ABSTRACT

This study investigated the mechanism of Danggui Shaoyao Powder(DSP) against mitophagy in rat model of Alzheimer's disease(AD) induced by streptozotocin(STZ) based on PTEN induced putative kinase 1(PINK1)-Parkin signaling pathway. The AD rat model was established by injecting STZ into the lateral ventricle, and the rats were divided into normal group, model group, DSP low-dose group(12 g·kg~(-1)·d~(-1)), DSP medium-dose group(24 g·kg~(-1)·d~(-1)), and DSP high-dose group(36 g·kg~(-1)·d~(-1)). Morris water maze test was used to detect the learning and memory function of the rats, and transmission electron microscopy and immunofluorescence were employed to detect mitophagy. The protein expression levels of PINK1, Parkin, LC3BⅠ/LC3BⅡ, and p62 were assayed by Western blot. Compared with the normal group, the model group showed a significant decrease in the learning and memory function(P<0.01), reduced protein expression of PINK1 and Parkin(P<0.05), increased protein expression of LC3BⅠ/LC3BⅡ and p62(P<0.05), and decreased occurrence of mitophagy(P<0.01). Compared with the model group, the DSP medium-and high-dose groups notably improved the learning and memory ability of AD rats, which mainly manifested as shortened escape latency, leng-thened time in target quadrants and elevated number of crossing the platform(P<0.05 or P<0.01), remarkably activated mitophagy(P<0.05), up-regulated the protein expression of PINK1 and Parkin, and down-regulated the protein expression of LC3BⅠ/LC3BⅡ and p62(P<0.05 or P<0.01). These results demonstrated that DSP might promote mitophagy mediated by PINK1-Parkin pathway to remove damaged mitochondria and improve mitochondrial function, thereby exerting a neuroprotective effect.

ABSTRACT

Objective@#To investigate the correlation between body composition and eating habits among medical students, and to provide evidence for health promotion.@*Methods@#In December 2021, stratified cluster random sampling method was used to conduct a questionnaire survey and body composition assessment among 445 students in grade one to grade four in Jining Medical University.@*Results@#There were 152 girls (53.3%) and 45 boys (28.1%) with low skeletal muscle mass. Totally 167 students ( 37.5% ) had lower muscle mass, including 115 females (40.4%) and 49 males (30.6%). High body fat percentage was found in 259 (58.2%) students, including 179 females (62.8%) and 80 males (50.0%). There were 192 students (43.1%) with abnormal waist to hip ratio, with 139 females (48.8%) and 53 males (33.1%). In addition, emotional eating score of female students was significantly higher than that of male students(6.85±2.24, 6.11±2.69, t =2.96, P <0.05). Cognitive restricted eating was positively correlated with skeletal muscle mass and musde mass( r=0.13, 0.13, P <0.05). Emotional eating was positively correlated with body fat percentage, body fat and waist hip ratio( r =0.20, 0.20, 0.16, P <0.05). Unrestricted eating was positively correlated with body fat percentage, body fat and waist hip ratio( r =0.15, 0.18, 0.15, P <0.05). Multiple linear regression analysis showed that gender, family residence, physical activity and cognitive eating were associated with skeletal muscle mass and muscle mass of medical students( P <0.05).@*Conclusion@#With low skeletal muscle mass, low muscle mass, body fat percentage and waist and hip high ratio, reasonable eating habits combined with resistance exercise should be adopted to improve their physical health.

ABSTRACT

Objective To analyze the epidemiological features and diagnosis of imported malaria cases in Zhejiang Province from 2017 to 2020, so as to provide the scientific evidence for the management of imported malaria in the province. Methods The data of malaria cases reported in Zhejiang Province were captured from the Information Management System for Parasitic Disease Control of China Information System for Disease Control and Prevention from 2017 to 2020, and the temporal, spatial and human distribution, and initial and definitive diagnosis of imported malaria cases were descriptively analyzed. Results A total of 593 malaria cases were reported in Zhejiang Province from 2017 to 2020, and all were overseas imported cases, including 532 men and 61 women, with a mean age of 41 years. There were 93.93% of the malaria cases from African countries, and the malaria parasites infecting these cases included Plasmodium falciparum, P. vivax, P. ovale, P. malariae and mixed infections, with P. falciparum as the predominant species (76.73%, 455/593). All malaria cases received totally correct initial diagnoses in county- and city-level centers for disease control and prevention (CDC) and entry-exit inspection and quarantine sectors, and the proportion of malaria cases with confirmation at the day of initial diagnosis was 41.48% (207/499) in medical institutions and 66.18% (45/68) in CDC (χ2 = 14.779, P < 0.001). In addition, the median interval [M (QR)] of malaria cases was 1 (2) d from onset to initial diagnosis and 1 (2) d from initial diagnosis to confirmation in Zhejiang Province from 2017 to 2020, and the median interval [M (QR)] of severe malaria cases was significantly longer than that of non-severe cases [2 (3) d vs. 1 (2) d; Z = −3.002, P < 0.05]. Conclusions Zhejiang Province faces great challenges of malaria control, and post-elimination surveillance of malaria still requires to be reinforced. Meanwhile, the awareness of seeking medical services requires to be improved among returners from malaria-endemic regions and the diagnostic capability of malaria requires to be improved among medical professionals.

