foi avaliado. As espécies identificadas foram Enterococcus faecium (56,25%), E. durans (31,25%) e E. casseliflavus (12,5%). Nenhuma outra espécie de bactéria ácido-láctica, como Lactobacillus sp., foi identificada. A maioria dos enterococos isolados foi resistente ao pH gástrico (2.0) e a 0,3% de oxgall. Todos os enterococos testados foram resistentes à ceftazidima, oxacilina e estreptomicina e sensíveis à clindamicina, eritromicina e penicilina. A resistência à ciprofloxacina, gentamicina, tetraciclina e vancomicina variou entre as amostras. Todos os enterococos testados inibiram fortemente (P<0,05) Escherichia coli e Listeria monocytogenes, inibiram moderadamente E. faecalis e Staphylococcus aureus e não inibiram Pseudomonas aeruginosa, Salmonella enterica var. Typhimurium e uma amostra de E. durans isolada de leite de ovelha. Quatro amostras de E. faecium, uma de E. durans e uma de E. casseliflavus apresentaram o melhor potencial probiótico...

ABSTRACT

El concepto de autorregulación del aprendizaje ha asumido una importancia creciente en la literatura, ya que la investigación ha sugerido que los alumnos participan activamente en su proceso de aprendizaje, monitorizando y regulando su proceso de estudio con el fin de alcanzar determinados objetivos. La información recogida en revistas indexadas en la base Scielo, en cuanto a la autorregulación, todavía no ha sido sistematizada convenientemente. Por ello, se ha realizado una revisión sistemática de la literatura, con base en datos SciELO.org, en el período comprendido entre el año 2001 y el año 2011, para analizar las evidencias recogidas en diferentes investigaciones sobre: a) la naturaleza del aprendizaje autorregulado, b) su evaluación, c) la promoción de habilidades para la autorregulación y su utilidad en el contexto educativo y d) la posibilidad de implicación de los profesores en la promoción de dicho aprendizaje.


The concept of self-regulation in learning has been assuming a rising importance in the literature once the research has suggested that students participate actively in their learning process, monitoring and regulating their study process to achieve self-set goals. Nevertheless, the information about self-regulation has not been yet systematized on the journals indexed in Scielo. Therefore, the current study performed a systematic review of the literature on Scielo.org, between the period 2001 and 2011 to analyze the existence evidence regarding: a) the nature of self-regulated learning, b) it's assessment, c) the promotion of self-regulated skills and their usefulness in the context of education, and d) the potential implication of teachers on the promotion of lifelong learning.

Context: Contrast sensitivity (CS) function is one of the most important tests available for evaluating visual impairment. Multiple sclerosis (MS) can produce highly selective losses in visual function and psychophysical studies have demonstrated CS deficits for some spatial frequencies. Aims: This work studies the differences in CS between a group of controls and a group of MS patients, focusing on the location of the maximum sensitivity peak, shape of the curve, and determination of the most affected spatial frequencies. Materials and Methods: Using a sinusoidal stimulus the authors assessed CS function in 28 subjects with definitive relapsing remitting MS, and in 50 controls with acuities of 20/25 or better. The peaks of the CS curves were studied by fitting third degree polynomials to individual sets of data. Results: Compared with the control group, the CS function curve for MS subjects showed more deficits in extreme points (low‑ and high‑spatial frequencies). Our results display significant CS losses, at the high‑frequencies band level, in the beginning of the disease. When the disease progresses and the disabilities appear, there are greater losses at the low‑frequencies band level. In average, the CS curve peaks for the MS group were shifted in relation to the control group. Conclusions: CS losses in the MS group suggest an association with ageing and disability level in the expanded disability status scale. The position of the CS function peak is influenced by MS, age, and degree of disability.

