ABSTRACT

The status change of brain functional region of functional magnetic resonance imaging (fMRI) of midnight-noon ebb-flow acupuncture method at stomach meridian was explored. Ten healthy volunteers were recruited. With identical sequence of magnetic resonance scans, fMRI data was collected respectively in the timely acupoint group, untimely acupoint group and timely non-acupoint group. Volunteers in the timely acupoint group were treated at Zusanli (ST 36) and Chongyang (ST 42) between 7:02:53 A.M. to 9:02:52 A.M.; volunteers in the untimely acupoint group were treated with Zusanli (ST 36) and Chongyang (ST 42) between 3:02 P.M. to 5:02 P.M.; volunteers in the timely non-acupoint group were treated at non-acupoints 1 to 2 cm outside Zusanli (ST 36) and Chongyang (ST 42) between 7:02:53 A. M. to 9:02:52 A.M.. All fMRI data was analyzed with SPM5 software, and intra-group comparison and among-group comparison of timely acupoint group, untimely acupoint group and timely non-acupoint group were performed. The tonsil of cerebellum, the right-side insular cortex showed strengthened function in the timely acupoint group; the right-side medial frontal gyrus showed weakened function in the untimely acupoint group; the right-sided insular cortex showed strengthened function in the timely non-acupoint group. The comparison among groups indicated that there was different brain change in three groups. The midnight-noon ebb-flow acupuncture method at stomach meridian is likely to activate the function of cerebellum and insular lobe, which can have a positive effect on patients with flaccidity syndrome and stomach diseases.

ABSTRACT

<p><b>BACKGROUND</b>Cervical keratinocytes are recovered at a low numbers and frequently associated with contaminating human fibroblasts which rapidly overgrow the epithelial cells in culture with medium supplemented with 10% fetal bovine serum (FBS). However, it is difficult to initiate keratinocyte cultures with serum-free keratinocyte growth medium alone because cell attachment can be poor. Therefore, the culture of these cells is extremely difficult. In this study, we described a modified culture medium and coated culture plastics for growing normal human cervical epithelial cells in vitro.</p><p><b>METHODS</b>Normal cervical epithelial tissue pieces were obtained and digested with type I collagenase to dissociate the cells and a single cell suspension produced. The cells were cultured on plastic tissue culture substrate alone or substrate coated with collagen type I from rat tail, with modified keratinocyte serum-free medium (K-SFM) supplemented with 5% FBS. After attachment, the medium were replaced with K-SFM without FBS. The expression of basal keratins of the ectocervical epithelium, K5, K14 and K19 were assayed by immunofluorescence with monoclonal antibodies to identify the cell purity.</p><p><b>RESULTS</b>Our results indicate that cells attached to the culture plastic more quickly in K-SFM supplemented with 5% FBS than in K-SFM alone, as well as to tissue culture plastic coated with collagen type I than plastic alone. The modified medium composed of K-SFM and 5% FBS combined with a specific tissue culture plastic coated with collagen type I from rat tail was the best method for culture of normal cervical epithelial cells. K5, K14 and K19 were assayed and keratinocyte purity was nearly 100%.</p><p><b>CONCLUSION</b>A novel, simple and effective method can be used to rapidly obtain highly purified keratinocytes from normal human cervical epithelium.</p>

ABSTRACT

<p><b>OBJECTIVE</b>Ischemic postconditioning effectively minimizes the ischemic/reperfusion injury, and the large series of case reports on its protective effects in cardiac surgery are limited. A randomized trial was conducted to investigate the effect of ischemic postconditioning on cardiopulmonary protection in children undergoing cardiac surgery for tetralogy of Fallot.</p><p><b>METHODS</b>One hundred and five-children with tetralogy of Fallot undergoing surgery were randomly assigned to control (n=58) and ischemic postconditioning groups (n=47). Ischemic postconditioning was performed by intermittent aortic clamping after reperfusion. After surgery, the duration of intensive care unit (ICU) stay, capacity of blood transfusion, hemodynamics, inotropic scores, respiratory function, and release of blood lactate were assayed.</p><p><b>RESULTS</b>There was a significant decrease in the ICU stay in the postconditioned group compared with the control group (37+/-21 hrs vs 54+/-26 hrs; P<0.05 ). The capacity of blood transfusion (308+/-230 mL vs 526+/-515 mL; P<0.05) and the inotropic scores (5.9+/-5.0 vs 10.3+/-7.7; P<0.05) in the postconditioned group were significantly reduced compared with those in the control group. Blood lactate contents in the postconditioned group was significantly lower that those in the control group 1, 3, 6, 9, 12 and 20 hrs after surgery. The postconditioned group showed more improved hemodynamics and respiratory function than the control group.</p><p><b>CONCLUSIONS</b>Ischemic postconditioning may provide clinical benefits with respects to myocardial and pulmonary protections in children undergoing repair for tetralogy of Fallot.</p>

ABSTRACT

OBJECTIVE@#To summarize the experience of mitral vavuloplasty for mitral insufficiency in 53 patients.@*METHODS@#Between January 2001 and September 2006,53 patients (31 males and 22 females) with mitral insufficiency underwent mitral valve repair at our hospital. The mean age was (23.8+/-10.4) years, and the mean weight was (43+/-12) kg,including 29 cases of mitral prolapse,17 congenital mitral insufficiency, 5 rheumatic mitral insufficiency, and 2 other etiology. All operations were performed under cardiopulmonary bypass, moderate hypothermia and under the surveillance of transesophagus echocardiography.@*RESULTS@#There were 2 early postoperative deaths, and another patient failed to repair and received valve replacement afterward. Postoperative echocardiography indicated that mitral insufficiency was completely corrected in 42 patients, and residual mitral regurginitation in the other 8 patients. During the follow-up with mean period 13.8 months,41 patients revealed 28 in NYHA classI,13 in Class II.@*CONCLUSION@#Mitral valve repair should be the preferred modes of surgical correction for regurgitate mitral valves. With proper vavuloplasty technique, patients with selective mitral insufficiency patients may achieve satisfactory effect.

ABSTRACT

OBJECTIVE@#To investigate the effect of diet-induced hyperlipidemia on TGF-beta/Smad signaling pathway in the kidney of diabetic rats, and to explore the mechanism by which hyperlipidemia leads to renal injury in diabetes.@*METHODS@#Diabetic rats and non-diabetic rats were fed with normal fat diet and high fat diet for 16 weeks, respectively. The expressions of TGF-beta1, TbetaRII, and Col-IV mRNA in the renal cortex were examined by reverse transcriptase-PCR,TbetaRII and p-Smad staining in glomerular cells were detected by immunohistochemical staining, and the expression of TGF-beta1 and Col-IV protein was determined by Western blot.@*RESULTS@#Diet-induced hyperlipidemia up-regulated the levels of TGF-beta1, TbetaRII, p-Smad, and Col-IV protein and mRNA in the renal cortex of diabetic rats compared with those of non-diabetic rats. However, feeding high fat diet to non-diabetic rats had no influence on the expression of TGF-beta1, TbetaRII, p-Smad2, and Col-IV in the renal cortex.@*CONCLUSION@#Hyperlipidemia induced by high fat diet ingestion leads to renal injury in diabetic rats through activating TGF-beta1 /Smad signaling pathway.