ABSTRACT

OBJECTIVE: To screen out suitable antibacterial agent and reasonable dosage for timolol maleate eye drops. METHODS: According to the method of bacteriostatic effect test in Chinese Pharmacopoeia (Ch.P) 2015, the antimicrobial effectiveness of antimicrobial preservatives at different concentrations was tested to screen out the optimal agent and dosage. RESULTS: The eye drops added with ethyl hydroxybenzoate met the B-level requirements of bacteriostatic efficacy, which was the same as the sample without antibacterial agent, so ethyl hydroxybenzoate was not suitable as the antimicrobial preservative. The growth of five kinds of test microorganisms was inhibited effectively by 0.03 mg·mL-1 benzalkonium bromide. The timolol maleate eye drops with 0.03 mg·mL-1 benzalkonium bromide met the requirement of the quality standard. CONCLUSION: Benzalkonium bromide can be used as the antimicrobial preservative for timolol maleate eye drops.

ABSTRACT

Objective: To explore the molecular mechanism of the supplemented Naotaifang (sNTF) in the treatment of vascular dementia (VD), and the mRNA expression profiles of hippocampal tissue of VD model rats before and after the intervention of modified sNTF were investigated by microarray analysis. Methods: VD model was established by bilateral common carotid artery ligation. VD rats were treated with sNTF for 30 days. HE staining and Morris water maze were used to evaluate the therapeutic effect of sNTF. The mRNA expression profiles data of VD model rats before and after intervention of sNTF were obtained by Agilent mRNA expression chip. The significantly differentially expressed genes were screened by microarray analysis, and the protein-protein interaction (PPI) network was constructed. The biological processes and signaling pathways in which differentially expressed genes were mainly involved and analyzed by GO and pathway enrichment. Immunohistochemistry and qRT-PCR were used to verify the chip analysis results. Results: HE staining and Morris water maze experiments showed that VD rats showed cerebral ischemia, hippocampal neuron damage, and decreased spatial learning and memory function, but sNTF can partially reverse this trend. 469 differential expression genes were screened by microarray analysis, including 180 up-regulated genes and 289 down-regulated genes. IL6, FGF2, TNF, and IL1b may be the main pharmacodynamic targets of sNTF in the treatment of VD rats, and the results were verified by immunohistochemistry and qRT-PCR. GO and pathway enrichment analysis showed that these genes were closely related to biological processes such as inflammation and apoptosis, and these genes were mainly involved in the regulation of TNF signaling pathway, toll-like receptor signaling pathway and the apoptosis pathway. Conclusion: The results suggested that the therapeutic effect of DSS on AD involves multiple genes and pathways, and and inhibition of hippocampal neuroinflammation may be one of the important mechanisms of its anti-VD.

ABSTRACT

This paper aimed to investigate the effects of Jinwu Jiangu recipe total extract on the IL-17/STAT3 signals in rheumatoid arthritis synovial fibroblasts(RASF). The primary RASFs were cultured by tissue piece method , and divided into blank control group, Jinwu Jiangu recipe low dose group, Jinwu Jiangu recipe middle dose group, Jinwu Jiangu recipe high dose group, and tripterygium glycosides control group. They were then treated with corresponding serum free medium, different doses of Jinwu Jiangu recipe total extract(0.06, 0.6, 6.0 g·L⁻¹), and tripterygium glycosides(0.03 g·L⁻¹) respectively for 24 hours. The gene expression levels of RORα, RORγt, and STAT3 mRNA were detected by polymerase chain reaction(PCR), and the protein activity of IL-17R and pSTAT3 were measured by Western blot assay. The results showed that as compared with blank control group, the expression levels of RORα, RORγt, IL-17R and STAT3 mRNA in RASF were significantly declined(<0.01). As compared with tripterygium glycosides control group, Jinwu Jiangu recipe total extract middle dose group and high dose group can down-regulate the expression levels of RORα, RORγt, IL-17R and STAT3 mRNA(<0.05), and the effect was more obvious in high dose group(<0.01). As compared with blank control group, the protein expression levels of IL-17R and pSTAT3 in each treatment group were obviously decreased(<0.01). As compared with tripterygium glycosides control group, Jinwu Jiangu recipe high dose group had more obvious effect in down-regulating the protein expression of pSTAT3(<0.01). Therefore, Miao medicine Jinwu Jiangu recipe total extract can down-regulate the expressions of RORα, RORγt, and STAT3 mRNA, and inhibit the protein activity of IL-17R and pSTAT3 in RASF.