ABSTRACT

Lactobacillus spp. isolated from different portions of chickens' gastrointestinal tract were evaluated concerning their ability to survive in a water-in-oil (W/0) emulsion containing sesame and sunflower oil. After sixty days of emulsion storage under refrigeration, three of five strains tested survived in number equal to or higher than 10(6)cfu/g. Lactobacillus reuteri 2M14C, which presented the highest survival in W/O emulsion (10(7)cfu/g), was tested for its capacity to resist throughout the passage through gnotobiotic mice gastrointestinal tract and for the ability to stimulate murine peritoneal macrophages phagocytosis. This strain remained at a number above 10(9)cfu/g feces during ten days of monoassociation, and monoassociated mice showed phagocytic activity significantly greater than the germ-free controls (P<0.05). The results suggest that the formulation can be used to incorporate viable Lactobacillus spp. cells in animal feed. Moreover, the results suggest that L. reuteri 2M14C is a strong candidate to be incorporated in probiotic formulations for use in chicken.


Lactobacillus spp. isolados de diferentes porções do trato gastrintestinal de frangos foram testados quanto à capacidade de se manterem viáveis em uma emulsão água/óleo (A/O) contendo óleos de gergelim e de girassol. Após sessenta dias de estocagem sob refrigeração, três de cinco linhagens testadas sobreviveram em concentração igual ou superior a 10 6 UFC/g. Lactobacillus reuteri 2M14C, que apresentou maior capacidade de sobrevivência na emulsão desenvolvida (10 7 UFC/g), foi testado quanto à sua capacidade de sobreviver às condições do trato gastrintestinal in vivo em camundongos gnotobióticos. Após dez dias de monoassociação com L. reuteri 2M14C, foi testada também a capacidade de estimulação da atividade fagocítica de macrófagos peritoneais. A linhagem permaneceu em número superior a 10 9 UFC/g de conteúdo fecal durante os dez dias de monoassociação, e os camundongos monoassociados apresentaram atividade fagocítica maior (P<0,05) que a do grupo controle isento de germe. Os resultados sugerem que a formulação proposta é capaz de manter a viabilidade de células de lactobacilos para adição em ração animal, necessitando, no entanto, de um acompanhamento dessa viabilidade por tempo maior de estocagem. Além disso, os resultados demonstram que Lactobacillus reuteri 2M14C é um forte candidato a ser adicionado em formulações probióticas para uso em frangos.

ABSTRACT

O potencial probiótico in vitro de 12 amostras de bactérias ácido-láticas (11 Lactobacillus spp. e uma Weissella paramesenteroides), isoladas de queijo-de-minas artesanal da Serra da Canastra, foi investigado. Essas amostras foram caracterizadas in vitro quanto à susceptibilidade a antimicrobianos, antagonismo contra microrganismos de referência patogênicos e não patogênicos e sensibilidade a pH gástrico e sais biliares. Apenas L. rhamnosus B4, W. paramesenteroides C10 e L. rhamnosus D1 apresentaram resistência a menor número de antimicrobianos (ceftazidima, oxacilina, estreptomicina e vancomicina). Todas as amostras apresentaram atividade antagonista frente a todas as bactérias patogênicas testadas e não inibiram as bactérias não patogênicas. Todas as amostras foram resistentes ao pH gástrico (2.0). Alguns microrganismos mostraram pouca inibição do crescimento em presença de sais biliares, enquanto outros foram moderadamente ou altamente inibidos. L. rhamnosus B4, W. paramesenteroides C10 e L.rhamnosus D1 apresentaram o melhor potencial probiótico de acordo com os testes in vitro realizados.


The in vitro probiotic potential of twelve samples of lactic acid bacteria (eleven Lactobacillus spp. and one Weissella paramesenteroides) isolated from Minas artisanal cheese from Serra da Canastra was evaluated for their antimicrobial susceptibility, antagonistic activity against pathogenic and lactic acid bacteria, as well as for their tolerance to gastric pH and biliary salts. Only L. rhamnosus B4, W. paramesenteroides C10, and L. rhamnosus D1 showed resistance to tested antimicrobials (ceftazidime, oxacillin, streptomycin, and vancomycin). All samples showed antagonism against all pathogenic bacteria tested and did not inhibit the growth of nonpathogenic samples. Regarding gastric pH tolerance, all samples showed low inhibition at pH 2. The tolerance to biliary salts varied from low to great among tested samples. L. rhamnosus B4, W. paramesenteroides C10, and L. rhamnosus D1 showed the best probiotic potential revealed by their in vitro test results.

