ABSTRACT

Objetivo. Evaluar la capacidad in vitro e in vivo de Lactobacillus acidophilus1, aislado en este estudio, para prevenir enfermedad diarreica causada por Vibrio cholerae 01 OGAWA, en conejos lactantes. Materiales y métodos. Se aisló Lactobacillus acidophilus1 a partir de heces de niños sanos, se seleccionó por su capacidad amilolítica a partir de un grupo de bacterias ácido lácticas (BAL). El mejor sustrato amiláceo para el crecimiento de la BAL fue estandarizado previamente. Con L. acidophilus1 se realizó un ensayo in vitro e in vivo de inhibición antagónica sobre el enteropatógeno y se evaluó la prevención de la colonización por V. cholerae 01 OGAWA en conejos lactantes. Resultados. De acuerdo con el análisis de varianza L. acidophilus1 presentó la mejor capacidad amilolítica respecto a las otras BAL aisladas, p<0.5. Se encontró que un inóculo con una densidad celular de 35x10(6) bacterias/ml en el fermento láctico es capaz de ejercer el mayor efecto antagónico in vitro sobre V. cholerae. Se demostró el efecto probiótico in vivo, ya que los conejos enfrentados con el patógeno y sin recibir probiótico tuvieron una probabilidad de sobrevida menor de 0.25 respecto al grupo de animales retados con el patógeno y simultáneamente alimentados con el probiótico cuya probabilidad de sobrevida fue de 0.95. Conclusiones. L. acidophilus1 se considera un microorganismo probiótico, capaz de sobrevivir a su paso por el tracto gastrointestinal en un modelo animal y prevenir la colonización intestinal por V. cholerae 01 OGAWA en conejos lactantes.


Objective. To assess the capacity in vitro and in vivo of Lactobacillus acidophilus1 isolated in this study, capable of preventing diarrheal disease caused by Vibrio cholerae 01 OGAWA in rabbits newborn. Materials and methods. Lactobacillus acidophilus1 was isolated from faeces of healthy children and was selected from a group of lactic acid bacteria (LAB). The better starchy substrate for the growth of the LAB was previously standardized. Using the L. acidophilus1, there was carried out an in vitro and in vivo assay of antagonistic inhibition on the enteropathogen and it was evaluated the prevention of colonization by V. cholerae 01 Ogawa in rabbits newborn. Results. L. acidophilus1 present the better amylolytic capacity, p<0.5 in the ANDEVA. It was found that an inoculum with a cell density of 35x10(6) bacteria/ml in the lactic ferment is able to exert the greatest antagonistic effect in vitro over V. cholerae. The probiotic effect was demonstrated in vivo, since rabbits exposed to the pathogen without having received probiotic had a lower survival probability of 0.25 regarding the group of animals exposed with the pathogen and simultaneously fed with the probiotic whose probability of survival was of 0.95. Conclusions. L. acidophilus1 is considered a probiotic microorganism capable of surviving when passing through the gastrointestinal tract in an animal model and preventing intestinal colonization by V. cholerae 01 Ogawa in rabbits newborn.

ABSTRACT

La obesidad en la infancia está asociada con dislipemia, hipertensión arterial, intolerancia a la glucosa y predisposición temprana a enfermedad cardiovascular. La obesidad abdominal juega un rol central en el Síndrome Metabólico (SM), entidad que predispone a los individuos afectados al desarrollo de diabetes y enfermedad cardiovascular en la adultez. La insulinorresistencia que acompaña al SM puede evaluarse a través de distintos marcadores El objetivo de este trabajo fue evaluar la presencia de SM y de factores de riesgo cardiovascular (RCV) (dislipemias, hipertensión arterial y obesidad) y de marcadores metabólicos (Apo B y PCR ultrasensible) en nuestra población de adolescentes. Fueron evaluados 943 estudiantes adolescentes (429 mujeres y 541 varones), entre 11 y 14 años, de escuelas secundarias de la Universidad de Buenos Aires. Se midieron parámetros antropométricos y se determinaron las concentraciones plasmáticas de glucemia, lípidos, apolipoproteína B (apo B) y PCR ultrasensible (PCRus). Los varones presentaron mayor peso (p= 0.004), mayor circunferencia de cintura (p= 0.0002) y mayor presión arterial sistólica y diastólica (p= 0.008 y p= 0.002 respectivamente) con respecto a las mujeres. También presentaron mayor nivel de glucemia (p= 0.04) y menor nivel de HDL-colesterol (p= 0.004). No hubo diferencia significativa en los niveles plasmáticos de triglicéridos, PCRus y apo B entre ambos sexos. La presencia de SM fue de 5,45% en varones y de 1,63% en mujeres. El grupo de adolescentes con SM presentó mayor peso, IMC, circunferencia de cintura, presión sistólica y diastólica, como también valores más elevados de glucemia, triglicéridos y menor HDL-colesterol, siendo las diferencias significativas. Los niveles séricos de ApoB fueron mayores en el grupo con SM (p= 0.001). En conclusión, este estudio muestra la presencia de SM y de factores de riesgo cardiometabólico con incremento de apoB en edades tempranas, por lo cual su detección precoz en niños y adolescentes, ayudaría a implementar medidas preventivas.


