ABSTRACT

Abstract Thirty-six isolates of psittacid herpesvirus (PsHV), obtained from 12 different species of psittacids in Brazil, were genotypically characterized by restriction fragment length polymorphism (RFLP) analysis and PCR amplification. RFLP analysis with the PstI enzyme revealed four distinct restriction patterns (A1, X, W and Y), of which only A1 (corresponding to PsHV-1) had previously been described. To study PCR amplification patterns, six pairs of primers were used. Using this method, six variants were identified, of which, variants 10, 8, and 9 (in this order) were most prevalent, followed by variants 1, 4, and 5. It was not possible to correlate the PCR and RFLP patterns. Twenty-nine of the 36 isolates were shown to contain a 419 bp fragment of the UL16 gene, displaying high similarity to the PsHV-1 sequences available in GenBank. Comparison of the results with the literature data suggests that the 36 Brazilian isolates from this study belong to genotype 1 and serotype 1.

ABSTRACT

Objetivo: Apesar do grande número de publicações em cirurgia oncoplástica e reconstrutiva da mama, diversas questões permanecem controversas. Assim, o objetivo desta Reunião de Consenso, foi desenvolver um guia prático de recomendações baseadas nas melhores evidências disponíveis na literatura. Método: Os painelistas foram os membros da Comissão de Cirurgia Oncoplástica e Reconstrutiva da Sociedade Brasileira de Mastologia. A reunião foi realizada em agosto de 2015 em Bento Gonçalves (RS). Cada painelista recebeu e respondeu previamente um questionário com 46 itens, com base na melhor evidencia cientifica e em sua experiência. Foi considerado consenso a concordância de 75% entre painelistas. Resultados: Houve consenso em 25 itens, dos quais para oito houve concordância de 100%. O mais importantes foram: comprometimento das margens em cirurgia oncoplástica pode ser resolvido com ampliação de margens na maioria dos casos; tumores multifocais não são contraindicação para cirurgia oncoplástica; idade >70 anos não representa contraindicação para uso de técnicas oncoplásticas; reconstrução imediata pode ser indicada com segurança para a maioria das candidatas à mastectomia; pacientes com indicação de radioterapia pós-mastectomia podem ser submetidas à reconstrução imediata, devendo ter ciência dos riscos maiores para mau resultado estético; mastectomia com preservação do complexo areolopapilar é segura nos casos de câncer; radioterapia após a mastectomia com preservação do complexo areolopapilar não está indicada fora dos critérios clássicos de irradiação do plastrão; tela abdominal reduz chances de hernia no caso de reconstrução com TRAM. Conclusão: através desta reunião foi possível estabelecer importantes pontos consensuais de acordo com a opinião dos especialistas, que poderão auxiliar os mastologistas na tomada de decisões em cirurgias oncoplásticas e reconstrutivas da mama.


Objective: Despite the large number of publications in oncoplastic and breast reconstructive surgery, several issues remain controversial. The aim of this Consensus Meeting was to develop a practical guide of recommendations based on the best evidence in the literature. Method: All panelists were members of the Oncoplastic Commission of the Brazilian Society of Mastology. The Consensus Meeting was held in Bento Gonçalves (RS), in August 2015. Each panelist received and answered a questionnaire with 46 items, based on the best evidence in the literature and in their expertise. It was considered consensus the agreement of 75% between panelists. Results: There was consensus on 25 items, of which eight were for 100% agreement. The most important of these topics were: involvement of the margins in oncoplastic surgery can be solved by resection of margins in most cases; multifocal tumors is not a contraindication for oncoplastic surgery; age >70 years is not a contraindication for use of oncoplastic techniques; immediate reconstruction can be performed safely to most candidates for mastectomy; patients for post-mastectomy radiotherapy may be subject to immediate reconstruction and should be aware of the risks for poor aesthetic result; mastectomy with preservation of the nipple and areola complex is safe in cancer; radiotherapy after mastectomy with preservation of the nipple and areola complex is not indicated outside the classical criteria of irradiation chest wall; abdominal mash reduces chances of hernia in TRAM flaps. Conclusion: In this meeting it was possible to establish important consensus points according to the opinion of experts, which can help breast surgeons in their decision-making in oncoplastic and reconstructive surgery of the breast.