ABSTRACT

A osteopetrose é uma doença rara, caracterizada pelo aumento generalizado da densidade óssea. Tem como característica principal, a reabsorção osteoclástica defeituosa, resultando no acúmulo de massa óssea. Além disso, pode ocorrer retardo do crescimento, desnutrição progressiva, anemia e caquexia. O presente relato descreve o caso de uma cadela, com aproximadamente nove meses de idade, sem raça definida, com histórico de apatia e disorexia. Hemogramas seriados demonstraram pancitopenia persistente. Vários exames laboratoriais foram realizados para excluir doenças como erliquiose, leishmaniose e cinomose, porém todos foram negativos. O mielograma constatou hipocelularidade relativa por provável aplasia/hipoplasia medular. Exames radiográficos evidenciaram o aumento da radiopacidade óssea e hipertrofia da região cortical, e a necropsia confirmou a redução do canal medular. O exame histopatológico confirmou a osteopetrose. Conclui-se que a osteopetrose pode causar comprometimento na produção de células sanguíneas, sendo uma causa rara de anemia mielotísica, além de alterações neurológicas secundárias à má-formação dos ossos do crânio.


Osteopetrosis is a rare disease characterized by generalized increase in bone density. The defective osteoclastic resorption results in the accumulation of bone mass. Furthermore, there may be growth delay, progressive malnutrition, anemia and cachexy. This report describes the case of a nine month old, mixed breed, female dog presented with apathy and disorexia. Laboratory tests were performed to rule out Ehrlichiosis, Leishmaniasis, and Canine distemper, but all were negative. Hemogram showed persistent pancytopenia and myelogram showed relative hypocellularity, probably due to spinal cord aplasia/hypoplasia. Radiographs showed increased bone opacity and hypertrophy of the cortical region, and the necropsy confirmed the reduction of the medullary canal. Histopathological examination confirmed osteopetrosis. In conclusion, osteopetrosis can compromise blood cell production, and is a rare cause of mielotisic anemia. It can also cause neurological deficits due to cranium bone deformation.

ABSTRACT

Avaliou-se a influência da temperatura de descongelação na integridade de espermatozoides criopreservados de cães. Foram utilizados reprodutores das raças Basset Hound (n=3) e Rottweiler (n=3), submetidos a colheitas de sêmen por manipulação peniana. As amostras de sêmen foram descongeladas a 37ºC/1min (G1) ou 70ºC/6s (G2) e avaliadas quanto à motilidade progressiva, vigor e integridade do acrossoma após 0, 30 e 60 minutos de incubação (37ºC), e ultraestrutura espermática imediatamente após a descongelação. Em todos os tempos de incubação, a motilidade progressiva dos espermatozoides descongelados a 70ºC por 6s (74,6%) foi mais alta (P<0,05) que a dos descongelados a 37ºC por 1min (64,6%). O vigor espermático não diferiu (P>0,05) entre os grupos, e o porcentual de gametas com acrossomas íntegros foi maior (P<0,05) nos espermatozoides do G1 do que no G2. Lesões ultraestruturais foram identificadas nos espermatozoides descongelados de ambos os grupos, em maior quantidade nos gametas do G2. Conclui-se que amostras congeladas de sêmen de cães devam ser descongeladas a 37ºC por 1min.


