ABSTRACT

ObjectiveTo investigate whether menaquinone-4 (MK-4) can exert a protective effect against carbon tetrachloride (CCl4)-induced acute liver injury (ALI) in mice by alleviating ferroptosis. MethodsAfter adaptive feeding, adult male ICR mice, aged 8 weeks, were divided into Control group, MK-4 group, CCl4 model group (6-hour, 12-hour, and 24-hour), and MK-4+CCl4 group (6-hour, 12-hour, and 24-hour), with 6 mice in each group. The mice in the Control group were given intraperitoneal injection of an equal dose of corn oil; the mice in the MK-4 group were given intraperitoneal injection of 40 mg/kg MK-4 solution, followed by an equal dose of corn oil after 1 hour; the mice in the MK-4+CCl4 group (6-hour, 12-hour, and 24-hour) were given intraperitoneal injection of 40 mg/kg MK-4 solution, and after 1 hour, the mice in this group and the CCl4 model group (6-hour, 12-hour, and 24-hour) were given intraperitoneal injection of 0.3 mL/kg CCl4 solution, with samples collected at 6, 12, and 24 hours. HE staining was used to observe the pathological changes of mouse liver; Prussian blue staining was used to observe iron accumulation in liver tissue; a biochemical analyzer was used to measure the serum levels of aspartate aminotransferase (AST) and alanine aminotransferase (ALT); related kits were used to measure the levels of tissue iron content and the oxidative stress indices malondialdehyde (MDA) and glutathione (GSH) in liver homogenate; RT-PCR was used to measure the expression levels of ferroptosis marker genes (acyl-CoA synthetase long-chain family member 4 [ACSL4], prostaglandin-endoperoxide synthase 2 [PTGS2], and glutathione peroxidase 4 [GPX4]) and iron metabolism-related genes (hemojuvelin [HJV], transferrin receptor 1 [TFR1], and ferroportin [FPN]), and Western blot was used to measure the protein expression level of GPX4. A one-way analysis of variance was used for comparison of continuous data between multiple groups, and the least significant difference t-test was used for further comparison between two groups. ResultsIn the aging study, compared with the Control group, the CCl4 model group (6-hour, 12-hour, and 24-hour) had significant increases in liver weight coefficient and the serum levels of ALT and AST (all P<0.05), and HE staining also showed that liver injury gradually aggravated over time. Meanwhile, compared with the CCl4 model group (6-hour, 12-hour, and 24-hour), the MK-4+CCl4 (12-hour) group had significant reductions in liver weight coefficient and the serum levels of ALT and AST (all P<0.05), with a reduction in the necrotic area of liver tissue, and therefore, 12-hour mouse tissue samples were used for detection in the following study. Compared with the Control group, the CCl4 group had a significant increase in MDA and a significant reduction in GSH (both P<0.05), and compared with the CCl4 group, the MK-4+CCl4 group had a significant reduction in MDA and a significant increase in GSH (both P<0.05). Compared with the Control group, the CCl4 group had significant increases in the key ferroptosis indices ASCL4 and PTGS2 and a significant reduction in GPX4 (all P<0.05); compared with the CCl4 group, the MK-4+CCl4 group had significant reductions in the mRNA expression levels of ASCL4 and PTGS2 and a significant increase in the mRNA expression level of GPX4 (all P<0.05). Western blotting showed that compared with the Control group, the CCl4 group had a significant reduction in the protein expression level of GPX4 (P<0.05), and compared with the CCl4 group, the MK-4+CCl4 group had a significant increase in the protein expression level of GPX4 (P<0.05). Prussian blue staining showed that compared with the Control group, the CCl4 group had a significant increase in iron accumulation; after MK-4 intervention, compared with the CCl4 group, the MK-4+CCl4 group had a significant reduction in iron accumulation. As for the measurement of iron metabolism genes in mouse liver, compared with the Control group, the CCl4 group had a significant increase in iron content, significant reductions in the mRNA expression levels of FPN and HJV, and a significant increase in the mRNA expression level of TFR1 (all P<0.05); after protection with MK-4, there was a significant reduction in iron content, significant increases in the mRNA expression levels of FPN and HJV, and a significant reduction in the mRNA expression level of TFR1 (all P<0.05). ConclusionMK-4 intervention in advance can alleviate CCl4-induced ALI in mice, possibly by inhibiting ferroptosis and improving the expression of iron metabolism-related genes in mouse liver.

