ABSTRACT

Objective:To study effects of neuroligin (NLG) on the proliferation and apoptosis in human neuroblastoma SH-SY5Y cells. Methods:The SH-SY5Y cells were cultured in vitro for 24 hours, and then transfected with NLG siRNA at dose of 50,100,200 μmol/L,respectively.MTT procedure was used to detect the cell proliferation, and expression levels of apoptosis gene including Bax or Bcl-2 and Bcl-xl were measured by RT-PCR.Results:Compared to control groups proliferation of SH-SY5Y cells were distinctly inhibited after NLG siRNA transfection accompany with a dose-dependent,which was caused by activation of apoptosis. Conclusions:NLG protect neuron by inhibiting apoptosis.

ABSTRACT

Objective To investigate the protective effects of curcumin on the cellular model of Alzheimer's disease(AD) and the expression of growth associated protein-43(GAP-43). Methods Aβ25~35 was used to treat the hippocampus neurons of rat and the cellular model of AD was established. The survival rate was detected by MTT assay. The cells were randomly divided into blank control, model, curcumin 10 and 20μmol/L groups. The effect of curcumin on apoptosis was assayed by flow cytometry. The pro?trusive length and GAP-43 positive cell rate were detected by immunocytochemistry. The expression of GAP-43 was detected by West?ern blot. Results Compared with the model group, curcumin significantly reduced the toxicity of Aβ25~35,increased the survival rate and decreased the apoptosis rate of the cells(P<0.05). It also significantly increased the average protrusive length,GAP-43 positive cell rate and GAP-43 expression(P<0.05 or P<0.01). Conclusion The protective effect of curcumin on the cellular model of AD was likely related to the up-regulation of GAP-43 expression.

ABSTRACT

ObjectiveTo investigate the relationship between nonalcoholic fatty liver disease(NAFLD)and the severity of coronary artery disease (CAD).MethodsRetrospective cross-sectional studies were conducted to examine the relationship between NAFLD and the severity of CAD. CAD was diagnosed by coronary angiography and NAFLD by type-B ultrasound.ResultsThe logistic regression analysis showed that hypertension, NAFLD, diabetes, smoking, and high level of γ-glutamyltransferase were risk factors of multiplevessel lesions and occlusion of CAD( P<0.05 or P<0.01 ) , and high levels of total cholesterol and alanine aminotransferase were risk factors of multiple-vessel lesions ( P<0. 05 or P<0.01 ) , family history was a risk factor of occlusion of CAD, whereas, high level of high-density lipoprotein cholesterol was an important protective factor of multiple-vessel lesions and occlusion of CAD ( P<0. 01 ). NAFLD was a dependent risk factor of multiple-vessel lesions and occlusion of CAD( P<0.05 or P<0. 01 ). ConclusionsNAFLD could be regarded as a main risk factor of the severity of CAD. Clinicians should pay more attention to the prevention and treatment of NAFLD in order to improve the outcome of CAD.
