ABSTRACT

Survivin is the most recent member of the IAP family, which shows a higher expression in almost all tumor cells, but not detected in most normal cells. Our attention is drawn by survivin due to its selective expression, which was taken as one molecule sign of tumor cells. Accordingly, survivin is examined in most leukemia cell line and leukemia specimen, whose expression also showed the association with leukemia. As in other tumor cells, survivin also plays an important role in the leukemogenesis. Meanwhile the detection of survivin can provide an useful prognosis information. This article is a brief overview of the development of survivin and its relationship with leukemia.

ABSTRACT

Objective: To observe the change of blood cell,marker of vascular endothelium and lung of rabbits during experimental severe decompression sickness(DCS) and to compare each parameter between survival group and dead group.Methods: 14 rabbits were put into the chamber.They stayed for 35mins under 0.55 MPa,and another 35 mins under 0.7 MPa,then decompressed to 0.1 MPa in 4 minutes.Samples were drew before the compression and after decompression for testing count of white blood cell(WBC),neutrophilic leukocyte,lymphocyte and platelet(PLT) and the concentration of ET-1 and the vWF.The tissue of lung of all the rabbits was observed under light and electron microscope.The changes of above parameters of survival rabbits were compared with that of the dead.Results: After rapid decompression,there were 8 rabbits died within 30 minutes,while the other 6 rabbits survived without any symptom of decompression sickness after 24 hours observation.Before the compression,the total count of WBC of dead rabbits was?(11.13?2.37)?10~(9)/L,which was higher the that of survival rabbits,(8.87?2.11)?10~(9)/L.After decompression,the decrease of PLT of dead group,(273?(63)?)10~(9)/L,was more than that of survival group,(148?63)?10~(9)/L.Ttissuedamage of lungs of both dead and survived rabbits was observed as partial alveoli collapse,interstitial edema,local emphysema,various emboli in microcirculation and thickening of respiratory membrane.The damage of lung of dead rabbits was much worse than that of the survival.Conclusion:Experimental DCS induced the increasing of circulation tension,WBC obturation,various emboli,thickening of respiratory membrane,consumption of surface active substance in the body of rabbits.Those rabbits who reacted too severe and had bad compensation ability would die,on the contrary,those rabbits who had a good compensation could survive.

ABSTRACT

Objective: To investigate the change of functional marker and morphology of vascular endothelium induced by simulating diving condition. Methods:14 rabbit were put in the chamber. They stayed for 35 min under 0.55 MPa, and another 35 min under 0.7 MPa, then decompressed to 0.1 Mpa following a stage decompression schedule. Blood sample were draw at 30 min before compression and after decompression for testing endothelin-1(ET-1) and von Willebrand factor (vWF). These parameters were compared to see the change from pre-compression to post-decompression. The morphological change of vascular endothelium was observed under electrical microscope after decompression. Results: After decompression, plasma levels of ET-1 was increased from (1.33?0.23)ng/L to (2.99? 0.35)ng/L and activity of vWF was reduced from (2.35?0.47)% to (1.89?0.34)%. Swelling and defluvium of vascular endothelium was found under electrical microscope. Conclusion: Compression-decompression can cause the damage of vascular endothelium.

ABSTRACT

Objectives To explore the effects of hyperosmosis of sea water and low temperature on hemorrhage and coagulation sys- tem of dogs after open abdominal injury followed by sea water immersion,and to observe the efficiency of plasma in addition to routine first aid,so as to provide a theoretical basis for the early treatment of open abdominal wound in naval warfare.Methods 24 dogs with open in- jury of the abdomen were randomly divided to three groups with 8 animals for each group.The dogs in control group were not subjected to sea water immersion and received routine treatments;dogs in conventional therapy group were immersed in sea water and then given the routine first aid;dogs in plasma group were immersed with aqua marina and then given a combined treatment of routine first aid plus an in- fusion of plasma.The following parameters were measured and recorded:survival time,body temperature,mean arterial blood pressure, osmotic pressure of plasma,prothrombin time,partial thromboplastin time,d-dimer and factorⅡ.Results Prothrombin time and partial thromboplastin time were significantly prolonged,D-Dipolymer increased and factorⅡdecreased after sea immersion with open abdominal wound.The survival time of dogs in plasma group(45.6?12.5h)was significantly longer than that of dogs in both control and conven- tional therapy groups(P

ABSTRACT

Objective To observe the changes in the relative indexes of vascular endothelium, blood coagulation and fibrinolysis system of rabbits with experimental severe decompression sickness (DCS), and to compare the above indexes of surviving rabbits with those of dead in order to analyze the mechanism of causes of death. Methods 14 rabbits were put into a decompression chamber. They stayed for 35 min under an atmospheric pressure of 0.55 MPa, followed by a pressure of 0.7MPa for 35min. Then they were subjected to a pressure of 0.1MPa for 4 minutes. Blood samples were drawn before the compression, under high pressure, and after decompression to determine endothelin-1 (ET-1), von Willebrand factor (vWF), fibrinogen (FIB), D-Dimer, blood coagulation factor Ⅷ(FⅧ), plasminogen (PLG), plasmin inhibitor (PL-IN), prothrombin time (PT), and activated partial thromboplastin time (APTT). The changes of above parameters of surviving rabbits were compared with those of the dead. Results After rapid decompression, 8 rabbits died within 30 minutes, while the other 6 rabbits survived and all symptoms of decompression sickness disappeared 24 hours after decompression. The plasma level of ET-1 increased from 1.33?0.33pg/ml to 2.74?0.87pg/ml after a 30min stay under 0.55MPa, while the activity of vWF increased from 2.62?0.69% to 3.64?1.48%. Compared with the surviving rabbits, those dead rabbits showed significant reduction in FIB (0.92?0.12g/L) and D-Dimer (55.63?12.12ng/ml), after rapid decompression. Conclusions There was a release of vasoactive substance in the blood of rabbits during the period when they stayed under high pressure. After rapid decompression, the most important difference between the survivors and the dead was that there were stronger coagulation activation, more consumption of FIB and weaker secondary fabrinolysis in the latter compared with the survivors.

ABSTRACT

Objective:To study the effects of human CBS(Cord Blood Serum) on hematopoietic cells culture in vitro.Methods:CBS and/or different combinations of cytokines were added to the culture system derived from human bone marrow mononuclear cells.Colony forming capacity of colony forming unit-granulocyte/ monocyte (CFU-GM)?colony forming unit-granulocyte/ erythrocyte/ megakaryocyte/ monocyte (CFU-GEMM)?cobble-stone area forming cells (CAFC)?long-term culture initiating cells (LTC-IC) were observed.Results:CBS had certain cytokine-like stimulating activity that could promote the proliferation and differentiation of hematopoietic cells of bone marrow in vitro. By comparison with cytokines, its GM-CSF-like activity was equal to 65.6 ?g/L rhGM-CSF, and its IL-3-like activity was equal to 2.9 ?g/L rhIL-3.Conclusion:CBS contains stimulating activities for proliferating and differentiating of hematopoietic cells. CBS alone can maintain hematopoietic cells culture in vitro. This study showed the prospect of clinical application of CBS.