Aiming to evaluate the influence of the thawing temperature on the viability of canine cryopreserved sperm, Basset Hound (n=3) and Rottweiler (n=3) dogs were used, submitted to semen collected through manual manipulation. Semen samples were thawed at 37ºC during 1min (G1) or at 70ºC during 6s (G2), and evaluated for progressive motility, vigor and acrosome integrity, after 0, 30 e 60 minutes of incubation (37ºC), and sperm ultrastructure immediately after thawing. In all incubation times, the average of progressive motility was higher (P<0.05) in samples from G2 Group (74.6%) than from G1 (64.6%). Sperm vigor had no difference (P>0.05) between groups, and the percentage of gametes with intact acrosome was higher (P<0.05) on sperm cells from G1 than from G2. Ultrastructural changes were identified on dog sperm from both groups, and were observed in higher quantity in gametes from G2 Group. It can be concluded that samples of frozen dog sperm must be thawed at 37°C for 1min.

Reportamos un caso de un paciente joven que ingresó para estudio y manejo de una hipocalcemia grave. Durante su evolución el paciente presentó dolor abdominal (cólico vesicular) relacionado con la presencia de un ejemplar de Ascaris lumbricoides en la vesícula biliar, que respondió a terapia médica conservadora.


We report a case of a young patient who was admitted for study and management of severe hypocalcemia. During evolution of the condition the patient presents abdominal pain (biliary colic) related to the presence of Ascaris lumbricoides in the gallbladder, responding to conservative medical therapy.

ABSTRACT

Puba or carimã is a Brazilian staple food obtained by spontaneous submerged fermentation of cassava roots. A total of 116 lactobacilli and three cocci isolates from 20 commercial puba samples were recovered on de Man, Rogosa and Sharpe agar (MRS); they were characterized for their antagonistic activity against foodborne pathogens and identified taxonomically by classical and molecular methods. In all samples, lactic acid bacteria were recovered as the dominant microbiota (7.86 ± 0.41 log10 CFU/g). 16S-23S rRNA ARDRA pattern assigned 116 isolates to the Lactobacillus genus, represented by the species Lactobacillus fermentum (59 isolates), Lactobacillus delbrueckii (18 isolates), Lactobacillus casei (9 isolates), Lactobacillus reuteri (6 isolates), Lactobacillus brevis (3 isolates), Lactobacillus gasseri (2 isolates), Lactobacillus nagelii (1 isolate), and Lactobacillus plantarum group (18 isolates). recA gene-multiplex PCR analysis revealed that L. plantarum group isolates belonged to Lactobacillus plantarum (15 isolates) and Lactobacillus paraplantarum (3 isolates). Genomic diversity was investigated by molecular typing with rep (repetitive sequence)-based PCR using the primer ERIC2 (enterobacterial repetitive intergenic consensus). The Lactobacillus isolates exhibited genetic heterogeneity and species-specific fingerprint patterns. All the isolates showed antagonistic activity against the foodborne pathogenic bacteria tested. This antibacterial effect was attributed to acid production, except in the cases of three isolates that apparently produced bacteriocin-like inhibitory substances. This study provides the first insight into the genetic diversity of Lactobacillus spp. of puba.

ABSTRACT

Amostras de queijo de minas artesanal foram coletadas em 18 queijarias localizadas em propriedades rurais da região da Serra da Canastra, Minas Gerais, com o objetivo de avaliar a influência da altitude sobre a população de bactérias acidolácticas. As queijarias estavam distribuídas nas altitudes de 600 a 900m, 900 a 1000m e mais de 1000m. Observaram-se populações mais elevadas de bactérias acidolácticas nas amostras de queijo da altitude de 600 a 900m. Lactobacillus rhamnosus, Lactobacillus casei e Lactobacillus plantarum foram os principais microrganismos isolados e identificados por PCR ARDRA 16S-23S rDNA, além de Enterococcus spp., Lactococcus spp. e outras espécies de Lactobacillus. Sugere-se que estas espécies estejam adaptadas ao ambiente de produção do queijo de minas artesanal produzido na região, o que resultaria em características sensoriais próprias do produto.