ABSTRACT

Purpose: To investigate the expression of alkaline phosphatase (ALP) activity in gingival fibroblasts from individuals with chronic periodontitis (CP) and drug-induced gingival hyperplasia (DGH) induced by diphenylhydantoin. Methods: Gingival fragments were obtained from 13 patients (8 women and 5 men, from 22 to 74 years of age), with 4 fragments from clinically normal gingiva (NG), 5 from biopsy of periodontal pockets with CP, and 4 from DGH induced by diphenylhydantoin. Using an enzymatic digestion procedure, gingival cell suspensions containing ALP-positive fibroblasts were prepared without affecting ALP activity. Cytochemistry and histochemistry analyses were performed. Results: Fibroblasts from NG presented low levels of ALP when compared to CP and DGH, which showed elevated and intermediate levels of ALP, respectively. Little cell proliferation was observed for fibroblasts from CP and DGH as compared to NG. However, the quantity of cells recovered from the subcultures was similar to the quantity recovered from the initial cell culture for the three sources. Conclusion: The expression of ALP is increased in CP and DGH, and fibroblasts in CP and DGH show low proliferation. This suggests that periodontal inflammation and diphenylhydantoin may influence ALP expression and human gingival fibroblast expansion. Other studies are necessary to better assess the importance of ALP in the development and progression of CP and DGH.


Objetivo: Investigar a expressão da atividade de fosfatase alcalina (ALP) em fibroblastos gengivais de sujeitos com periodontite crônica (PC) e hiperplasia gengival medicamentosa (HGM) induzida por difenilhidantoína. Metodologia: Os fragmentos gengivais foram obtidos de 13 pacientes (8 mulheres e 5 homens, 22 a 74 anos), sendo 4 fragmentos procedentes de gengiva clinicamente normal (GN), 5 de biópsias de bolsas periodontais com PC e 4 de HGM induzida pela difenilhidantoína. Através de digestão enzimática, suspensões de células gengivais foram preparadas contendo fibroblastos ALP-positivos sem afetar a atividade da enzima ALP. Análises citoquímica e histoquímica foram realizadas. Resultados: Fibroblastos de GN apresentaram fraca positividade para ALP quando comparados com PC e HGM. PC e HGM exibiram níveis elevados e intermediários, respectivamente, de ALP. Observou-se menor proliferação celular na cultura de células em PC e HGM que em GN. Entretanto, a quantidade de células recuperadas das sub-culturas foi similar à quantidade recuperada na cultura inicial para as três linhagens. Conclusões: A expressão de ALP na PC e na HGM encontra-se aumentada e ocorre baixa proliferação de fibroblastos na PC e na HGM. Isto sugere que a inflamação periodontal e a difenilhidantoína podem influenciar a expressão de ALP e a multiplicação de fibroblastos gengivais humanos. Outros estudos são necessários para melhor avaliar a importância de ALP no desenvolvimento e na progressão de PC e HGM.

ABSTRACT

An outbreak of Malignant Catarrhal Fever (MCF) resulted in death of five female buffaloes and one domestic cow from the same farm. Four buffaloes died 10-15 days after the appearance of clinical signs, while the fifth was euthanized in extremis, after similar clinical signs. Histopathological lesions included multifocal histiolymphocytic epicarditis, myocarditis and lymphocytic interstitial pneumonia, which are commonly seen in cases of MCF in buffaloes. Furthermore, lymphocytic vasculitis centered in the adventitia, with occasional fibrinoid necrosis in the muscular layer, was found in the kidneys, liver, spleen, lymph nodes and brain. Nucleotide sequencing of DNA fragments from the central nervous system amplified by PCR revealed 98 percent similarity with known OHV-2 sequences from Genbank. Additionally, PCR analysis also revealed the presence of OHV-2 DNA in the peripheral mononuclear blood cells of two clinically healthy buffaloes. The diagnosis of MCFwas based on epidemiological, clinical, gross and histopathological findings and on the results of a semi-nested PCR followed by nucleotide sequencing.


