ABSTRACT

Background: Endoscopic ultrasonography (EUS) can display the different layers and structures of the intestinal wall, however, its value in diagnosis and assessment of disease activity of inflammatory bowel disease (IBD) is still in exploration. Aims: To investigate the value of colonoscopy and EUS in diagnosis and evaluation of disease severity of IBD. Methods: A retrospective study was conducted in patients with active IBD admitted at the First Affiliated Hospital of Kunming Medical University from September 2017 to September 2019. The results and characteristics of colonoscopy and EUS were analyzed. Results: One hundred and five active IBD patients were enrolled, including 68 cases of ulcerative colitis (UC) and 37 cases of Crohn's disease (CD). ABSTRACT

Objective MicroRNAs are differentially expressed in colorectal cancer tumor tissues and adjacent normal tissues, which play an important role in the development of colorectal cancer. This study aims to investigate the expression of microRNA-7-5p in colorectal cancer patients and its influence on the proliferation and apoptosis of colon cancer cell CaCo2. Methods The high-throughput microarray was used to screen differentially expressed microRNAs, and real-time quantitative PCR (RT-PCR) was used to detect the expression of microRNA-7-5p in 10 cases of colorectal cancer patients and corresponding adjacent normal tissues. Western blot was performed to detect the expression of intestinal trefoil factor (TFF3) in different tissues and CaCo2 cells after transfection with microRNA-7-5p. The expression of TFF3 in different tissues and CaCo2 cells transfected with microRNA-7-5p was detected. TUNEL combined with flow cytometry was used to detect the apoptosis of CaCo2 cells after transfection. The CCK-8 assay was used to detect the proliferation of CaCo2 cells after transfection. Results The relative expression of MicroRNA-7-5p in colorectal cancer tissue was 0.409 ± 0.095, which was significantly lower than that of normal tissue adjacent to cancer (1.000 ± 0.014), and the difference was statistically significant (P <0.01). The relative expression of TFF3 in colorectal cancer tissues was significantly higher than that in normal adjacent tissues (P <0.01). The relative expression of TFF3 in CaCo2 cells decreased in the overexpressed microRNA-7-5p of the control group (0.729 ± 0.041). The proliferation ability (0.930 ± 0.007) was significantly lower in the blank control group (0.990 ± 0.005) (P <0.01), and it could increase the proportion of early apoptotic cells (50.700 ± 0.989) and late apoptotic cells (40.525 ± 0.515). Conclusion MicroRNA-7-5p is lowly expressed in colorectal cancer and is associated with the occurrence and development of colorectal cancer. Up-regulated microRNA-7-5p inhibits the proliferation of colon cancer cell CaCo2 and promotes its apoptosis.

Colorectal cancer screening and monitoring are effective Methods to reduce the incidence and mortality of colorectal cancer. Endoscopic screening is indeed the most effective method at present. However, endoscopy is not perfect, and it may miss some precancerous lesions and colorectal cancer, or incomplete resection of some malignant lesions. Thereafter these lesions were diagnosed before the next endoscopic examination of cancer, that is interval Colorectal cancer. This article focuses on the latest findings on the etiology ,classification ,clinical molecular biology characteristics of I-CRCs,and also sums up the correct identification and monitoring of high-risk populations.

ABSTRACT

Objective To compare the clinical efficacy of endoscopic ligation between using intensive ligation and non-intensive ligation methods for severe esophageal varices.Methods A total of 89 patients with cirrhosis and esophageal varices in our hospital from 2007 to 2013 were selected and divided into two groups.Patients in the observation group were treated with endoscopic ligation using intensive ligation (39 cases),and patients in the control group were treated with endoscopic ligation using non-intensive ligation (50 cases).Sixshooter multi-band ligator was used for endoscopic therapy.If the number of loop ligature collar was larger than 6 and less than or equal to 12,it was regarded as intensive ligation;if the number of loop ligature collar was smaller or equal to 6,it was regarded as non-intensive ligation.The clinical efficacy was compared between the two groups.Results The average number of ligation in the observation group was greater than that in the control group,there was statistically significant difference (t=29.11,P=0.000).The cure rate at the first time in the observation group was higher than that in the control group,there was statistically significant difference (x2=5.002,P=0.025).No statistically significant difference was found in efficient rate at the first ligation (x2 =2.268,P=0.132).The total effective rates in the observation group half a year,one year,two years and three years after operation was significantly higher than those in the control group (P<0.05).Conclusion Intensive ligation is a safe,reliable and effective approach for esophageal varices,which could have a high effective rate in the treatment of esophageal varices and reduce the average number of ligation,patients recover well after surgery and have a good long-term prognosis.