Samples of minas artisanal cheese were collected in 18 small-scale producer properties located in the rural region of Serra da Canastra, Minas Gerais state, aiming to evaluate the influence of three altitudes, from 600 to 900m, 900 to 1000m, and higher than 1000m, on the lactic acid bacteria (LAB) population. High populations of LAB were observed in the cheese samples, mainly in the lowest altitude. Lactobacillus rhamnosus, Lactobacillus casei, and Lactobacillus plantarum were the major LAB isolated from the cheese samples and identified according to PCR ARDRA 16S-23S rDNA. Enterococcus spp., Lactococcus spp., and other species of Lactobacillus genus were also found. It is suggested that these microorganisms are adapted to the production environment of the minas artisanal cheese which result in the unique sensorial properties of the product.

ABSTRACT

It is well known that eosinophilia is a key pathogenetic component of toxocariasis. The objective of the present study was to determine if there is an association between peritoneal and blood eosinophil influx, mast cell hyperplasia and leukotriene B4 (LTB4) production after Toxocara canis infection. Oral inoculation of 56-day-old Wistar rats (N = 5-7 per group) with 1000 embryonated eggs containing third-stage (L3) T. canis larvae led to a robust accumulation of total leukocytes in blood beginning on day 3 and peaking on day 18, mainly characterized by eosinophils and accompanied by higher serum LTB4 levels. At that time, we also noted increased eosinophil numbers in the peritoneal cavity. In addition, we observed increased peritoneal mast cell number in the peritoneal cavity, which correlated with the time course of eosinophilia during toxocariasis. We also demonstrated that mast cell hyperplasia in the intestines and lungs began soon after the T. canis larvae migrated to these compartments, reaching maximal levels on day 24, which correlated with the complete elimination of the parasite. Therefore, mast cells appear to be involved in peritoneal and blood eosinophil infiltration through an LTB4-dependent mechanism following T. canis infection in rats. Our data also demonstrate a tight association between larval migratory stages and intestinal and pulmonary mast cell hyperplasia in the toxocariasis model.

In the present study, the effects of Polybia paulista venom (PPV) on renal and vascular tissues were investigated. Isolated kidneys perfused with PPV (1 and 3 ìg/mL) had increased perfusion pressure, renal vascular resistance, urinary flow, and glomerular filtration rate; and reduced sodium tubular transport. Histological evaluation demonstrated deposits of proteins in Bowman's space and tubular lumen, and focal areas of necrosis. The venom promoted a cytotoxic effect on Madin-Darby canine kidney (MDCK) cells. A significant increase in lactic dehydrogenase levels was observed in response to venom exposure. In isolated mesenteric vascular beds, pressure and vascular resistance augmented in a dose-dependent manner. PPV increased the contractility of aortic rings maintained under basal tension. This contractile response was inhibited when preparations were maintained in Ca2+-free medium. Likewise, verapamil, a voltage-gated calcium channel blocker, also inhibited the contractile response. In this study, phentolamine, a blocker of á-adrenergic receptor blocker, significantly reduced the contractile effect of PPV in the aortic ring. In conclusion, PPV produced nephrotoxicity, which suggests a direct effect on necrotic cellular death in renal tubule cells. The vascular contractile effect of PPV appears to involve calcium influx through voltage-gated calcium channels via adrenergic regulation.

ABSTRACT

Este trabalho teve como objetivo determinar a prevalência de Escherichia coli produtoras de Shigatoxinas (STEC) e E. coli dos sorogrupos O157, O111 e O113 em rebanhos leiteiros do Município de Jaboticabal, SP, Brasil. A presença de sequências gênicas stx1, stx2e eae foi detectada pela reação em cadeia da polimerase (PCR) em amostras de fezes. Todas as amostras stx e eae positivas foram submetidas a uma nova reação de PCR para detecção das sequências rfb O157, O111 e O113. Observou-se uma alta prevalência (72,16%) de sequências stx nas fezes dos bovinos, sendo que o perfil genotípico encontrado com maior frequência foi o stx1 associado à stx2. Os coeficientes de prevalência das sequências rfb O157, O111 e O113 foram, respectivamente, 14,77%, 0,2% e 30,83%. Animais de todos os rebanhos (100%) apresentaram em suas fezes STEC e E. coli O113 e os sorogrupos O157 e O111 foram observados em 60,0% e 10,0% dos rebanhos, respectivamente. Concluiu-se que a alta prevalência de STEC detectada em rebanho leiteiro evidenciada nas fezes de bovinos desempenham um papel importante na contaminação ambiental e podem oferecer risco de agravo à saúde pública.