ABSTRACT

Objective:To retrospectively analyze the bone marrow smears of neuroblastoma and rhabdomyosarcoma in children and summarize the morphological characteristics of the two types of tumors invading the bone marrow to provide reference for the identification and differential diagnosis.Method:A total of 908 bone marrow specimens were collected from the outpatient and inpatient children who were diagnosed as Neuroblastoma and Rhabdomyosarcoma by pathological tissue or lymph node biopsy in Sun Yat-sen University Cancer Center from January 2013 to July 2020. Of which, 231 cases of tumor bone marrow metastasis were detected. Bone marrow smears were observed and analyzed, classified and summarized according to the morphological characteristics of tumor cells.Result:A total of 231 cases of bone marrow metastases were detected, including two types of tumors, 217 cases Neuroblastoma, with an invasion rate of 34.23%; 14 cases Rhabdomyosarcoma, with an invasion rate of 5.11%. The tumor cells of neuroblastoma were arranged in a pseudo-chrysanthemum or wall-like arrangement and most of them were surrounded by nerve fibers. According to cell size, they could be divided into large cell type and small cell type. Rhabdomyosarcoma cells were mainly medium in size, with vacuoles in the nucleus, and double, triple and multinucleated cells can be seen. The cytoplasm was gauze-like, with bead-like vacuoles at the edges. According to the morphological characteristics of neuroblastoma and rhabdomyosarcoma, they can be differentiated from acute leukemia.Conclusion:Among two malignant solid tumors in children, Neuroblastoma had a higher bone marrow invasion rate, while Rhabdomyosarcoma had a lower bone marrow invasion rate. Rhabdomyosarcoma can be initially divided into embryonal type and acinar type according to whether tumor cells were fused or not.

ABSTRACT

Chromium is a harmful contaminant showing mutagenicity and carcinogenicity.Therefore,detection of chromium requires the development of low-cost and high-sensitivity sensors.Herein,blue-fluorescent carbon quantum dots were synthesized by one-step hydrothermal method from alkali-soluble Poria cocos polysaccharide,which is green source,cheap and easy to obtain,and has no pharmacological ac-tivity due to low water solubility.These carbon quantum dots exhibit good fluorescence stability,water solubility,anti-interference and low cytotoxicity,and can be specifically combined with the detection of Cr(Ⅵ)to form a non-fluorescent complex that causes fluorescence quenching,so they can be used as a label-free nanosensor.High-sensitivity detection of Cr(Ⅵ)was achieved through internal filtering and static quenching effects.The fluorescence quenching degree of carbon dots fluorescent probe showed a good linear relationship with Cr(Ⅵ)concentration in the range of 1-100 μM.The linear equation was F0/F=0.9942+0.01472[Cr(Ⅵ)](R2=0.9922),and the detection limit can be as low as 0.25 μM(S/N=3),which has been successfully applied to Cr(Ⅵ)detection in actual water samples herein.

ABSTRACT

Objective: To evaluate the safety and efficacy of Compound Huangdai Tablets (Realgar-Indigo Naturalis formula, RIF) combined with all-trans retinoic acid (ATRA) to treat acute promyelocytic leukemia (APL). Methods: This study was registered in PROSPERO (CRD42018108118). The relevant literatures on RIF treatment of APL were systematically searched in the following databases: China National Knowledge Infrastructure, Wanfang, VIP Medical Information System, Chinese Biomedical Database, EMBASE, Cochrane Library, and PubMed. The quality of the included studies was evaluated and Review Manager 5.3 software and Stata 13.0 software were used to perform the Meta-analysis. In addition, this study used the method of network pharmacology to conduct a preliminary exploration of the mechanism of RIF on APL. Results: The study included 12 studies involving 775 APL patients. The Meta-analysis showed that there was no significant difference (P 0.05) between the RIF group and the arsenic trioxide (ATO) group for primary outcomes, secondary outcomes apart from liver dysfunction. The incidence of liver dysfunction (P = 0.006) in the RIF group were significantly lower than those in the ATO group. In addition, the cost of maintenance therapy in the RIF group was significantly lower (P 0.05) than the ATO group. Besides, the active ingredients in RIF mainly act on targets proteins such as ACHE, NCOA2, RXRA, and then play a role in the treatment of APL through regulating multiple molecular mechanisms, such as TP53 regulates transcription of cell cycle genes, nuclear receptor transcription pathway. Conclusion: There was no significant difference in efficacy of oral RIF combined with ATRA compared with intravenous ATO combined with ATRA for the treatment of APL. The oral RIF exposed patients to less risk, offered more convenience and had lower prices. RIF can treat APL by multi-target and multi-pathway interventions that inducing apoptosis of APL cells and inhibiting the proliferation of APL cells, and so on. Therefore, oral RIF in the treatment of APL is worthy of further research and development.