ABSTRACT

A detection method for furazolidone and florfenicol in soil with various environmental matrices was established using ultra performance liquid chromatography-tandem spectrometry (UPLC / MS / MS) technique. Extracting solution of a mixture of phosphate buffer (pH = 3) and acetonitrile (3 : 7, V/ V) was used in this experiment. The extracted water samples were enriched by SAX-HLB solid phase extraction column before the process of nitrogen blowing ( high purity nitrogen). The enriched antibiotics were desalted with 8 mL of methanol. Waters BEH C18(2. 1 × 100 mm) column was used for the sample separation. UPLC / MS / MS was carried out for qualitative and quantitative analysis under multi-reaction monitoring mode. The detection limit of the method was determined by 3 times of signal-to-noise ratio, and the limit of determination of the method was determined by 10 times of signal-to-noise ratio. The results showed that the detection limits of furazolidone and florfenicol were 1. 19 and 0. 41 μg / kg, respectively, and the limits of quantitation of furazolidone and florfenicol were 3. 40 and 1. 37 μg / kg, respectively. Besides, recovery experiment showed that, for the soil samples spiked with 50 μg / L furazolidone and florfenicol, the recoveries were 79% for florfenicol and 92% for furazolidone. Similarly, for the soil samples spiked with 200 μg / L furazolidone and florfenicol, the recoveries were 96% for furazolidone and 86% for florfenicol.

ABSTRACT

Objective To evaluate the bacteriostatic effect of recombinant human lactoferrin(rhLF) on Helicobacter(H .) py‐lori and its influence on CagA ,Ure and gastric mucosal IL‐8 .Methods The minimum inhibitory concentration(MIC)and the influ‐ence of different drug concentrations on the proliferation of H .pylori were detected .The effects of rhLF on the mRNA and protein expressions of CagA and Ure in H .pylori were detected by RT‐PCR and Western blot ,respectively .The animal study :Balb/c mice were adopted and assigned randomly into four groups ,including the standard triple+rhLF(group A) ,rhLF(group B) ,standard tri‐ple(group C) and normal saline(group D) .The histopathological HE staining was used to observe the gastric inflammation and ELISA was used to detect the IL‐8 level of gastric tissue in each group .Results MIC was 0 .5 mg/mL ,moreover rhLF inhibited the bacterial growth and proliferation with a concentration‐dependent manner .rhLF could reduce the expression of H .pylori major viru‐lence factor CagA ,mRNA and protein of Ure .Comparing the group A with the group B ,C and D ,the gastric mucosal inflammation score and the IL‐8 levels of gastric tissue homogenates had statistically significant differences(P<0 .05) .Conclusion rhLF inhibits the growth and proliferation of H .pylori ,moreover inhibit the expression of major virulence factor CagA in H .pylori ,mRNA and protein of Ure in different degrees ,weakens its pathogenicity ,meanwhile reduces the IL‐8 level in mice gastric mucosa ,and allevi‐ates H .pylori related gastric mucosal inflammatory response .

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the prevalence of Helicobacter pylori (H.pylori) resistance to metronidazole among three populations in Yunnan.</p><p><b>METHODS</b>Susceptibilities to metronidazole among 109 H. pylori strains (33 H. pylori strains from Han, 31 H. pylori strains from Bai and 45 H. pylori strains from Naxi ethnic populations) were tested by Epsilometer test (E-test).</p><p><b>RESULTS</b>In 109 H. pylori strains, the overall metronidazole resistance rate was 67.89%. There were no significant difference in the metronidazole resistant rates of H. pylori among Han, Bai, Naxi populations Yunnan in terms of the distribution on age and upper gastroduodenal diseases. In the facet of gender, metronidazole resistant rate of H. pylori was significantly lower in Han males than in females (chi2=5.304, P=0.027), but not seen in the Bai or Naxi peoples.</p><p><b>CONCLUSION</b>Metronidazole resistance rate of H. pyloriin Yunnan was high, but no significant difference was found among Han, Bai, Naxi peoples in the province.</p>

ABSTRACT

Objective To search the feasibility of mutation,RdxA gene was analyzed by the metronidazole induction experiment.Method The non-multiple,sensitive H.pylori strains,were selected and subjected to metronidazole induction experiment,which made H.pylori strains transform susceptibility to resistance.Extracted DNA of metronidazole sensitive and resistant pairs of H.pylori strains before and after induced,amplified by PCR and sequenced rdxA gene.Result 4 of 12 H.pylori strains were induced successfully with metronidazole whose MICs increased by 16~64 times.Strains were transferred three times on metronidazole-free medium and the MICs redetermined were the same as those after induced.There was 0.2~0.8% difference between pairs of metronidazole sensitive and resistant H.pylori strains.There was 2.2~2.7% difference between unrelated H.pylori strains.Conclusion The metronidazole induction experiment could make H.pylori acquire stable resistance to metronidazole in a short period and exclude that the natural polymorphism of rdxA gene influenced on mutation analysis.There was good hope that the experiment would become a good method to research metronidazole resistance mechanism