ABSTRACT

To analyze the medication features and regularity of prescriptions of Chinese medicine in treating patients with dementia based on ancient medical records. In the article, we retrieved the ancient medical records related to the treatment of dementia (from the Han Dynasty to the late Qing period) in Chinese Medical Classics, Chinese Ancient Medical Books and digital library, and then set up a medical records normalized database. The medication features and prescription rules for dementia were analyzed by frequency statistics and association rules (Apriori algorithm, improved mutual information algorithm and complex system entropy clustering). Finally, a total of 156 prescriptions were selected, involving 123 Chinese herbs, with a total frequency of 11 747 for the herbs, and 8 core prescriptions were mined. After the association rules between the frequency and prescriptions for the treatment of dementia were determined, we found that the most commonly used herbs included Fuling (Poria), Yuanzhi (Polygalae Radix), Renshen (Ginseng Radix et Rhizoma), Shichangpu (Acori Tatarinowii Rhizoma), Gancao (Glycyrrhizae Radix et Rhizoma), Danggui (Angelicae Sinensis Radix), Maidong (Ophiopogonis Radix), Baizhu (Bletillae Rhizoma), Dihuang (Rehmanniae Radix) and Ganjiang (Zingiberis Rhizoma); the frequently-used drugs compatibility was mainly for tonifying Qi-blood, regulating Yin and Yang and inducing resuscitation. The drugs were mainly of warm nature and sweet (mild) flavor, and the channel tropism of drugs mainly distributed to the heart, liver, spleen and kidney. The core prescriptions were composed of Renshen (Ginseng Radix et Rhizoma), Fuling (Poria), Yuanzhi (Polygalae Radix), Shichangpu (Acori Tatarinowii Rhizoma), and Baizhu(Bletillae Rhizoma). In conclusion, high frequency herbs and core prescriptions reflect the prescriptions by ancient physicians mainly focus on Qi-replenishing, spleen-invigorating and heart-nourishing, but also reflect the prescription rules of nourishing Yin, enriching blood, eliminating phlegm and warming Yang for the treatment of dementia. The medication features and prescription rules for the treatment of dementia obtained by association rules are useful to guide the clinical practice of Chinese medicine in treatment of dementia.

Objective: To establish a method for the determination of eucalyptol, camphor and menthol in compound menthol camphor eucalyptus oil solution by GC.Methods: An HP-INNOWAX 19091N-216 capillary column(60 m× 0.32 mm , 0.50 μm)was used.The carrier gas was nitrogen with the flow rate of 30 ml·min-1 , the gas was hydrogen with the flow rate of 40 ml·min-1 and the oxidant gas was air with the flow rate of 400 ml· min-1.The detector was FID and the inlet temperature was 250℃.The temperature program was as follows: the initial column temperature was 50℃, and then risen to 160℃ with a rate of 10℃·min-1 and kept for 5 min, and finally risen to 220℃ with a rate of 20℃·min-1 and kept for 3 min.The split ratio was 15∶1 and the injection volume was 1 μl.Results: The linear range of eucalyptol, camphor and menthol was 0.031 9-2.550 0 mg·ml-1 (r=1.000 0), 0.041 3-3.305 0 mg·ml-1 (r=1.000 0) and 0.053 7-4.294 0 mg·ml-1 (r=1.000 0), respectively.The average recovery was 98.24% (RSD=0.3% , n =9), 98.97% (RSD=0.4% , n =9) and 98.98% (RSD=0.5% , n =9), respectively.Conclusion: The method is sensitive and accurate with good stability, which can be used to determine the contents of eucalyptol, camphor and menthol in compound menthol camphor eucalyptus oil solution.