ABSTRACT

OBJECTIVE:To study the current situation of pharmacovigilance work in large ,medium and small-scale pharmaceutical manufacturers in Jiangsu Province ,and to provide reference for the pharmacovigilance development of drug manufacturers with different scales. METHODS :The situation of pharmacovigilance work in 108 manufacturers in Jiangsu province was investigated through a questionnaire survey and related websites. The pharmacovigilance work (organization,personnel and training,document,computer system ),drug safety monitoring (case report ,regular safety update report ,post-marketing safety research) and drug risk management (signal management , risk management plan , risk control measures , drug safety communication)of different manufacturers were investigated to put forward the suggestions. RESULTS & CONCLUSIONS :There was no significant difference in the organizational structure (independently established specialized agencies )among manufacturers of different scales (P=0.60). Most of the manufacturers had less than 50% of the proportion in the independent establishment of specialized institutions for pharmacovigilance. There was significant difference in personnel and training (situation and number of full-time staff in charge ,medical and clinical pharmacy personnel number ),document(formulating training management system , entrusted management and key monitoring procedures ),computer system (P<0.05). There was no difference in the main collection ways of case reports among manufacturers of different scales ;however,the number of independent reports in 2019(P< 0.01),the proportion of quality control process for regular safety update reports (P=0.01),and the proportion of carrying out post-marketing safety research in recent five years (P<0.01)in large-scale manufacturers were all significantly higher than small- and medium-scale manufacturers. The proportions of large-scale manufacturers (70.00%) and medium-scale manufacturers (84.38%),which considered “lack of technical guidelines ”as an important factor affecting signal management ,were higher than that of small-scale manufacturers (53.57%)(P=0.01);the proportions of large-scale manufacturers (60.00%)and medium-scale manufacturers(50.00%),which had carried out risk management plans in the past five years ,were higher than that of small-scale manufacturers(30.36%)(P=0.04);the proportion of large-scale manufacturers (50.00%),which adopted the measures in recent 5 year,was higher than medium-scale manufacturers (37.50%)and small-scale manufacturers (25.00%);the proportions of large-scale manufacturers (70.00% ) and medium-scale manufacturers (59.38% ), which carried out communication for pharmacists,were higher than small-scale manufacturers (32.14%)(P<0.01). Large-scale manufacturers outperformed small- and medium-scale manufacturers in terms of pharmacovigilance system ,drug safety monitoring and drug risk management. Large-scale manufacturers had a certain degree of lack of initiative on performing risk management plans ,medium-scale manufacturers on full-time staffs in charge ,and small-scale manufacturers on pharmacovigilance system. So ,it is recommended that large-scale manufacturers take effective control of variety risk as the goal and actively risk management ;medium-scale manufacturers should continuously enhance the awareness of responsibility and improve the investment of resources on pharmacovigilance work ; small-scale manufacturers should pay more attention to improving the pharmacovigilance system and the compliance of specific work.