É relatado um surto de febre catarral maligna (FCM) em Minas Gerais, que resultou na morte de 5 búfalas e uma vaca de uma mesma propriedade. Quatro búfalas morreram com 10-15 dias após o início dos sinais clínicos e uma búfala foi sacrificada in extremis, após manifestar sinais clínicos semelhantes. O exame histopatológico revelou lesões comumente observadas em búfalos com FCM como epicardite e miocardite histiolinfocítica multifocal e pneumonia linfocítica intersticial. Além disso, vasculite linfocítica, principalmente na camada adventícia, com necrose fibrinóide da camada muscular, foi observada no rim, fígado, baço, linfonodos e cérebro. A seqüência de nucleotídeos amplificada pela técnica de PCR revelou 98 por cento de homologia entre o fragmento de DNA amplificado da amostra do sistema nervoso central (SNC) da búfala com seqüências de OHV-2 previamente depositadas no Genbank. Adicionalmente, a técnica de PCR revelou a presença do DNA viral no sangue total de 2 búfalas, clinicamente sadias. O diagnóstico de FCM foi baseado em dados epidemiológicos, clínicos, patológicos, histopatológicos e semi-nested PCR.

ABSTRACT

Justificar a reconstrução da pars interarticularis pela técnica modificada de Buck como método efetivo no tratamento de lombalgia por espondilólise ou espondilolistese tipo 1, refratárias ao tratamento conservador. MÉTODOS: avaliação retrospectiva de oito pacientes com idade entre 11 e 34 anos, operados e acompanhados no período de outubro de 2004 a maio de 2008 com a técnica de Buck utilizando-se parafusos canulados de titânio. Nenhum paciente foi imobilizado no pós-operatório. Os pacientes foram selecionados após serem submetidos a estudos por imagem utilizando radiografias e ressonância magnética. Também foram avaliados no pós-operatório, segundo os critérios de Henderson e quanto à satisfação com o tratamento, assim como o índice de consolidação. RESULTADOS: sete pacientes foram classificados como excelentes, retornando ao nível de prática esportiva e laboral prévia. Dentre eles, um evoluiu com parestesia transitória, com recuperação completa. A oitava paciente apresentou uma pseudartrose bilateral, mas com bom resultado do ponto de vista clínico. Todos os pacientes ficaram satisfeitos com o resultado. CONCLUSÃO: a reconstrução ístmica com parafusos de titânio atingiu excelentes resultados ao aliar biomecânica estável, dissecção mínima da musculatura, com preservação da anatomia e mobilidade do segmento.


To justify the modified Buck's technique of pars reconstruction as an effective method to spondylolysis and low grade spondylolisthesis treatment. METHODS: eight patients with ages 11 to 34 years were retrospectively evaluated from October 2004 to may 2008. They were operated with Buckïs technique using titanium canulated screws. Immobilization wasn't used in any patient in post-operative period. All patients were studied with magnetic resonance imaging. All patients were assessed in post-operative period with Henderson's criteria of functional capacity. RESULTS: seven patients were assessed as excellent. They didnït complain pain and were able to return to former occupation and sportive activities. One patient presented temporary paresthesia and leg weakness, and one patient bilateral pseudoarthrosis that cursed with good grade functional capacity. All patients were satisfied with treatment. CONCLUSION: the isthmic reconstruction using titanium screws allowed excellent results allied with stable biomechanics, minimal injury to back muscles, preservation of anatomy and segmental mobility, and better operated spinal imaging.