ABSTRACT

Objective To investigate the efficacy of ranibizumab combined with pars plana vitrectomy (PPV) in treatment of patients with diabetic retinopathy together with neovascular glaucoma.Methods A retrospective analysis of 16 patients (16 eyes) with diabetic retinopathy accompanied by neovascular glaucoma underwent vitrectomy combined with ranibizumab between June 2013 and June 2017 was conducted.Data were collected on patients' intraocular pressure (IOP),best-corrected visual acuity (BCVA),iris neovascularization,chamber angle changes before and after surgery,and the postoperative complications were recorded.Results No significant difference was approached in IOP before surgery and 1 day after ranibizumab injection [(42.54 ± 16.52) mmHg vs.(38.78 ± 11.38)mmHg,1 kPa =7.5 mmHg] (P>0.05).And there was also no significant difference in the IOP before surgery and 1 week after vitrectomy [(45.30 ± 12.20) mmHg vs.(42.54 ± 16.52) mmHg] (P >0.05);The IOP at 1 month,3 months and 6 months after surgery was (27.56 ± 19.38) mmHg,(20.49 ± 18.66) mmHg and (19.79 ± 16.53) mmHg,respectively,which were significantly different from preoperative IOP (all P < 0.05).At the end of the follow-up,iris neovascularization completely disappeared in 10 eyes (62.5%) and iris neovascularization was significantly lessened in 6 eyes (37.5%).Conclusion Ranibizumab with PPV for diabetic retinopathy and neovascular glaucoma can effectively reduce intraocular pressure and postoperative complications as well as improve visual acuity slightly.

ABSTRACT

Objective To evaluate the sensitivity to 5 clinically commonly used anticancer drugs in vivo using the zebrafish xenotransplantation models of human lung cancer,stomach cancer,and liver cancer cells,respectively. Methods Zebrafish xenotransplantation models of A549 lung cancer cells,SGC-7901 stomach cancer cells and HepG2 liver cancer cells were established. The xenograft models of A549 cells were treated with three different doses of cis-platinum, paclitaxel, vinorelbine, endostar and bevacizumab, respectively. The SGC-7901 model was treated with three concentrations or doses of paclitaxel, irinotecan, hydroxyurea, cis-platinum and 5-fluorouracil, respectively. And the HepG2 model was treated with three concentrations or doses of adriamycin,gemcitabine,hydroxyurea,cis-platinum and 5-fluorouracil. The tumors were analyzed and quantified in vivo by fluorescence microscopy,and the inhibition rates of tumor growth with each drug were calculated and compared with the model control group for statistical significance. Results All of the tested anticancer drugs showed inhibitory effect on tumor cells in the zebrafish xenograft models with statistical significance in a dose-dependent manner. During the drug sensitivity test,the inhibition rate of bevacizumab on A549 lung cancer cells decreased in the order(65%)> cis-platinum(55%)> vinorelbine(40%)> endostar(39%)>paclitaxel(27%). As for the SGC-7901 stomach cancer cells, the tumor growth inhibition rate decreased in the order hydroxyurea(46%)> 5-FU(31%)= irinotecan(31%)> paclitaxel(26%)> cis-platinum(24%). And the therapeutic effect of cis-platinum on the HepG2 liver cancer cells decreased in the order(64%)> hydroxyurea(56%)>gemcitabine(46%)> adriamycin(45%)> 5-FU(38%). Conclusions Zebrafish xenotransplantation models of cancer cells are suitable for in vivo sensitivity test of anticancer drugs.

Objective To investigate preventive and therapeutic effects ofAngong Niuhuang Pills (ANP) on cerebrovascular diseases in Zebrafish models.Methods Zebrafish cerebral hemorrhage was induced by treatment with simvastatin,thrombosis by arachidonic acid,blood vessel loss by simvastatin,and Alzheimer's disease (AD) by A1C13.Zebrafish models were treated with ANP by direct drug soaking at various concentrations.Results ANP had a significantly preventive effect on Zebrafish cerebral hemorrhage with an efficacy of 11%,41%,and 48% at concentrations of 27.8,83.3,and 250 μg/mL,respectively,and the incidence of cerebral hemorrhage in 83.3 and 250 g/mL groups decreased significantly compared with model group (P < 0.001).Compared with model group,the red cell staining intensity of Zebrafish in 333 and 1 000 g/mL groups increased significantly (P < 0.001),and the thrombus prevention rates of 111,333,and 1000 g/mL groups were 5%,33%,and 64% respectively.Compared with model group,the intestinal vascular area of Zebrafish in ANP 11.1,33.3,and 100.0 μg/mL groups increased significantly (P < 0.001),and the rate of angiogenesis was 21%,25% and 26% respectively.ANP promoted dyskinesia recovery by 48%,88%,and 78%,and improved response efficiency by 15%,82% and 75%,respectively in AD zebrafish at concentrations of 111,333,and 1000 μg/mL.Conclusions Our results in this study support ANP as a preventive and therapeutic medicine for cerebral hemorrhage,cerebral ischemia,thrombosis and AD.