ABSTRACT

Objective:To observe the effect of functional electrical stimulation (FES) combined with an ankle-foot orthosis (AFO) and gait training on lower limb motor function, gait parameters and the joint angles of hemiplegic stroke survivors.Methods:Thirty-two stroke survivors who met the inclusion criteria were selected and randomly divided into a control group ( n=10), an orthosis group ( n=10), and a combination therapy group ( n=12). In addition to routine medication and rehabilitation, the control group received only gait training, the orthosis group received gait training and an AFO and the combination therapy group was given FES, an AFO and gait training. All three groups were treated for four weeks. Then, the Fugl-Meyer lower extremity assessment (FMA-LE), the Brunnstrom lower extremity assessment (BRL), and Functional Ambulation Categories (FACs) were used to evaluate the lower limb motor function and walking ability of the three groups. The gait parameters of the three groups were quantified using a three-dimensional gait analyzer, and the changes in the hemiplegic gait before and after treatment were compared among the three groups. Results:After the treatment the average FMA-LE, FAC and BRL scores, time-space parameters, and joint angle parameters of all three groups had all improved significantly. After the intervention the average indicators in the combined therapy group (including stride frequency, stride length and walking speed) were all significantly better than in the other two groups.Conclusions:Adding FES to gait training with an AFO can effectively improve lower limb motor function and the walking ability of hemiplegic stroke survivors.

Objective@#To understand prevention and control of infectious disease among primary schools in Xi’an. @*Methods@#A total of 93 primary schools were selected through multi-stage stratified sampling method from 14 counties in Xi’an, then these schools are surveyed to meaure the classroom ventilation.@*Results@#About 28.0% (26/93) of these schools met the national standard for health staffs, with 7.4%(4/54) in urban primary schools and 56.4% in rural primary schools (22/39), the difference between urban and rural areas is statistically significant(P<0.01). Urban primary schools were better than rural primary schools in morning inspection, registration and tracking for illness, verification of vaccination certificates, surveillance and reporting of epidemic outbreaks, verification of school recovery(P<0.05). The average per capita area among the 92 primary schools was (1.22±0.34) m2, with (1.08±0.26) m2 in urban primary schools and (1.42±0.33) m2 in rural primary schools, the difference between urban and rural areas was statistically significant (P<0.01). The average per capita ventilation area of 92 primary schools was (0.15±0.07) m2 with (0.13±0.04) m2 in urban primary schools and (0.19±0.08) m2 in rural primary schools, the difference between urban and rural areas was statistically significant (P<0.01).@*Conclusion@#School-based infectious diseases prevention and control needs to be improved in Xi’an, especially in health personnel. Urban schools appear superior in the prevention and control of infectious diseases and sanitary conditions, while rural schools appear superior in student density and classroom ventilation.

ABSTRACT

The root cause of tumor recurrence or metastasis is a small part of the tumor with unlimited proliferation of tumor stem cells,which can exist for a long time and continue to self-renewal causing rapid tumor proliferation and invasion, metastasis capacity, drug sensitivity, eventually resulting in the problem of weakened clinical treatment. Therefore,it is of great importance to in-depth understanding of tumor stem cells and then to seek strategies to suppress tumor stem cells in clinical targeted tumor therapy. In this paper,the discovery process of tumor stem cells,biological characteristics,identification and culture methods were described to summary a va-riety of methods to inhibit tumor stem cells according to their characteristics so as to treat tumor. Moreover,it focu-ses on the research progress on antibiotic treatment of cancer stem cells.

Objective To analyze mRNA expression of silent information regulator 6 (SIRT6) gene in the blood of population with family history of longevity in Bama county of Guangxi and to explore its association with SIRT6 gene polymorphism and its protein Methods One hundred and thirty-seven people (aged 30 ~ 106, 70 males, 67 females, 6.57% Han nationality, 93.43% Zhuang and Yao nationalities) with family history of longevity (long-lived family history group), and 91 people (aged 22~89, 51 males, 40 females, all Zhuang and Yao nationalities) without family history of longevity were recruited in the study (non-long-lived family history group). Real-time fluorescence quantitative RT-PCR was used to detect mRNA expression of SIRT6 gene in two groups. Results SIRT6 mRNA expression of total and femals in long-lived family history group were higher than those in non-long-lived family history group (P0.05). Conclusion The expression of mRNA in SIRT6 gene may be associated with familial aggregation of longevity in Bama County of Guangxi and high expression of mRNA of SIRT6 in G allele carriers contributes to longevity.

