ABSTRACT

Objective To evaluate the automatic biochemical analyzer when used to detect urinary vanilmandelic acid (VMA), and to compare it with manual method. Methods The automatic biochemical analyzer using homogenous enzyme immunoassay technology was compared with the manual method on accuracy, precision, linear range, recovery rate, anti-interference capability and etc when used to detect VMA.The comparison was also carried out on positive rate and etc when the two methods were used to test the urine specimens of the healthy subjects and suspected patients of hypertension, hyperthyroidism and hypothyroidism. Results The two methods both had the results on accuracy, precision, linear range, recovery rate, anti-interference capability meet the requirements described in the instruction of reagent kit, while the analyzer gained advantages over the manual method.The positive rates by the two methods for testing urine specimens were similar,while the analyzer behaved better in diagnosing the patient with critical value.Conclusion The analyzer proves better than the manual method when used to detect VMA,and thus is worthy promoting in clinical trial.

ABSTRACT

OBJECTIVE: To purify butin and butein from the Vernonia anthelmintica Willd, and preliminary study their effects on proliferation and melanogenesis of A375 human melanoma cells. METHODS: Selica gel and Sephadex LH-20 column chromatography methods were used to separate butin and butein; their effects on proliferation and melanogenesis of A375 human melanoma cells were studied by MTT method, enzyme method and NaOH method. RESULTS: Butin and butein were isolated and identified. In the concentration range of 0.50-10.00 μg · mL-1, butin and butein enhanced the proliferation of A375 human melanoma cells, and the effect of butein was stronger than butin(P < 0.05) ; in the concentration range of 0.10-1.00 μg · mL-1, both butin and butein increased the activity of tyrosinase; in the concentration range of 0.50-5.00 μg · mL-1, butein stimulated melanogenesis, but butin inhibited melanogenesis. CONCLUSION: Butin and butein may be the active components of Vernonia anthelmintica Willd; butein promotes the proliferation and stimulates the melanogenesis of A375 human melanoma cells, however, butin mildly inhibites the melanogenesis.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the effect of acteoside on SK-N-SH nerve cell injury induced by okadaic acid (OA).</p><p><b>METHOD</b>SK-N-SH nerve cells were processed with 20 nmol * L OA to establish the Alzheimer's disease (AD) cellular model, and 5, 10, 20 mg . L-1 acteoside was used to antagonize against its effect. Cell morphology was observed under inverted microscope. The cell survival rate was detected with MTT, and the LDH release rate was measured by enzyme label kit. Western blot was applied to determine the expression of phosphorylation tau proteins in nerve cells.</p><p><b>RESULT</b>The acteoside could significantly improve SK-N-SH cell morphology, enhance the cell survival rate, decrease the cell LDH release rate and the expression of phosphorylated tau proteins at p-Ser 199/202 and p-Ser 404 sites, up-regulated the expression of at non-phosphorylated tau proteins at Ser 202 site and Ser 404 sites.</p><p><b>CONCLUSION</b>Acteoside has significant protective effect on nerve cell injury induced by OA.</p>