ABSTRACT

BACKGROUND:Mesenchymal stem cel s are multipotent progenitor cel s that can be isolated from the bone marrow, adipose tissue, umbilical cord blood and so on. Mesenchymal stem cel s possess strong immunosuppressive effects on both innate and adaptive immunity. OBJECTIVE:To summarize the immunomodulatory properties of mesenchymal stem cel s for T lymphocytes, B lymphocytes, natural kil er cel s and dentritic cel s and prospect its therapeutic implication. METHODS:PubMed and CNKI databases were searched for relevant literatures published from 2000 to 2015. The key words were“mesenchymal stem cel s, immunomodulator, T cel s, B cel s, NK cel s, dendriticc cel s”in Chinese and English, respectively. Then, 61 papers were included and further analyzed. RESULTS AND CONCLUSION:The immunomodulatory effects of MSCs on lymphocytes, B lymphocytes, natural kil er cel s and dentritic cel s were elicited by cel-cel contact and soluble cytokines. But mesenchymal stem cel s from different sources hold different immunomodulatory effect on immune cel s. When we use mesenchymal stem cel s in clinic, some important factors, such as isolation methods, cel sources, colonization sites, should be taken into account. In order to ensure the clinical safety and effectiveness, there are stil many problems to be further studied before mesenchymal stem cel s can be widely used in clinic.

ABSTRACT

<p><b>OBJECTIVE</b>To observe the therapeutic angiogenesis effect of naotai recipe (NR) on local ischemia/reperfusion (I/R) injury of rats by observing signaling pathway of hypoxia-inducible factor-lα (HIF-1α) and vascular endothelial growth factor (VEGF).</p><p><b>METHODS</b>Totally 120 Sprague-Dawley (SD) rats were randomly divided into 4 groups, namely, the normal control group (n =12), the sham-operation group (n =12), the I/R model group (n =48), and the NR group (n =48). Cerebral I/R injury models were established using thread suture method. Rats in the I/R model group and the NR group were sub-divided into 4 sub-groups according to the 1st, 3rd, 5th, and 7th I/R day (n =12). The phenomenon of neovasculization was observed by immunofluorescence staining. The protein and mRNA expression levels of HIF-la, VEGF-A, and VEGFR II receptor were detected by RT-PCR.</p><p><b>RESULTS</b>There were a large amount of labels for neovasculization in the ischemic area of the NR group. Double-immunofluorescence labeling [vWF (red) and BrdU (green)] was observed in the NR group. Compared with the model group, the HIF-1α protein expression was obviously enhanced on the 1 st day of I/R (P <0.01), and the VEGF protein expression started to enhance on the 3rd day in the NR group (P <0.01). The VEGFR protein expression level was the highest in the NR group on the 5th day of I/R (P <0.01). The protein expression of VEGF and HIF-1α started to decrease on the 7th day of I/R.</p><p><b>CONCLUSION</b>NR could strengthen angiogenesis after I/R by elevating the expression of HIF-lα and activating HIF-lα/VEGF signaling pathway.</p>

ABSTRACT

Objective: To detect the mRNA levels of AK097082 and BC031001 encoded by psiTPTE22 gene in order to determine whether psiTPTE22 gene is the microarray-detected over-expressed gene in colon cancer and explore the role of psiTPTE22 in carcinogenesis. Methods: RT-PCR and real-time fluorescence quantitative PCR were used to detect the transcription of AK097082 and BC031001 encoded by psiTPTE22 gene in kidney, liver, stomach, lung, and colon cancer tissues and adjacent normal tissues as well as nine cancer cell lines and placenta tissues. Results: RT-PCR showed that only the mRNA of BC031001 encoded by psiTPTE22 was expressed in colon tissues. Real-time quantitative PCR assay and F test confirmed that there was no significant difference between colon cancer and adjacent normal tissues (P > 0.05). So psiTPTE22 was not the specific up-regulated gene in colon cancer detected by microarray. Real-time quantitative PCR assays also confirmed that BC031001 was over-expressed in kidney, liver, stomach, and lung normal tissues but had low expression in corresponding cancer tissues. BC031001 had no expression or weak expression in several cancer cell lines but had relatively high expression in placenta tissues. F tests found that the difference was significant in the expression level of BC031001 between kidney, liver, stomach, and lung adjacent normal tissues and the corresponding cancer tissues (P < 0.05). Conclusions: The mRNA expression of BC031001 encoded by psiTPTE22 gene has negative correlation with several kinds of cancer. Further studies are needed to reveal the mechanism underlying its down-regulation and its role in a carcinogenesis.