ABSTRACT

Objective To study the effects of case-based learning (CBL) teaching method combined with network on orthodontic postgraduate clinical ability training. Methods Totally 40 ortho-dontic postgraduates were randomly divided into 2 groups (20 per group) named as experimental group (EG) and control group(CG). EG involved in CBL combined with network teaching method,while CG with CBL method only. after 5 teaching cycles,test scores of two groups were analyzed with SPSS 22.0. Results Average scores were (86.60 ±9.71) in EG and (85.45 ±10.79) in CG ,without statistical differences (P=0.73). Pass rate of two groups were 95.00% and 90.00% ,without statistical differences (P=1.00); As for the questionnaire survey, most of the students (90.00%) in EG considered CBL com-bined with network teaching method had no confliction with the clinical time,and helped to carry out in-depth clinical topic discussion. What was more,all of them (100.00%) in EG believed network facilitate their learning. Only part of them (60.00%) in EG thought that they spent a lot of time on reference material preparation. Conclusions Both CBL combined with network teaching method and CBL method can improve the clinical ability of orthodontic postgraduates significantly. However, CBL combined with network teaching method has advantages over time management, knowledge learning and resource sharing.

ABSTRACT

<p><b>OBJECTIVE</b>The aim of this study was to analyze the initial displacement of the maxillary first molar by tip back bend in Tip-Edge force system and to explore the relationships between the displacement and the degree or position of tip back bend.</p><p><b>METHODS</b>The Tip-Edge three-dimensional finite element model was constructed by CT scan of a dry skull with all teeth. The conditions of five types of degree (20 degrees, 30 degrees, 40 degrees, 50 degrees, 60 degrees) and four types of position (2, 4, 6, 8 mm) of tip back bend were added to the three-dimensional finite element model and the initial displacements of the maxillary first molar were calculated.</p><p><b>RESULTS</b>Under the effect of tip back bend, distal and extrusive displacement of the maxillary first molar occurred. The distal and extrusive movement increased as the degree of tip back bend increased or the position of tip back bend moved distally.</p><p><b>CONCLUSION</b>The initial displacement of the maxillary first molar could be controlled by the precise control of the degree and position of tip back bend.</p>

ABSTRACT

Objective To study the protective role of pre-resolving mediator lipoxin A4(LXA4) in the NA+ -K+-ATPase in alveolar type Ⅱ (AT Ⅱ ) epithelial cells of rats exposed to lipopolysaccharide (LPS). Method The AT Ⅱ cells were isolated and purified, and divided randomly into control group (PBS), vehiculum (alcohol 0.7 μL/mL) group, LPS (1 μg/mL) group, LXA4(1/10 mol/mL) group and LPS (1 μg/mL LPS) + LXA4(1/10 mol/mL) group. After exposure to LPS and/or LXA4 for4 hours, NA+-K+ -ATPase and β1-subunits mRNA in AT Ⅱ epithelial cells were detected by using RT-PCR, and ATP, ADP, AMP, total adenine nucleotides (TAN) and energy charge (EC) were measured by using high performance liquid chromatography (HPLC), and then the activities of Na+-K+-ATPase were calculated accordingly. Results The NA+-K+-ATPase α-subunit and β-subunit mRNA were significantly decreased in LPS group ( P < 0.05 vs. control group). However, the expressions of NA+ -K+-ATPase mRNA were significantly enhanced by application of LXA4 to AT Ⅱ epithelial cells exposed to LPS (P <0.05 vs. LPS group). The activities of NA+ -K+ -ATPase were enhanced in LPS group (P <0.05 vs. control group). Compared with control group and LPS group, the activities of NA+-K+-ATpase in LPS + LXA4 group were significantly increased (P <0.01 vs. control group; P <0.05 vs. LPS group). The EC of AT Ⅱ epithelial cells were higher in LPS group ( P < 0.01 vs. control group). There were no significant differences in EC between control group and LPS + LXA4group(P >0.05). Conclusions The pro-resolving mediator LXA4 can enhance the expressions of NA + -K + -ATPase α-subunit and β-subunit mRNA, and the activities of NA + -K + -ATPase in AT Ⅱ epithelial cells or rats exposed to LPS, and ca also balance the metabolism of AT Ⅱ epithelial cells. These findings suggest that LXA4 plays an important role in lung edema clearance in lung injury induced by endotoxin, and the role is likely associated with the enhancement of the expressions of Na+ -K+ -AT-Pase α-subunit and β-subunit, and the activities of Na+ -K* -ATPase, maintaining the balance of metabolism of AT Ⅱ epithelial cells.