Obesity in the infancy is associated with dyslipidemia, hypertension, glucose intolerance and early development of cardiovascular risk. Abdominal obesity associated with metabolic syndrome predisposes to diabetes mellitus and cardiovascular disease in adulthood. Insulin resistance is present in this syndrome and should be evaluated. The aim of the study was to evaluate the presence of metabolic syndrome (MS) and cardiovascular risk (dyslipidemia, hypertensión and obesity) and metabolic risk markers (apolipoprotein B and high sensitivity C-reactive protein) in this group of adolescents. We evaluated 943 adolescents from high school (429 women y 541men), ages between 11 and 14. Weight, height, body mass index (BMI), waist circumference (WC) and blood pressure were determined in all subjects. Fasting serum concentrations of glucose, lipids, apolipoprotein B (apoB) and high sensitivity C-reactive protein (hs-CRP) were measured. Boys showed higher weight (p= 0.004), higher WC (p= 0.0002) and higher systolic and diastolic blood pressure (p= 0.008 y p= 0.002)) than girls. They also showed higher glycemia (p= 0.04) and lower HDLcolesterol levels (p= 0.004). There were no differences in triglycerides, hs-CRP and apo B levels between both sexes. The frequency of MS was 5.45% in males and 1.63% in females. The group with MS had higher weight, BMI, WC, and systolic and diastolic blood pressure as well as higher glucose and triglycerides and lower HDL-cholesterol levels Although no differences in hs-CRP were observed between groups, hs-CRP correlated with WC (r=0.14, p<0.001) and BMI (r=0.17, p< 0.001). ApoB levels were higher in the group with MS (p= 0.001). In conclusion this study shows the presence of MS and cardiovascular risk factors with high apo B levels at early age. Early detection of children and adolescents with metabolic abnormalities should be useful to implement strategies to avoid the development of cardiovascular disease in later life.

ABSTRACT

In many countries, photodynamic therapy (PDT) has been recognized as a standard treatment for malignant conditions (for example, esophageal and lung cancers) and non-malignant ones such as age-related macular degeneration and actinic keratoses. The administration of a non-toxic photosensitizer, its selective retention in highly proliferating cells and the later activation of this molecule by light to form reactive oxygen species that cause cell death is the principle of PDT. Three important mechanisms are responsible for the PDT effectiveness: a) direct tumor cell kill; b) damage of the tumor vasculature; c) post-treatment immunological response associated with the leukocyte stimulation and release of many inflammatory mediators like cytokines, growth factors, components of the complement system, acute phase proteins, and other immunoregulators. Due to the potential applications of this therapy, many studies have been reported regarding the effect of the treatment on cell survival/death, cell proliferation, matrix assembly, proteases and inhibitors, among others. Studies have demonstrated that PDT alters the extracellular matrix profoundly. For example, PDT induces collagen matrix changes, including cross-linking. The extracellular matrix is vital for tissue organization in multicellular organisms. In cooperation with growth factors and cytokines, it provides cells with key signals in a variety of physiological and pathological processes, for example, adhesion/migration and cell proliferation/differentiation/death. Thus, the focus of the present paper is related to the effects of PDT observed on the extracellular matrix and on the molecules associated with it, such as, adhesion molecules, matrix metalloproteinases, growth factors, and immunological mediators.

ABSTRACT

The effects of Photodynamic Therapy using 2nd generation photosensitizers have been widely investigated aiming clinical application treatment of solid neoplasms. In this work, ultrastructure changes caused by the action of two 2nd generation photosensitizers and laser irradiation on CHO-K1 and HeLa (neoplastic) cells were analyzed by transmission electron microscopy. Aluminum phthalocyanine chloride, aluminum phthalocyanine tetrasulfonate chloride and radiation from a semiconductor laser at a fluency of 0.5 J/cm2 (Power=26 mW; lambda=.670 nm) were used. The results showed induction of apoptosis. Such alterations where observed in HeLa but not in CHO-K1 cells after Aluminum phthalocyanine tetrasulfonate chloride (AlPcS4, photodynamic treatment. The Aluminum phthalocyanine chloride (AlPc) photodynamic treatment induced necrosis on the neoplastic cell line, and cytoplasm and nuclear alterations on the normal cell line.
