ABSTRACT

Objective@#To discuss the subjects of Chinese geriatrics in recent ten years by analyzing the keywords of Chinese Journal of Geriatrics.@*Methods@#Subjects and subject network(subject-specific network)of Chinese geriatrics were built by the statistical analysis and complex network method based on the analysis of keywords in Chinese Journal of Geriatrics from 2007 to 2018.@*Results@#In recent ten years, 3808 articles were published in Chinese Journal of Geriatrics with a total of 4467 keywords.The keywords frequency was 1 in 3284 keywords.Keywords frequency was not less than 20 in 79 keywords, which was published in two-thirds of all published articles.The keywords frequency was not less than 10 in 186 keywords, which was published in 80% of all published articles, forming an interrelated thematic network.@*Conclusions@#Subjects of Chinese geriatrics have a diversity and relative centrality, and correlations between the subjects are the complexly interdisciplinary subject network.

ABSTRACT

(E)-Methyl-4-aryl-4-oxabut-2-enoate (YH-8) is a novel PKnB protein kinase inhibitor with good anti-tuberculosis activity. To evaluate its pharmacokinetics in rats, a sensitive and selective high performance liquid chromatography-tandem mass spectrometric (LC--MS/MS) method has been developed and validated for the quantification of YH-8 in rat plasma for the first time. Samples were pre-treated using a liquid--liquid extraction with ethyl acetate and the chromatographic separation was performed on a C18 column by gradient elution with methanol--water as the mobile phase. YH-8 was detected using a tandem mass spectrometer in positive selected reaction monitoring (SRM) mode. Method validation revealed good linearity over the range of 1-500 ng/mL for YH-8 with a lower limit of quantification (LLOQ) of 1 ng/mL. Intra- and inter-day precision of YH-8 assay in rat plasma samples were 2.0%-6.8%, with accuracy of the method being 100.69%-106.18%. Stability test showed that when spiked into rat plasma, YH-8 was stable for 12 h at room temperature, for up to 15 days at -70 °C, and after three freeze-thaw cycles. Extracted samples were found to be stable over 12 h in an auto-sampler. The method was successfully applied to the pharmacokinetic study of YH-8 in rats after oral administration at 100 mg/kg and 200 mg/kg.

ABSTRACT

Objective To compare the dosimetry between three -dimensional conformal radiotherapy (3DCRT)and intensity -modulated radiotherapy(IMRT)in the treatment of upper thoracic esophageal carcino-ma,and to provide references to choose radiotherapy program for clinical physician .Methods twenty-five cases with upper esophageal carcinoma (clinical stageⅠ~Ⅲstage)were treated by 3DCRT and IMRT at the concentra three-dimensional radiation treatment planning system .The different exposure doses between target area and effected organs were compared by dose volume histogram ( DVH) with the planed target volume ( PTV) ,which must reach 95% of the prescriptive doses.Results Two different radiotherapy plans of IMRT and 3DCRT:V95, (99.91 ±0.14)%,(95.73 ±4.14)% respectively,P0.05;targeting minimum dose(Dmin)were(5 458.88 ±184.06) cGy,(4541.60 ±599.0)cGy,P0.05;Lung V10 (35.39 ±11.41)%,(29.0 ±8.80)%,P<0.05,Lung V5(44.95 ±15.55)%,(37.27 ±11.93)%,P<0.05. Conclusion Intensity-modulated radiotherapy is better than 3DCRT technology in showing PTV volume ,target conformal degrees and the mean index ,spinal cord protection ,However ,The risk of lung injury could be increased with the enlarged area of low -dose irradiation in lung .

ABSTRACT

Objective To investigate AXIN2 mRNA expression level in hepatocellular carcinoma (HCC) , and to analyze the effect of AXIN2 gene methylation status on its mRNA expression and HCC genesis and development. Methods Fifty-three surgical excised HCC specimens and paired adjacent non-cancerous specimens, seven normal liver specimens and five HCC cell lines were collected. The expression of AXIN2 at mRNA level and the methylation status of AXIN2 gene promoter were determined by quantitative PCR. Results The expression of AXIN2 mRNA was lower in HCC tissues (0.1629 + 0.0679) than that in adjacent non-cancerous tissues (0. 4155 + 0. 2330), and there was significant difference (Z= -2. 567, P = 0. 010). The methylation level of AXIN2 gene in HCC and adjacent non-cancerous tissues (39. 77% ±3. 89%, and 36. 92% ±2. 81%) was significantly higher than that in normal liver tissues (7. 38% ±2. 40% , t=-3. 663 ,P = 0. 009;t= -4. 591 ,P = 0. 007).AXIN2 gene was hypermethylated in all five HCC cell lines. There was a negative correlation between AXIN2 mRNA expression level and the degree of methylation ( r = -0. 458, P = 0. 032). The methylation level was higher in TNM Ⅲ patients of HCC than that in TNM Ⅰ and Ⅱ patients (P =0.008). Conclusion The down-regulation of AXIN2 gene mRNA expression is correlated with its hypermethylation status. The low expression of AXIN2 mRNA and the abnormal methylation of promoter may be one of the important mechanism of HCC genesis and development.