The purpose of this study was to determine the prevalence of Shigatoxigenic Escherichia coli (STEC) and serogroups O157, O111 and O113 in dairy cattle from Jaboticabal, state of São Paulo, Brazil. Feces samples were collected from 10 herds and assessed for the presence of the virulence genes stx1, stx2 and eae by polymerase chain reaction (PCR). All samples positive for stx and eae were submitted to a second PCR reaction targeting the sequences rfb O157, rfb O111 and rfb O113. A high prevalence of stx (72.16%) was detected in the fecal samples, the most frequent being stx1 associated to stx2. The prevalence of sequences rfb O157, rfb O111 and rfb O113 was 14.77%, 0.2% and 30.83%, respectively. STEC and serogroup O113 was identified in all herds (100%), and serogroups O157 and O111 were observed in 60% and 10% of the herds. In conclusion, the high STEC prevalence detected in dairy herds evidences that bovine feces might play an important role as a contamination source in the region of Jaboticabal.

O objetivo deste trabalho foi determinar o número de coliformes totais, termotolerantes, Staphylococcus coagulase positivo e a presença de bactéria do gênero Salmonella no músculo, no tecido superficial, no trato gastrintestinal de peixes e na água de cultivo de pesque-pagues situados na microbacia do Córrego Rico, SP. Não foi detectado Staphylococcus coagulase positivo em nenhuma amostra de água e peixe. O número mais provável (NMP) de coliformes totais no peixe variou de 2,0 x 10 a >1,1 x 104 NMP.100 mL-1, na água de cultivo variou de 4,2 x 104 a > 2,4 x 105 NMP.100 mL-1. A contagem de coliformes termotolerantes no peixe variou de < 3 a 5,1 x 103 NMP.g-1, na água de cultivo variou de 3,8 x 102 a 2,0 x 104 NMP.100 mL-1. Não houve diferença estatística (P > 0,05) entre as populações de micro-organismos pesquisados na água, pele e trato gastrintestinal, o que reflete uma relação entre a presença dos micro-organismos na água e nesses dois locais analisados. Foi isolada Salmonella sp. em uma amostra de músculo e em duas amostras de trato gastrintestinal. O pescado pode ser veículo de contaminação cruzada, tendo como fonte dos micro-organismos a pele e o trato gastrintestinal para sua própria musculatura. Portanto, o monitoramento da qualidade da água é de suma importância para garantir a produção de peixes com boa qualidade.


The objective of this study was to determine the number of total and thermotolerant coliforms, coagulase-positive Staphylococcus, and the presence of bacteria of the genus Salmonella in the muscle, surface tissue, and gastrointestinal tract of fish, and in the pond water of fee-fishing establishments located in the Córrego Rico microwatershed in the state of São Paulo, Brazil. Coagulase-positive Staphylococcus was not detected in any sample of pond water or fish. The most probable number (MPN) of total coliforms in fish ranged from 2.0 x 10 to > 1.1 x 104 NMP.100 mL-1, while in pond water it ranged from 4.2 x 104 to > 2.4 x 105 NMP.100 mL-1. The count of thermotolerant coliforms in fish ranged from < 3 to 5.1 x 103 NMP.g-1, while in pond water it ranged from 3.8 x 102 to 2.0 x 104 NMP.100 mL-1. There was no statistical difference (P > 0.05) among the studied populations of microorganisms in pond water, surface tissue and gastrointestinal tract, which reflects a relationship between the presence of microorganisms in the water and in the two tissues analyzed. Salmonella sp. was isolated in one sample of muscle and in two samples of gastrointestinal tract. The fish can be a vehicle for cross contamination, having the gastrointestinal tract and the surface tissue as a source of microorganisms for its muscles. Therefore, monitoring the water quality is important to ensure the production of fish with good quality.