Justificar la reconstrucción de la pars interarticularis por la técnica modificada de Buck como método efectivo en el tratamiento de la lumbalgia por espondilosis o espondilolistesis tipo 1, refractarias al tratamiento conservador. MÉTODOS: evaluación retrospectiva de ocho pacientes con edad entre 11 a 34 años, operados y acompañados en el periodo de Octubre de 2004 hasta Mayo de 2008 con la técnica de Buck usando tornillos canulados de titanio. Ningún paciente fue inmovilizado en el postoperatorio. Los pacientes fueron seleccionados después de ser sometidos a estudios por imagen utilizando radiografías y resonancias magnéticas. También fueron evaluados en el postoperatorio, según los criterios de Henderson y el grado de satisfacción con el tratamiento, así como también el índice de consolidación. RESULTADOS: siete pacientes fueron clasificados como excelentes, retornando al nivel de práctica deportiva y laboral previa. Dentro estos siete pacientes, uno evolucionó con parestesias transitorias, con recuperación completa. La octava paciente presentó una pseudoartrosis bilateral, pero con buen resultado desde el punto de vista clínico. Todos los pacientes quedaron satisfechos con el resultado. CONCLUSIÓN: la reconstrucción ístmica con tornillos de titanio tuvo excelentes resultados al juntar biomecánica estable, disección mínima de la musculatura, con preservación de la anatomía y modalidad del segmento.

During the period of October 2004 until February 2006, 75 samples of central nervous system (CNS) obtained from horses that died with neurological signs in the state of Minas Gerais were sent to the Laboratory of Compared Virology in ICB/UFMG for diagnosis of equine herpesviruses. All samples were previously diagnosticated negative for rabies virus by the Laboratório de Saúde Animal of the Instituto Mineiro de Agropecuária (IMA). Among the analyzed samples, 39 (52%) were positive for the equine herpesvirus 1(EHV-1) through the polymerase chain reaction assay (PCR). In most cases, the histopathological examination of the CNS revealed a mild vasculitis with perivascular mononuclear cuffing, congestion, arterial thrombosis and degeneration of nervous tissue. The positive CNS samples for EHV-1 were obtained from horses. sampled from 30 municipalities of Minas Gerais state. The cases occurred in an isolated form in different periods of the year, not presenting a seasonal character. The clinical course duration was acute, varying between one and four days. The most frequently observed neurological signs were ataxia, unsteadiness in the hind limb, paralysis of hind limbs and recumbency. According to information provided by IMA, the infections caused by EHV-1 were as frequent as the ones caused by rabies virus in horses of Minas Gerais state during the studied period. Hence, it became important to include EHV-1 encephalitis in the differential diagnosis from other diseases of the central nervous system in horses of Minas Gerais state.


Durante o período de Outubro de 2004 a Fevereiro de 2006, 75 amostras de sistema nervoso central (SNC) oriundas de eqüinos que morreram com sinais neurológicos no estado de Minas Gerais foram enviadas ao Laboratório de Virologia Comparada no ICB/UFMG para o diagnóstico de herpesvírus eqüino. Essas amostras foram previamente diagnosticadas negativas para o vírus da raiva através dos testes de imunofluorescência direta e inoculação em camundongos,no Laboratório de Saúde Animal (LSA) do Instituto Mineiro de Agropecuária (IMA). Dentre as amostras analisadas, 39 (52%) foram positivas para o herpesvírus eqüino 1 (EHV-1) através da técnica de reação em cadeia pela polimerase (PCR). Na maioria dos casos, o exame histopatológico do SNC revelou uma discreta vasculite com infiltrado perivascular de células mononucleares, congestão, trombose arterial e degeneração do tecido nervoso central. As amostras de SNC positivas para o EHV-1 foram coletadas de eqüinos oriundos de 30 municípios de Minas Gerais. Os casos de EHV-1 ocorreram de forma isolada não apresentando caráter sazonal. Na maioria dos casos(71,8%), a evolução dos sinais clínicos foi aguda, sendo que os sinais clínicos observados com mais freqüência foram ataxia, instabilidade dos membros posteriores, paralisia dos membros posteriores e decúbito. De acordo com informações relatadas pelo IMA, as infecções causadas pelo EHV-1 foram tão freqüentes como as infecções causadas pelo vírus da raiva em eqüinos no estado de Minas Gerais durante o período estudado. Portanto, torna-se importante a inclusão da encefalite pelo EHV-1 no diagnóstico diferencial de outras doenças do SNC de eqüinos no estado de Minas Gerais.

ABSTRACT

A labelled avidin-biotin ELISA (lab-ELISA) using repeated serum samples of goats showed a progressive seroconversion with higher seroconversion rate at the period going from the beginning of the breeding up to the last half of lactation (35.0 percent), compared to that recorded at the beginning of breeding (17.8 percent)(p<0.05). Furthermore, the seroreactivity pattern, evaluated by a lab-ELISA standard-curve with serum samples collected at 30-40 days intervals during 12 months, was caracterized by high individual variability. No seroreversion was observed and there were higher titers in the group of animals which delivered kids and established a lactation period (n=6; mean titre=913.4 units) compared to the group of goats that failed to conceived (n=4; mean titre=261.2 units) (p<0.01)

ABSTRACT

Acute and latent infections with the Brazilian LA031 strain of Aujeszky's disease virus (ADV) were established in mice. Ultraviolet irradiated ADV administered subcutaneously was a successful way to establish latent infection. The presence of ADV was detected by PCR. Two sets of 22-mer primers were synthesized and used to amplify gG glycoprotein gene sequences in acute and latent infected trigeminal nerve ganglia. The specificity of the amplification was verified by dot-blot hybridization.

ABSTRACT

Monoclonal antibodies (MABs) were produced against an ethylenediaminetetraacetate (EDTA) extract of Leptospira interrogans serovar icterohaemorrhagiae being characterized by gel precipitation as IgM and IgG (IgG1 and IgG2b). The EDTA extract was detected as several bands by silver staining in SDS-PAGE. In the Western blot the bands around 20 KDa reacted with a monoclonal antibody, 47B4D6, and was oxidized by periodate and was not digested by pronase, suggesting that the determinant is of carbohydrate nature. Immunocytochemistry, using colloidal gold labeling, showed that an EDTA extract determinant recognized by monoclonal antibody 47B4D6, is localized under the outer envelope of serovar icterohaemorrhagiae. The MAB raised against the EDTA extract was not able to protect hamsters from lethal challenge with virulent homologous leptospires.

ABSTRACT

Estudou-se a ocorrência de infecçäo por rotavírus suíno no Brasil entre janeiro de 1987 a dezembro de 1989. Examinaram-se, pelas técnicas de ELISA e PAGE, 330 amostras de fezes de suínos, 197 diarréicas e 133 normais, provenientes de 70 granjas em regime de criaçäo intensivo, localizadas em 17 municípios dos Estados de Minas Gerais, Säo Paulo, Paraná e Santa Catarina. O rotavírus suíno foi encontrado em 26,4//das amostras analisadas. Das 87 amostras positivas, 84 (96,5//) foram colhidas de leitöes com diarréia. O rotavírus também foi identificado com 61,8//(34/55) e 57,9//(11/19) das amostras de fezes diarréicas colhidas de lotes de animais com três e cinco semanas de idade, respectivamente, evidenciando sua importância na terceira semana e após o desmame dos leitöes. Os resultados mostram ampla distribuiçäo da rotavirose suína no Brasil, detectada em 46 granjas localizadas em 12 municípios, distribuídos nos quatro estados pesquisados

ABSTRACT

Estudou-se a presença de anticorpos anti-vírus da influenza aviária A em frangos de corte das regiöes do Alto Säo Francisco e Metalúrgica de Minas Gerais, no período de 1985 a 1986. Para tanto, testaram-se 2000 aves procedentes de 175 lotes pertencentes a 96 granjas comerciais daquelas regiöes. Os soros revelaram-se negativos ao teste de imunodifusäo em ágar gel, indicando que o vírus näo havia infectado as aves das granjas estudadas

ABSTRACT

Intestinal contents from 54 chicken flocks with diarrhea were examined for reovirus by genome eletropherotyping. Only 5 of the 54 specimens produced genome electropherotype characteristic of reovirus. No genome variations among reovirus strains were found, but they revealed extensive differences from a control avian atypical rotavirus. Three-day-old chickens could be infected with one reovirus isolate. Chickens developed diarrhea, decrased weight gain, some mortality and the viral genome could recovered from their droppings
